Characteristics of Taste Compounds of Red Snow Crab Cooker Effluent and Hepatopancreas for Developing a Crab-like Flavorant (게향 소재 개발을 위한 붉은 대게 자숙액 및 내장의 정미 성분 특성)

  • In order to develop materials of crab-like flavorant, taste compounds including physicochemical characteristics were analyzed in red snow crab cooker effluent(RSCCE) and hepatopancreas. The $30\;^{\circ}Brix$ was a suitable condition in from 1.5 to $40\;^{\circ}Brix$ RSCCE by sensory evaluation. Lactic acid and succinic acid were major compounds in non-volatile organic acids detected in both $30\;^{\circ}Brix$ RSCCE and hepatopanceras. The 5 compounds such as AMP, HxR, IMP, ATP and GMP were major in ATP related compounds of $30\;^{\circ}Brix$ RSCCE, whereas 3 compounds including IMP, GMP and Hx in hepatopanceras. The content of total free amino acids in hepatopancreas was 5.6 times higher than in $30\;^{\circ}Brix$ RSCCE. The major compounds in $30\;^{\circ}Brix$ RSCCE were followed by methionine, lysine, arginine, valine, histidine, alanine, hydroxy proline, and glycine in that order, whereas methyl histidine, leucine, alanine, glutamic acid, glycine, valine, threonine, taurine, isoleucine, and serine were followed in hepatopancreas. By adding 0.5%(w/w) hepatopancreas in $30\;^{\circ}Brix$ RSCCE, crab meat-like odor was kept high level by sensory evaluation.

  • Enzymatic browning is considered desirable in tea and tobacco processing but undesirable in many fruits processing at the present time. It is necessary to understand the nature of the enzyme, phenoloxidase, in order to control browning reactions, and extend its effects to formation of browning products as antioxidants in ginseng. Ginseng exhibits antioxidant activity when incorporated with turkey dark meat patties. The activity in red ginseng showed about two times stronger than white ginseng. One of the phenolic antioxidants from fresh, white and reprocessed white ginseng was identified as phenol 2.6 Bis(1.1 dimethyl ethyl) 4-methyl among several unknown compounds by GC/mass spectrometer. In red ginseng, no phenol 2.6 Bis (1.1 dimethyl ethyl) 4-methyl was detected, the compound may be polymerized by phenoloxidase and form some higher molecular compounds which may possess high antioxidant activity. Phenoloxidase isozymes in fresh Korean ginseng (panax ginseng C.A. Meyer) were extracted with phosphate buffer at pH 7.3. The isozymes were purified through ammonium sulfate fractionation, dialysis and chromatography on a DEAE-cellulose column. Two groups of phenoloxidase were shown to be present, one in the floating agglomerated group and the other in the precipitate. group from the 0.85 saturation ammonium sulfate. The DEAE-cellulose column chromatography, the phenoloxidase isozyme present in the precipitate appears as the first peak (I), and that in the agglomerate in the second peak (II). Isozyme I showed higher activity with catechin and catechal, and isozyme II showed higher activity with p-cresol. The isozyme showed two optimum pH activity one at pH 4.5 and the other at 8.5 with catechin as substrate. Korean ginseng phenoloxidase has high heat stability. When heated at $75^{\circ}C$ for 2 hours, its activity remained $90\%\;and\;80\%$ on phenoloxidase I and II respectively. Phenoloxidase I was most active on (+) catechin followed by p-cresal, catechol and epicatechin. Phenoloxidase II was most active on p-cresal followed by (+) catechin, catechol, p-coumanic acid and epicatechin. Sodium bisulfite, sodium cyanide, ascorbic acid glutachion in the oxidized form, sodium diethyl dithiocarbomate and ethylendiamine tetra acetate (EDTA) acted as inhibitors. Red ginseng color development was initiated by phenoloxidase and finished by a followed sun drying process. The antiaging activity of ginseng may be initiated by the antioxidant in the ginseng.

Effects Gamma irradiation and Fumigation on Pigment and Pungent Components of Red Pepper during Storage (건고추의 저장 중 색소 및 신미성분에 대한 감마선과 훈증 처리의 영향)

  • In order to study the quarantine and sanitization methods for dried red pepper, comparative effects of commercial fumigation (methyl bromide/MeBr, phosphine gas/$PH_3$) and gamma irradiation (5, 10 kGy) were investigated in terms of soluble pigment capsanthin, and capsaicin content of pepper during storage at mom temperature for 8 months. In water-soluble pigment, the degree of browning was highest in the fumigated samples, particularly in pericarp part, as compared to the control and irradiated samples. In general, the changes in capsanthin content were not apparent by treatment groups, but some reduced content were found in 10 kGy-irradiated pericarp group, Eight months of storage resulted in the significant decrease in capsanthin contents of pericarp part by $26.76{\sim}38.08%$ depending on treatment groups. The contents of capsaicin and dihydrocapsaicin were not different between the control and 5 kGy groups, while their contents decreased in both fumigated and 10 kGy-irradiated groups(p<0.05). The reduction in pungent component was observed in all the sooted samples, which was mote apparent in teated groups than in the control.

