We evaluated microbial populations and aflatoxin production in unhulled and white rice from rice processing complexes of the National Agricultural Cooperative Federation in five regions in Korea and identified three predominant Aspergillus species. Fungal and bacterial populations in rice samples were significantly different between regions in 2007. Aflatoxins were also detected and varied at the levels of 2.45 - 3.43 ng per g unhulled rice grain and 1.29 - 2.09 ng per g white rice grain. Unhulled rice generally detected higher level of aflatoxins than white rice regardless of sampling regions; however, no significant differences were found in Anseong and Cheonan in 2005 and Cheonan and Gimpo in 2007. Aflatoxin production between sampling regions was not different regardless of rice type and sampling year. Although the fungal diversity was highly distinct from region to region, three Aspergillus isolates were predominant in the rice samples; thus, representative isolates AC317, AF57, and AF8 were selected and identified based on their morphological and molecular characteristics. Consequently, isolates AC317, AF57, and AF8 were identified as A. candidus, A. flavus, and A. fumigatus, respectively. These fungi can produce mycotoxins that are harmful for consumers and thus it is important to detect and reduce the population of storage fungi in rice.
Silver nanoparticles have been used in agricultural practice because of their biocide effect. However, limited information is available for the effect of silver nanoparticles on soil quality. Therefore, the main purpose of this study was to evaluate effect of silver nanoparticle application on soil especially when fertilizer is applied. To simulate potassium fertilizer, potassium chloride was mixed with silver nanoparticles in soil. Concentration of silver and chloride was measured and result showed that concentration of both compounds was decreased at the range of $3.4mg\;kg^{-1}$ and 78-84% respectively after treatment. In addition, analysis of microbial population after treatment showed that microbial population was increased when silver nanoparticles and KCl were mixed. Those results indicated that application of fertilizer has impact on biocide effect of silver nanoparticles in soil.

This study was conducted to investigate the effects of supplementation of a probiotics complex on growth performance, nutrient digestibility, diarrhea score and microbial population in pigs weaned at 21 days of age. Treatments were 1) control A (0.2% antibiotics, Avilamycin), 2) control B (0.1 % $Ractocom^{(R)}$), 3) 0.1%, 4) 0.2% and 5) 0.3% probiotics complex; 80 pigs were used and each treatment had 4 replicates with 4 pigs per replicate (16 pigs per treatment). During phase I period (d 0 to 14), although there was no significant difference, pigs fed control B diet showed higher ADG (average daily gain) and better F/G (feed/gain) than any other treatments. During late experimental period (d 15 to 28), pigs fed diet supplemented with 0.2% probiotics complex showed slightly higher ADG. Overall (d 0 to 28) the diet that contained 0.2% probiotics complex gave slightly higher ADG and ADFI (average daily feed intake) than the other diets. In a metabolic trial using 20 piglets, nutrient digestibility showed the best results in pigs fed 0.2% probiotics complex diet, but not significantly different from other groups. Diarrhea score and microbial population status in intestine, colon and feces were not affected by dietary treatments. In conclusion, this study suggested that a newly developed probiotics complex can replace antibiotics in weaned pigs.
Objective: This study aimed at studying the potential use of Flemingia (Flemingia macrophylla) as a protein source fodder to improve nutrients digestibility and ruminal fermentation efficiency in beef cattle. Methods: Four, Thai native beef cattle were randomly assigned in a 4×4 Latin square design. Four levels of Flemingia hay meal (FHM) were used to replace soybean meal (SBM) in the concentrate mixtures in four dietary treatments replacing levels at 0%, 30%, 60%, and 100% of SBM. Results: The experimental findings revealed that replacements did not effect on intake of rice straw, concentrate and total dry matter (DM) intake (p>0.05). However, the apparent digestibilities of DM, organic matter, crude protein, acid detergent fiber, and neutral detergent fiber were linearly increased up to 100% replacement levels. Moreover, the production of total volatile fatty acids, and propionate concentration were enhanced (p<0.05) whereas the concentration of acetate was reduced in all replacement groups. Consequently, the CH4 production was significantly lower when increasing levels of FHM for SBM (p<0.05). Furthermore, rumen bacterial population was additionally increased (p<0.05) while protozoal population was clearly decreased (p<0.05) in all replacement groups up to 100%. In addition, microbial nitrogen supply and efficiency of microbial nitrogen synthesis were enhanced (p<0.05), as affected by FHM replacements. Conclusion: The findings under this experiment suggest that 100% FHM replacement in concentrate mixture enhanced rumen fermentation efficiency, nutrients digestibilities, bacterial population, microbial protein synthesis, and subsequently reduced CH4 production in beef cattle fed on rice straw.

