토양생태계는 정량화하기 어려운 다양한 규칙이 다중적으로 조화되어 끊임없이 변하고있어 어떤 현상을 획일적으로 평가하기 어렵다. 그러나, 미생물은 환경에 대한 감수성이 빨라 토양의 생물적인 특성 변화를 가장 먼저 나타내기 때문에, 미생물은 생태계의 가변적 변동상의 지표가 될 수 있다. 또한 토양에 생명력을 부여해주는 기능을 가지고 있어 토양미생물의 작용 및 기능 평가는 토양 건전성 관리에 있어 중요한 역할을 한다. 따라서 본 연구는 생물학적 토양 건전성 평가 지표요인 개발하고자 주요 모재별 미경지 토양의 생물학적 토양특성을 평가하고자 수행하였다. 주요 모재별 미경지 토양의 미생물 밀도는 현무암유래 토양에서 낮고 석회암유래 토양에서 높은 경향이었으나, 그람 음성균은 비슷한 수준을 유지하고 있었다. 토양 건조처리시 미생물의 생존율은 형광성 Pseudomonas속이 0.1%로 가장 낮았으나 고온성 Bacillus속과 호알카리성균은 90%이상으로 높았다. 토양미생물체량과 미생물 유fo 유기탄소량 간에는 정의 상관관계를 나타냈으며, 토양 건조처리 효과는 토양별 9.7~95.0% 범위로 평균 39.8%였다. 건토를 재습윤하였을 때 중온성 Bacillus속은 원래 균수보다 높게 복원되는 토양이 많았다. 토양 pH는 미생물수 복원율에 정의 영향을 주었다. 건토의 재습윤화에 의한 토양 미생물체량 복원율은 평균 65.3%였으며, 재습윤토의 미생물체 복원량과 형광성 Pseudomonas속간에 유의한 정의 상관관계가 있었다.
The study of microbial communities in sediments is impaired by the lack of reliable extraction methods. This study reports on the efficiency of recovery of a method for extracting bacteria, protists and diatoms from sandy sediments using a modified decant/fix method. The best extractions were achieved after fixation with a microtubule stabilising fixative and subsequent sonication for 80 seconds. We estimate that the efficiencies of recovery of bacteria and mixed heterotrophic flagellates were $96.4{\pm}3.5\%$ and $96.9{\pm}4.6\%$, respectively. Diatoms were recovered with an efficiency of 38-83% and varied considerably from species to species. This study suggests that the decant/fix method is effective in extracting small cells such as bacteria and heterotrophic flagellates, and that the efficiency of recovery of the method varies due to cell length and different types of organisms. When microbial carbon biomass had been underestimated by up to 32%, with much of that relating to larger cells such as microalgae and ciliates. We note that the corrected abundances may be still a subset of the total numbers present.
경남지역 논토양의 글로말린 함량과 미생물 특성과의 관계를 분석하기 위하여 2011년에 20개소를 선정하여 분석한 결과는 다음과 같다. 토양 글로말린 함량은 토양 유기물 함량 (r=0.595, p<0.01) 및 탈수소효소 활성 (r=0.663, p<0.01)과 정의상관 관계를 나타냈다. 또한, 글로말린 함량은 미생물 생체 탄소 함량 (r=0.710, p<0.001) 및 총 세균군집 (r=0.616, p<0.01)과 정의상관을 보였다. 그리고 글로말린 함량은 그람음성 세균의 군집 (r=0.561, p<0.05) 및 cy19:0/18:$1{\omega}7c$ (r=0.487, p<0.05)와 정의상관 관계를 나타냈다. 따라서 논토양에서 글로말린 함량은 지속가능한 친환경농업을 위해 토양 건강성의 지표로 활용할 수 있을 것으로 생각된다.
