Supplementation of Essential Oil Extracted from Citrus Peel to Animal Feeds Decreases Microbial Activity and Aflatoxin Contamination without Disrupting In vitro Ruminal Fermentation

  • Long-term storage of feeds or feedstuffs in high temperature and humid conditions can be difficult because of microbial contamination. Essential oil isolated from industrial waste citrus peel could be used as a preservative because it is likely to have anti-bacterial and anti-fungal activity. Our objective was to determine whether different levels (0.028, 0.056 and 0.112 g/kg) of citrus essential oil (CEO) would provide anti-microbial activity and enhance preservation of animal feed without influencing rumen fermentation. At 0.112 g/kg, CEO inhibited growth of Escherichia coli (ATCC 25922) and Salmonela enteritidis (IFO 3313). Growth of E. coli recovered after 24 h of incubation, but S. enteritidis continued to be inhibited for 72 h. Preservation of antibiotic-free diets for swine was assessed by observing anti-aflatoxin activity. Aflatoxin was detected in control feed samples on days 16 (8 ppb) and 21 (8 ppb) and in anti-fungal agent (AA) treated samples on days 16 (2 ppb) and 21 (4 ppb). However, aflatoxin was not detected in feed samples treated with CEO. Treatment with CEO and AA did not influence ruminal pH, dry matter digestibility (DMD) or organic matter digestibility (OMD) over 48 h of incubation in rumen fluid. Acetate and propionate were slightly higher with CEO treatment (p<0.05), but total concentration of volatile fatty acid (VFA) was not significantly affected by treatment. Ammonia-N concentration was slightly higher for the control treatment (p<0.05). This study showed that treating feed with CEO enhances preservation of animal feed without influencing in vitro rumen fermentation.

Changes of Physico-Chmical Properties and Microbial Activity During the Early Stage of Composting with Pig and Chicken Manure (돈분 및 계분의 초기 퇴비화 과정중 이화학적 특성과 미생물 활성변화)

  • This study was carried out to investigate changes of physico-chemical properties and microbial activity during the early stage of composting with pig and chicken manure. The results were as follows; 1. The temperature was rapidly increased from the 3rd to the 7th day, and especially the pig manure compost preparing with enzyme was maintained $56^{\circ}C{\sim}69^{\circ}C$. 2. The pH range was shown $7.7{\sim}9.3$, and the pH level increased from the 3rd day to 25th day. Also after the 25th day the pH level decreased gradually. 3. The C/N ratio in the pig manure compost decreased 16.8 at the 30th day, while the compost containing enzymes decreased 19.2 at the 30th day. Chicken manure compost showed similar results at the 28 of C/N ratio at the 30th day with enzyme treatment. 4. The total ammount of sugar in pig manure compost was $6,000{\sim}7,000mg/kg$, while the chicken manure compost was $2,000{\sim}4,000mg/kg$. However, there was no significant difference in view point of enzyme treatment. 5. Cellulase, phosphatase and xylanase activity were continually increased, however amylase and urease activity were not changed during composting.

Metabolite Profiling and Microbial Community of Traditional Meju Show Primary and Secondary Metabolite Differences Correlated with Antioxidant Activities

  • Meju, a type of fermented soybean paste, is used as a starter in the preparation of various Korean traditional soybean-based foods. In this study, we performed Illumina-MiSeq paired-end sequencing for microbial communities and mass spectrometry analysis for metabolite profiling to investigate the differences between 11 traditional meju products from different regions across Korea. Even though the bacterial and fungal communities showed remarkable variety, major genera including Bacillus, Enterococcus, Variovorax, Pediococcus, Weissella, and Aspergillus were detected in every sample of meju. The metabolite profile patterns of the 11 samples were clustered into two main groups: group I (M1-5) and group II (M6-11). The metabolite analysis indicated a relatively higher amino acid content in group I, while group II exhibited higher isoflavone, soyasaponin, and lysophospholipid contents. The bioactivity analysis proved that the ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical-scavenging activity was higher in group II and the FRAP (ferric reducing antioxidant power) activity was higher in group I. The correlation analysis revealed that the ABTS activity was isoflavonoid, lipid, and soyasaponin related, whereas the FRAP activity was amino acid and flavonoid related. These results suggest that the antioxidant activities of meju are critically influenced by the microbiome and metabolite dynamics.

Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora (작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.

Dyeing Properties and Bio-Functions of Cotton Fabrics Dyed with Naturally Fermented Ecklonia Cava Extract (자연 발효 감태 추출물로 염색한 면직물의 염색 특성과 바이오 기능성)

  • This study investigated the dyeing properties and bio-functions of cotton fabrics dyed with naturally fermented Ecklonia cava extract in order to compare it with a comparison of unfermented extract. Hot water-extracted Ecklonia cava was fermented naturally under the various conditions of a fermenting period (2-8 days) and amount of molasses (0.1-1.8% v/v); in addition, it was also tested for characterization by FT-IR, antioxidant activity, total polyphenol content, and anti-microbial activity. For dyed cotton fabrics, color strength (K/S), physical color properties, dyeing fastness, sun protective property, and anti-microbial activity were evaluated considering dyeing conditions. As a result, the fermented dye under fermentation condition of 0.1% v/v with molasses during 4 days was revealed as having a similar chemical structure to the unfermented one and showed a total polyphenol content with 32.88mg/g and better antioxidant activity than the unfermented one. As for dyed fabrics, the color strength value by K/S was the highest under the condition of 0.1% v/v of molasses during 4 days among all fermenting conditions. The dyed fabrics had a reasonably good fastness (except for light). Anti-microbial activity against K. pneumoniae was better for the fermented extract-dyed fabric especially with lower dye concentrations.

