  • The present study was to conducted the sexing efficiency and accuracy of bovine embryo by LAMP (Loop-mediated isothermal amplification) method, the development of the biopsied embryos into re- reformation and the freezability of these blastocysts by slow-freezing and vitrification. In vivo embryos were superovaluted with gonadotropin(Antorin R-10) for 4 days combined with progestrone releasing intravaginal(CIDR) insertion in Hanwoo donors, and in vitro embryos were used blastocyst embryos at Day 7 or Day 8 after post-insemination in vitro. The biopsy of bovine embryo was carried out in a 80μl drop with Ca2+-Mg2+ free D-PBS and the viability of biopsied embryos were evaluated in IVMD medium at over 12 h culture time in 5% CO2 incubator. For embryo sexing, about five or seven blastomeres were isolated from in vitro and in vivo embryos at blastocysts with microblade. and were then subjected to LAMP. The survivability of biopsied embryos were no difference in the development rate to re-formation of blastocysts between in vivo and in vitro embryos(100% and 90% respectively). The rates of sexed embryos were compared according to two groups, the female rate was lower than that the male in the in vivo and in vitro embryos(46% vs, 54% and 40% vs, 60%, respectively). However, there were no difference in the overall sexing ratio between the two groups. The survivability of frozen-thawed sexed embryos were lower in the in vitro than in vivo embryos in the slow-freezing(Group 1) and vitrification method(Group 2), (41.7% vs. 58.8% and 57.1% vs, 77.8. respectively).

  • This study was conducted to evaluate the effects of the fog-cooling system on the growth and yield characteristics of two large-fruited paprika cultivars during summer cultivation season. The temperature inside the greenhouse equipped with fog-cooling system was $2-3^{\circ}C$ lower than that in the control. The results of study show the possibilities of maintaining indoor temperatures below $35^{\circ}C$ and relative humidity at the level of 80% using fogcooling system during hot seasons of the year. Plant height, fruit weight and number of fruits per plant were higher for both cultivars in the fog-cooling treatment compared to those in control. Mean fruit weight and yield per unit area were higher in the fog-cooling treatment than those in the control. However there were no significant differences in sugar content, flesh thickness and locule number of fruits due to fog-cooling system. Number of fruits with epidermal cracking was decreased in the fog-cooling system for both paprika cultivars. Mineral contents of plants such as nitrogen (N), potassium (K), calcium (Ca), magnesium (Mg), were not affected due to fog-cooling treatment.

  • Experiment was conducted to elucidate the effect of various nitrogen fertilizers on the growth and yield of soybean cv. Backwoon in field, and to examine soybeans produced with different nitrogen fertilizers for their nutritional constituents and protein digestibility. Soybean plants were generally similar in growth among fertilizers applied, and plants with non-nitrogen and with ammonium sulfate showed tendency to have fewer internodes. Nodule number and nodule weight also were lower in plants with non-nitrogen and with ammonium sulfate. Nitrogen fixation ability per nodule number were similar at an early growth stage and at flowering stage treatments and were relatively higher with newly developed complex fertilizer at pod elongation stage. With newly developed complex fertilizer and with non-nitrogen, there were slight increase and in 100 grain weight and yield per 10a, respectively. Crude protein contents in soybeans with nitrogen fertilizers were higher than that with non-nitrogen, and the reverse were true for crude fat. Crude ash contents were relatively similar among the treatment. Crude fiber contents were higher with readily used complex fertilizer than with the others. The contents of inorganic constituents were of the same sort among the treatments, that of Fe with non-nitrogen being much lower than with the others. Protein digestibility was the highest in the non-nitrogen treated soybean.

  • The objectives of this study were to investigate the contamination of the soils around the major roadsides in Seoul and to accumulate base-line data. Fifty topsoils ($0{\sim}5$ cm) and 48 subsoils ($20{\sim}50$ cm) were collected from 50 sampling sites near Manguro in Chungnanggu. The samples were air-dried, passed through 2-mm sieves, and analyzed to determine the physicochemical properties and the contents of heavy metals (Cd, Cu, Pb, Zn) and anions (Cl, $NO_3$, $SO_4$). Soil textures of top- and subsoils were mainly loamy sand and sandy loam, respectively. Average $_PH$ of top- and subsoil was approximately 7.5, much higher than that of the forest soils in Seoul. Average heavy metal contents were lower than the levels allowed by the Soil Environment Conservation Act of Korea, whereas much higher than those of the forest soils. Contents of some heavy metals were higher than the maximum allowed levels. Anion concentrations were also much higher than those of the forest soils. A careful management to prevent the aggravation of the present contamination level and the diffusion of contaminants is recommended.

