The effect on methanogens attached to the surface of rumen ciliate protozoa by the addition of plant extracts (pine needles and ginkgo leaves) was studied with particular reference to their effectiveness for decreasing methane emission. The plant extracts (pine needles and ginkgo leaves) were added to an in vitro fermentation incubated with rumen fluid. The microbial population including bacteria, ciliated-associated methanogen, four different groups of methanogens and Fibrobacter succinogenes were quantified by using the real-time PCR. Gas profiles including methane, carbon dioxide and hydrogen, and runinal fermentation characteristics were observed in vitro. The methane emission from samples with an addition of individual juices from pine needles, ginkgo leaves and 70% ethanol extract from ginko leaves was significantly lower (p<0.05, 27.1, 28.1 and 28.1 vs 34.0 ml/g DM) than that of the control, respectively. Total VFAs in samples with an addition of any of the plant extracts were significantly lower than that of the control (p<0.05) as well. The order Methanococcales and the order Methanosarcinales were not detected by using PCR in any incubated mixtures. The ciliate-associated methanogens population decreased from 25% to 49% in the plant extacts as compared to control. We speculate that the supplementation of juice from pine needles and ginkgo leaves extract (70% ethanol extract) decreased the protozoa population resulting in a reduction of methane emission in the rumen and thus inhibiting methanogenesis. The order Methanobacteriales community was affected by addition of all plant extracts and decreased to less than the control, while the order Methanomicrobiales population showed an increase to more than that of the control. The F. succinogenes, the major fibrolytic microorganism, population in all added plant extracts was increased to greater than that of the control. In conclusion, pine needles and ginkgo leaves extracts appear to have properties that decrease methanogenesis by inhibiting protozoa species and may have a potential for use as additives for ruminants.
This study was conducted to investigate the effects of Acanthopanax senticosus extract (ASE) as a dietary additive on gut microflora in weaned piglets. A total of sixty pigs were weaned at 21 d of age (BW = $5.64{\pm}0.23kg$) and allocated on the basis of BW and litter to three dietary treatments in a randomized complete block design. The dietary treatments were: control group (basal diet), antibiotics group (basal diet+0.02% colistin), and ASE group (basal diet+0.1% ASE). On d 7, 14 and 28 after consuming the experimental diets, five piglets per group were sacrificed and then the contents from the jejunum, ileum and cecum were collected to determine changes in the microbial community by using a polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) technique and estimating the contents of Lactobacillus and E. coli by in vitro culturing methods. The results showed that the ASE promoted the microflora diversity in the cecum. Enumeration of bacteria in the gut contents showed that the number of Lactobacillus increased (p<0.05), while that of E. coli decreased (p<0.05) when compared with the other 2 groups as the days of age progressed post-weaning. These findings suggested that the ASE, as a substitute for dietary antimicrobial products, could improve the development of the normal gut microflora and suppress bacterial pathogens, and effectively promote a healthy intestinal environment.
The objective of this study was to investigate the effect of banana flower powder (BAFLOP) supplementation on gas production kinetics and rumen fermentation efficiency in in vitro incubation with different ratios of roughage to concentrate in swamp buffalo and cattle rumen fluid. Two male, rumen fistulated dairy steers and swamp buffaloes were used as rumen fluid donors. The treatments were arranged according to a $2{\times}2{\times}3$ factorial arrangement in a Completely randomized design by using two ratios of roughage to concentrate (R:C; 75:25 and 25:75) and 3 levels of BAFLOP supplementation (0, 2 and 4% of dietary substrate) into two different kinds of rumen fluid (beef cattle and swamp buffalo). Under this investigation, the results revealed that the rumen ecology was affected by R:C ratio. The pH declined as a result of using high concentrate ratio; however, supplementation of BAFLOP could buffer the pH which led to an improvement of ruminal efficiency. BAFLOP supplementation affected acetic acid (C2) when the proportion of concentrate was increased. However, there were no effect on total volatile fatty acid (TVFA) and butyric acid (C4) by BAFLOP supplementation. The microbial community was affected by BAFLOP supplementation, especially the bacterial population. As revealed by real-time PCR, the populations of F. succinogenes and R. albus were reduced by the high concentrate treatments while that of R. flavafaciens were increased. The populations of three dominant cellulolytic bacteria were enhanced by BAFLOP supplementation, especially on high concentrate diet. BAFLOP supplementation did not influence the ammonia nitrogen ($NH_3$-N) concentration, while R:C did. In addition, the in vitro digestibility was improved by either R:C or BAFLOP supplementation. The BAFLOP supplementation showed an effect on gas production kinetics, except for the gas production rate constant for the insoluble fraction (c), while treatments with high concentrate ratio resulted in the highest values. In addition, BAFLOP tended to increase gas production. Based on this study, it could be concluded that R:C had an effect on rumen ecology both in buffalo and cattle rumen fluid and hence, BAFLOP could be used as a rumen buffering agent for enhancing rumen ecology fed on high concentrate diet. It is recommended that level of BAFLOP supplementation should be at 2 to 4% of total dry matter of substrate. However, in vivo trials should be subsequently conducted to investigate the effect of BAFLOP in high concentrate diets on rumen ecology as well as ruminant production.
