  • Many of the diseases that occur in a life being are either closely related to water, or they occur by loss or deterioration of water metabolism. There are six parts of study on this subject in ${\ulcorner}$Dongeubogam${\lrcorner}$. The parts are, the part of Jinaek the part of Dameum the part of Sobyeon the part of Bujong the part of Changman and the part of Seub. In these parts, it mentions loss of perspiration, abnormal urination, edema, abdominal dropsy, formation of abnormal body fluid and intrusion of dampness into the body and etc as the abnormal water metabolism. Loss of perspiration and urination is a process of eliminating the dampness in the body. Perspiration would be the abnormality of yanghwa[陽化]. Urination would be the loss of eumhwa[陰化]. Eum[飮] is the fluid accumulated in the body that failed to go through the process of Cihwa[氣化]. Dam[痰] is formed when the body fluid is heated by the smoking-fire. Meanwhile, the dampness occurs when the water penetrates into the bones, muscles and joints. Edema and abdominal dropsy are both outcomes of accumulated body fluid. Edema is the liquified body fluid congested on the surface or the peripheral ends of the body. Abdominal dropsy is congestion of fluid, that lost the characteristic of blood due to blood deterioration, in the abdominal part.

  • The aim of this study was to compare two formulations of film-coated pellets containing c1arithromycin after single oral dose study in healthy male volunteers. Two formulations with different coating polymers were prepared: formulation-1 (F-1) was prepared by incorporating three kinds of pH-dependent gradient-release coated pellets into capsules and formulation-2 (F-2) was prepared by coated with an insoluble semiosmotic film. Release profiles of filmcoated pellets were evaluated using paddle method under different conditions. Pharmacokinetic profiles of these formulations were obtained in three healthy male volunteers and compared to commercially available immediate release (IR) tablets. The relative bioavailability based on the $AUC_{0-24h}$ was found to be $96.2\%\;and\;58.7\%$ for F-1 and F-2 compared with IR, and the $T_{max}$ was delayed.

  • This study was carried out to investigate the effect of whole grape, grape pomace and grape juice intake on cadmium (Cd) metabolism during aging in thirteen-month-old Sprague-Dawley male rats. One hundred and twenty rats weighing 548.8 $\pm$ 4.3 g were assigned to eight groups according to body weight and were raised for 3, 5 or 7 months on diets containing 2 % (w/w) dried powders of three different parts of the grape (Campbell Early) and 0.02 % (w/w) CdC12. Food intake tended to decrease with aging, and body weight and epididymal fat pad (EFP) weights of Cd-exposed groups were lower than those of Cd-free groups. Cadmium accumulated in the blood and tissues and Cd concentration was the lowest in the pomace group among Cd-exposed animals. Metallothionein (MT) concentration in the tissues increased through Cd administration. Grape diets, especially grape pomace diets, were effective in decreasing Cd absorption in the tissues by increasing Cd excretion through feces. The intake of grape pomace alleviated the decrease in bone density caused by Cd administration and prevented a decrease in glomerular filtration rates (GFR) with aging. Among the parts of grape, grape pomace, which had highest content of dietary fiber and flavonoids, was the most effective. The results of this study suggest the possibility of using grape pomace as a functional food material, a prospect that previously has been discarded.

  • [Purpose] In the present pilot study, we aimed to investigate the effects of the Silverrobics exercise program, which is similar to aerobic dance, on the factors related to glucose metabolism and liver enzymes. [Methods] Eight elderly women with obesity and impaired fasting glucose participated in the Silverrobics exercise program (60 minutes per session for five times a week for 8 weeks). The program was conducted at 50-60% of the heart rate reserve at 1 to 2 weeks and at 60-80% of the heart rate reserve at 3 to 8 weeks. To verify the effect of this 8-week exercise program on glucose metabolism and liver enzymes, blood analysis at pre- and post-training was performed. [Results] After the Silverrobics exercise program, there were significant decreases in the glucose (p<0.05), glycated hemoglobin A1c (p<0.05), 1,5-anhydroglucitol (p<0.05), and insulin levels (p<0.01) and homeostatic model assessment of insulin resistance score (p<0.05). However, there were no significant effects on the liver enzymes, except for alkaline phosphatase. The alkaline phosphatase level increased after the Silverrobics exercise program (p<0.05). [Conclusion] Although the Silverrobics exercise program had no beneficial effects on the liver enzymes, it may play an important role in preventing liver diseases considering the effects on glucose metabolism.