The objective of this study was to determine the effect of rain tree pod meal with palm oil supplementation on feed intake, digestibility, microbial protein synthesis and microbial populations in dairy cows. Four, multiparous early-lactation Holstein-Friesian crossbred (75%) lactating dairy cows with an initial body weight (BW) of $405{\pm}40$ kg and $36{\pm}8$ DIM were randomly assigned to receive dietary treatments according to a $4{\times}4$ Latin square design. The four dietary treatments were un-supplementation (control), supplementation with rain tree pod meal (RPM) at 60 g/kg, supplementation with palm oil (PO) at 20 g/kg, and supplementation with RPM at 60 g/kg and PO at 20 g/kg (RPO), of total dry matter intake. The cows were offered concentrates, at a ratio of concentrate to milk production of 1:2, and chopped 30 g/kg of urea treated rice straw was fed ad libitum. The RPM contained condensed tannins and crude saponins at 88 and 141 g/kg of DM, respectively. It was found that supplementation with RPM and/or PO to dairy cows diets did not show negative effects on feed intake and ruminal pH and BUN at any times of sampling (p>0.05). However, RPM supplementation resulted in lower crude protein digestibility, $NH_3$-N concentration and number of proteolytic bacteria. It resulted in greater allantoin absorption and microbial crude protein (p<0.05). In addition, dairy cows showed a higher efficiency of microbial N supply (EMNS) in both RPM and RPO treatments. Moreover, NDF digestibility and cellulolytic bacteria numbers were highest in RPO supplementation (p<0.05) while, supplementation with RPM and/or PO decreased the protozoa population in dairy cows. Based on this study, supplementation with RPM and/or PO in diets could improve fiber digestibility, microbial protein synthesis in terms of quantity and efficiency and microbial populations in dairy cows.
This study was carried out to investigate the change of rhizosphere microflora in paddy soils of long-term application of same fertilizer by single cropping. Treatments consisted of no fertilizer, NPK, PK, NK, NP and NPK + compost and the same fertilizer has been applied to the soil of each treatment for thirty five years. Any remarkable difference of the microbial population structure was not detected from the rhizosphere of Hwayeongbyeo and that of Hwasambyeo. However, slight difference of dominant microbial species in the rhizosphere was recognized between different fertilization practice. The bacterial population in the rhizosphere was steadily reduced over time after rice plant setting out, while actinomycetal population was increased over time. The increase of actinomycetal population was the highest in the NPK + compost treatment, suggesting the effect of organic matters in the change of actinomycetal population. As for the aerobic culturable bacterial diversity, Bacillus megaterium, B. mycoides, B. licheniformis and B. subtilis were dominants in genus Bacillus, and Streptomyces spororaveus, S. canus, S. tauricus and S. galbus in genus Streptomyces and genus Micromonospora was another dominant in actinomycetes.

The objective of this study was to investigate the effect of levels of cottonseed meal with various carbohydrate sources in concentrate on feed intake, nutrient digestibility, rumen fermentation and microbial protein synthesis in dairy bulls. Four, 6 months old dairy bulls were randomly assigned to receive four dietary treatments according to a $2{\times}2$ factorial arrangement in a $4{\times}4$ Latin square design. Factor A was carbohydrate source; cassava chip (CC) and cassava chip+rice bran in the ratio of 3:1 (CR3:1), and factor B was cotton seed meal levels in the concentrate; 109 g CP/kg (LCM) and 328 g CP/kg (HCM) at similar overall CP levels (490 g CP/kg). Bulls received urea-lime treated rice straw ad libitum and were supplemented with 10 g of concentrate/kg BW. It was found that carbohydrate source and level of cotton seed meal did not have significant effects on ruminal pH, ammonia nitrogen concentration, microbial protein synthesis or feed intake. Animals which received CC showed significantly higher BUN concentration, ruminal propionic acid and butyric acid proportions, while dry matter, organic matter digestibility, populations of total viable bacteria and proteolytic bacteria were lower than those in the CR3:1 treatment. The concentration of total volatile fatty acids was higher in HCM than LCM treatments, while the concentration of butyric acid was higher in LCM than HCM treatments. The population of proteolytic bacteria with the LCM treatments was higher than the HCM treatments; however other bacteria groups were similar among the different levels of cotton seed meal. Bulls which received LCM had higher protein digestibility than those receiving HCM. Therefore, using high levels of cassava chip and cotton seed meal might positively impact on energy and nitrogen balance for the microbial population in the rumen of the young dairy bull.