  • Bacterium capable of using biphenyl as a sole source of carbon and energy were isolated from soil, and based on the results of 16S rDNA sequence, strain BK10 identified as a Sphingobium yanoiktiyae. The optimum cultural conditions were as follows; $NH_4NO_3$ 1g, $K_2HPO_4$ 1g, $MgSO_4{\cdot}7H_2O$ 0.5g, $CaCO_3$ 0.2 g per 1 liter of distilled water. The Sphingobium yanoikuyae BK10 strain was completely utilized biphenyl in mineral salt media containing biphenyl at concentration 500 $\mu$g/ml of biphenyl as a sole carbon and energy source within 48 hours. Optimumal pH and temperature for biphenyl degradation and cell growth of strains were 6.0$\sim$8.0 and 20$\sim$50$^{\circ}C$, respectively. Especially, at 30$^{\circ}C$, cell-growth were higher than other temperature. Cell grown on biphenyl has been shown to have a higher removal rate for biphenyl than grown on sucrose. This study shows that Sphingobium yanoikuyae BK10 strain had a high biodegradation capability of biphenyl and can be simulate a candidate compounds the bioremediation of PCBs (Polychlorinated biphenyl) contaminant soil and water.

  • The purpose of this study was to extensively identify $PM_{10}$ sources and to estimate their contributions to the study area, based on the analysis of the $PM_{10}$ mass concentration and the associated inorganic elements, ions, and total carbon. The contribution of $PM_{10}$ sources was estimated by applying a receptor method because identifying air emission sources were effective way to control the ambient air quality. $PM_{10}$ particles were collected from May to November 2007 in the Yongin-Suwon bordering area. $PM_{10}$ samples were collected on quartz filters by a $PM_{10}$ high-volume air sampler. The inorganic elements (Al, Mn, V, Cr, Fe, Ni, Cu, Zn, Cd, Pb, Si, Ba, Ti and Ag) were analyzed by an ICP-AES after proper pre-treatments of each sample. The ionic components of these $PM_{10}$ samples ($Cl^_$, $NO_3^-$, $SO_4^{2-}$, $Na^+$, $NH_4^+$, $K^+$, $Ca^{2+}$, and $Mg^{2+}$) were analyzed by an IC. The carbon components (OC1, OC2, OC3, OC4, OP, EC1, EC2 and EC3) were also analyzed by DRI/OGC analyzer. Source apportionment of $PM_{10}$ was performed using a positive matrix factorization (PMF) model. After performing PMF modeling, a total of 8 sources were identified and their contribution were estimated. Contributions from each emission source were as follows: 13.8% from oil combustion and industrial related source, 25.4% from soil source, 22.1% from secondary sulfate, 12.3% from secondary nitrate, 17.7% from auto emission including diesel (12.1%) and gasoline (5.6%), 3.1% from waste incineration and 5.6% from Na-rich source. This study provides information on the major sources affecting air quality in the receptor site, and therefore it will help us maintain and manage the ambient air quality in the Yongin-Suwon bordering area by establishing reliable control strategies for the related sources.