우리나라의 전통 발효 식품인 젓갈로부터 배양 분리법과 분자생물학적 분석법을 이용하여 원핵 세균 군집을 분석하고, 신규 미생물 분리를 목표로 하였다. 젓갈은 생산 지역과 주재료를 고려하여 17 종을 선정하였으며, 이들 젓갈 시료를 적정 희석배수로 희석하여 12종류의 미생물 선택배지에 도말, 배양한 후 나타난 집락(colony)을 형태학적 특성에 따라 무작위로 308개를 선정하여 분리하였다. 순수 분리된 미생물은 PCR 방법을 이용하여 16S rRNA 유전자의 염기서열을 분석한 후, 기존에 보고된 미생물 database와 비교함으로서 17종의 젓갈 내 미생물 군집을 확인하였다. 젓갈의 발효 및 숙성 과정에 관여하는 lactic acid bacteria (Leuconostoc 속, Weisella 속, Lactococcus 속, Lactobacillus 속, Carnobacterium 속, Marinilactibacillus 속, Tetragenococcus 속)와 Bacillus 속, Pseudomonas 속, Micrococcus 속, Brevibacterium 속, Microbacterium 속과 Kocuria 속이 17가지 젓갈에서 광범위하게 분리되었으며, Salinicoccus 속, Halomonas 속, Cobetia 속, Lentibacillus 속, Paracoccus 속, Psychrobacter 속이 소수 분리되었다. 또한 분리된 미생물의 계통학적 분석을 통하여 기존에 보고된 적이 없는 신규 미생물 14종을 분리하였다.
1. 고온 CSTR은 비교적 짧은 start-up 기간과 높은 $H_2$ 수율을 나타내었다. $H_2$ 생산속도와 $H_2$ 수율의 안정화를 근거로 판단컨대 start-up 기간은 30일 이내이었으며, 최고 $H_2$ 수율은 2.4 mol $H_2/mol$ glucose이었다. 2. 비교적 긴 HRT와 침전조를 이용한 biomass의 재순환에도 불구하고, 유입 포도당의 농도가 낮아 biomass 농도는 다른 중온 반응기에서 보고된 것에 비해 낮은 편이었다. 3. 운전 초기에 $CH_4$이 발생하였으나 8일 이후부터는 pH를 1.0 이하로 유지하였더니 14일 이후로는 거의 검출되지 않는 것으로 봐서 메탄생성균이 식종균에 남아 있더라도 반응기 운전조건을 통해 $CH_4$ 발생을 억제할 수 있었다. 4. 식종 미생물과 반응기로부터 취한 시료의 DGGE band 패튼이 다른 것으로 보아 고온 CSTR 조건에서 식종된 미생물 군집의 조성이 변화하였음을 알 수 있었다. 5. DGGE 분석결과 초기 43일간의 운전기간 동안에 관찰된 미생물 군집조성은 동적인 변화를 나타내었다. 약 14일부터는 biogas 조성이 거의 일정하였으나 미생물 군집은 동적 변화를 나타내었다. F. gondwanens와 T. Thermoanaerobacterium과 계통발생학적으로 가장 연관이 있는 개체군들이 운전 21일과 41일째에 각각 우점으로 나타났다. 6. 본 연구에서 식종 슬러지를 열처리하는데 사용한 조건은 메탄생성균을 완전히 제거하는데 불충분하다는 것은 운전 초기에 $CH_4$이 biogas에서 검출되었고, 식종 슬러지와 반응기로부터 취한 시료에서 메탄생성균이 가지는 mcrA 유전자가 PCR로 증폭되었으므로 알 수 있었다. 7. 메탄생성균의 주요 목에 특이적인 primers를 사용하여 PCR을 실시한 결과 식종슬러지에 있는 메탄생성균들은 주로 Methanosarcinales와 Methanomicrobiales 목에 속하였으며, $CH_4$이 발생했던 때의 반응기에 있는 메탄생성균들은 주로 Methanobacteriales 목에 해당되는 것으로 나타났다.
