In vivo activity of recombinant human erythropoietin (rh-EPO) has been examined using polycythemic model in mice and acute hemorrhage model in rats. The number of reticulocytes in blood stream was increased after a single injection of rh-EPO depending on the dosage of rh-EPO in polycythemy model. It seemed that optimal dose of rh-EPO for polycythemic mice was around 1-10 U/kg. Rh-EPO also showed the effectiveness for increase of reticulocyte numbers both in male and female rats after bleeding. The number of reticulocytes and the change of hemoglobin concentration in the blood stream of normal rats has been examined after injection of rh-EPO. The maximum value of reticulocyte was observed on the 6th day of the injection in these normal rats. In addition, the increase of reticulocyte and the concentration of hemoglobin were dependent on the dosage of rh-EPO. The increase of hemoglobin concentration was continued to the 9th day after injection. In this study, the efficacy of rh-EPO was confirmed in both mice and rats.
본 연구는 MICE 산업(회의, 포상여행, 컨벤션, 전시 및 이벤트)에서 소셜네트워크서비스(SNS)의 이용의도에 영향을 미치는 요인과 성별차이를 분석하는 것이 목적이다. 본 연구에서는 MICE 산업, SNS 특성 그리고 기술수용모형(TAM)의 이론적 배경을 바탕으로 연구모형과 가설을 설정하였다. 본 연구에서는 MICE 참여경험을 가지고 있는 SNS 사용자를 대상으로 설문조사를 하였다. 가설검증 결과는 다음과 같다. 첫째, 정보제공성, 편리성, 서비스 품질, SNS 이용경험은 SNS 이용태도에 긍정적인 영향을 미치는 것으로 나타났다. 둘째, SNS 이용태도는 SNS 이용의도에 긍정적인 영향을 미치는 것으로 나타났다. 마지막으로, 정보제공성과 서비스 품질은 남성이 여성보다 SNS 이용태도에 미치는 영향력이 더 크게 나타났고, 편리성과 SNS 이용경험은 여성이 남성보다 SNS 이용태도에 미치는 영향력이 더 큰 것으로 나타났다. 그리고 SNS 이용태도는 남성이 여성보다 SNS 이용의도에 더 큰 영향력이 있는 것으로 나타났다. 본 연구의 결과는 MICE 산업에서 SNS 이용의도와 관련된 다양한 시사점을 제공할 것이다.
Purpose: Synthesis of regulated on activation, normal T-cells expressed and secreted (RANTES) in the airway has previously been shown to be elevated after respiratory syncytial virus (RSV) infection. However, since few studies have examined whether RSV-infected asthma patients express a higher level of RANTES than do normal individuals, we used a murine model of asthma to address this question. Methods: We prepared Dermatophagoides farinae-sensitized mice as an asthma model, and then infected them with RSV and analyzed the changes in airway responsiveness and the cell populations and cytokine levels of bronchoalveolar lavage fluid. Results: RANTES synthesis increased in response to RSV infection in both control mice and in asthma model (D. farinae) mice. However, there was no significant difference in the amount of RANTES produced following RSV infection between control and D. farinae mice. RSV infection affected neither interferon-${\gamma}$ synthesis nor airway responsiveness in either control or D. farinae mice. Conclusion: RSV infection did not induce more RANTES in a murine model of asthma than in control mice.

The pharmacological interest coupled with traditional uses (antidiarrhoeal, antiseptic, analgesic etc) prompted us to test for anti-inflammatory, antinociceptive and diuretic activitities of Trema (T.) orientalis Linn. The crude methanolic leaves extract of T. orientalis was investigated for its possible anti-inflammatory activities using carrageenin induced rat paw edema model and cotton pellet implantation method in mice. Then the extract analyzed for its antinociceptive activities by acetic acid induced writhing model in mice. The extract possessed significant anti-inflammatory activity in both models at the doses of 200 and 400 mg/kg body weight of mice. Moreover, the extract showed significantly reduced the number of acetic acid-induced abdominal constriction in mice of 200 and 400 mg/kg body weight. The extract also showed positive diuretic activity in albino mice.
