  • The purpose of this study was to search for a mixture of stevioside and sucrose, the sweetness of which in soft drink such as cocoa and ginger tea were almost equal the sweetness of commomly used 7% sucrose in the drinks. Results obtained by the present experiment were as fellows: 1) Mixture of 50% stevioside 40mg (0.04%) (A) and 2.0% sucrose, 95% stevioside 40mg (0.04%) (B) and 1.0% sucrose, and 50% and 95% stevioside 60mg (0.06%) (A,B) respectively, the sweetness of which in ginger tea corresponded to that of reference in the drink. 2) Mixture of 50% stevioside 40mg (0.04%) (A) and 2.0% sucrose, 95% stevioside 20mg (0.02%) (B) and 2.0% sucrose, and 50% stevioside 40-60mg (0.04-0.06%) (A) and 95% stevioside 40-60mg (0.04-0.05%) (B), the sweetness of which in cocoa is equal to that of standard sucrose in the drink. 3) The mixture contains low energe, and also the after-taste of stevioside was not detected in the above drinks.

Improvement of Transformation Efficiency Through In Vitro Methylation and SacII Site Mutation of Plasmid Vector in Bifidobacterium longum MG1

  • The different cleavage patterns of pYBamy59 plasmid isolated from E. coli $DH5{\alpha}$ and B. longum MG1 by the cell extract of B. longum MG1 suggested that the main reason for its low transformation efficiency was related to the restriction modification (R-M) system. To confirm the correlation between the R-M system and transformation efficiency, in vitro methylation and site-directed mutagenesis were performed in pYBamy59. Sequence analysis of pYBamy59 fragments digested by the cell extract of B. longum MG1 revealed that all fragments were generated by restriction of the sequence recognized by SacII endonuclease. When pYBamy59 from E. coli was methylated in vitro by CpG or GpC methyltransferase, it was protected from SacII digestion. Site-directed mutagenesis, which removed SacII sites from pYBamy59, or in vitro methylation of pYBamy59 showed 8- to 15-fold increases in the transformation efficiency over intact pYBamy59. Modification of the SacII-related R-M system in B. longum MG1 and in vitro methylation in pYBamy 59 can improve the transformation efficiency in this strain. The results showed that the R-M system is a factor to limit introduction of exogenous DNA, and in vitro modification is a convenient method to overcome the barrier of the R-M system for transformation.

A COMPARATIVE EVALUATION OF ORAL CHLORAL HYDRATE VS. ORAL MIDAZOLAM IN SEDATIVE EFFECT (Chloral hydrate와 Midazolam의 경구투여시 진정효과에 대한 비교)

  • The purpose of present study was to compare the sedative effect of chloral hydrate and oral midazolam treatment. The study was conducted on twenty two child patients who required at least two visits. All the patients showed a good physical status (ASA-I). The patient was randomly assigned to receive drug either chloral hydrate 60mg/kg and hydroxyzine 25mg(Group I) or midazolam 0.5mg/kg and hydroxyzine 25mg(Group II) at each visit. Treatment procedures were divided into six stages. In each stage, sleep, crying, movement and overall behavior score were evaluated. The overall results can be summarized as follows: 1. Success rate was 59.9% in group I and was 77.8% in Group II. 2. Mean sleep score was $1.81{\pm}0.93$ in group I and was $1.44{\pm}0.69$ in group II (P<0.05). 3. Through all treatment procedures, no significant difference was observed between group I and group II in terms of crying, movement and overall behavior score(P>0.05). 4. There was no significant difference between the sex and the age(P>0.05). In regard to above results, oral midazolam considered to be a valuable drug as sedative agent in pediatric dentistry.

