The mechanisms leading to ontogenetic diet shift in a microcanivore, Pterogobius elapoides(Gobiidae)

  • A variety of fish species undergo an ontogenetic change in prey selectivity, and several potentially interacting factors, including nutrient requirement, microhabitat change, and foraging ability, may account for the occurrence of the shift. Here we examine the foraging ecology and ontogenetic diet shift of a micro-carnivorous goby, Pterogobius elapoides (serpentine goby), dominant component of fish assemblage in shallow rocky areas off the coast in Korea and Japan. Although most other gobies are primarily benthic carnivores, P. elapoides is a semipelagic fish; however, little is known about how those species change their foraging tactics with growth. In our diet analyses, the most common diet was pelagic copepods and benthic amphipods, and diet shift was observed from pelagic to benthic with growth. The ontogenetic diet shift seems to be the result of the preference for energetically more profitable prey in larger-size classes as well as the results of different prey availability due to among-habitat variation in diet. However, differential food preference does not appear to affect individual scope for searching food. Several factors such as predation pressures and interspecific resource partitioning might contribute to the changes in diet observed among size classes, which were included in our ongoing tests.

성장 중인 쥐에서 음식물의 경도가 하악 과두의 해면골에 미치는 영향 : 미세전산화 단층촬영을 이용한 연구 (THE EFFECTS OF DIETARY CONSISTENCY ON THE TRABECULAR BONE ARCHITECTURE IN GROWING MOUSE MANDIBULAR CONDYLE : A STUDY USING MICRO-CONFUTED TOMOGRAPHY)

  • 하악 과두의 발달과 증식은 측두하악관절 부위의 생역학적 환경의 변화에 따라 변경될 수 있다. 이 부위에 전달되는 생역학적 하중은 섭취하는 음식물의 경도를 다르게 함으로써 변화시킬 수 있다. 이번 연구의 목적은 성장 중인 쥐에서 부드러운 음식물의 섭취에 의해 저작력을 변화시키는 것이 하악 과두의 해면골의 형태를 변화시킬 수 있는지 미세전산화 단층촬영을 이용하여 분석하는 것이었다. 생후 21일 된 C57BL/6 쥐 36마리를 무작위로 두 그룹으로 나누었다. 8주 동안 대조군의 쥐들은 일반적인 딱딱한 덩어리의 사료를, 실험군의 쥐들은 덩어리의 사료를 잘게 갈은 후 물과 섞어 부드럽게 만들어 먹였다. 또한 실험군의 쥐들의 하악 절치를 일주일에 두 번씩 잘라서 짧게 만들었다. 8주 후 모든 쥐들을 희생하여 우측 하악 과두를 준비하였다. 미세전산화 단층촬영과 삼차원 영상 분석프로그램을 이용하여 하악 과두 해면골의 bone volume(BV), bone surface(BS), total volume(TV) bone volume fraction(BV/TV), surface to volume ratio(BS/BV), trabecular thickness(Tb. Th.), structure model index(SMI)와 degree of anisotropy(DA)를 측정하고 이들 값으로부터 trabecular number(Tb. N.)와 trabecular separation (Tb. Sp.)을 계산하였다 미세전산화 단층영상을 얻은 후 하악 과두의 조직 표본을 만들었다. 연구 결과는 다음과 같았다. 1. Bone volume fraction(BV/TV), trabecular thickness(Tb. Th.)와 trabecular number(Tb. N.)가 대조군에 비해 부드러운 음식을 먹인 실험군에서 유의하게 감소되었다(p<0.05). 2. Trabecular separation(Tb. Sp.)은 부드러운 음식을 먹인 실험군에서 유의하게 증가하였다(p<0.05). 3. Surface to volume ratio(BS/BV), structure model index(SMI), degree of anisotropy (DA)는 두 군 사이에 유의한 차이를 보이지 않았다(p>0.05). 4. 조직 절편을 관찰한 결과, 부드러운 사료를 먹인 실험군에서 하악 과두의 연골층의 증식 층과 전체 두께가 상당히 감소하였다.