Purpose: C57BL/6 mice, which are among the most common backgrounds for genetically engineered mice, are resistant to the induction of periodontitis by oral infection with periodontal pathogens. This study aimed to develop a periodontitis model in C57BL/6 mice using coaggregation between human pathogens and the mouse oral commensal Streptococcus danieliae (Sd). Methods: The abilities of Porphyromonas gingivalis ATCC 33277 (Pg33277), P. gingivalis ATCC 49417 (Pg49417), P. gingivalis KUMC-P4 (PgP4), Fusobacterium nucleatum subsp. nucleatum ATCC 25586 (Fnn), and F. nucleatum subsp. animalis KCOM 1280 (Fna) to coaggregate with Sd were tested by a sedimentation assay. The Sd-noncoaggregating Pg33277 and 2 Sd-coaggregating strains, PgP4 and Fna, were chosen for animal experiments. Eighty C57BL/6 mice received oral gavage with Sd once and subsequently received vehicle alone (sham), Fna, Pg33277, PgP4, or Fna+PgP4 6 times at 2-day intervals. Mice were evaluated at 5 or 8 weeks after the first gavage of human strains. Results: Fnn, Fna, and PgP4 efficiently coaggregated with Sd, but Pg33277 and Pg49417 did not. Alveolar bone loss was significantly higher in the PgP4 group at both time points (weeks 5 and 8) and in all experimental groups at week 8 compared with the sham group. The PgP4 group presented greater alveolar bone loss than the other experimental groups at both time points. A higher degree of alveolar bone loss accompanied higher bacterial loads in the oral cavity, the invasion of not only PgP4 but also Sd and Fna, and the serum antibody responses to these bacteria. Conclusions: Periodontitis was successfully induced in C57BL/6 mice by oral infection with a P. gingivalis strain that persists in the oral cavity through coaggregation with a mouse oral commensal bacterium. This new model will be useful for studying the role of human oral bacteria-host interactions in periodontitis using genetically engineered mice.
본 연구는 호텔 MICE의 적정인력 산정을 위한 지표를 개발하는 목적에서 시작되었다. 따라서 본 연구는 국내외 다양한 MICE 행사들을 유치하고 운영하고 있는 서울의 특급호텔 MICE 관련 전문가들을 대상으로 적정 인력을 산정할 때 중요하게 고려해야 하는 항목들을 도출하고 지표의 신뢰성과 타당성을 시도하였다. 연구결과 및 내용은 다음과 같다. 첫째, 도출 결과의 일반화를 위하여 최종 개발된 지표는 호텔 MICE 행사와 관련한 289명의 종사원들을 대상으로 실증 분석을 하여 지표의 신뢰성과 타당성을 검토하였다. 둘째, 그 결과 호텔의 인력 산정 및 운영에 매우 큰 영향을 미치는 호텔의 인사 및 경영전략을 3가지로 분류(서비스 지향, 안정성 지향, 효율성 지향)하였다. 셋째, 도출된 지표는 실무적으로 측정할 수 있도록 정량지표(15개)와 정성 지표(19개)로 구분하였으며, 호텔 MICE의 적정인력 산정은 정량지표와 정성 지표와 의 총계이다. 본 연구는 도출된 적정인력 지표를 정량 및 정성 지표로 구분하고 향후 실측할 수 있는 산식의 모형을 제시하였다는 점에서 향후 실무적으로 크게 기여할 것으로 기대한다.
Objective: The aim of this study was to investigate the wound healing effect of herbal ointment, Gamijaungo, on the burn-induced model. Reports about Gamijaungo on the wound healing effect by local application in mice model or human study have published in the several domestic or internationally, but most are anecdotal and lack solid scientific evidence. Method: We observed the morphologic and histologic changes in the burn-induced mice model. we counted white blood cell and platelet changes. we confirmed VEGF, PI3K and pAkt protein expression by Western blot analysis. Result: In this study, we observed that Gamijaungo showed strong wound healing effects in the morphologic and histologic changes in the burn-induced mice model. Also we found that the significant changes of white blood cell and platelet changes by the treatment of Gamijaungo. In molecular mechanism, we got the strong positive effect by Gamijaungo treatment on angiogenesis, a key process in the formation of the granulation tissue during wound healing. Conclusion: These findings suggest the potential use of Gamijaungo as a therapeutic in thermal burn-induced skin injuries.