  • This study examined the effects of nicotinamide on proliferation, differentiation, and energy metabolism in a primary culture of bovine adipocytes. After treatment of cells with 100-500 $\mu{M}$ nicotinamide, cell growth was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), and cellular lipid content was assessed by Oil Red O staining and a triglyceride (TG) assay. Several factors related to energy metabolism, namely adenosine triphosphatase (ATPase) activity, nitric oxide (NO) content, nitric oxide synthase (NOS) activity, the number of mitochondria and the relative expression of glyceraldehydes-3-phosphate dehydrogenase (GAPDH), peroxisome proliferator-activated receptor-$\gamma$ ($PPAR_{\gamma}$) and inducible NOS (iNOS), were also investigated. Results showed that nicotinamide induced both proliferation and differentiation in bovine preadipocytes. Nicotinamide decreased NO production by inhibiting NOS activity and iNOS mRNA expression, and controlled lipolytic activity by increasing ATPase activity and the number of mitochondria. The present study provides further evidence of the effects of nicotinamide on lipid and energy metabolism, and suggests that nicotinamide may play an important role in the development of bovine adipose tissue in vivo. This emphasizes the importance of investigating bovine adipose tissue to improve our understanding of dairy cow physiology.

  • This study investigated the effects of xylooligosaccharide on feces bifidobacteria proliferation, lactic acid concentration and lipid metabolism in healthy woman. Fourteen volunteers were randomly assigned to 2 groups : 1.4 g/day xylooligosaccharide intake group, 2.8 g/day xylooligosaccharide intake group. The duration of the study was 28 days. The amount of feces and excretion time were not affected by xylooligosaccharide intake. The color of feces changed to yellow brown, and hardness of stool and effort to evacuation were reduced by xylooligosaccharide intake. Xylooligosaccharide intake reduced the fecal pH significantly after 14 days in 2.8 g/day intake group (p < 0.05). The number of fecal bifidobacteria were significantly increased after 28 days in 1.4 g/day intake group (p < 0.05), and in 2.8 g/day intake group, the number of fecal bifidobacteria significantly increased after 14 days (p < 0.05). Water contents of feces were not affected by xyloolgosacchride intake. The fecal triglyceride and cholesterol concentrations were increased in 2.8 g/day intake group (p < 0.05), and in 1.4 g/day intake group, fecal cholesterol concentration only was increased (p < 0.05). The fecal lactic acid concentration was significantly increased in 2.8 g/day intake group (p < 0.05). Serum trigly-ceride, cholesterol and glucose concentration were significantly decreased in 2.8 g/day intake group (p < 0.05). In conclusion, xylooligosaccharide dietary supplementation may be beneficial to gastrointestinal health and lipid metabolism, and 2.8 g/day intake was more effective than 1.4 g/day intake.

