Microalgal cultivation using wastewater is now regarded as essential for biodiesel production, as two goals can be achieved simultaneously; that is, nutrient removal efficiency and biomass production. Therefore, this study examined the effects of carbon sources, the N:P ratio, and the hydraulic retention time (HRT) to identify the optimal conditions for nutrient removal efficiency and biomass production. The effluent from a 2nd lagoon was used to cultivate microalgae. Whereas the algal species diversity and lipid content increased with a longer HRT, the algal biomass productivity decreased. Different carbon sources also affected the algal species composition. Diatoms were dominant with an increased pH when bicarbonate was supplied. However, 2% $CO_2$ gas led to a lower pH and the dominance of filamentous green algae with a much lower biomass productivity. Among the experiments, the highest chlorophyll-a concentration and lipid productivity were obtained with the addition of phosphate up to 0.5 mg/l P, since phosphorus was in short supply compared with nitrogen. The N and P removal efficiencies were also higher with a balanced N:P ratio, based on the addition of phosphate. Thus, optimizing the N:P ratio for the dominant algae could be critical in attaining higher algal growth, lipid productivity, and nutrient removal efficiency.

Exposure of cells to ultraviolet B (UVB) radiation can induce production of free radicals and reactive oxygen species (ROS), which damage cellular components. In addition, these agents can stimulate the expression of matrix metalloproteinase (MMP) and decrease collagen synthesis in human skin cells. In this study, we examined the anti-photoaging effects of extracts of Tetraselmis suecica (W-TS). W-TS showed the strongest scavenging activity against 2,2-difenyl-1-picrylhydrazyl (DPPH) and peroxyl radicals, followed by superoxide anions from the xanthine/xanthine oxidase system. We observed that the levels of both intracellular ROS and lipid peroxidation significantly increased in UVB-irradiated human skin fibroblast cells. Furthermore, the activities of enzymatic antioxidants (e.g., superoxide dismutase) and the levels of non-enzymatic antioxidants (e.g., glutathione) significantly decreased in cells. However, W-TS pretreatment, at the maximum tested concentration, significantly decreased intracellular ROS and malondialdehyde (MDA) levels, and increased superoxide dismutase and glutathione levels in the cells. At this same concentration, W-TS did not show cytotoxicity. Type 1 procollagen and MMP-1 released were quantified using RT-PCR techniques. The results showed that W-TS protected type 1 procollagen against UVB-induced depletion in fibroblast cells in a dose-dependent manner via inhibition of UVB-induced MMP-1. Taken together, the results of the study suggest that W-TS effectively inhibits UVB-induced photoaging in skin fibroblasts by its strong anti-oxidant ability.
2010년 영산강 섬진강 수계의 29개 호소 45개 지점의 환경조사에서 동정된 식물플랑크톤은 총 371종으로 규조강과 녹조강이 주를 이루었다. 호소의 규모와 식물플랑크톤의 분포 양상 사이에 상관성이 거의 없는 것으로 나타났다. 호소의 유역면적과 식물플랑크톤 현존량은 상관성을 보이지 않았고, 유역면적과 출현종수 사이에도 상관성이 매우 낮았다. 수표면적과 식물플랑크톤 출현종수와 현존량 사이에는 낮은 상관성이 나타났다. 본 조사 호소들에서 호소의 규모와 식물플랑크톤 군집 사이에 상관성이 높지 않은 것은 호소의 규모가 작고 수량의 연중 변동이 크며 먹이망과 인위적인 간섭이 식물플랑크톤의 자연적인 분포를 왜곡하였기 때문으로 생각된다. 호소의 축조연대와 식물플랑크톤의 분포 사이에도 상관성이 없는 것으로 나타났다. 식물플랑크톤의 출현 현존량은 호소의 형성연대와 상관관계가 없는 분포를 보여주었다. 축조연대와 클로로필 a의 농도 사이에는 더 낮은 상관성을 보여주었다. 본 조사가 이루어진 호소들에서는 생성연대의 차이에 따른 부영양화 정도에 차이가 있지만 다른 요소들(체류시간, 수심, 생물상)에 의한 효과로 호소 생성연대의 영향을 찾을 수 없었다.