강화도 장화리의 펄 갯벌에 서식하는 저서 원생동물 군집의 계절 분포 및 섭식률을 파악하기 위하여 2002년 4월부터 2004년 4월까지 총 3개의 정점을 선정하여 2-3개월 간격으로 조사를 수행하였다. 저서 원생동물의 현존량과 생물량은 섬모충류와 종속영양 편모류에 의해 98% 이상 우점하여 나타났으며, 그 외 종속영양 와편모류와 아메바 그룹이 일부 시기에 관찰되었으나 중요하지 않은 그룹으로 나타났다. 종속영양 편모류의 현존량과 탄소량은 각각 $0.2-5.9{\times}10^5\;cells\;cm^{-3}$와 $0.02-9.2\;{\mu}C\;cm^{-3}$로 분포하였으며, 생물량은 춘계와 추계에 비교적 높은 분포를 보였다. 종속영양 편모류는 표충 2.5 mm 이내에서 다른 층에 비해 높은 분포를 보였으며, 종속영양 편모류의 수평적 분포는 정점별로 차이가 없는 것으로 나타났다. 종속영양 편모류는 $10{\mu}m$이하의 크기에 의하여 주로 우점하여 나타났으나, 2월과 4월 사이에 $20{\mu}m$ 이상의 종속영양 유글레나가 높게 출현하였다 섬모충류의 현존량과 생물량은 각각 $0.1-17.8{\times}10^3\;cells\;cm^{-3}$와 $0.02-9.1\;{\mu}gC\;cm^{-3}$ 범위로 분포하였으며 춘계와 추계에 높은 분포를 보였다. 섬모충류 그룹은 주로 하모류에 의해 우점 하였으며, 계절별로 두드러진 우점 종들은 관찰되지 않았다. 섬모충류의 수직적 분포는 표층 2.5 mm이내에서 높은 분포를 보였으며, 정점별 분포는 조간대 상부 정점에 비해 조간대 하부 정점에서 높은 분포를 보였다. 섬모충류의 시.공간적 분포는 저서 미세조류의 생물량 분포와 유사한 양상을 보였다. 조사기간동안 저서 원생동물 그룹의 탄소량은 섬모충류에 의해 평균 66%의 높은 우점률을 보였으며, 저서 원생동물의 계절변동은 저서 미세조류의 변동과 유사한 양상을 보였다. 본 연구에서 저서 원생동물의 섭식률을 측정한 결과 박테리아 생산력과 저서 미세조류 생산력의 각각 13.4-40.7%와 20.1-36.4%를 제거하였으며, 박테리아에 대한 섭식률은 섬모충류에 비해 종속영양 편모류에 의해 더 높은 것으로 나타났다. 박테리아와 돌말류에 대한 저서 원생동물의 섭식률은 춘계에 가장 높았으며 저서 미세생물 먹이 망에서 저서 원생동물은 미세생물 군집의 생물량이 높은 춘계에 박테리아와 돌말류의 포식자로서 중요한 역할을 하고 있음을 시사하였다.
The indiscriminate use of the fossil fuel has caused serious environmental pollutions such as the shortage of energy and global warming. Microalgae have being emphasized as $3^{rd}$ generation biomass which makes the carbon dioxide reduce effectively as well as produces the biofuel. Large scale production of microbial biomass by continuous culture is a quite challenging issue, because off-line optimization strategies of a microbial process utilizing a model-based scheme give rise to many difficult problems. In this paper, the static and simple control method which was able to be applied in time-variant growth environment and large scale of algae culture was studied. The significant disturbances in on-line measurement of cell density were reduced by Savitzky-Golay FIR smoothing filter. Dunaliella salina was cultivated continuously in a flat panel photobioreactor by the on-off control of the turbidostat process.
Carbon catabolite repression (CCR) is a key regulatory system found in most microorganisms that ensures preferential utilization of energy-efficient carbon sources. CCR helps microorganisms obtain a proper balance between their metabolic capacity and the maximum sugar uptake capability. It also constrains the deregulated utilization of a preferred cognate substrate, enabling microorganisms to survive and dominate in natural environments. On the other side of the same coin lies the tenacious bottleneck in microbial production of bioproducts that employs a combination of carbon sources in varied proportion, such as lignocellulose-derived sugar mixtures. Preferential sugar uptake combined with the transcriptional and/or enzymatic exclusion of less preferred sugars turns out one of the major barriers in increasing the yield and productivity of fermentation process. Accumulation of the unused substrate also complicates the downstream processes used to extract the desired product. To overcome this difficulty and to develop tailor-made strains for specific metabolic engineering goals, quantitative and systemic understanding of the molecular interaction map behind CCR is a prerequisite. Here we comparatively review the universal and strain-specific features of CCR circuitry and discuss the recent efforts in developing synthetic cell factories devoid of CCR particularly for lignocellulose-based biorefinery.
