Immunomodulating Effect of the Lectin from Allomyrina dichotoma (장수풍뎅이(Allomyrina dichotoma) 렉틴의 면역기능 증강효과)

  • A lectin was purified from Allomyrina dichotoma (ADL) by physiological saline extraction, ammonium sulfate fractionation, anion exchange column chromatography on DEAE Sephadex A-50 and gel filtration column chromatography on Sephadex G-200. Several biochemical properties of ADL were characterized as follows: ADL from gel filtration column chromatography showed single band on SDS-PAGE. ADL agglutinated the erythrocytes of rabbit and human A, B, O, AB. Agglutinability was relatively stable at basic pH, and was stable at temperature below $40^{\circ}C$. Agglutinability was not affected by metal ions and EDTA. This lectin was proved to be a glycoprotein which contains 0.47% of sugars. The molecular weight of ADL was estimated to be 97,000 dalton by SDS-PAGE. By amino acid analysis, ADL exhibited high amounts of aspartic acid. The lectin's immunomodulating effect was measured as cytokine production. The productions of 5 cytokines $(IL-1{\alpha},\;IL-2,\;IL-6,\;IFN{\gamma}\;and\;TNF{\alpha})$ from peripheral blood mononuclear cells were measured by ELISA. The lectin induced the highest secretion of IL-2 at 8 hr, $TNF{\alpha}$ at 4 hr, and $IFN{\gamma}$ at 24hr, respectively. These results suggest that ADL can elicit the production of detectable cytokines from PBMC.

  • It is well known that PCB (Printed Circuit Board) is a complex mixture of various metals. In this study, pyro-metallurgical process was investigated to extract valuable metallic components from the PCB scrap. PCB scrap was shredded and oxidized to remove plastic materials, and then, quantitative analysis were made. 15 mass %$Al_2O_3-45$ mass %CaO-40 mass %$SiO_2$ and 32 mass %$SiO_2-20$ mass %$Al_2O_3-38$ mass %CaO-10 mass %MgO, were chosen as basic slag compositions which are determined based on the quantitative analysis of PCB scrap. During experiments a super kanthal rotating furnace was used to melt and separate metallic components. Moreover the revolution effect on was the recovery of valuable metals from PCB scrap also investigated.

Extraction of Representative Emotions for Evaluations of Tactile Impressions in a Car Interior (자동차 인테리어의 촉감 평가를 위한 대표감성 추출)

  • There are few that evaluate tactile emotion as it pertains to car interior parts, while studies on visual evaluations of car interiors as well as usability tests in a visual sense are numerous. The purpose of this study is to determine typical in-vehicle tactile emotions so that they can be used to evaluate tactile impressions of car interior parts. 52 words related to tactile impressions of car interiors were gathered from a survey in conjunction with an in-vehicle test, interviews with the car salespersons, and an analysis of car reviews. After a factor analysis with 52 words, 10 categories of major tactile emotions were clustered. These were roughness, toughness, friction, comfortability, stiffness, softness, temperature, sleekness, familiarity, and flexibility. These representative tactile emotions regarding a car interior can be used to evaluate tactile impressions of surfaces such as leather, plastic, metal and wood when used as parts in car interiors.

Use of Li-K-Cd Alloy to Remove MCl3 in LiCl-KCl Eutectic Salt (Li-K-Cd 합금을 이용한 LiCl-KCl 용융염에서 금속염화물의 제거)

  • In this study, we prepared Li-K-Cd alloy, which meets the requirement of eutectic ratio of Li:K, to maintain the operating temperature of the drawdown process at $500^{\circ}C$ and to achieve the reuse of LiCl-KCl molten salt. The prepared Li-K-Cd alloys were added to LiCl-KCl salt bearing U and Nd at $500^{\circ}C$ to investigate the removal of $UCl_3$ in the salt. The reduction of $UCl_3$ in the salt was examined by measuring the OCP value of salt and analyzing the salt composition by ICP-OES. Reduction was also visually confirmed by change of salt color from dark purple to white. The experimental results reveal that the prepared Li-K-Cd alloy has reductive extractability for $UCl_3$ in salt. By improving the preparation method, the Li-K-Cd alloy can be applied to the drawdown process.

Desmutagenic Activity of Heated Mountain Herb Juices (산채류(山菜類) 가열즙(加熱汁)의 돌연변이 억제 작용에 관(關)한 연구(硏究))

  • Potential mutagenicity of ten heated edible mountain herbs were examined with spore recassay, Ames test and DNA breaking test. Samples of edible mountain herbs were prepared with water extraction at $100^{\circ}C$ for 20 minutes. With the rec-assay, no significant mutagengic activity could be obtained from all of the samples, but among the eight of metal ions added to sample solution, $Pb^{2+}$ to R. crispus heated juice, $Zn^{2+}$ to L. fischeri and S. bracycarpa heated juice increased mutagenic activity of the samples. With the Ames test and DNA breaking test, all of the samples did not show mutagenicity. However, breaking action was activated on heated L. fischeri, P. japonicus. A. triphylla and A. tataricus juices in the presence of 25mM $Cu^{2+}$. But heated A. elata, H. aurantiaca, A. triphylla, S. bracycarpa and A. scaber juices were inactivated in the presence of 25mM $Fe^{2+}$. Desmutagenic activities against benzo$({\alpha})$pyrene significantly increased as increasing concentration of the heated edible mountain herb juices.