  • The enzyme xylose isomerase (E.C. 5.3.1.5, XI) is responsible for the conversion of an aldose to ketose, especially xylose to xylulose. Owing to the ability of XI to isomerize glucose to fructose, this enzyme is used in the food industry to prepare high-fructose corn syrup. Therefore, we studied the characteristics of XI from Anoxybacillus kamchatkensis G10, a thermophilic bacterium. First, the gene coding for XI (xylA) was inserted into the pET-21a(+) expression vector and the construct was transformed into the Escherichia coli competent cell BL21 (DE3). The expression of recombinant XI was induced in the absence of isopropyl-thio-${\beta}$-galactopyranoside and purified using Ni-NTA affinity chromatography. The optimum temperature of recombinant XI was $80^{\circ}C$ and measurement of the heat stability indicated that 55% of residual activity was maintained after 2 h incubation at $60^{\circ}C$. The optimum pH was found to be 7.5 in sodium phosphate buffer. Magnesium, manganese, and cobalt ions were found to increase the enzyme activity; manganese was the most effective. Additionally, recombinant XI was resistant to the presence of $Ca^{2+}$ and $Zn^{2+}$ ions. The kinetic properties, $K_m$ and $V_{max}$, were calculated as 81.44 mM and $2.237{\mu}mol/min/mg$, respectively. Through redundancy analysis, XI of A. kamchatkensis G10 was classified into a family containing type II XIs produced by the genera Geobacillus, Bacillus, and Thermotoga. These results suggested that the thermostable nature of XI of A. kamchatkensis G10 may be advantageous in industrial applications and food processing.

  • The benthic environmental impact experiment addresses environmental impacts at a specific site related to deep-sea mineral resource development. We have acquired several tens of multi- or box core samples at 31 sites within the Benthic environmental Impact Site (BIS) since 2010, aiming to examine the basic properties of surficial deep-sea sediment as a potential source for deep-water plumes. In this study, we present the geochemical properties such as major elements, rare earth elements (REEs), and heavy metal contents at the BIS. Such proxies vary distinctly according to the Facies association. The lithology of all core sediments in the BIS corresponds to both Association Ib and Association IIIb. The vertical profiles of some major elements ($SiO_2$, $Fe_2O_3$, CaO, $P_2O_5$, MgO, MnO) show noticeable differences between Association Ib and IIIb, while others ($Al_2O_3$, $TiO_2$, $Na_2O$, and $K_2O$) do not vary between Association Ib and IIIb. REEs are also distinctly different for Associations Ib and IIIb; in Association Ib, REY and HREE/LREE are uniform through the sediment section, while they increase downward in Association IIIb like the major elements; below a depth of 8 cm, REY is over 500 ppm. The metal enrichment factor (EF) evaluates the anthropogenic influences of some metals (Cu, Ni, Pb, Zn, and Cd) in marine sediments. In both Associations, the EF for Cu is over 1.5, the EF for Ni and Pb ranges from 0.5 to 1.5, and the EF for Zn and Cd are less than 0.5, indicating Cu is enriched but Zn and Cd are relatively depleted in the BIS. The vertical variations of geochemical properties between Association Ib and IIIb are shown to be clearly different, which seems to be related to the global climate changes such as the shift of Intertropical convergence zone (ITCZ).

  • The study was compared for growth and yield of cherry tomato (Lycopersicon esculentum var. cerasiforme) crops as affected by various homemade liquid fertilizer (LF), commonly applied in the environmentally friendly farmhouses. LF treatment included UT (untreatment, water), OC (oil cake), BF (bone+fish meal), FA (fish amino meal), SO (sesame oil meal), and SF (starfish). Seasonal pH and EC in SO- and SF-LF rapidly decreased at 30 days after the storage, which were the highest EC of 0.6 - 0.8 dS/m, followed by BF-LF with 0.4 dS/m EC. T-N concentration in LF was the highest on the SF (0.0062%), followed by SO (0.0059%) and BF materials (0.0030%), which were all the great for the K concentration in the LF. P and Ca concentrations were the highest on the FA-LF, with the highest Mg concentration observed on the vegetable SO-LF. Soil EC was the highest on the SF-LF plots of 0.74 dS/m, with no significant differences between the treatments observed on the macro-nutrient concentrations in the soil and leaf. Leaf dry weight, leaf temperature, stem diameter, and plant height were investigated at once per 15 days. UT-LF reduced the leaf dry weight at all the measurement time while the plant height was low at an initial measurement but increased and similar to the other homemade LF treatments at a later measurement. Fruit yield and average fruit weight were the lowest on the UT-LF plots at 75 days after fertigation. Fruit diameter was increased by the BF-LF and SF-LF, with the highest fruit soluble solid contents and fruit coloring observed on the FA-LF. BF-LF maintained high fruit firmness.