To elucidate the trophic role of heterotrophic nano- and microplankton (HNMP), we investigated their biomass, community structure, and herbivory in three different water masses, namely, south of Polar Front (SPF), Polar Front Zone (PFZ), the Sub-Antarcitc Front (SAF) in the Drake Passage in the Southern Ocean, during the austral summer in 2002. We observed a spatial difference in the relative importance of the dominant HNMP community in these water masses. Ciliates accounted for 34.7% of the total biomass on an average in the SPF where the concentration of chlorophyll-a was low with the dominance of pico- and nanophytoplankton. Moreover, the importance of ciliates declined from the SPF to the SAF. In contrast, heterotrophic dinoflagellates (HDFs) were the most dominant grazers in the PFZ where the concentration of chlorophyll-a was high with the dominance of net phytoplankton. HNMP biomass ranged from 321.9 to 751.4 $mgCm^{-2}$ and was highest in the PFZ and lowest in the SPF. This result implies that the spatial dynamic of HNMP biomass and community was significantly influenced by the composition and concentration of phytoplankton as a food source. On an average, 75.6%, 94.5%, and 78.9% of the phytoplankton production were consumed by HNMP in the SPF, PFZ, and SAF, respectively. The proportion of phytoplankton grazed by HNMP was largely determined by the composition and biomass of HNMP, as well as the composition of phytoplankton. However, the herbivory of HNMP was one of the most important loss processes affecting the biomass and composition of phytoplankton particularly in the PFZ. Our results suggest that the bulk of the photosynthetically fixed carbon was likely reprocessed by HNMP rather than contributing to the vertical flux in Drake Passage during the austral summer in 2002.
Macroalgae has been strongly touted as an alternative biomass for biofuel production due to its higher photosynthetic efficiency, carbon fixation rate, and growth rate compared to conventional cellulosic plants. However, its unique carbohydrate composition and structure limits the utilization efficiency by conventional microorganisms, resulting in reduced growth rates and lower productivity. Nevertheless, recent studies have shown that it is possible to enable microorganisms to utilize various sugars from seaweeds and to produce some energy chemicals such as methane, ethanol, etc. This paper introduces the basic information on macroalgae and the overall conversion process from harvest to production of biofuels. Especially, we will review the successful efforts on microbial engineering through metabolic engineering and synthetic biology to utilize carbon sources from red and brown seaweed.

In a recently developed strategy for in-situ treatment of polychlorinated biphenyls (PCB), bioaugmentation was used in conjunction with a surfactant, sorbitan trioleate, as a carbon source for the degrader bacteria, along with the monoterpene, carvone, and salicylic acid as inducing substrates. Two bacteria were used for soil inoculants, including Arthrobacter sp. st. B1B and Ralstonia eutrophus H850. This methodology achieved 60% degradation of PCBs in Aroclor 1242 after 18 weeks in soils receiving 34 repeated applications of the degrader bacteria. However, an obvious limitation was the requirement for soil mixing after every soil inoculation. In the research reported here, bioaugmentation and biostimulation treatment strategies were modified by using the earthworm, Pheretima hawayana, as a vector for dispersal and mixing of surface-applied PCB-degrading bacteria and soil chemical amendments. Changes in microbial biomass and microbial community structure due to earthworm effects were examined using DNA extraction and PCR-DGGE of 16S rDNA. Results showed that earthworms effectively promoted biodegradation of PCBs in bioaugmented soils to the same extent previously achieved using physical soil mixing, and had a lesser, but significant effect in promoting PCB biodegradation in biostimulated soils treated with carvone and salicylic acid. The effects of earthworms were speculated to involve many interacting factors including increased bacterial transport to lower soil depths, improved soil aeration, and enhanced microbial activity and diversity.