  • The effect of cardiopulmonary bypass (CPB) on cerebral physiology during heart surgery remains incompletely understood. This study was carried out to investigate changes of cerebral metabolism and the association between the changes and clinical factors during heart surgery. Seventy adult patients (n=70) scheduled for elective cardiac surgery were participated in the present study. Middle cerebral artery blood flow velocity (V$_{MCA}$), cerebral arteriovenous oxygen content difference (C(a-v)O$_2$), cerebral oxygen extraction (COE), and modified cerebral metabolic rate for oxygen (MCMRO$_2$) were measured during six phases of the operation; Pre-CPB, CPB-10 min, Rewarm-1 (nasopharyngeal temperature 34$^{\circ}C$), Rewarm-2 (nasopharyngeal temperature 37$^{\circ}C$), CPB-off, and Post-OP (at skin closure after CPB-off). Each relationship of age, arterial blood gas parameters, or other variables to V$_{MCA}2$, C(a-v)O$_2$, COE, or MCMRO$_2$ was evaluated. V$_{MCA}$ increased (P<0.0001) whereas C(a-v)O$_2$ decreased (P<0.01) throughout the five phases of the operation compared to Pre-CPB value (control). COE diminished at CPB-10, Rewarm-1, and CPB-off (P<0.05) while MCMRO$_2$ reduced at CPB-10 and Rewarm-1 (P<0.05) compared to Pre-CPB value. Positive correlation was found between age and cerebral metabolic parameters (V$_{MCA}$, C(a-v)O$_2$, COE, or MCMRO$_2$) during CPB (range r=0.24 to 0.38, p<0.05). Four cerebral metabolic parameters had partially negative or positive correlation with arterial blood gas parameters and other variables (arterial blood pH, $O_2$ tension, $O_2$ content, $CO_2$ tension, blood pressure, blood flow, temperature, or hematocrit) during the operation. In conclusion, CPB led to marked alterations of cerebral metabolism and age, pH, and $CO_2$ tension profoundly influenced the changes during cardiac surgery.

  • The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. • Objective : The rat middle cerebral artery thread-occlusion model has been widely used to investigate the pathophysiological mechanisms of stroke and to develop therapeutic treatment. This study was conducted to analyze energy metabolism, apoptotic signal pathways, and genetic changes in the hippocampus of the ischemic rat brain. Methods : Focal transient cerebral ischemia was induced by obstructing the middle cerebral artery for two hours. After 24 hours, the induction of ischemia was confirmed by the measurement of infarct size using 2,3,5-triphenyltetrazolium chloride staining. A cDNA microarray assay was performed after isolating the hippocampus, and was used to examine changes in genetic expression patterns. Results : According to the cDNA microarray analysis, a total of 1,882 and 2,237 genes showed more than a 2-fold increase and more than a 2-fold decrease, respectively. When the genes were classified according to signal pathways, genes related with oxidative phosphorylation were found most frequently. There are several apoptotic genes that are known to be expressed during ischemic brain damage, including Akt2 and Tnfrsf1a. In this study, the expression of these genes was observed to increase by more than 2-fold. As energy metabolism related genes grew, ischemic brain damage was affected, and the expression of important genes related to apoptosis was increased/decreased.Conclusion : Our analysis revealed a significant change in the expression of energy metabolism related genes (Atp6v0d1, Atp5g2, etc.) in the hippocampus of the ischemic rat brain. Based on this data, we feel these genes have the potential to be target genes used for the development of therapeutic agents for ischemic stroke.

  • The dietary fiber alginic acid has no nutritional value; however, it decreases the utilization of nutrients by the body and can prevent obesity, hyperlipidemia, arteriosclerosis, constipation, and colon cancer. The low-molecular-weight alginic acid polymannuronate improves serum and liver lipid metabolism by decreasing cholesterol levels in high-cholesterol groups and may be used to control high blood pressure. Previously, we showed that polymannuronate, a physiologically active agent from seaweed, has a lipid-lowering effect and preventative role in colon cancer. In addition, the differentiation of polymannuronate-treated adipocytes was inhibited, triggering decreased leptin expression. This study examined ways to increase dietary satisfaction with and improve the nutritional quality of polymannuronate using hamburger buns supplemented with polymannuronate. Twenty male Sprague-Dawley rats were divided into two groups: the control group was fed hamburger buns containing 6% gluten, while the experimental group was fed hamburger buns containing 6% gluten and 10% polymannuronate. The serum triglyceride, phospholipid, and total and free cholesterol levels of the rats in the experimental group were decreased compared with those of the controls. The serum aspartate aminotransferase activity levels did not differ between the two groups. This study demonstrates that polymannuronate has beneficial effects on lipid metabolism and may be commercially useful.