1. $H_2$ 생산속도와 $H_2$ 수율의 안정화를 근거로 판단컨대 start-up 기간은 30일 이내로 나타나 중온 CSTR에 비해 짧은 편이었다. 2. 고온 CSTR의 최고 $H_2$ 수율은 2.4 mol $H_2/mol$ glucose로 나타나 보고 된 중온의 것에 비해 우수한 편에 속하였다. 3. 운전 초기에 $CH_4$이 발생하였으나 14일 이후부터는 pH를 5.0 이하로 유지하면 거의 검출되지 않는 것으로 봐서 메탄생성균이 식종균에 남아 있더라도 반응기 운전 조건을 통해 $CH_4$ 발생을 억제할 수 있었다. 4. 고온 CSTR은 초기 운전 후에 적용한 운전조건의 변화(유입 포도당 농도, pH, 및 온도)에 민감한 것으로 나타났다. 특히 pH 및 온도변화에 대해 $H_2$ 생산속도와 $H_2$ 수율, 포도당 제거율 면에서 반응기 성능의 감소 및 불안정이 나타나, 운전 조건 변화 후에 나타난 고온 CSTR의 성능회복이 쉽지 않음을 알 수 있었다. 5. 문헌에 보고 된 중온 CSTR과는 달리 고온 CSTR는 일정한 조건에서도 불안정 한 성능을 나타내기도 하였다. 6. 불안정한 반응기 성능은 lactate 농도 증가와 더불어 n-butyrate와 acetate 농도 감소를 동반하였다. 생산된 n-butyrate와 acetate의 농도는 lactate의 농도변화와 반대의 경향을 나타내었다. 7. 비교적 긴 HRT와 침전조를 이용한 biomass의 재순환에도 불구하고, 유입 포도당의 농도가 낮아 biomass 농도는 다른 중온 반응기에서 보고된 것에 비해 낮은 편이었다. 8. T. thermosaccharolyticum와 계통발생학적으로 관련된 개체군이 반응기 운전 후 약 40일부터 우점으로 나타나 반응기 성능과 상관없이 그 이후로 계속 우세한 것으로 나타났다.
Klebsiella pneumoniae는 인간의 피부, 구강, 호흡기, 요로, 장관 등에서 일반적으로 존재하는 상재세균이다. 하지만 지역사회획득 폐렴의 주요 원인이 되는 기회감염균이기도 하다. 옥수수수염은 K. pneumoniae, Staphylococcus aureus, Bacillus subtilis, Shigella spp., Salmonella spp., Escherichia coli, Pseudomonas aeruginosa 등 병원성 세균에 대해 항균활성이 있는 것으로 알려져 있다. 본 연구에서는 옥수수수염 농축액의 K. pneumoniae에 대한 항균능을 확인하고자 하였다. 이를 위해 K. pneumoniae ATCC 13883, broad-spectrum β-lactamase (BSBL) 생성균주, exteded-spectrum β-lactamase (ESBL) 생성균주 및 carbapenemase 생성균주를 수집하였다. 항균제 감수성 검사는 디스크 확산법을 사용하였고 내성유전자는 PCR 증폭과 염기서열 분석을 통해 확인하였다. 또한 항균활성 실험을 위하여 옥수수수염 농축액을 이용하여 MacConkey agar 평판배지(50%, 100%)를 제조하였고 균주의 성장억제를 확인하기 위하여 제조된 배지에 K. pneumoniae를 평판도말하여 37℃ 항온기에서 18시간동안 배양하였다. 수집된 K. pneumoniae는 SHV-1 (n=8), SHV-2a (n=8), SHV-5 (n=2), SHV-11 (n=2), SHV-12 (n=18), TEM-1 (n=10), CTX-M-3 (n=2), CTX-M-14 (n=2), CTX-M-15 (n=1), GES-5 (n=5), KPC-2 (n=6), KPC-3 (n=4) 및 NDM-1 (n=2)을 보유하고 있었다. 시험에 사용된 K. pneumoniae 균주 가운데 K. pneumoniae ATCC 13883균주에 대해 항균활성이 있었으나 BSBL, ESBL 및 carbapenemase 생성균주에 대한 항균활성을 확인할 수는 없었다. 따라서 옥수수수염 추출액은 K. pneumoniae에 의한 지역사회획득 감염증 예방과 회복에 도움을 줄 수 있을 것으로 생각된다.
본 연구에서는 생리활성물질을 생성하는 해양미생물을 분리하기 위하여 연근해역의 해면을 조사하였다. 해면 중의 미생물은 $1.2{\times}10{\sim}2.1{\times}10^3/g$ 범위에 있었고, 이들 중 방선균은 $0.01{\sim}0.5\%$(data 생략)로 나타났다. 분리된 미생물은 각각 배양하여 항균물질을 검토하였으며, 검토하던 중에 비교적 강한 항균물질을 생산하는 균주 No. 101을 분리, 선발하였다. 선발된 균의 특징은 sclerotia로 형태가 다양하였고, 다양한 배지에서 생육이 가능하였고, 기균사의 색깔은 회백색, 배면의 색깔은 주로 회백색에서 갈색으로 나타났으며, 젤라틴 액화능과 전분 분해능을 가지나 myo-inositol은 이용하지 못하였다. 이 균의 배양학적 특성은 최적 pH 및 온도가 $6.5{\sim}7.0$ 및 $30^{\circ}C$였으며, 탄소원으로는 starch가 30g/l, 질소원으로는 casamino acid 10g/l가 적합한 것으로 나타났다. 방선균 No. 101의 균체 및 균배양액의 추출물에 대한 항균 및 항진균성에서는 Staphylococcus aureus와 Bacillus subtilis에서 강한 항균력을, 효모와 곰팡이에 대하여는 낮은 항진균성을 나타내었다.
