Study of Antioxidation Action of Lenonuri herba Extract (익모초 추출물의 항산화 작용에 관한 연구)

  • Motherwort (Leonurus sibiricus L), a hemp nettle (Labiatae) plant, grows about 1.5${\~}$2 m high spontaneously to wildness in Korea, China, Japan, and other Asian place. Its medical applications includes women's uterine disease, urination, bloody stool, bloody urine, and hemorrhoids. It's also effective for high blood pressure, heart stimulation, and anti-cancer activity. We first prepared chlroform and methanol extracts of motherwort and then they were fractionated using water, $30\%$ methanol, $60\%$ methanol, and $100\%$ methanol, respectively. Each fractionates is assayed for free radical scavenging activities against DPPH and anti-oxidant activity by TBARS assay measuring lipid peroxidation using LDL. The $30\%$ and $60\%$ methanol fractionates of methanol extracts showed strong anti-oxidant activity compared to vitamin C. They also had more Potent SOD activity using pyrogallol at 250 ppm than that of vitamin C. These results suggest that anti-oxidant activities of motherwort may be applicable to development of natural anti-oxidant cosmetics. Possibility of nature anti-oxidation ability cosmetics is seen possibility low official.

Effect of O2 Plasma Treatments of Carbon Supports on Pt-Ru Electrocatalysts

  • In the present study, carbon supports mixed with purified multi-walled carbon nanotubes (MWNTs) and carbon blacks (CBs) were used to improve the cell performance of direct methanol fuel cells (DMFCs). Additionally, the effect of $O_2$ plasma treatment on CBs/MWNTs supports was investigated for different plasma RF powers of 100, 200, and 300 W. The surface and structural properties of the CBs/MWNTs supports were characterized by FT-IR, X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), and inductive coupled plasma-mass spectrometer (ICP-MS). The electrocatalytic activity of PtRu/CBs/MWNTs catalysts was investigated by cyclic voltammetry measurement. In the experimental results, the oxygen functional groups of the supports were increased with increasing plasma RF power, while the average Pt particle size was decreased owing to the improvement of dispersibility of the catalysts. The electrochemical activity of the catalysts for methanol oxidation was gradually improved by the larger available active surface area, itself due to the introduction of oxygen functional groups. Consequently, it was found that $O_2$ plasma treatments could influence the surface properties of the carbon supports, resulting in enhanced electrocatalytic activity of the catalysts for DMFCs.

Antioxidant Activity of the Halophyte Ligustrum japonicum (염생식물 광나무(Ligustrum japonicum)의 항산화 활성)

  • Dried samples of Ligustrum japonicum were extracted twice: with methylene chloride and with methanol (MeOH), respectively. The combined crude extracts were successively fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water fractions by liquid-liquid partition. Antioxidant activities of crude extract and its solvent fractions were evaluated by measuring authentic $ONOO^-$, and $ONOO^-$ generated from 3-morpholinsydnonimine (SIN-1) as well as degree of occurrence of intracellular ROS in HT 1080 cells, and genomic DNA oxidation. The 85% aq.MeOH and n-BuOH fractions exhibited the good antioxidant activity. Further purification of the 85% aq.MeOH fracition led to the isolation of Oleanolic acid (1), Maslinic acid (2), and Ursolic acid (3). All compounds showed the significant antioxidant effects in all assay systems.

Antioxidant Effect of Alisma plantago-aquatica var. orientale and Its Main Component (택사 메탄올 추출물과 주성분의 항산화작용)

  • Reactive oxygen species(ROS) or free radicals are produced in the pathogenesis of human diseases including atherosclerosis, diabetes, cancer, and aging. Antioxidants are associated with the prevention of ROS-induced tissue and cellular damage in the various diseases. This study investigated the antioxidative activities of the methanol extract of Alisma plantagoaquatica var. orientale and its main component under conditions of radical generation using allophycocyanin and ferric-thiocyanate assay. Alisol B 23-acetate as a main component was isolated from the methanol extract of Alisma plantago-aquatica var. orientale. In results, the extract of Alisma plantago-aquatica var. orientale showed inhibitory activity on AAPH [2,2'-azobis(2-amidinopropane)dihydrochloride]-induced protein oxidation. Also, the extract of Alisma plantago-aquatica var. orientale and alisol B 23-acetate inhibited lipid peroxidation. These results indicate that Alisma plantago-aquatica var. orientale and alisol B 23-acetate show promise as therapeutic agents for various damages involving free radical reactions.

Tyrosinase Inhibitory and Antioxidant Activities of Korean Mistletoe (Viscum album var. coloratum) Extract and Its Fractions

  • The copper-containing enzyme, tyrosinase, catalyzes the oxidation of tyrosine into dihydroxy phenylalanine (DOPA) and subsequently DOPAquinone. It is responsible, not only for the pigment melanin biosynthesis in human skin, but also for browning in foods. In the present study, tyrosinase inhibitory and antioxidant activities of Korean mistletoe extract and its fractions were investigated. As a result, both water and methanol (MeOn) extracts inhibited the tyrosinase activity. Among the fractions, the fraction eluted with $95\%$ MeOn significantly inhibited the tyrosinase activity. The fraction was further purified, and the purified fraction C strongly inhibited the enzyme activity up to $92\%$. In addition, water and methanol extracts exerted radical scavenging effects. The fractions eluted with $70\%\;MeOn\;and\;95\%$ Me on showed high radical scavenging activities. In conclusion, these results suggest that Korean mistletoe extract and its fractions might be useful for the treatment of various dermatological disorders such as epidermal hyperpigmentation and for improving food quality.