Microalgae hold promise as producers of sustainable biomass for the production of biofuels and other biomaterials. However, the selection of strains with efficient and robust production of desirable resources remains challenging. In this study, we isolated a green microalga from Korea and analyzed its morphological, molecular, and biochemical characteristics. Microscopic and phylogenetic analyses demonstrated that the isolate could be classified into the genus Chlamydomonas, and we designated the isolate Chlamydomonas sp. KIOST -1. Compositions of protein, lipid, and carbohydrate in the microalgal cells were estimated to be 58.8 ± 0.2%, 22.7 ± 1.2%, and 18.5 ± 1.0%, respectively. Similar to other microalgae belonging to Chlorophyceae, the dominant amino acid and monosaccharide in Chlamydomonas sp. KIOST-1 were glutamic acid and glucose. On the other hand, the proportions of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids clearly differed from other species in the genus Chlamydomonas, and monounsaturated fatty acids accounted for a large portion (41.3%) of the total fatty acids in the isolate. Based on these results, Chlamydomonas sp. KIOST-1 has advantageous characteristics for biomass production.

Omega-3 fatty acids, including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are polyunsaturated fatty acids beneficial to human health. A limited number of microalgae have been used for commercial omega-3 production, which necessitates the identification of new microalgae with high omega-3 contents. We explored the fatty acid composition and EPA and DHA contents of the mixotrophic dinoflagellate Gymnodinium smaydae fed with the optimal algal prey species Heterocapsa rotundata. Cells of G. smaydae were found to be rich in omega-3 fatty acids. In particular, the DHA content of G. smaydae was 21 mg g-1 dry weight, accounting for 43% of the total fatty acid content. The percentage of DHA in the total fatty acid content of G. smaydae was the highest among the reported microalgae except for Crypthecodinium cohnii. Moreover, to determine if the prey supply interval affected the growth rate of G. smaydae and its fatty acid content, three different prey supply intervals (daily, once every 2 d, and once for 4 d) were tested. Daily prey supply yielded the highest total fatty acid and DHA contents in G. smaydae. Furthermore, we successfully produced high-density G. smaydae cultures semi-continuously for 43 d with daily prey supply. During the semi-continuous cultivation period, the highest density of G. smaydae was 57,000 cells mL-1, with an average growth rate of 0.7 d-1. Taken together, the percentage of EPA and DHA in the total fatty acid content was maintained in the range of 54.2-56.9%. The results of this study support G. smaydae as a promising microalgal candidate for commercial DHA production and demonstrate that daily supply of prey can efficiently produce high-density G. smaydae cultures for more than a month.
A total of 646 strains, including green algae and diatoms, were isolated from 220 samples to screen microalgae with high lipid productivity (LP). The samples were obtained from nine habitats in Northern Xinjiang, China in June 2013. This study initially identified eight lipidrich strains, namely, Desmodesmus intermedius XJ-498, D. intermedius XJ-145, D. intermedius XJ-99, Monoraphidium pusillum XJ-489, M. dybowskii XJ-435, M. dybowskii XJ-151, Mychonastes homosphaera XJ-488, and Podohedriella falcata XJ-176, based on 18S rDNA sequencing. The strains were cultured in a photobioreactor for the same period. Results showed that the specific growth rate (day-1) of M. pusillum XJ-489 was the highest (1.14 ± 0.06), and the biomass concentration (g/l) of D. intermedius XJ-99 was the highest (2.84 ± 0.3). Futhermore, the lipid content (%) of M. dybowskii XJ-151 was the highest (33.5 ± 4.38), and the lipid productivity (mg l-1 day-1) of My. homosphaera XJ-488 was the highest (86.41 ± 9.04). C16 to C18 accounted for 86% to 98% of the total lipid, and the biodiesel qualities of the selected algae corresponded to international standards. This study suggests that My. homosphaera XJ-488, D. intermedius XJ-99, and M. dybowskii XJ-151 are the most potential strains for biodiesel production among all the isolated strains.

A unicellular green alga was axenically isolated from the Port of Jeongja, Ulsan, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Heterochlorella luteoviridis. This is the first report of this species in Korea. The microalgal strain was named as H. luteoviridis MM0014 and its growth, lipid composition, and biomass properties were investigated. The strain thrived over a wide range of temperatures ($5-30^{\circ}C$) and withstood up to 0.5 M NaCl. The results of gas chromatography/mass spectrometry analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids. Its major fatty acids were linoleic acid (35.6%) and ${\alpha}$-linolenic acid (16.2%). Thus, this indigenous marine microalga is a potential alternative source of ${\omega}3$ and ${\omega}6$ polyunsaturated fatty acids, which are currently obtained from fish and plant oils. Ultimate analysis indicated that the gross calorific value was $19.7MJ\;kg^{-1}$. In addition, the biomass may serve as an excellent animal feed because of its high protein content (51.5%). Therefore, H. luteoviridis MM0014 shows promise for applications in the production of microalgae-based biochemicals and biomass feedstock.
