Capsaicin Ameliorates Cisplatin-Induced Renal Injury through Induction of Heme Oxygenase-1

  • Cisplatin is one of the most potent chemotherapy agents. However, its use is limited due to its toxicity in normal tissues, including the kidney and ear. In particular, nephrotoxicity induced by cisplatin is closely associated with oxidative stress and inflammation. Heme oxygenase-1(HO-1), the rate-limiting enzyme in the heme metabolism, has been implicated in a various cellular processes, such as inflammatory injury and anti-oxidant/oxidant homeostasis. Capsaicin is reported to have therapeutic potential in cisplatin-induced renal failures. However, the mechanisms underlying its protective effects on cisplatin-induced nephrotoxicity remain largely unknown. Herein, we demonstrated that administration of capsaicin ameliorates cisplatin-induced renal dysfunction by assessing the levels of serum creatinine and blood urea nitrogen (BUN) as well as tissue histology. In addition, capsaicin treatment attenuates the expression of inflammatory mediators and oxidative stress markers for renal damage. We also found that capsaicin induces HO-1 expression in kidney tissues and HK-2 cells. Notably, the protective effects of capsaicin were completely abrogated by treatment with either the HO inhibitor ZnPP IX or HO-1 knockdown in HK-2 cells. These results suggest that capsaicin has protective effects against cisplatin-induced renal dysfunction through induction of HO-1 as well as inhibition oxidative stress and inflammation.

  • Objectives:This study was designed to investigate the effects of herbal composition and fermented Cervi Pantotrichum Cornu on the growth of longitudinal bone in the adolescent male rats. Methods: Longitudinal bone growth was measured by fluorescence microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate and the induction of local insulin-like growth factor-1 (IGF-1) and bone morphogenetic protein-2 (BMP-2) were measured. Results: The herbal composition treatment enhanced longitudinal bone growth and total heght of growth plate and promoted the induction of local IGF-1 and BMP-2 of growth plate. Conclusions: This study showed that the herbal composition have the effect of bone growth in adolescent rats and might be used for the growth delayed adolescent and inherent growth failure patient.

Methods: Longitudinal bone growth was measured by fluorescent microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate, the induction of local insulin-like growth factor-1 (IGF-1), and bone morphogenetic protein-2 (BMP-2) were measured. Results: Horse bone powder extract enhanced longitudinal bone growth and total height of the growth plate. Also, it promoted the induction of local IGF-1 and BMP-2 of the growth plate. Conclusions: This study shows that the horse bone powder extract effects longitudinal bone growth in adolescent rats and might be used for both stunted adolescents and inherent growth failure patients.

Methods : Longitudinal bone growth was measured by fluorescent microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate, the induction of local insulin-like growth factor-1 (IGF-1), IGF-1 receptor, bone morphogenetic protein-2 (BMP-2), BMPR-1A, indian hedgehog (IHH), and parathyroid hormone-related protein (PTH-rP) were measured. Results : Cheunggyeongsamul-tang extract enhanced longitudinal bone growth and total height of the growth plate. Also, it promoted the induction of local IGF-1, BMP-2, IHH and PTH-rP of the growth plate. Conclusions : This study shows that the Cheunggyeongsamul-tang extract effects longitudinal bone growth in adolescent rats and might be used for both stunted adolescents and inherent growth failure patients.

Single-cell RNA sequencing reveals the heterogeneity of adipose tissue-derived mesenchymal stem cells under chondrogenic induction

  • This study investigated how adipose tissue-derived mesenchymal stem cells (AT-MSCs) respond to chondrogenic induction using droplet-based single-cell RNA sequencing (scRNA-seq). We analyzed 37,219 high-quality transcripts from control cells and cells induced for 1 week (1W) and 2 weeks (2W). Four distinct cell clusters (0-3), undetectable by bulk analysis, exhibited varying proportions. Cluster 1 dominated in control and 1W cells, whereas clusters (3, 2, and 0) exclusively dominated in control, 1W, and 2W cells, respectively. Furthermore, heterogeneous chondrogenic markers expression within clusters emerged. Gene ontology (GO) enrichment analysis of differentially expressed genes unveiled cluster-specific variations in key biological processes (BP): (1) Cluster 1 exhibited up-regulation of GO-BP terms related to ribosome biogenesis and translational control, crucial for maintaining stem cell properties and homeostasis; (2) Additionally, cluster 1 showed up-regulation of GO-BP terms associated with mitochondrial oxidative metabolism; (3) Cluster 3 displayed up-regulation of GO-BP terms related to cell proliferation; (4) Clusters 0 and 2 demonstrated similar up-regulation of GO-BP terms linked to collagen fibril organization and supramolecular fiber organization. However, only cluster 0 showed a significant decrease in GO-BP terms related to ribosome production, implying a potential correlation between ribosome regulation and the differentiation stages of AT-MSCs. Overall, our findings highlight heterogeneous cell clusters with varying balances between proliferation and differentiation before, and after, chondrogenic stimulation. This provides enhanced insights into the single-cell dynamics of AT-MSCs during chondrogenic differentiation.