  • The aims of this studies were carried out to investigate the antioxidant activity on low density lipoprotein(LDL) using substances extracted from Bacillus sp. RH-5. The antioxidative substances produced extracellular in the culture broth by Bacillius sp. RH-S was obtained by elution of chloroform : methanol from silicagel column (80cm x100cm) chromatography. Band 4 eluted from fraction 3 by TLC was appeared at highest level of antioxidative activity using thiocyanate methed. Band 4 at a concentration of 100 or 200$\mu$g/$m\ell$ inhibited oxidation of LDL induced by the mouse transformed macrophage. According to IR. NMR or GC/MASS, the antioxidant substance was identified as 5-hydroxyindole.

A Study on Some Antioxidative Effects of Stewed Pumpkin Juice on Lipid (호박즙의 유지에 대한 항산화 효과)

  • This study was carried out to investigate some antioxidative effects of stewed pumpkin juice (SPJ) and the SPJs added with ginger (SPJ-G), onion (SPJ-O), jujube (SPJ-J) or boxthorn (SPJ-B), respectively, on the lipid oxidation of soybean oil (S) and mackerel pike boiled ground (MPBG). Antioxidative substances of SPJ were extracted by using the two solvent layers of n-hexane:acetone (1:1)(H) and methanol:water (7:3)M. The antioxidative effects of the extracts were observed by the peroxide value (P.O.V.) and thiobabituric acid (T.B.A.) value. The P.O.V. and T.B.A. value of two solvent extracts of S and MPBG showed significantly lower than those of S and MPBG. The SPJ showed very distingnished antioxidative effects. It was considered that melanoidine extracted by using methanol:water (7:3) solvent as well as carotenoid extracted by using n-hexane:acetone (1:1) solvent had an antioxidative effect. During storage, T.B.A. value of MPBG significantly increased, while that of MPBG-SPJ significantly decreased more than that of MPBG. T.B.A. value of MPBG-SPJs decreased much than that of MPBG-SPJ, SPJ-O, SPJ-G, SPJ-J and SPJ-B, in order of abundance, showed higher level of antioxidative effects than SPJ.   • 2,5-dimethoxy-2,5-dihydrofurfuryl alcohol was electrosynthesized from furfuryl alcohol in methanol solution by using three kinds of metal oxide anode. The electrods were prepared by the following process : Thin layer of semiconducting material such as tin(IV)oxide and antimony(III)oxide was made on the titanium base metal in an electric furnace. The titanium metal block with the layer was coated with ${\alpha}-PbO_2$, ${\beta}-PbO_2$, and $MnO_2$ in each electrolytes by anodic deposition, respectively. The lead dioxide electrodes showed better anti-corrosive property than the manganase dioxide electrode. The yield of the product was 92% which is almost the same as the one with conventional platinum electrodes.

  • The effect of antioxidant activity of Platycodon grandiflorum (PG) on the liposomal phospholipid membrane was investigated by spectrophotometry. Membrane oxidation causes damage to the membrane fluidity and permeability. It brings further destruction to the sustenance of biological homeostasis. In addition, it is related to several diseases, aging and carcinogenesis. The sample PG was extracted and fractionated to five different types; butanol (PGMB), ethylacetate (PGMEA), ethylether (PGMEE), hexane (PGMH) and methanol (PGMM). The oxidation indices of PGMEA and PGMEE fractions in oxidized dilinoleoylphosphatidylcholine (DLPC) liposomes had stronger antioxidant activities than that of ${\alpha}$-tocopherol and were similar to antioxidant activities compared with butylated hydroxy toluene (BHT), a well-known potent antioxidant, in oxidized DLPC liposomes. The oxidation indices of PGMM extract, PGMB and PGMH fractions exhibited weak antioxidant activity compared with ${\alpha}$-tocopherol in oxidized DLPC liposomes. The oxidation indiex of PGMEE fractions added with vitamin C showed even strong antioxidant activity in the oxidized DLPC liposomes. The oxidation activity of BHT with vitamin C also proved to be stronger than BHT without vitamin C. Therefore vitamin C evidently helps to improve the effect of antioxidant in DLPC liposomes. These results indicate that potentially bioactive substances in PGMEE fraction has a function as potent antioxidant against phospholipid membrane oxidation.

Antioxidant Activities of Water or Methanol Extract from Cherry (Prunus yedoensis) and Its Utilization to the Pork Patties (버찌(Prunus yedoensis) 추출물의 항산화 활성 평가 및 돈육 패티에 이용)

  • This study was performed to investigate the antioxidant activity of cherry added into meat products. Water and methanol were used to extract the antioxidant compounds from cherry. Total phenolic compounds of the methanol and water extract of cherry were 2.17 g/100 g and 2.77 g/100 g, respectively. The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of methanol extract showed similar activities to those with ascorbic acid at all concentrations (from 0.1% to 2.0%). Especially, water extract of cherry showed similar activity to those of ascorbic acid (AA), and methanol extract, when 2% of cherry extract was added. The reducing power of cherry was not comparable to those with AA, however no differences in reducing power were observed between the water and methanol extract. The iron chelating ability of cherry was observed in the range of 17.8-94.0% at both water and methanol extracts. An increased iron chelating ability was observed with increased concentration up to 2%. Iron chelating ability for water extract of cherry tended to be lower than those with methanol extract. After pork patties were manufactured with methanol extract of cherry at 0.5 and 1.0%, physicochemical properties, lipid oxidation and microbial changes of patties were measured. The addition of methanol extract of cherry reduced pH, brightness, redness, yellowness and thiobarbituric acid reactive substance (TBARS). During 14 d of storage, pH, TBARS and microbial counts were increased, while redness and yellowness values were decreased. Since the addition of methanol extract of cherry lowered TBARS during storage, it could be used as a natural antioxidant in meat products.