미립자사료에 효모, Kluyveromyces fragilis와 Candida utilis의 첨가가 은어 자어의 성장과 체성분에 미치는 영향 (Growth and Body Composition of Larval Ayu (Plecoglossus altivelis) Fed the Micro-diets Containing Kluyveromyces fragilis and Candida utilis)

  • 은어 자어 배합사료에 효모의 첨가효과를 조사하기 위해 K. fragilis, protoplasted K. fragilis, C. utilis, protoplasted C. utilis 및 맥주효모를 각각 $5{\%}씩$ 첨가한 6종류의 실험사료를 미립자 크럼블 형태로 제조하여 평균체중 l00 mg 전후의 자어를 대상으로 사육 실험하였다. 사료별 3반복으로 7주간 사육 실험한 후의 생존율은 protoplasted K. fragilis, C. utilis 및 protoplasted C. utilis 첨가구가 $93{\~}94{\%}$로 효모 무첨가구인 대조구의 $87{\%}$보다 유의하게 높은 경향이었으며 (P<0.05), K. fragilis 및 맥주효모 첨가구는 대조구와 유의한 차이를 보이지 않았다 (P>0.05). 증체율은 protoplasted K. fragilis 첨가구가 $149{\%}$로 대조구의 $78{\%}$보다 유의하게 높았고 (P<0.05), 그 외 실험구들과는 유의한 차이가 없었다 (P>0.05). 전어체의 단백질과 회분 함량은 모든 실험구간에 유의한 차이는 없었고 (P>0.05), 지질 함량은 K. fragilis와 맥주효모 첨가구들이 대조구보다 낮은 값을 보였다 (P<0.05). 전어체의 아미노산 조성중 Asp는 효모 첨가구들이 대조구보다 유의하게 높은 값을 보였으며 (P<0.05), 그 외 아미노산들은 실험구간에 유의한 차이가 없었다 (P>0.05). 이상의 결과로부터 C. utilis보다는 K. fragilis를 외벽 처리하여 첨가하는 것이 은어 자어의 성장과 어체 지질 성분을 더 개선시킬 수 있을 것으로 전망된다.

Effect of vegetable oils with different fatty acid composition on high-fat diet-induced obesity and colon inflammation

  • BACKGROUND/OBJECTIVES: Different fatty acids exert different health benefits. This study investigated the potential protective effects of perilla, olive, and safflower oils on high-fat diet-induced obesity and colon inflammation. MATERIALS/METHODS: Five-week old, C57BL/6J mice were assigned to 5 groups: low-fat diet (LFD), high-fat diet (HFD) and high-fat diet supplemented with-perilla oil (HPO), olive oil (HOO), and safflower oil (HSO). After 16 weeks of the experimental period, the mice were sacrificed, and blood and tissues were collected. The serum was analyzed for obesity- and inflammation-related biomarkers. Gene expression of the biomarkers in the liver, adipose tissue, and colon tissue was analyzed. Micro-computed tomography (CT) analysis was performed one week before sacrifice. RESULTS: Treatment with all the three oils significantly improved obesity-induced increases in body weight, liver weight, and epididymal fat weight as well as serum triglyceride and leptin levels. Treatment with perilla oil (PO) and safflower oil (SO) increased adiponectin levels. The micro-CT analysis revealed that PO and SO reduced abdominal fat volume considerably. The mRNA expression of lipogenic genes was reduced in all the three oilsupplemented groups and PO upregulated lipid oxidation in the liver. Supplementation of oils improved macroscopic score, increased colon length, and decreased serum endotoxin and proinflammatory cytokine levels in the colon. The abundance of Bifidobacteria was increased and that of Enterobacteriaceae was reduced in the PO-supplemented group. All three oils reduced proinflammatory cytokine levels, as indicated by the mRNA expression. In addition, PO increased the expression of tight junction proteins. CONCLUSIONS: Taken together, our data indicate that the three oils exert similar anti-obesity effects. Interestingly, compared with olive oil and SO, PO provides better protection against high-fat diet-induced colon inflammation, suggesting that PO consumption helps manage inflammation-related diseases and provides omega-3 fatty acids needed by the body.

