Dead insects infected with fungi were collected from mountains and islands in several locations in Korea from 2003 to 2007. Morphological characteristics of the fungal fruiting bodies produced on the insect samples were examined. A total of 542 entomopathogenic fungal isolates were obtained from the insect samples and identified based on their morphological and cultural characteristics. Three species of Beauveria, eight species of Cordyceps and four species of Isaria were mainly isolated from the insect samples. The other entomopathogenic fungi isolated from the insect samples were Metarhizium anisopliae, Nomuraea rileyi, Paecilomyces sp. and Verticillium sp.

The value of silkworms as functional health food materials has increased, as has the interest in its disease control for stable production, and in the economic value of entomopathogenic microorganisms. In this study, we isolated and identified disease-causing fungi from white muscardine silkworms, and confirmed whether this strain could produce white muscardine silkworms. For the analysis of the cause of white muscardine disease in the infected silkworms, the fungi and prokaryotes causing the disease were identified, isolated, and identified using metagenome analysis. Metagenomic analysis detected a large amount of the fungus Metarhizium rileyi in silkworms, and a large amount of the bacterium Enterococcus mundtii, which was presumed to be the causative agent of the disease. For accurate identification of the fungi, these were purified by culture medium, and sequencing and phylogenetic tree analyses were performed using an internal transcribed spacer. As a result, M. rileyi, Cladosporium cladosporioides, and C. tenuissimum were identified. In general, M. rileyi is known to form green conidia, but in this study, white-yellow conidia were formed, indicating that the exact causative agent of the fungal disease cannot be estimated by diagnosing the symptoms. Thus, a diagnostic method is necessary for the continuously collection of required pathogens, and identifying their morphological and genetic characteristics.

Biological control of lepidopterous insect pests, Agrotis segetum, Artogeia rapae, Mamestra brassicae, Plutella xylostella, Spodoptera exigua, and S. litura with entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae isolated from Gyeongbuk province were investigated. Mean lethal concentration ($LC_50$) values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A. segetum larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of A, rapae larvae were $1.2\times10^7\;conidia/m\ell$ and $5.2\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of M. brassicae and P. xylostella, larvae were $1.5\times10^6\;conidia/m\ell$, $9.7\times10^5\;conidia/m\ell$, $3.0\times10^6\;conidia/m\ell$ and $1.4\times10^6\;conidia/m\ell$, respectively. $LC_50$ values of B. bassiana and M. anisopliae against $2\cdot3rd$ instar of S. exigua, and S. litura larvae were $6.3\times10^6 \;conidia/m\ell$, $2.6\times10^6\;conidia/m\ell$, $1.6\times10^7\;conidia/m\ell$ and $3.4\times10^6\;conidia/m\ell$ respectively.
DNA fragments homologous to chitin synthase gene were amplified from the genomic DNA of Beauveria brongniartii by PCR using degenerate primers. Cloning and sequencing of the PCR-amplified fragments led to the identification of a gene, designated BbCHSl. Comparison of the deduced amino acid sequence of BbCHSl with those of other Euascomycetes revealed that BbCHSl is a gene for class II chitin synthase. The Blastp search of the deduced amino acid sequence of BbCHSl displayed the highest rate of similarity, 95.8%, with CHS2 of Metarhizium unisopliae. Phylogenetic analysis of the amino acid sequences confirmed the taxonomic and evolutionary position of B. brongniartii, which was previously derived by traditional fungal classification based on morphological features.
We isolated nine fungal strains from different environmental materials collected from different locations during a survey of fungal diversity in Korea. Using molecular phylogenetic analyses and morphological characteristics, nine previously undescribed strains were identified and assigned to the species Collariella robusta, Fusicolla acetilerea, Hongkongmyces pedis, Hongkongmyces snookiorum, Mariannaea fusiformis, Metarhizium pemphigi, Pallidocercospora crystallina, Scopulariopsis candida, and Volutella citrinella. Diverse environmental samples may thus be a promising source for isolating and investigating novel fungal species, thus sampling efforts should be increased in future studies. This study also reports identification of some rare fungal species belonging to the genera Hongkongmyces and Pallidocercospora from Korea.
The silverleaf whitefly, Bemisia tabaci, is a major pest distributing worldwide damaging over 900 host plant species, and is highly resistant to chemical pesti- cides. Due to the high pesticide resistance of whitefly, there is a need for alternatives to chemical control. Entomopathogenic fungi are candidates for biological pesticide that can overcome the resistance problem of chemical pesticide. Therefore, in this study, we tested pathogenicity of the entomopathogenic fungi to select high insec- ticidal activity against whitefly. As a result, IPBL-C (Cordyceps fumosorosea) and IPBL-F (Metarhizium pinghaense) isolates showed high insecticidal activity against whitefly. Additionally, as a result of culturing the selected isolates on spent coffee grounds medium, the conidia of IPBL-F produced on coffee grounds medium showed five times higher heat stability after heat treatment at 45℃ for one hour than conidia produced on PDA medium.
Purpose: The aim was to collect and classify the Cordyceps parasitized in cicadas from July to October every year from 1990 to 2016 in Korea. And they were frequently collected in Nepal, Vietnam, Japan, China, and Thailand. Methods: Cordyceps parasitizing cicadas collected in mountains and fields. Results: A total of 1,104 specimens were collected that belonged to 10 different species under nine genera. The highest number of samples belonged to Ophiocordyceps (563 specimens), followed by Isaria (361 specimens), Polycephalomyces (73 specimens), Cordyceps (70 specimens), Beauveria (25 specimens), Perennicordyceps (8 specimens), Metarhizium (2 specimens) and Purpureocillium (2 ones). Among Ophiocordyceps spp. O. longissima was most frequently collected with a total of 426 samples out of 563, followed by O. heteropoda with 120 ones and O. sobolifera with 17 specimens. The species mainly collected in Korea, but C. ishikariensis was collected in Nepal only. The new characteristic was that Isaria cicada-like synnemata were found growing together with C. ishikariensis stromata on the same host. In Korea, the collected 691 specimens in total out of 1,104 were found in Mt. Halla in Jeju Island. Other mountains in Korea where the samples were collected were Mountains Daeryong, Jiri, Yongmoon, Samag, Seolag, Gujeol, Duryun, Baegam, Chilgap, Chundeung, Naejang, Welchul, and Daeryong. The three samples were not identified. Conclusions: A total of 1,104 specimens belonged to 10 different species under nine genera, and the collected 691 samples were found in Mt. Halla in Jeju Island.