Cross-resistance of Colletotrichum acutatum s. lat. to Strobilurin Fungicides and Inhibitory Effect of Fungicides with Other Mechanisms on C. acutatum s. lat. Resistant to Pyraclostrobin (Strobilurin계 살균제에 대한 고추탄저병균의 교차저항성과 Pyraclostrobin 저항성균에 대한 다른 기작 살균제의 억제 효과)

  • Colletotrichum acutatum s. lat. 20JDS8 sensitive and 20CDJ6 resistant to pylaclostrobin were used to investigate the cross-resistance with fungicides belonging to the strobilurins and the characteristics of fungicidal controlling activities with different mechanisms against the isolate resistant to the fungicide. The resistant isolate of 20CDJ6 also showed the resistance to azoxystrobin, trifloxystrobin, and kresoxim-methyl, suggesting that there is a cross-resistance relationship. All fungicides with different action mechanisms inhibited mycelial growth of both susceptible and resistant isolates of C. acutatum s. lat., but their disease control effects in fruits were different according to the fungicides. The disease control effect of isopyrazam against 20JDS8 and 20CDJ6 was very low, and fluazinam showed a control effect of 91.9% and 88.1% against 20JDS8 and 20CDJ6 only when it was treated before inoculation by spraying spore suspensions on pepper fruits without wounds. Tebuconazole and prochloraz effectively inhibited not only the mycelial growth of 20JDS8 and 20CDJ6 on potato dextrose agar medium, but also disease incidence in red pepper fruits. As a result of this study, C. acutatum s. lat. 20CDJ6 resistant to pyraclostrobin showed cross-resistance with other strobilurin fungicides. In addition, we think that fluazinam, tebuconazole, and prochloraz can be recommended as alternative fungicides for the control of red-pepper pyranthracnose pathogens resistant pyraclostrobin. However, fluazinam can be effective only if it is treated protectively before the occurrence of the disease.

Development of Measuring Technique for Somatic Cell Count in Raw Milk by Spectroscopy (분광분석법을 이용한 우유의 체세포수 측정기술 개발)

  • The objective of this study was to develop models to predict SCC (somatic cell count) in unhomogenized milk by visible and near-infrared (NIR) spectroscopic technique. Total of 100 milk samples were collected from dairy farms and preserved to minimize propagation of bacteria cells during transportation. Reductive reagents such as methyl red, methylene blue, bromcresol purple, phenol red and resazurin were added to milk samples, and then colors of milk were changed based on SCC of milk. For optimal reductive reagents, reaction time was controlled at 3 level of reaction time. A spectrophotometer was used to measure reflectance spectra from milk samples. The partial least square (PLS) models were developed to predict SCC of unhomogenized milk. The PLS results showed that milk samples with reductive reagents had a good correlation between predicted and measured SCC at 5 minutes of reaction time in the visible range. The PLS models with resazurin reagent had the best performance in $400{\sim}600\;nm$. The prediction results of milk samples with resazurin had 0.86 of correlation coefficient and 14,184 cell/mL of SEP.

Study of Dye Encapsulated Microcapsule Polymerization Using Polyurethane Prepolymer Synthesis and Textile Finishing (폴리우레탄 프리폴리머 합성을 통한 색소 담지 마이크로캡슐 제조 및 섬유가공)

  • In this study, dye encapsulated microcapsules were produced by polyurethane prepolymer synthesis method using hexamethylene diisocyanate, ethylene glycol and methyl ethyl ketone. The study showed that the average size of microcapsules were $4.697{\mu}m$ in normal distribution. These microcapsules were induced red color by thermochromic fluoran red dye with showing color change as temperature. After the textile finishing of microcapsules, durability of microcapsules were checked as crocking times and lightfastness. The microcapsules were pressed at protrusion of textile weave in 10 crocking times which meant that the microcapsules not fallen off. Lightfastness was acceptable giving rating 4. It means that the polyurethane microcapsules not affect to light durability.