최근 괭생이모자반(Sargassum horneri)은 제주의 해안가와 해변, 연근해 양식장 등 매년 대량으로 속출되어 인근 양식업자 및 주민에게 막대한 피해를 주고 있는 추세이다. 본 연구에서는 제주 인근 연안으로 흘러 들어온 괭생이 모자반에 서식하고 있는 미생물의 다양성을 탐색하고 동정을 통한 미생물의 기능을 파악함으로써 생태학적 문제에 관한 기초자료를 제공하고자 하였다. Proteobacteria는 분리 된 균주 중 88%를 차지한 우점문으로 ${\alpha}-proteobacteria$강은 6속 10종으로 Pseudorhodobacter속이 40%를 차지하고 Paracoccus속 20%, Rhizobium, Albirhodobacter, Skermanella 및 Novosphingobium 속은 각각 10%를 차지했다. ${\beta}-proteobactera$강은 5속 10종으로 Hydrogenophaga속이 50%, Azoarcus 20%, 나머지 Oxalicibacterium, Duganella, Xenophilus속은 각각 10%를 차지했다. ${\gamma}-proteobacteria$강은 13속 57종으로 Proteobacteria문에서 74%로 우점강을 차지했고, Shewanella는 23%, Pseudomonas속 12%, Cobetia 19%, Rahnella, Vibrio 및 Serratia속은 4%, 나머지 Rheinheimera, Raoultella, Pantoea, Acinetobacter, Moraxella 및 Psychrobacter속은 2%를 차지했다. Actinobacteria는 1속 2종으로 Gordonia와 Nocardioides속이 각각 50%를 나타냈다. Bacteroidetes문은 3속 5종으로 Lacihabitans, Mariniflexile속은 33%, 나머지 Dyadobacter, Cellulophaga와 Ferruginibacter속은 11%를 차지했다.
Although microorganisms are, in fact, the most diverse and abundant type of organism on Earth, the ecological functions of microbial populations remains poorly understood. A variety of bacteria including marine Vibrios encounter numerous ecological challenges, such as UV light, predation, competition, and seasonal variations in seawater including pH, salinity, nutrient levels, temperature and so forth. In order to survive and proliferate under variable conditions, they have to develop elaborate means of communication to meet the challenges to which they are exposed. In bacteria, a range of biological functions have recently been found to be regulated by a population density-dependent cell-cell signaling mechanism known as quorum-sensing (QS). In other words, bacterial cells sense population density by monitoring the presence of self-produced extracellular autoinducers (AI). N-acylhomoserine lactone (AHL)-dependent quorum-sensing was first discovered in two luminescent marine bacteria, Vibrio fischeri and Vibrio harveyi. The LuxI/R system of V. fischeriis the paradigm of Gram-negative quorum-sensing systems. At high population density, the accumulated signalstrigger the expression of target genes and thereby initiate a new set of biological activities. Several QS systems have been identified so far. Among them, an AHL-dependent QS system has been found to control biofilm formation in several bacterial species, including Pseudomonas aeruginosa, Aeromonas hydrophila, Burkholderia cepacia, and Serratia liquefaciens. Bacterial biofilm is a structured community of bacterial cells enclosed in a self-produced polymeric matrix that adheres to an inert or living surface. Extracellular signal molecules have been implicated in biofilm formation. Agrobacterium tumefaciens strain NT1(traR, tra::lacZ749) and Chromobacterium violaceum strain CV026 are used as biosensors to detect AHL signals. Quorum sensing in lactic acid bacteria involves peptides that are directly sensed by membrane-located histidine kinases, after which the signal is transmitted to an intracellular regulator. In the nisin autoregulation process in Lactococcus lactis, the NisK protein acts as the sensor for nisin, and NisR protein as the response regulator activatingthe transcription of target genes. For control over growth and survival in bacterial communities, various strategies need to be developed by which receptors of the signal molecules are interfered with or the synthesis and release of the molecules is controlled. However, much is still unknown about the metabolic processes involved in such signal transduction and whether or not various foods and food ingredients may affect communication between spoilage or pathogenic bacteria. In five to ten years, we will be able to discover new signal molecules, some of which may have applications in food preservation to inhibit the growth of pathogens on foods.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