Humanized mice, containing engrafted human cells and tissues, are emerging as an important in vivo platform for studying human diseases. Since the development of Nod scid gamma (NSG) mice bearing mutations in the IL-2 receptor gamma chain, many investigators have used NSG mice engrafted with human hematopoietic stem cells (HSCs) to generate functional human immune systems in vivo, results in high efficacy of human cell engraftment. The development of NSG mice has allowed significant advances to be made in studies on several human diseases, including cancer and graft-versus-host-disease (GVHD), and in regenerative medicine. Based on the human HSC transplantation, organ transplantation including thymus and liver in the renal capsule has been performed. Also, immune reconstruction of cells, of the lymphoid as well as myeloid lineages, has been partly accomplished. However, crosstalk between pluripotent stem cell derived therapeutic cells with human leukocyte antigen (HLA) mis/matched types and immune CD3 T cells have not been fully addressed. To overcome this hurdle, human major histocompatibility complex (MHC) molecules, not mouse MHC molecules, are required to generate functional T cells in a humanized mouse model. Here, we briefly summarize characteristics of the humanized mouse model, focusing on development of CD3 T cells with MHC molecules. We also highlight the necessity of the humanized mouse model for the treatment of various human diseases.
The neuropeptide vasopressin (VP) is a nine- amino acid hormone synthesized as preprohormone in the cell bodies of hypothalamic magnocellular neurons. The tumor in magnocellular neurons of the hypothalamus is associated with disfunctions of the cell bodies, leading to the diabetes insipidus. In order to study with the diabetes insipidus caused by a defect in VP synthesis and its secretion, we have produced the transgenic mice regulated by vasopressin promoter inserted to SV40 T antigen coding sequence (pVPSV.IGR2.1). One transgenic line expressing high levels of SV40 T antigen was propagated. The founder and all transgene positive adult animals have appeared with shorten mortality or apparent phenotypic abnormalities, including immune complex disease, and eventually die between 4 and 8 months of age. The mRNA and protein of SV40T antigen transgene were detected in brain of fetus as well as in brain, spleen, lung and lymph node in moribund at the age of 20 weeks. Histological analysis of transgenic mice showed that tumor developed in brain similar to primitive neuroectodermal tumors (PNET) in man. We also detected lymphomas in spleen and lymph node, and consequent tumor formation in various tissues of the transgenic mice. In pVPSV.IGR2.1, 21% mice showed brain tumor (PNET) at 5 weeks and 100% mice showed brain tumor after 15 weeks. In addition, Expression of apoptosis related genes (Bcl-28 & Bax) was increased over their age in mice with PNET as compared to control mice. Apoptosis related gene expression might be deregulated in mice with brain tumor. However, transgenic mice were not developed with the diabetes insipidus. These mice represent the first disease model to exhibit primitive neuroectodermal tumor in brain, as well as a unique model system for exploring the cellular pathogenesis of lymphomas.
골은 일생에 걸쳐 지속적인 재형성과장을 거치면서 유지되고 이러한 기전에 대한 연구는 골다공증을 비롯한 골대사 질환의 병태생리와 치료에 있어 큰 진전을 이루고 있다. 특히 생체 내 골형성 및 재생과정을 연구하는데 있어 형광표지자를 이용하는 방법이 널리 알려져 있는데, 그 중 calcein은 칼슘 킬레이터로 골이 새롭게 형성하는 부위에 녹색을 띔으로써 유용한 마커로 사용된다. 그러나 대부분의 골형성 연구에서 실험동물의 경우 표본제작을 할 때 크기가 작고 뼈가 부숴지기 쉬워 rat이나 rabbit을 이용하였으며, mice의 femur를 cross-section해서 관찰한 연구는 거의 없는 실정이다. 그래서 본 연구에서는 어린 mice를 실험동물로 이용하였으며, 생체 내 calcein을 4주간, 8주간 투여한 후 골 형성 변화를 형광현미경으로 관찰한 결과 8주차 쥐에서 4주차보다 진하고 골 형성 간격도 넓게 관찰된 것을 확인 할 수 있었다. Mice는 빠른 시일 내에 결과를 얻을 수 있고 부작용이 적은 장점이 있어서 앞으로 knock-out mice를 이용한 생체 내 실험에 활용하기 적합하다고 생각되며, 골형성 속도 평가 등 다양한 분야에서의 골 형성과 재생연구에 있어 기초 정보를 제공할 것으로 기대한다.