Manganese removal by KMnO4: Effects of bicarbonate and the optimum conditions (과망간산칼륨을 이용한 용해성 망간 제거: 중탄산염 영향 및 최적조건)

  • This study is focused on manganese (Mn(II)) removal by potassium permanganate ($KMnO_4$) in surface water. The effects of bicarbonate on Mn(II) indicated that bicarbonate could remove Mn(II), but it was not effectively. When 0.5 mg/L of Mn(II) was dissolved in tap water, the addition of $KMnO_4$ as much as $KMnO_4$ to Mn(II) ratio is 0.67 satisfied the drinking water regulation for Mn (i.e. 0.05 mg/L), and the main mechanism was oxidation. On the other hand, when the same Mn(II) concentration was dissolved in surface water, the addition of $KMnO_4$, which was the molar ratio of $KMnO_4/Mn(II)$ ranged 0.67 to 0.84 was needed for the regulation satisfaction, and the dominant mechanisms were both oxidation and adsorption. Unlike Mn(II) in tap water, the increasing the reaction time increased Mn(II) removal when $KMnO_4$ was overdosed. Finally, the optimum conditions for the removals of 0.5 - 2.0 mg/L Mn(II) in surface water were both $KMnO_4$ to Mn(II) ratio is 0.67 - 0.84 and the reaction time of 15 min. This indicated that the addition of $KMnO_4$ was the one of convenient and effective methods to remove Mn(II).

Synthesis of Pt(II) Complexes containing Flavin mononucleotide as Leaving Ligand and their Anticancer Activity (Flavin mononucleotide를 탈리기로한 백금 (II) 착체의 합성과 그 항암활성)

  • A series of vitamin-containing Pt(II) complexes of the type [Pt (FMN) (L)] (FMN=flavin mononucleotide, L=ethylenediamine, 1,3-propanediamine, 1,4-bu-tanediamine) was synthesizd and characterized by IR, electronic absorption, elemental analysis and FAB=Mass. The coordination sites of FMN to Pt(II) ions were determined to be N(5) and O(6) with resultant chelate ring formation. Theses compounds have much better water solubility (30-35 mg/ml) than cisplatin (1 mg/ml). The anticancer activity of this vitamin-containing Pt(II) series was investigated by MTT assay against mouse and human leukemia cell lines in vitro. Among these compounds, FMN (1,4-butanediamine) Pt(II) having seven-membered ring structure as amine ligand showed moderate anticancer activity.

Cucumber (Cucumis sativus L.) Fruit and Combination with Losartan Attenuate the Elevation of Blood Pressure in Hypertensive Rats Induced by Angiotensin II

  • Objectives: Cucumis sativus L. (C. sativus) is vegetable commonly used for managing blood pressure and often consumed in combination with standard antihypertensive therapy, despite lack of scientific evidence supporting their use. Combination of herbs and standard medication could have positive or negative effects. Therefore, this study aimed to evaluate the antihypertensive activity of C. sativus and the combined effect with losartan in the hypertensive rat model induced by angiotensin II. Angiotensin II is a component of the renin-angiotensin-aldosterone system that, upon binding to its receptor, constricts blood vessels leading to elevation of blood pressure. Methods: In an antihypertensive study, rats received C. sativus orally at doses of 9, 18, 27, and 36 mg/kg (full dose); while in a combination study, animals received losartan 2.25 mg/kg combined by either with C. sativus 9 or 18 mg/kg. The standards group received losartan 2.25 mg/kg or 4.5 mg/kg (full dose). Results: Blood pressure was measured using the tail-cuff method. C. sativus significantly attenuated angiotensin II-induced hypertension as observed in groups receiving C. sativus at 9, 18, 27, and 36 mg/kg at 30 minutes after induction showed the average change (Δ) of systolic blood pressure (SBP) and diastolic blood pressure (DBP) with respect to time zero were 28.8/18.3, 24.8/15.8, 22.8/15.5, and 11.5/9.0 mmHg, respectively. Whereas the average change (Δ) of SBP and DBP in the rats receiving the combination of half doses of C. sativus and losartan were 8.8/9.0 mmHg, respectively. These diminished effects were better than a full dose of C. sativus and comparable with a full dose of losartan (6.5/7.8 mmHg). Conclusion: The present findings indicate that C. sativus dose-dependently blocks blood pressure elevation induced by angiotensin II. The combination of half dose of C. sativus and losartan has an additive effect in lowering blood pressure.

