Inhibition of Osteoclast Differentiation by Wheat Bran Butanol Fraction (밀기울 부탄올 분획물이 파골세포의 분화억제에 미치는 효과)

  • Osteoporosis is a disease of bones that leads to an increased risk of fracture. In osteoporosis, the bone mineral density is reduced, bone microarchitecture deteriorates, and the amount and variety of proteins in bone are altered. $It^{\circ}{\emptyset}s$ caused by the imbalance between born resorption and born formation. Recently natural products from plants have been extensively studied as therapeutic drugs to treat and prevent various diseases. Wheat bran is the hard outer layers of wheat grain and produced as a by-product of milling in the production of refined grains. In oriental medicines, Bu So Maek (Tritici Immaturi Semen) with wheat bran has been used as bronchitis, sedatives and anti-sweating effects. However effects of wheat bran butanol fraction (WBB, 50 ${\mu}g/ml$) in osteoclast differentiation remains unknown yet. Thus we investigated the effects of WBB on RANKL induced osteoclast differentiation. WBB inhibited osteoclast differentiation by downregulating the RANKL-induced activations of MAP kinases. Moreover mRNA expression of osteoclast-mediating molecules such as c-Fos, NFATc1 and DC-STAMP were attenuated by WBB during osteoclast differentiation. The finding of this study show that WBB and its components might prevent osteoclast-related bone loss.

Machine Learning-Based Detection of Cache Side Channel Attack Using Performance Counter Monitor of CPU (Performance Counter Monitor를 이용한 머신 러닝 기반 캐시 부채널 공격 탐지)

  • Recently, several cache side channel attacks have been proposed to extract secret information by exploiting design flaws of the microarchitecture. The Flush+Reload attack, one of the cache side channel attack, can be applied to malicious application attacks due to its properties of high resolution and low noise. In this paper, we proposed a detection system, which detects the cache-based attacks using the PCM(Performance Counter Monitor) for monitoring CPU cache activity. Especially, we observed the variation of each counter value of PCM in case of two kinds of attacks, Spectre attack and secret recovering attack during AES encryption. As a result, we found that four hardware counters were sensitive to cache side channel attacks. Our detector based on machine learning including SVM(Support Vector Machine), RF(Random Forest) and MLP(Multi Level Perceptron) can detect the cache side channel attacks with high detection accuracy.

Fractal dimension, lacunarity, and cortical thickness in the mandible: Analyzing differences between healthy men and women with cone-beam computed tomography

  • Purpose: The objective of this study was to assess the fractal dimension, lacunarity, trabecular microarchitecture parameters, and cortical linear measurements in the mandibles of male and female individuals to identify differences between them. Materials and Methods: In total, 116 cone-beam computed tomography scans of healthy individuals of different ages (57 men and 59 women, aged between 20 and 60 years) were selected. The following bone parameters were measured: 1) buccal, lingual, and basal cortical bone thickness in 5 standard parasagittal sections (the midline, the left and right sides of the lower lateral incisors, and the left and right sides of the lower canines); 2) the bone volume fraction of 10 sequential axial sections from each patient by creating a volume of interest in the area between the lower canines; and 3) fractal dimension and lacunarity using grayscale images of the same region of the volume of interest in the anterior mandible. Spearman correlation coefficients and the Mann-Whitney test were used. Results: A significant and positive correlation was found between age and cortical thickness, especially in the region of the central incisors. Significant differences between sexes in terms of fractal dimension, lacunarity, and bone volume were found. Women revealed lower fractal dimension values and higher lacunarity and bone volume ratio values than men. Conclusion: Fractal dimension, lacunarity, trabecular bone volume, and cortical thickness were different between men and women of different ages.

Dependencies of Group Velocity and Attenuation Coefficient on Structural Properties in Copper and Nickel Foams with an Open-Celled Structure as Trabecular-Bone-Mimicking Phantoms (해면질골 팬텀으로서 개포된 구조를 갖는 구리폼 및 니켈폼에서 구조적 특성에 대한 군속도 및 감쇠계수의 의존성)

  • In the present study, copper and nickel foams with an open-celled structure as trabecular-bone-mimicking phantoms were used to investigate the dependencies of group velocity and attenuation coefficient on structural properties such as trabecular thickness (Tb.Th) and trabecular separation (Tb.Sp) in trabecular bone. The group velocity and attenuation coefficient of the copper and nickel foams were measured by a through-transmission method in water, using a pair of broadband, unfocused transducers with a diameter of 12.7 mm and a center frequency of 1.0 MHz. The separation of the Biot's fast and slow waves was consistently observed in the ultrasonic signals transmitted through the copper and nickel foams. The group velocities of the copper and nickel foams showed highly positive correlations with Tb.Th and Tb.Sp. The attenuation coefficient of the copper foam showed a highly negative correlation with Tb.Th and Tb.Sp, whereas that of the nickle foam showed a highly positive correlation with Tb.Th and Tb.Sp. These results advance our understanding of those previously reported by other researchers using trabecular bone samples or phantoms.