In Indonesia, Coptotermes curvignathus Holmgren and Cryptotermes cyanocephalus Light termite attacks can damage wood and causing losses of 8.7 trillion rupiah per year. Wood treatment and soil barrier are very important to protect structures and their components from termite infestation. The application of synthetic chemicals that pose risks to the environment and human health. The growing movement to replace these chemicals with new termiticides that are safe for the environment in place of persistent organic pollutants. Efficacy performance in entomopathogenic fungi spores such as that produce decanoic acid, hexadecanoic acid, palmitic acid, and octadecadienoic acid are potential compounds that provide for ecofriendly termite control. Entomopathogenic fungal spores from Metarhizium anisopliae, Trichoderma harzianum, and Beauveria bassiana, as the active ingredient were formulated with nanocellulose was added as an inert ingredient to which helped to deliver the active ingredients on controlling the target pest, and enhance the utility ability of the product fungi to control termites. The mortality of these termites successively entomopathogenic fungi was the main cause of death. The higher concentrations being associated with of nanocellulose used affect increased the number of termites mortality. M. anisopliae paired with 60% nanocellulose concentrate was the best percentage for yielded the greatest C. curvignathus and C. cynocephalus termite control. In the field testing, M. anisopliae provided better, it showed the finest result of termite control rather than B. bassiana and T. harzianum. The results of the research indicate that entomopathogenic fungi can be used for Pest Control Management as the subterranean termite and drywood termite control.
To develope a microbial pesticide for the control of agricultural and forestal pests in Korea, the mass culture system of entomopathogenic fungi was studied. Previously, we have developed the mass culture system which was adaptable for the culture of Beauveria bassiana. In this study, we determined the efficacy of this mass culture system for other entomopathogenic fungi, B. bassiana, Beauveria brongniartii, Metarhizium anisopliae, and Verticillium lecanni. To determine the efficacy of mass culture system, we examined the growth rate of entomopathogenic fungi in this system which was composed of 1st liquid media for growth of blastospore and 2nd pellet media for growth of conidia. As the result, we obtained that the blastopore numbers increased 103-104 times in liquid media at 72 hrs post inoculation. The results showed that this mass culture system for the growth of entomopathogenic fungi was effective.
Dokdo island is located in the northeastern part of Ulleungdo, known as volcanic island. In total, 53 fungal isolates were isolated from Dokdo island soil sample, using dilution plate technique. The isolates were identified on the basis of morphological characteristics and rDNA ITS sequence analysis. Out of them, 41 isolates were identified at the level of species. The dominant fungal species and genera included Fusarium spp., Mucor sp., Clonostachys spp., and Trichoderma sp. The % sequence identity (the number of matches/the complete alignment length) values via NCBI BLAST searching of EML-IF9, EML-MF30-1 and EML-DDSF4 represented 97.19% (485/499) with Clonostachys cf. rosea (GenBank accession no. KC313107), 98.33% (472/480) with Metarhizium guizhouense (GenBank accession no. HM055445), and 100% (350/350) with Mortierella oligospora (GenBank accession no. JX976032), respectively. Three species of C. rosea, M. guizhouense and M. oligospora represented new records of fungi from Dokdo island, Korea. The antimicrobial activities of the fungal strains varied with tested. Two isolates (EML-MFS30-1 and EML-IF9) showed antifungal activity against several fungi including Fusarium oxysporum and Rhizotonia solani. Clonostachys rosea (EML-IF9) showed strong hydrolytic enzyme activity. Our results showed that the antagonistic fungi including Clonostachys rosea will be used as potential biocontrol agents for control of fungal diseases.
