To isolate a novel antifungal compound, we obtained 571 kinds of endolichenic fungi from Lichen Bioresources Center and examined their antifungal abilities. Four fungi Stereocaulon sp. (1429), Stereocaulon sp. (1430), Cryptosporiopsis sp. (0156), and Graphis sp. (1245) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these fungi with chloroform and then ethyl acetate. The chloroform fraction exhibited the highest anti-fungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. To see information for the inhibitor present in chloroform fraction we employed GC-MS for the fractions of Stereocaulon sp. (1429). We found that hexamethylcyclotrisiloxane, decanoic acid, hexadecanonic acid-methyl ester, 14-octadecenoic acid-methyl ester, and octadecenoic acid-methyl ester were present more in chloroform fraction than in ethylacetate fraction. This indicates that those compounds could be possible antifungal candidates since antifungal activity of chloroform extract was two times higher than that of ethyl acetate extract.
4-Methyl-2,6,7-trioxa-1-phosphabicyclo [2,2,2] octane 1-sulfide (BPS), 4-Methyl-2,6,7-trioxa-1-phosphabicyclo[2,2,2] octane 1-oxide (BPO) and related monocyclic methylphosphates were prepared and the oxidation of BPS with MCPBA in chlorofrom or methanol was carried out to investigate the formation of reactive intermediates and reaction mechanism. BPO was the only product in chloroform while in methanol isomeric monocyclic methylphosphates were formed through opening of bicylic structure with subsequent phosphorylation of methanol by reactive intermediate formed in reaction. Formation of little amount of BPO was also observed. The structure of phosphorylating intermediate was probed with various spectroscopic methods and monocyclic methyl sulfenyl ester was suggested as a possible structure.
The needles of torreya(Torreya nucifera S. et Z) were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. The ethylacetate soluble mixture was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Most of the needle extractives were flavan and its methyl ether compounds such as (+)-catechin, (-)-epicatechin, (+)-gallocatechin, (-)-epigallocatechin, 3'-O-methyl-(+)-catechin and 3'-O-methyl-(-)-epicatechin including protocatechuic acid, one of benzoic acids. $^1H$- and $^{13}C$-NMR, NOE and CI-MS analyses were performed to elucidate the structures of the isolated compounds.
Dissolution profiles of ketoconazole in solid dispersion system (SDS) were investigated in the second fluid (pH 6.8) for the dissolution test (KPV). PVP K-30 and methyl cellulose were used as carriers in SDS, and chloroform as a solvent. Computer optimization technique was applied to obtain an optimum formula of SDS, and the following results were obtained; 1) Dissolution rate of ketoconazole in SDS was larger than that of pure ketoconazole in the second fluid of pH 6.8. 2) PVP K-30 and methyl cellulosr were good carriers for SDS of ketoconazole. Moerover PVP K-30 was better than methyl cellulose. 3) The optimum formula of ketoconazole SDS obtained from computer optimization technique was chloroform 175 ml, PVP K-30 5g and methyl cellulose 0.2g per 1g of ketoconazole. 4) The experimental value of $A_{60}$ (Amount released from ketoconazole tablet during 60 minutes) obtained from SDS by optimum formula agreed well with the value calculated by polynomial regression equation.

Methanol extract of garlic was fractionated to chloroform and aqueous fractions. The cholorform fraction possessed the highest antimutagenic activity in Salmonella typhimurium TA 100 and TA98, and was further fractionaed into four Allium sativum chromatography fractions (ASC F1, 2, 3 and 4) by column and thin layer chromatographies. The ASC F2 exhibited the higher antimutagenic activity and contained 18 chemical compounds tentatively identifed by GC-MS, NMR and FT-IR. Among the 18 compounds, methyl linoleate was a major compound to exhibit the antimutagenicity.

Ixeris dentata was extracted with methanol and then the methanol extract was further fractionated to hexane, chloroform, ethyl acetate, butanol and aqueous fraction. The methanol extract of lxeris dentata had the strong antimutagenic effect on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest. Among the solvent extracted fractions from the methanol extract, the chloroform fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity of AFB1 with inhibition rate of 74 percent. The methanol extract of Ixeris dentata also revealed the inhibitory effect on the growth of MG-63 human osteosarcoma cells after 6 days of breeding at 37℃. The chloroform fraction and the ethyl acetate fraction from the methanol extract of lxeris dentata were most effective and inhibited the growth of MG-63 cells by 97 and 93 percent, respectively. It is suggested that the inhibitory effects of lxeris dentata on the mutagenicity and the growth of MG-63 human osteosarcoma cells are strong in the lipid soluble fractions.
Methanol extract of dried persimmon leaves was refractionated using hexane, chloroform, ethylacetate, n-butanol aqueous fractions. Among these chloroform fraction showed the highest inhibition rate on the mutagenicities of aflatoxin B₁(AFB₁) and 3,2' -dimethyl-4-amino-bipheny1(DMAB) in Salmonella typhimurium TA 100. Chloroform fraction was further fractionated into eight fractions by silica gel column chromatography and thin layer chromatography(TLC). The fraction 5 on TLC exhibited the highest antimutagenic activities in AFB₁and DMAB. 2,4-Decadienal, dihydro-4-methyl-2(3H)-furanone, hexanoic acid 1,4-bis(1-methy1 ethy1)benzene, heptanoic acid, phenol, octanoic acid, nonanoic acid and benzoic acid were tentatively identified from this antimutagenic fraction by GC-MS.
Novel antimicrobial substance was isolated from seed coat of Canavalia gladiata by extraction with 75% methanol. Isolation and purification were conducted with solvent fractionation and chromatography on silica gel and sephadex LH-20 columns. Each fraction of antimicrobial activity was tested by paper disc method. Single compound was obtained from the 4th fraction of sephadex LH-20 column chromatography using chloroform/methanol (1:4, v/v), and identified as 3,4,5-trihydroxybenzoic acid methyl ester (methyl gallate) based on HPLC, GC/MS, FT-IR, $^1H$ NMR, and $^{13}C$ NMR analyses. This is the first report describing the presence of methyl gallate in C. gladiata.
The antioxidative action of petroleum ether, chloroform, chloroform-methamol (2 : 1, v/v), methanol and methanol-water extracts of Panax ginseng roots in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUFA) was investigated in vitro. All of the extracts of Panax ginseng roots possessed antioxidative activity and inhibited the formation of hydroperoxides in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUEA). The induction period in the autoxidation of MEUFA was extended by the addition of each extract. The antioxidative activity was more prominent with chloroform and chloroform-methanol (2 : 1, v/v) extracts than with other extracts. The antioxidative activity of each extract estimated by Olcott's oven test and the measurement of peroxide value (POV) showed a similar tendency. From the results obtained, it was conclude that extracts of Panax ginseng roots had remarkable antioxidative activity in the autoxidation of MEUFA in vitro.
