Characteristics of Adsorption and Desorption of Metalaxyl in the Green Soil of Golf Course (골프장 그린 토양에서 Metalaxyl의 흡ㆍ탈착 특성)

  • Laboratory experiments were conducted to examine the behavior of metalaxyl in environment which was used as pesticide in green soil of golf course and as functions of the characteristics of adsorption, desorption and degradation in soil texture and organic matter contents. Acid water containing metalaxyl was conducted to evaluate the effects on adsorption, desorption and degradation. The adsorption of metalaxyl played more significant role in organic contents than clay contents, and pH Increases more pH 2.5 than pH 5.6. The desorption of metalaxyl from contaminants soil decreased higher organic contents LS-soil than S-soil, but the desorption amount of metalaxyl increased more pH 5.6 than pH 2.5. The rate of degradation of metalaxyl in green soil environmental increased higher organic contents LS-soil than S-soil and decreased more pH 2.5 than pH 5.6. These results indicated that the behavior of metalaxyl of the green soil was affected the soil texture of the golf course. Increasing of organic contents, the adsorption amount of metalaxyl on soil increased. Moreover the decrease of the pH of solution increased adsorption amounts and decreased desorption amounts. As the results, the transportation of metalaxyl in soil decreased the acidic rates. The acidification of soil by the acid rain increased the adsorption amount of metalaxyl, but the degradation of metalaxyl decreased. Therefore, it is possible to sustain contamination in run-off the stream and ground water by residuals in soil.

The Change of Resistance of Phytophthora infestans to Metalaxyl and the Relationship with the Pathogenicity on Pepper Plants (국내 고추 역병균의 metalaxyl 저항성 변화 및 metalaxyl 저항성과 고추에 대한 병원성과의 상관 관계)

  • Through the agar dilution method on V-8 juice agar, sensitivity of Phytophthora capsici causing pepper Phytophthora blight to metalaxyl was investigated by using isolates obtained from infected pepper plants during 3 years from 2005 to 2007. By the lapse of time, $EC_{50}$ value to metalaxyl was decreased, showing 1.45, 0.83, and $0.32{\mu}g\;mL^{-1}$ in 2005, 2006, and 2007. None of 2007 isolates was found to be resistant to metalaxyl. Compared the sensitivity of P. capsici isolates to metalaxyl with those to mandipropamid and dimethomorph, there is not a cross resistance response between metalaxyl and mandipropamid/dimethomorph. The resistance to metalaxyl in pepper Phytophthora blight pathogen was not related with the mycelial growth on V-8 agar medium and the pathogenicity on pepper plants.

  • Five commercial chemicals, Dimethomorph, Foseyl-Al, Oxadixyl + Mancozeb, Propamocarb hydrochloride, and Metalaxyl, were tested for control of tobacco black shank (Phytophthora nicotianae) and/or delaying buildup of the resistant population to Metalaxyl which has been used for control of the disease in Korean tobacco farms. Propamocarb hydrochloride seemed to have a cross-resistance to the Metalaxyl resistant isolates showing similar response with Metalaxyl in vitro. Meanwhile, Dimethomorph+Mancozeb and Oxadixyl+Mancozeb were selected to be the promising chemicals which are able to be alternative to Metalaxyl for the black shank control in accordance with in vitro and in field trial.

Changes in Frequencies and Distribution of A2 Mating Type and Metalaxyl-Resistant Isolates of Phytophthora infestans in Korea (우라나라 감자 역병균 A2 교배형 및 Metalaxyl 저항성균의 빈도 및 분포의 변화)

  • Phytophthora infestans populations collected from various geographical locations of Korea in 1991 and 1993 were analyzed for mating types and responses to metalaxyl. Both A1 and A2 mating type isolates were detected in 1991. The majority of the isolates were A2 mating type, but no A1 mating type was detected in 1993. About 40% of the isolates collected in 1991 were resistant to metalaxyl, and the distribution of metalaxyl-resistant isolates of P. infestans was strongly associated with their geographic origins in Korea. Metalaxyl-resistant isolates with EC50 values > 50$\mu\textrm{g}$/ml were collected from the northern provinces of Kangwon, Kyungbuk, and Chonbuk, but not from the southern provinces of Kyungnam, Chonnam, and Jeju in 1991. The drastic increase in the degree of quantitative resistance to metalaxyl was detected among the isolates from the southern provinces during 1991~1993. More than 50% of the isolates collected from the southern provinces of Kyungnam and Chonnam in 1993 had EC50 values >50$\mu\textrm{g}$/ml. The province of Kangwon had isolates with the greatest resistance to metalaxyl. this alpine areas might be the origin of metalaxyl-resistant isolates of P. infestans in Korea. The A2 genotype with metalaxyl resistance appears to be displacing the A1 genotype which is presently the predominant genotype in Korea.