미세조류는 통상적인 에너지 작물에 비하여 빠른 성장속도와 높은 오일함량으로 바이오디젤 생산의 원료로 관심을 받고 있다. 미세조류의 지질은 주로 트리글리세라이드인 중성지방으로 에스테르교환반응을 통하여 바이오디젤인 지방산 메틸에스테르로 전환할 수 있다. 본 논문에서는 영양분의 제한, 염도 및 금속 성분의 변화와 같이 미세조류의 지질 생산을 촉진할 수 있는 배양환경 스트레스 조건의 영향들을 비교 고찰하였다. 사용하는 미세조류 종에 따라 스트레스에 대응하여 지질의 양이 변하거나 구성하는 지방산의 조성이 변화될 수 있다. 비록 질소원 제한 조건이 가장 많이 사용되는 지질생산 촉진조건이긴 하지만, 미세조류로부터 바이오디젤 생산성의 향상을 위해서는 그 외에도 영양분 과잉 조건, 염도의 변화, pH, 온도, 금속 성분 농도 변화 등의 다른 조건들도 고려되어야 한다.
2011년 영산강 섬진강 수계 29개 호소조사에서 동정된 식물플랑크톤은 규조강 161종, 녹조강 157종, 남조강 39종, 기타 67여종으로 총 424종이었다. 조사기간 동안 4차 조사시기에 상대적으로 더 다양한 종이 출현하였으며 대동호, 대동제, 소포제, 군곡제, 개초제, 둔전제 지점에서 다양한 종들이 출현하였고, 영산호를 비롯한 하천형 호소에서는 비교적 빈약한 종이 출현하였다. 각 지점에서 20% 이상의 우점율을 보인 종은 대부분 규조류였고 2차와 3차 조사에서 녹조류와 남조류도 우점의 빈도가 높아졌다. 개체군 밀도는 조사 호소나 시기에 따른 뚜렷한 패턴이 없었고 특정 종의 성장에 크게 의존하는 특성을 나타냈다. 클로로필-$a$의 농도는 조사호소와 시기에 따른 편차가 매우 컸으며 식물플랑크톤의 개체수와 일치하지 않는 것으로 나타났다. 총질소와 총인의 농도와 클로로필-$a$의 농도 사이에는 매우 낮은 상관성이 나타났다.
적조 다발지역인 진동만에서 살조세균의 동태를 파악하기 위하여, 1998년 5월 19일부터 7월 7일까지, chlorophyll-a 및 살조세균의 계수$\cdot$분리하였고, 분리한 살조세균의 살조특성을 조사하였다. 본 연구기간동안 살조세균의 밀도는 $6.0\times10^1\~6.4\times10^5$cell $\ell^{-1}$의 범위에서 변동하였으며, chlorophyll-a의 농도의 증가 및 감소에 대하여 각각 1주일의 lag time을 두고 이와 동일한 변동양상을 나타내었다. 현장시료에서 분리된 3종의 살조세균 H519S5-4, H605S5-15 및 H605S5-22 균주의 살조범위를 침편모조류 Heterosigma akashiwo와 Chattonelia sp., 와편모조류 Gymnodinium catenatum, Gyrodinium impudicum 및 Cochlodinium polyklikoides, 규조류 Chaetoceros sp., Cescinodiscus granii, Ditylum brightwellii 및 Thalassiosira rotula를 대상으로 조사하였다. H60585-22 균주는 실험에 사용된 9종의 미세조류를 전부 사멸시켜 광범위한 살조범위를 나타내었다. 또한, 균주 H605S5-15는 H. akashiwo, G. catenatum, T. rotula 및 Chattonella sp.에 대하여, H519S5-4는 H. akashiwo, Chattonella sp., Chaetoceros sp. 및 G. catenatum에 대하여 살조력을 나타내었다. 살조유형은 3종 모두 세포 외 물질을 분비하여 살조하는 살조인자분비형 세균으로 나타났다. 세균이 나타내는 살조력은 식물성 플랑크톤의 군집동태에 영향을 미치는 중요한 요인이며, 연안에서 적조의 급격한 소멸에 관여하는 것으로 사료된다.