  • Experiments were performed to investigate the effect of Panax ginseng petroleum ether fraction on delayed type hypersensitivity, rosette formation, phagocytic activity and histophathological influence in lead acetate treated mice. Lead acetate was administered in the drinking water and ginseng pet. ether fraction was injected i.p.. Mice were sensitized and challenged with sheep red blood cells. Erythrocyte(I) rosette formation and DTH reaction were significantly depressed in lead acetate treated mice, and those were restored administration of ginseng fraction. Ginseng pet. ether fraction administration did not have any effect on decreased phagocytic activity. Follicular and parafollicular areal destruction of spleen, and destruction of thymus were finded in lead acetate exposed-mice. Small dose of ginseng pet. ether fraction (5 mg/kg, 10 mg/kg), administraction inhibited those histopathological changes, but large dose (20 mg/kg) didn't.

Adsorption Characteristic of Mg(II), Al(III), Pb(II) Metal Ions on Cryptand Ion Exchange Resin from Water Fire Extinguishing Agent (물 소화약제로부터 Cryptand 이온교환수지의 Mg(II), Al(III) 및 Pb(II) 흡착특성)

  • Resins were synthesized with 1-aza-15-crown-5 macrocyclic ligand attached to styrene(dangerous matter) divinylbenzene(DVB) copolymer with crosslink of 1, 2, 6 and 15% by substitution reaction. The synthesis of these resins was confirmed by content of chlorine, element analysis, and IR-spectrum. The effects of pH, time and crosslink on adsorption of metal ion from water fire extinguishing agent by the synthetic resin adsorbent were investigated. The metal ion was showed fast adsorption on the resins above pH 3. The optimum equilibrium time for adsorption of metallic ions was about two hours. The adsorption selectivity determined in water was in increasing order of $Mg^{2+}>Al^{3+}>Pb^{2+}$. The adsorption was in the order of 1, 2, 6, and 15% crosslink resin.

Effects of taurine on cadmium exposure in muscle, gill, and bone tissues of Carassius auratus

  • This study was performed in order to investigate the effects of taurine on cadmium poisoning in muscle, gill, and bone tissues of wild goldfish. For this experiment, 80 wild goldfish were divided into four experimental groups: 0.3 mg/L of cadmium and 0 mg/L of taurine (Group I), 0.3 mg/L of cadmium and 20 mg/kg of taurine (Group II), 0.3 mg/L of cadmium and 40 mg/L of taurine (Group III), and 0.3 mg/L of cadmium and 80 mg/L of taurine (Group IV). The results were as follows: The cadmium concentration in muscle tissue of wild goldfish was 0.65-3.21 mg/kg wet wt in Group I, whereas it decreased in Group IV. Levels of cadmium in gill tissue of wild goldfish were 16.57-42.39 mg/kg wet wt in Group I, 15.23-43.01 mg/kg wet wt in Group II, 15.11-39.56 mg/kg wet wt in Group III, and 13.15-38.55 mg/kg wet wt in Group IV (P < 0.05), suggesting that the cadmium concentration decreased in the experimental groups compared to control. The cadmium concentration in bone tissue of wild goldfish after 28 days was 0.52-9.75 mg/kg in Group II, whereas it increased in Group III (P < 0.05). In conclusion, taurine may have a preventive effect against cadmium accumulation in biological tissues.

Production of Transgenic Petunia hybrida cv. Rosanpion Using Agrobacterium-mediated Transformation

  • Transgenic Petunia hybrida cv. Rosanpion was produced by Agrobactepium tumefaciens LBA4404 harboring a binary vector pBI 121 containing $\beta$-glucuronidase (gus) and neomycin phosphotransferase (nptII). For genetic transformation, leaf discs were precultured on MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA (MNB) for 2 days and cocultured for 15 mins with A. tumefaciens. For selection of transformant, leaf discs were transferred to fresh MNB containing 50 mg/L kanamycin and 500 mg/L cefotaxime. Eighteen plants were regenerated and four were confirmed by PCR for detection of gus and nptII gene integrated into the nuclear genome of petunia ‘Rosanpion’. Using this transformation system, we expect that transgenic petunia ‘Rosanpion’ incorporating a useful gene can be produced.