Effect of Exogenous Metalaxyl Seed-Soaking on Root Initiation and Elongation of Young Rice Seedling (벼 어린모 뿌리의 분화 및 신장에 미치는 Metalaxyl의 종자침청처리 효과)

  • The metalaxyl seed-soaking treatment markedly increased the root-mat formation of the young rice seedling(8- to l0-day-old) for machine transplanting. This experiment was conducted to find out a possible rooting mechanism of the young rice seedling in relation to the exogenous metalaxyl seed-soaking treatment. The rice seeds of Hwaseongbyeo variety were soaked in the solution of metalaxyl for 24 hours at room temperature. Metalaxyl(25% wettable powder), a fungicide, was used at the concentration of 200ppm as a promoting substance on the root-mat formation of the young rice seedling. Seeding rate of the young rice seedling was 220g per seed tray(30${\times}$60${\times}$depth 3cm). Metalaxyl seed treatment was highly effective in root initiation and elongation of the young rice seedling in terms of root activity, the number and length of root, and amino acid content of roots. The exogenous metalaxyl application would also increase the chlorophyll content and photosynthetic rate in the young rice seedling. Those factors resulted in vigorous root-mat formation of the young rice seedling. In particular, alanine is a lead compound of metalaxyl which biochemically plays an important role in energy intermediate of the rice plant. Alanine conjugation with a glucose (N-${\beta}$-glucoside) is probably the mechanism in enhancement of initiation and elongation of rice roots of young rice seedling.

Production of Polycolonal Antibody and Optimum Conditions in ELISA for Metalaxyl (Metalaxyl의 다클론 항체생산과 최적 ELISA 조건)

  • A competitive indirect enzyme-linked immunosorbent assay(ELISA) was developed to detect and quantify levels of the fungicide metalaxyl in crops. Antiserum against metalaxyl was demonstrated in rabbits immunized with metalaxyl-human serum albumin(HSA) conjugate. Metalaxyl-protein conjugate was prepared by mixed anhydride and peptide coupling method with EDC. In this assay, metalaxyl-ovalbumin(OA) was $coated(8{\mu}g/ml)$ on the microtiter plate, which was incubated for 1 hr at $4^{\circ}C$ or 4 hr at $37^{\circ}C$ with diluted antiserum(1:2,000). The optimum volume ratio of antigen and antibody mixture was 0.5: 1, which was incubated for 1 hr at $20^{\circ}C$. The detection of metalaxyl bound on the surface of wells was determined by the reaction(30 min) of antirabbit Ig G-peroxidase conjugate with its substrate.

Occurrence of Metalaxyl-Resistant Isolates of Pythium spp. Isolated from Turfgrasses of Golf Courses in Korea (우리나라 골프장 잔디에서 분리한 Metalaxyl 저항성 Pythium spp.의 발생)