Increased Antimutagenic and Anticancer Activities of Chinese Cabbage Kimchi by Changing Kinds and Levels of Sub-Ingredient (부재료 첨가 배추김치의 항돌연변이 및 항암성 증진효과)

  • To enhance the antimutagenic and anticancer activities of chinese cabbage kimchi, 13 kinds of kimchi, which were different kinds and levels of sub-ingredient added kimchi, were prepared and fermented at 15℃ for 1 day and then at 5℃ up to pH 4.3. The antimutagenic effects of the methanol extracts of the kimchi were studied by using Ames mutagenicity test in Salmonella typhimurium TA100 and SOS chromotest in E. coli PQ37. Among the kimchi samples, high ratio of red pepper powder(7%) and garlic(2.8% or 5.2%) added kimchi, 1% chinese pepper powder added kimchi and organic cultivated chinese cabbage kimchi significantly reduced(p<0.05) the mutagenicity induced by aflatoxin B1(AFB1) in Ames test and SOS response against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in SOS chromotest, and also the kimchi inhibited more effectively(p<0.05) the survival and growth of AGS human gastric adenocarcinoma cells than the standarized kimchi on the SRB assay, MTT assay and growth inhibition test. These results suggest that the antimutagenic and anticancer activites of kimchi can be increased by the sub-ingredients such as organic cultivated chinese cabbage, red pepper powder, garlic and chinese pepper powder.

Studies on Chemical Composition of Red Algae (국내산 홍조류의 구성성분에 관한 연구)

  • Chemical components of red algae (Gelidium amansii, Gracilaria verrucosa, Gigartina tenella, Carpopeltis cornea, Plocamium ovicornis, Callophyllis adnata, Lomentaria hakodatensis, Gymnogongrus flabelliformis, Chondrus pinnulatus, Adinotrichia fragilis, Gloipeltis tenax and Campylaephora hypnaeoides) were examined. The contents of carbohydrate, ash, crude protein and fat in dried red algae are $40.38\~69.88\%,\;8.57\~30.23\%,\;18.11\~33.90\%\;and\;0.11\~0.90\%$ on dry base, respectively. Yields of water-soluble compound in dried red algae and contents of sulfate in water-soluble compound extracted red algae were $4.32\~55.78\%\;and\;1.65\~19.48\%$, respectively. The principal sugars of water-soluble compound extracted from red algae were galactose, 3, 6-anhydrogalactose, glucose and xylose.

Effect of Inoculum Concentration of Rhizoctonia solani and Pythium ultimum Causing Damping-off of Pepper and Cucumber on the Efficacy of the Mixture of Etridiazole and Thiophanate-methyl (Rhizoctonia solani와 Pythium ultimum의 접종 농도가 etridiazole과 thiophanate-methyl 합제의 모잘록병 방제 효과에 미치는 영향)

  • This study was performed to investigate the inhibiting activity of etridiazole and thiophanate-mthyl on mycelial growth of Rhizoctonia solani and Pythium ultimum and the effect of inoculum density of each pathogen on the control efficacy of the mixture of etridiazole and thiophanate-methyl in a seedling assay test. In mycelial growth inhibition test, $EC_{50}$ values of etridiazole and thiophanate-methyl against R. solani were 15.87 and 9.34 ${\mu}g\;mL^{-1}$, while those were 0.2 and more than $500\;{\mu}g\;mL^{-1}$ against P. ultimum, respectively. Controlling activity of the mixture of etridiazole and thiophanate-methyl against damping-off of pepper and cucumber, caused by R. solani and P. ultimum was tested in a greenhouse. With the inoculation of R. solani, disease incidences of seedling in pepper were 73% and 95% at 0.5% and 1.0% of inoculum concentration, while in cucumber those were 55% and 62% at 1.0% and 2.0%. When P. ultimum was inoculated into soil by 2.0% of inoculum concentration, those in pepper and cucumber were 66.7% and 96.8%, respectively. The efficacy of the mixture was somehow affected by the concentration of R. solani. While each control value of the mixture was 94.4% and 90.7% in pepper and cucumber at low inoculum concentration (0.05%), the efficacy of the fungicide decreased in pepper and cucumber by 70.7% and 72.9% at high concentration of R. solani (0.1% in pepper and 0.5% in cucumber). However, the control value of the mixture was 100% in pepper and cucumber, irrespective of the inoculum concentration of P. ultimum, however, the increase of inoculum concentration in soil did not result in the decrease of the fungicide efficacy.

Analysis of Soil Microbial Communities Formed by Different Upland Fields in Gyeongnam Province

  • The present study investigated variations in soil microbial communities by fatty acid methyl ester (FAME) and the chemical properties at 24 sites of upland soils in Gyeongnam Province. The electrical conductivity of the soil under potato cultivation was significantly higher than those of the red pepper and soybean soils (p < 0.05). The gram-negative bacteria community in potato soil was significantly lower than those in the garlic and soybean soils (p < 0.05). The communities of actinomycetes and arbuscular mycorrhizal fungi in the red pepper soil were significantly higher than those in the potato soil (p < 0.05). In addition, the cy17:0 to 16:$1{\omega}7c$ ratio was significantly lower in red pepper, soybean, and garlic soils compared with potato soil, indicating that microbial stress decreased. Consequently, differences in soil microbial community were highly associated with cultivated crop species, and this might be resulted from the difference in soil chemical properties.