고콜레스테롤 식이 섭취 쥐에서 quercetin의 간 AMPK 및 microRNA-21 조절을 통한 지질대사 개선 효과 (Effects of quercetin on the improvement of lipid metabolism through regulating hepatic AMPK and microRNA-21 in high cholesterol diet-fed mice)

  • Quercetin의 지질대사 개선 효과에 대한 작용기전을 확인하기 위해 C57BL/6J mouse를 사용하여 실험을 수행하였다. 고콜레스테롤혈증을 유도하기 위해 6주간 1% 콜레스테롤과 0.5% cholic acid를 함유하는 고콜레스테롤 식이를 급여하였으며, quercetin은 0.05%와 0.1%의 수준으로 고콜레스테롤 식이에 추가하여 같은 기간 동안 제공하였다. Quercetin은 혈청과 간의 중성지방 및 콜레스테롤 수준을 용량 의존적으로 감소하는 것으로 나타났다. 고콜레스테롤 식이를 섭취한 쥐의 간에서 지방 합성을 촉진하는 SREBP-1c, ACC1 및 FAS 유전자 발현이 quercetin 섭취에 의해 억제되는 것을 확인하였다. Quercetin은 간세포 내에서 에너지 대사를 조절하는 AMPK 활성을 증가시켰다. 이에 반해 암세포 증식을 촉진하고 지방간에서 높게 발현되는 miR-21 발현은 quercetin 섭취에 의해 감소되었다. 본 연구의 결과는 quercetin이 고콜레스테롤 식이 섭취 쥐에서 혈청과 간의 지질 수준을 낮추는 지질대사 개선 효과가 있으며, 이러한 효과의 일부는 간 내 지방합성 유전자 (SREBP-1c, ACC1 및 FAS) 발현, AMPK 활성 및 miR-21 조절을 통해 매개된다는 것을 시사한다.

Methionine, Threonine, Tryptophane을 添加한 砂糖飼料에 의한 白쥐의 體重增加 및 Plasm, 肝臟中의 遊離아미노酸에 대한 硏究 (Studies on the body growth and free amino acids in plasm and liver of rat fed on sucrose diet adding Methionine, Threonine and Tryptophane)

  • By the micro-bioassay method, quantitative studies were made on the increment of body weight and changes in free-lysine and threonine contents in the liver and plasm of rats, when they were fed on sucrose diet added with 0.3% DL-methionine, 0.36% DL-threonine and 0.1% L-tryptophane. The results of the experiment were as follows: (1) The weight gain and growth gain of rats decreased more when they were fed on 0.36% DL-threonine diet (Sth-group) than 0.3% DL-methionine diet (Sm-group). But those gain were increased when they were fed on 0.1% L-tryptophane diet (Str-group). (2) The contents of free-lysine and threonine in plasm and liver of rats were decreased more when they were fed on 0. 36% DL-threonine diet than 0.3% DL-methionine diet. And, those free amino acids were increased greatly when they were fed on 0.1% L-tryptophane diet. (3) The increment of body weight was the biggest when the free amino acid pattern showed that the contents of free-lysine decreased and threonine increased in the plasm and liver of rats being fed on 0.l% L-tryptophane diet.

고지방식이 수컷 마우스 비만모델에서 micro-CT를 이용한 마황(麻黃)과 마우(魔芋)의 복부비만 조절효과 (Herba Ephedrae and Rhizoma Amorphophalli modulates visceral obesity in micro-CT of high fat induced obese male mice)