  • Of 125 isolates collected from 35 golf courses, sensitivity of 44 isolates of Pythium species to metalaxyl was determined on corn meal agar with various concentrations of metalaxyl (0.1, 1.0, 10.0, 50.0, 100.0, and $250.0{\mu}g\;a.\;i/ml$). The isolates were able to be categorized into the sensitive and resistant groups based on hyphal growth measured in terms of colony diameters on the medium with $1.0\;and\;10.0{\mu}g\;a.\;i./ml$. When compared with hyphal growth on the medium without metalaxyl, hyphal growth of the sensitive group which included 31 isolates was inhibited by $66{\sim}98%$ on the medium with $1.0{\mu}g\;a.\;i./ml$, whereas that of the resistant group which included 13 isolates grew well and the hyphal growth was inhibited only by $6{\sim}26%$. When $10.0{\mu}g\;a.\;i./ml$ of metalaxyl was included in the medium, hyphal growth of the sensitive and resistant groups was inhibited by $82{\sim}99%\;and\;27{\sim}47%$, respectively. Occurrence of metalaxyl-resistant isolates of Pythium spp. not only from turfgrasses on golf courses but also from other crops was observed for the first time in Korea. Metalaxyl-resistant isolates occurred most frequently in P. graminicola. Control effects of metalaxyl were determined by applying metalaxyl after and before inoculation of 4 and 3 isolates of sensitive and resistant isolates of P. graminicola, respectively, to creeping bentgrass in pots. The minimum concentration of metalaxyl to control metalaxyl-sensitive isolates was $6.25{\mu}g\;a.\;i./ml$, whereas the disease caused by the metalaxyl-resistant isolates could not be controlled with $12.50{\mu}g\;a.\;i./ml$ of metalaxyl. The disease was controlled more effectively by an application of metalaxyl prior to inoculation than after occurrence of the disease.

Determination of Pre-Harvest Residue Limits of Pesticides Metalaxyl-M and Flusilazole in Oriental Melon (생산단계 참외 중 Metalaxyl-M 및 Flusilazole의 잔류허용기준 설정연구)

  • The present study was performed to investigate the pre-harvest residue limit (PHRL) of pesticides namely, metalaxyl-M and flusilazole in oriental melon, and to identify the biological half-life and characteristics of their residues. In this study, pesticides were sprayed once as single spray and double spray on oriental melon. The oriental melon samples were collected at 0, 1, 2, 3, 5, 7, 9 and 11 days before harvest and samples were extracted with QuEChERS method. The residues of both the pesticides were quantified using GC/NPD and LC/MS/MS. The limit of detection was found to be 0.02 mg/kg and 0.01 mg/kg and their recoveries were greater than 95% (95.7% ~ 103.2% for metalaxyl-M and 100.2% ~ 106.8% for flusilazole) for both pesticides. The biological half-lives of both metalaxyl-M and flusilazole were 12 days at single and double spray, respectively. The PHRL of metalaxyl-M and flusilazole was found 1.0 mg/kg and 0.3 mg/kg, respectively for 10 days before harvest. The results of the present study shows the residual level of both the pesticides metalaxyl-M and flusilazole in oriental melon were less than their maximum residual limits.

Evaluation in ELISA for the Residue Analysis of Metalaxyl in Crops (작물체중 Metalaxyl 의 잔류분석을 위한 ELISA의 응용)

  • The cross-reactivities of antibody derived from rabbit immunized with metalaxyl-HSA conjugate were identified to 44% and 28% for metalaxyl acid and metolachlor respectively. The detection range of metalaxyl in competitive ELISA was 5 ppb to 5 ppm. The recoveries of metalaxyl in ELISA for 6 crops; potato, sesame, pepper, cabbage, cucumber and onion was ranged 77.5-103.6%. In this test, CV% were calculated below 10% except for sesame sample as shown 21.5%.

Variation in Metalaxyl and Streptomycin Sulfate Resistance of Isolates Formed from Single Sporangium of Phytophora infestans (감자역병균(Phytophthora infestans)의 단일 포자낭으로부터 형성된 균총의 Metalaxyl과 Streptomycin Sulfate 내성 변화)

  • Parent isolates and $first-generation(F_1)$ progenies from single-sporangium of Phytophthora infestans were investigated for the acqusition of metalaxyl and streptomycin sulfate(SM) resistance. The parent isolates, KM10, U6, CDB6 showed resistance to metalaxyl, and F817, DNC303, DN107 to SM. Especially, the average mycelial growth of $F_1$ progenies of F817, CDB6, JD1 was more than 51% than parent isolates on 25ppm metalaxyl added V-8 juice agar, whereas those of KM10, U6, JD1 were extended more than 51% similar on 200ppm SM treatment. They acquired resistance on the basis of the standard in this experiment. There were positive correlations between parent isolates and $F_1$colonies in resistance. The $F_1$ colonies obtained from parent isolates showing above interinediate resistance on metalaxyl amended agar acquired much resistance.