Anti-osteoarthritis effect of Boswellia serrata gum resin extract in monosodium iodoacetate-induced osteoarthritic Sprague-Dawley rats (Monosodium iodoacetate 유도 골관절염 동물모델에서 보스웰리아 검레진 추출물의 항골관절염 효과 연구)

  • Purpose: The aim of this study was to investigate the anti-osteoarthritic effect of the ethanol extract of Boswellia serrata gum resin (FJH-UBS) enriched with keto-β-boswellic acid and 3-O-acetyl-11-keto-β-boswellic acid compared to the conventional Boswellia serrata extract by adding the process of removing oil with hexane, in the monosodium iodoacetate (MIA)-induced osteoarthritis rat model. Methods: Sprague-Dawley (SD) rats were orally administered 0, 40, or 80 mg of FJH-UBS/kg body weight (BW)/day for 5 weeks and injected with MIA intra-articularly into right knee joints on day 15 to induce osteoarthritis. Changes in the knee joint microarchitecture, cartilage degradation, the expression of inflammatory mediators, cytokines, and matrix metalloproteinases (MMPs) in serum and synovia were observed. Results: Oral administration of FJH-UBS (80 mg/kg BW/day) reduced MIA-induced knee swelling and cartilage degradation and increased the expression of type II collagen and aggrecan in articular cartilage. Furthermore, FJH-UBS administration reduced MIA-induced increases in the serum levels of prostaglandin E2, leukotriene B4, interleukin (IL)-1β, IL-6, and MMP-13, and MIA-induced increases in the mRNA expressions of inducible nitric oxide synthase, cyclooxygenase-2, 5-lipoxygenase, IL-1β, IL-6, TNF-α, MMP-2, MMP-9, and MMP-13 in the synovia of knee joints. Conclusion: These results indicate that FJH-UBS exerts its anti-osteoarthritic effects by suppressing the expressions of inflammatory cytokines and MMPs and, thus, cartilage degradation. Furthermore, they suggest that FJH-UBS has potential use as a functional food that improves joint and cartilage health.

Effects of Resistance Training on BMD and Bone Metabolism Related Markers in Aging Rats (저항성 훈련이 노화흰쥐의 골밀도 및 골대사 관련 지표에 미치는 영향)

  • The purpose of this study was to investigate effect of resistance training on BMD and bone metabolism related markers in aging rats. Thirty male Spraugue-Daweley rats were divided into sedentary (CON; n=10 ) non-load resistance trained(NLRTG; n=10), and load resistance trained(LRTG; n=10) groups at the age of 64 weeks. The rats in the resistance training groups((NLRTG and LRTG) performed the tower climbing exercise 4 times a week. The LRTG groups were conditioned to climb a vertical ladder with weights appended to their tail 4 days/wk for 12 wks. After 12 weeks of exercise, serum osteocalcin, bone mineral density (BMD), breaking force, ash, Ca, and P in the femur were measured. After training, serum osteocalcin (OC) was significantly (p < 0.05) higher in both LRTG and NLRTG when compared to Control. Right femur BMD was significantly (p < 0.05) greater for LRTG when compared to both NLRTG and Control with no significant difference between NLRTG and Conrtol. The breaking force of femur was significantly (p < 0.05) greater for LRTG and NLRTG when compared to Control. The Ash, Ca, content of femur were significantly increased in resistance training groups than control group. These results suggest that the increase in bone mineral density induced by resistance training is mediated by changes in bone microarchitecture as well as training intensity and osteocalcin.

Estimation of Structural Properties from the Measurements of Phase Velocity and Attenuation Coefficient in Trabecular Bone (해면질골에서 위상속도 및 감쇠계수 측정에 의한 구조적 특성 평가)

  • Trabecular-bone-mimicking phantoms consisting of parallel-nylon-wire arrays were used to investigate correlations of phase velocity and attenuation coefficient with structural properties in trabecular bone. Trabecular separation (Tb.Sp) of the 7 trabecular-bone-mimicking phantoms ranged from 300 to $900\;{\mu}m$ and volume fraction (VF) from 1.6% to 8.7%. Phase velocity and attenuation coefficient of the phantoms were measured by using a through-transmission method in water, with a matched pair of broadband unfocused transducers with a diameter of 12.7 mm and a center frequency of 1 MHz. Phase velocity and attenuation coefficient at 1 MHz decreased almost linearly with increasing Tb. Sp and increased almost linearly with increasing VF. The simple and multiple linear regression models with phase velocity and attenuation coefficient as independent vanables and Tb.Sp and VF as dependent variables demonstrated that the coefficients of determination for the prediction of VF were higher than those for the prediction of Tb.Sp. The results obtained in the trabecular-bone-mimicking phantoms consisting of parallel-nylon-wire arrays were consistent with those in human trabecular bone suggesting that the structural properties can be estimated from the measurements of phase velocity and attenuation coefficient in trabecular bone.