Effect of the Repeated Treatment of Xylene to the Rats on the Xylene Metabolism (흰쥐에 Xylene반복 투여가 Xylene의 대사에 미치는 영향)

  • To evaluate the effect of repeated treatment of xylene on its metabolism, m-xylene (0.25 ml of 50% in olive oi1/100 g body weight) has been intraperitoneally given to the rats 1, 4, 8, 12 and 16 times every other day. m-Xylene was once more administered to the animals after 24 hrs since last injection of it. And then the animals were sacrificed after 24 hrs. Four times xylene treated rats showed the significantly elevated urinary m-methylhippuric acid, compared to those treated with the single dose of m-xylene with the continued similiar high levels of urinary m-methylhippuric acid up to the animals pretreated 12 times and then those treated 16 times defined the significantly decreased urinary m-methylhippuric acid compared to those treated 12 times. On the other hand, hepatic aniline hydroxylase and alcohol dehydrogenase activities demonstrated a gradual increase from the first group to the 12 times xylene-treated animals, but those treated 16 times showed the significantly decreased value compared with the 12 times treated-group. And aldehyde dehydrogenase activities in rats treated with m-xylene 8, 12 or 16 times were significantly decreased compared to those pretreated one or four times. In the early stage of xylene administration, proliferation of SERs were seen whereas SERs were decreased and RERs were clearly increased in xylene-treated rats 16 times. These results indicate that the frequency of xylene injection may influence upon the changes in xylene metabolite, m-methylhippuric acid and it may be due to induction of xylene metabolizing enzymes.

Effects of Ephedra Herba ethyl-acetate fraction on high fat diet induced hyperlipidemic mice (마황 에틸아세테이트 분획물이 고지방 식이로 유발된 생쥐의 지질대사에 미치는 영향)

  • Objective : Ephedra Herba has been widely used for patients with common cold, asthma in eastern countries, especially china japan and korea. Recently it has been also used for obesity in clinic with high frequency. This study was designed to investigate the effects of Ephedra Herba ethyl-acetate fraction (EEAF) on hyperlipidemic mice. Method : Effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured in this experiment, and in addition, histopathological and gene expression changes in liver tissue was also observed. Results : In our study, EEAF did not affect weight gain in hyperlipidemic mice. Oral administration of EEAF lowered levels of total cholesterol which were elevated by induction of hyperlipidemia. And administration of EEAF lowered fasting blood glucose significantly. By carrying out ontological analysis, large numbers of genes were identified in up or down regulated genes. The expression of the genes that were altered in response to high-fat diet was restored to normal levels in EEAF treated mice, with a recovery rate of 49%. And it was considered that fatty acid metabolism was one of important key pathway of the recovery. Conclusion : Results in our study suggest that EEAF can prevent obese and through regulation of dyslipidemia and hyperglycaemia.

Effects of Paraquat on Ascorbic Acid and Malondialdehyde Contents, and Superoxide Dismutase Activity in Spinach Chloroplasts under Light and Dark

  • The amounts of ascorbic acid in chloroplasts treated with light and light+paraquat (PQ) were reduced by 81 and 82% of initial level, respectively at 24 hr at incubation. And those treated with dark and dark+PQ were decreased by W and 55% of the original level, respectively. Malondialdehyde (MDA) contents at 24 hr of dark and dark+PQ treatment were increased by 6 and 31% of the initial level, respectively. When chloroplasts were treated with light and light+PQ, MDA contents after 24 hr were increased by 88 and 146% of the initial level, respectively. SOD activities treated with light and light+PQ were increased by 10 and 20% of the initial level, respectively for 3 hr and thereafter reduced by 46 and 49% of the original level, respectively at 24 hr. However, the SOD activities treated with dark and dark+PQ were decreased by 37 and 30% of the initial level, respectively. It is considered that PQ triggers the oxidation of ascorbic acid, the induction of lipid peroxidation and the inactivation of SOD under light so that PQ has inhibitors effect on the pathway of plant metabolism. Decreased oral bioavailability of cyclosporin A at second administration in human

  • Sandimmune Neoral$\^$(R)/ and Neoplanta$\^$(R)/ capsules were administered to twenty four healthy Korean male subjects at a cyclosporin A (CsA) dose of 175 mg in a 2 ${\times}$ 2 crossover investigation with a two-week wash-out phase. Concentrations of CsA in blood were measured by RIA method for over a period of 48 h. Result : The two formulations were found bioequivalent, but analysis of variance (ANOVA) indicated that there is a significant (p<0.01) period effect in AUC$\_$0-LAST/ (area under the blood concentration above assay limit of quantification-time curve) and C$\_$MAX/ (maximum blood concentration) between the administrations. Paired t-test revealed 6 and 9% decreases in AUC$\_$0-LAST/ and C$\_$MAX/, respectively at the second administration. This period effect on the pharmacokinetics of CsA may be relevant for the patients who need consecutive administration of the drug. A number of mechanisms, such as induction of the enzymes responsible for metabolism of the drug in the gut wall and/or liver and modulation of P-glycoprotein upon the consecutive dosing, appear consistent with the change, and needs experimental proof.

Cloning and Expression of Serratia marcescens Coenzyme A(CoA) Transferase Gene in E. coli

  • We have got several clones from Serratia marcescens which stimulated the cells to use maltose as a carbon source in E. coli TP2139 (${\Delta}$lac, ${\Delta}$crp). One of the cloned genes, pCKB13, was further analyzed. In order to find whether the increased expression of the gene under the direction of maltose metabolism, we constructed several recombinant subclones. We have confirmed that the clone, pCKB13 codes Coenzyme A transferase gene by partial nucleotide sequencing in the terminal region. The enzyme activity of Coenzyme A transferase increased after introduction of the multicopy of the cloned gene in E. coli. The recombinant proteins expressed by multicopy and induction with IPTG, two polypeptide of 26-and 28-kDa, were confirmed by SDS-PAGE. Southern hybridization analysis confirmed that the cloned DNA fragment was originated from S. marcescens chromosomal DNA.