A study on recovery of rare earth oxide powders from waste NiMH batteries (폐니켈수소전지로부터 희토류 산화물 분말의 회수에 대한 연구)

  • For the recovering rare earths in the spent nickel-metal hydride batteries, 10 M NaOH is added to the solution leached with sulfuric acid. The rare earth powders were precipitated at rate of 98 % at the condition of pH 2.0 or less. The recovered rare earth complex precipitate increased the leaching rate to nitric acid by heat treatment at $800^{\circ}C$ for 4 hours. Subsequently secondary precipitation was performed by adding oxalic acid to the solution in which the rare earth complex precipitate was dissolved. The re-precipitated rare earth powders were converted into oxide form through heat treatment at $800^{\circ}C$ for 4 hours with purity of 99.5 %.

Determination of a Trace Amount of Copper, Lead, Cadmium and Zinc in Water by Solvent Extraction and Square Wave Polarography (溶媒抽出-矩形波폴라로그래피에의 물중의 미량 구리, 납, 카드뮴 및 아연의 定量)

  • The following new techniques have been developed: (A); To a 500ml of sample water, it was adjusted pH 10 with ammonia-anmonium citrate, added 10ml of 1${\%}$ sodium diethyldithiocarbamate and extracted three times with 10ml of CHCl3. The extract was shaken with 10ml of 0.05N $HCl-4{\times}10^{-4}M\;HgCl_2$. The aqueous solution was added 2ml of 2N KCl and washed two times with 10ml of pure $CHCl_3$, and then recorded square wave polarograms. (B); To a 500ml of sample water adjusted pH 10 with ammonia-ammonium citrate, it was added 2ml of 1${\%}$ 8-hydroxyquinoline and extracted three times with 10ml $CHCl_3$. The separated $CHCl_3$ phase was shaken with 10ml of 0.2 N HCl. The aqueous solution was recorded polarograms directly. These methods can be used for determination of the ppb order of metal in water with an error of ${\pm}10{\%}$. The method (B) can not be used for the determination of zinc on account of the free 8-hydroxyquinoline.

Analysis of Volatile Flavor Components of Pleurospermum kamtschaticum (누룩치의 휘발성 향미성분 분석)

  • Volatile flavor components in leaf and petiole of fresh Pleurospermum kamtschaticum H$\_$OFFM/ were extracted by SDE (simultaneous steam distillation and extraction) method using diethyl ether as solvent. Essential oils were analyzed by gas chromatography (GC) and combined gas chromatography-mass spectrometry (GC-MS). Identification of volatile flavor components was based on the Rl of GC and mass spectrum of GC-MS. A total of 31 components, including 15 hydrocarbons, 4 aldehydes, 1 ketone, 5 alcohols, 2 esters, 3 acids and 1 oxide were identified in the essential oils. (Z)-${\beta}$-Farnesene, (Z, E)-${\alpha}$-farnesene and farnesene were the major volatile flavor components in fresh Pleurospermum kamtschaticum. Volatile flavor patterns of Pleurospermum kamtschaticum were analyzed using electronic nose. Sensor T30/1 and PA2 that were sensitive to alcohols had the highest resistance for fresh Pleurospermum kamtschaticum. Resistance of six metal oxide sensors was decreased in dried sample compared with fresh one.

A Novel Test Structure for Process Control Monitor for Un-Cooled Bolometer Area Array Detector Technology

  • This paper presents the results of a novel test structure for process control monitor for uncooled IR detector technology of microbolometer arrays. The proposed test structure is based on resistive network configuration. The theoretical model for resistance of this network has been developed using 'Compensation' and 'Superposition' network theorems. The theoretical results of proposed resistive network have been verified by wired hardware testing as well as using an actual 16x16 networked bolometer array. The proposed structure uses simple two-level metal process and is easy to integrate with standard CMOS process line. The proposed structure can imitate the performance of actual fabricated version of area array closely and it uses only 32 pins instead of 512 using conventional method for a $16{\times}16$ array. Further, it has been demonstrated that the defective or faulty elements can be identified vividly using extraction matrix, whose values are quite similar(within the error of 0.1%), which verifies the algorithm in small variation case(${\sim}1%$ variation). For example, an element, intentionally damaged electrically, has been shown to have the difference magnitude much higher than rest of the elements(1.45 a.u. as compared to ${\sim}$ 0.25 a.u. of others), confirming that it is defective. Further, for the devices having non-uniformity ${\leq}$ 10%, both the actual non-uniformity and faults are predicted well. Finally, using our analysis, we have been able to grade(pass or fail) 60 actual devices based on quantitative estimation of non-uniformity ranging from < 5% to > 20%. Additionally, we have been able to identify the number of bad elements ranging from 0 to > 15 in above devices.

Optimization of Culture Conditions for the Bioconversion of Vitamin $D_3\;to\;1{\alpha}$,25-Dihydroxyvitamin $D_3$ Using Pseudonocardia autotrophica ID9302

  • We assessed the ability of a Pseudonocardia sp. from soil samples to bioconvert vitamin $D_3$. The optimal culture conditions for the bioconversion of vitamin $D_3$ to active $1{\alpha}$,25-dihydroxyvitamin $D_3$ were investigated by varying the carbon and nitrogen sources, the metal salt concentrations, the initial pH, and the temperature. Microbial transformations were carried out with the addition of vitamin $D_3$ dissolved in ethanol. They were sampled by extraction with methanol-dichloromethane and the samples were examined by HPLC. Optimum culture conditions were found to be 0.4% yeast extract, 1% glucose, 3% starch, 1% fish meal, 0.2% NaCl, 0.01% $K_2HPO_4$, 0.2% $CaCO_3$, 0.01% NaF, and pH 7.0 at $28^{\circ}C$. The optimal timing of the addition of vitamin $D_3$ for the production of calcitriol by Pseudonocardia autotrophica ID9302 was concurrent with the inoculation of seed culture broth. Maximum calcitriol productivity and the yield of bioconversion reached a value of 10.4mg/L and 10.4% respectively on the 7th day in a 75L fementer jar under the above conditions.