This study was conducted to obtain the information about injury caused by continuous cropping of peony (Paeonia lactiflora). Soil physico-chemical properties, characteristics of microbial distribution and diversities in the continuous cropped field with peony were analyzed. As the results, pH and organic matter content were higher in the continuous cropping soil than those in the first cropping soil. Bulk density was decreased but porosity was increased in the continuous cropping soil. As the cultivation period was lengthened in years, the populations of bacteria and actinomyces were gradually decreased, whereas fungal population was increased. It was shown that the metabolic diversity patterns of the microbial communities in the continuous cropping soil differed from that of the first cropping soil. These results indicate that deterioration of soil quality such as physico-chemical properties including a soil depth, bulk density, porosity and soil pH is related with a continuous cultivation periods, and also affect a microbial population, especially fungi.

Objective: This study aimed to determine the effects of different roughages in total mixed ration (TMR) inoculated with or without coculture of Lactobacillus acidophilus (L. acidophilus) and Bacillus subtilis (B. subtilis) on in vitro rumen fermentation and microbial population. Methods: Three TMRs formulations composed of different forages were used and each TMR was grouped into two treatments: non-fermented TMR and fermented TMR (F-TMR) (inoculated with coculture of L. acidophilus and B. subtilis). After fermentation, the fermentation, chemical and microbial profile of the TMRs were determined. The treatments were used for in vitro rumen fermentation to determine total gas production, pH, ammonianitrogen (NH3-N), and volatile fatty acids (VFA). Microbial populations were determined by quantitative real-time polymerase chain reaction (PCR). All data were analyzed as a 3×2 factorial arrangement design using the MIXED procedure of Statistical Analysis Systems. Results: Changes in the fermentation (pH, lactate, acetate, propionate, and NH3-N) and chemical composition (moisture, crude protein, crude fiber, and ash) were observed. For in vitro rumen fermentation, lower rumen pH, higher acetate, propionate, and total VFA content were observed in the F-TMR group after 24 h incubation (p<0.05). F-TMR group had higher acetate concentration compared with the non-fermented group. Total VFA was highest (p<0.05) in F-TMR containing combined forage of domestic and imported source (F-CF) and F-TMR containing Italian ryegrass silage and corn silage (F-IRS-CS) than that of TMR diet containing oat, timothy, and alfalfa hay. The microbial population was not affected by the different TMR diets. Conclusion: The use of Italian ryegrass silage and corn silage, as well as the inoculation of coculture of L. acidophilus and B. subtilis, in the TMR caused changes in the pH, lactate and acetate concentrations, and chemical composition of experimental diets. In addition, F-TMR composed with Italian ryegrass silage and corn silage altered ruminal pH and VFA concentrations during in vitro rumen fermentation experiment.
A feeding trial was conducted to study the effect of tannin rich Pakar (Ficus infectoria) leaves on microbial profile, rumen fermentation and nutrient utilization in goats. Eight goats divided in two groups were fed pakar leaves (experimental group) and green oats (control group) as sole roughage source along with a fixed quantity of concentrate mixture for a period of 3 months. Two metabolic trials of six days duration were conducted after 30 and 90 days of experimental feeding. The dry matter intake was significantly higher (p<0.05) and digestibility's of DM, OM, CP, EE, NDF and ADF were reduced in experimental as compared with the control group. The TDN intake was similar (236.52 vs. 240.39 g/d) in both the groups. All the animals were in positive nitrogen balance. The concentration of ammonia nitrogen, TVFA, lactic acid and activities of xylanase and protease were reduced in pakar leaves fed goats. The rumen microbial profile as obtained by MPN technique showed no change in total bacterial population but total fungi and cellulolytic bacteria were reduced (p<0.05), whereas, tannin degrading/tolerant bacteria increased with the feeding of pakar leaves. Real time PCR data revealed a decrease in Ruminococcus flavefaciens, an increase in methanogens and no change in the Fibrobacter succinogenes population by feeding of pakar leaves.