  • Objectives : We investigated the effects of Herba Ephedrae and Rhizoma Amorphophalli on high fat diet induced obese male mice. Methods : 8 weeks old, high fat diet induced obese male mice were divided into 5 groups: C57BL/6 normal control, obese vehicle control, GGEx55 (Herba Ephedrae), GGEx61 (Rhizoma Amorphophalli), GGEx62 (Herba Ephedrae + Rhizoma Amorphophalli). After mice were treated with GGEx for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, rectal temperature, fat weight, plasma leptin and lipid levels. We also took micro-computerized axial tomography (micro-CT) on the mice. Results : 1. GGEx55 and GGEx62 groups significantly decreased body weight gain and feeding efficiency ratio compared with vehicle control. But they significantly increased rectal temperature. 2. Plasma total cholesterol and LDL-cholesterol concentrations were significantly increased by GGEx55 groups, whereas were significantly decreased by GGEx62 groups compared with vehicle control. 3. GGEx55 and GGEx62 groups significantly decreased total, subcutaneous and visceral fat as well as fat areas in micro-CT analysis of abdomen compared with vehicle control. 4. Plasma GOT and GPT concentrations were significantly increased by GGEx55 groups compared with vehicle control. Conclusions : These results demonstrate that GGEx55 and GGEx62 effectively reduces body weight gain, feeding efficiency ratio in high fat diet induced obese mice, leading to the modulation of obesity. In addition, GGEx55 and GGEx62 decreases visceral adipose tissue mass and improves plasma lipids, suggesting that GGEx55 and GGEx62 may act as a therapeutic agent for obesity.

  • Wistar Strain male rats were divided into four different diet groups and the control group was fed on seven percent casein diet (C), the second with a mixture of 80% of rice and 20% of buckwheat (RW), the third with rice only (R), and the fourth with a mixture of rice and buckwheat of equal nitrogen level (RWS). Each diet group consisted of six rats was fed for three weeks by the adlibitum feeding method. The results of the experiment, i, e, the growth gain and change in the level of branch-amino acids in the plasma determined by the micro-bioassay method were as follows. 1. The group C showed the largest growth gain and the rest did in the order of RW group, R group, and RWS group. 2. It was shown that rats fed on diet of higher protein score tend to have higher level of free branch-amino acids in the plasma. 3. Thus, the protein score may be estimated based on the level of the free branch-amino acids in the plasma.

  • The livers of the wistar strain male rats which had been mentioned in our study were analyzed by the micro-bioassay method for comparison of the levels of free branch-amino acids(valine, leucine, isoleucine) and phenylalanine, methionine, tyrosine. Rats which were fed on diet of higher protein score contained correspondingly higher level of free branch-amino acid in their liver while the levels of phenylalanine, methionine, tyrosine showed unrecognizable relationship with the protein score of the diet used.

저단백영양(低蛋白營養)에 있어서 성숙(成熟)흰쥐의 Plasma 및 Erythrocytes Free Amino Acid에 대하여 (A Study of the Free Amino Acids in the Plasma and Erythrocytes in the Male Adult Rats Fed with the Low Protein Diets)

  • An analysis of the free amino acid contained in the plasma and erythrocytes of the six groups of Wistar Strain male adult rats(body weight 200-300g) having fasted for sixteen hours was made by means of the HITACHI Amino Acid Autoanalyzer and the result of which was corrected with RC-24 B TOMY Micro Hematocrit Centrifuge. There was a depression of the plasma and erythrocytes free amino acid level on the noprotein diet with ad libitum feeding. But on the 20% casein diet there was an elevation in the levels of free amino acid and consequently alanine, glysine, lysine, serine and arginine level in the erythrocytes and threonine glutamic acid and taurine level in the plasma increased on the high protein diet. There was more plasma and erythrocytes free amino acid level on the 5% casein-30% fat diet than on the 5% casein-no fat diet with pair-feeding. In comparison, on the low calorie diet more free amino acids were found in plasma than in erythrocytes, but on the higher calorie diet more free amino acids were found in the erythrocytes than in the plasma. On the 20% casein-30% fat diet with pair-feeding the erythrocytes free amino acids level increased but in plasma free amino acids level decreased. Such as an opposite result was given in plasma and erythrocytes free amino acids level. In the pair-fed four groups, erythrocytes per plasma generally increased in the rate of less than 10.0 as the calorie increased. The essential amino acid per non essential amino acid generally increased in the ratio as protein level and calorie increased, and that ratio range was from 0.2 to 0.7. And essential amino acid per non essential amino acid of plasma was higher than that of erythrocytes.