Recombinant Expression and Characterization of Thermoanaerobacter tengcongensis Thermostable $\alpha$-Glucosidase with Regioselectivity for High-Yield Isomaltooligosaccharides Synthesis

  • A novel thermostable $\alpha$-glucosidase (TtGluA) from Thermoanaerobacter tengcongensis MB4 was successfully expressed in E. coli and characterized. The TtgluA gene contained 2,253 bp, which encodes 750 amino acids. The native TtGluA was a trimer with monomer molecular mass of 89 kDa shown by SDS-PAGE. The purified recombinant enzyme showed hydrolytic activity on maltooligosaccharides, p-nitrophenyl-$\alpha$-D-glucopyranide, and dextrin with an exotype cleavage manner. TtGluA showed preference for short-chain maltooligosaccharides and the highest specific activity for maltose of 3.26 units/mg. Maximal activity was observed at $60^{\circ}C$ and pH 5.5. The half-life was 2 h at $60^{\circ}C$. The enzyme showed good tolerance to urea and SDS but was inhibited by Tris. When maltose with the concentration over 50 mM was used as substrate, TtGluA was also capable of catalyzing transglycosylation to produce $\alpha$-1,4-linked maltotriose and $\alpha$-1,6-linked isomaltooligosaccharides. More importantly, TtGluA showed exclusive regiospecificity with high yield to produce $\alpha$-1,6-linked isomaltooligosaccharides when the reaction time extended to more than 10 h.

  • To produce and characterize antineoplastic constituents in the submerged cultured­mycelia of Laccaria laccata, the mycelia were extracted with distilled water. Purification of the extract was carried out by acetone precipitation, by ion exchange chromatography using DEAE­Sephadex A-50, CM-Sephadex C-25 resins, and by gel filtration chromatography on Sephadex G-200. Each fraction obtained during the purification was examined for antineoplastic activity against sarcoma 180 in ICR mice. As the purification proceeded, the antineoplastic activity was markedly increased. The highly purified Fraction E showed 75% tumor inhibition ratio at a dose of 10mg/kg/day and contained 81% polysaccharide and 4% protein. The antitumor component of Fraction E stimulated an accumulation of peritoneal exudate cells including peritoneal macrophages, and is named laccaran.

  • To verify the usefulness of enzyme activity as a index for the stability or maturity of apple pomace composting. the succession of microbial populations using viable count procedure. and Vmax of ${\beta}$-glucosidase and cellobiohydrolase were measured. based on an increase in fluorescence as the nonfluorescent methylumbelliferyl substrates were enzymatically hydrolyzed, leading to the highly fluorescent methylumbelliferyl molecule 4-methylumbelliferone(MUF). The activities of these enzymes in the decomposition of carbohydrates were gradually decreased in the course of the time. Correlation between microbial population and enzyme activity was not significant with exception of fungi. and the fungi were represented in high density. This indicates that the fungi probably play a major role in composting of apple pomace.

Screening of Antagonistic Bacteria having Antifungal Activity against Various Phytopathogens (다양한 식물병원성 곰팡이에 항진균 활성을 갖는 길항미생물의 탐색)

  • The aim of this study was to isolate a potential multifunctional biocontrol agent from bacteria for control of multiple plant diseases as an alternative to fungicides. A total of 201 strains were isolated from soil undamaged by repeated cultivation in Sunchang and their ability to produce antibiotics, siderophores and extracellular enzymes such as protease, cellulase and amylase was investigated. Selected strain SCS3 produced cellulose, protease and amylase. This strain also produced siderophores and showed excellent antifungal activity against various phytopathogens. SCS3 was identified as Bacillus subtilis using 16S rRNA sequencing, and named Bacillus subtilis SCS3. Finally, physiological and biochemical characteristics of B. subtilis SCS3 were examined. From the results, B. subtilis SCS3 was found to be a useful multifunctional biocontrol agent against various phytopathogens.

Comparison of Soil Physico-chemical and Microbial Characteristics in Soil of 'Niitaka' Pear Orchards between Organic and Conventional Cultivations (유기농법과 관행농법에 의해 재배한 '신고'배 과원 토양의 물리화학적 및 미생물학적 특성 비교)

  • Consumers' interest and government's support for the fruits rapidly increased organic fruit productions. This study was examined to compare the soil physicochemical and microbial properties of orchards soil in conventionally and organically management systems. Organic cultivation had lower soil bulk density, solid phase, and penetration resistance than the conventional cultivation. Soil pH and organic matter contents increased from March to August, and the values were greater in the organic cultivation than the conventional cultivation. Total nitrogen (N) and phosphorous concentrations decreased from March to August, and the organic soils had greater N but lower phosphorous concentrations than the conventional soils. Soil microbial carbon biomass increased 36% and 15% for organic and conventional cultivations, respectively, from March to August. Soil microbial N biomass was greater in June than March or August, and the organic cultivation had a greater biomass N compared to the conventional cultivation. Soil dehydrogenase and chitinase activities were greater in June than in March or August. ${\beta}$-glucosidase activity declined in both cultivations, while the phosphatase activity increased. Organic cultivation had greater enzyme activities in March, June, and August, except for the acid phosphatase activity in June.
