Objective: This study was undertaken to investigate how magnolia bark extract affects ob/ob mouse in terms of metabolic inflammation and insulin resistance. Methods: Leptin-deficient ob/ob mice were divided into 2 groups (n=5): a normal saline treatment (=control) and magnolia bark treatment. Wild type mice were the lean group (n=5). After 5 weeks, we measured fasting blood sugar (FBS) and conducted oral glucose tolerance tests (OGTTs) in each group. After 6 weeks, we measured body weight, epididymal fat pad weight, liver weight, serum glucose, serum insulin, and gene expression of tumor necrosis factor-${\alpha}$, interferon-${\gamma}$, and interleukin-6. We characterized the phenotype of adipose tissue macrophages (ATMs) and analyzed fractions of the phenotype in each group by flow cytometry. Results: In the magnolia bark group, fasting blood sugar, oral glucose tolerance levels, and insulin resistance (HOMA-IR) were significantly decreased. The population and proportion of ATMs among leukocytes in adipose tissue were significantly decreased in the magnolia bark group. The population and proportion of M1 type ATMs among ATMs were significantly decreased in the magnolia bark group. Gene expression of tumor necrosis factor-${\alpha}$ was significantly decreased in the magnolia bark group. Conclusions: These results support a positive effect of magnolia bark on metabolic diseases such as insulin resistance and metabolic inflammation in leptin-deficient ob/ob mice.

Oriental herbal medicines were examined for mutagenicity in the reverse mutation test on Salmonella typimurium T A98/100 and chromosomal aberration test on cultured mammalian cells (Chinese hamster cell lines). The reverse mutation test was performed by a plate incorporation method with and without a metabolic activation system (S9 mix). The tested herbal medicines did not significantly increase revertible colonies on any of the test strains with and without a metabolic activation system (S9 mix) at concentrations of 1 mg/ml. In the chromosomal aberration test, most tested herbal medicines did not significantly increase the number of aberrant cells on any of the test strains with a metabolic activation system (S9 mix) at concentrations of 1 mg/ml, compared with the vehicle control. However. Ansu Semen significantly increased the number of aberrant cells without a metabolic activation system (S9 mix). Paeoniae Radix. Hoelen, Aurantii nobilis Pericarpium, Cnidii Rhizoma, Angeliacae gigantis Radix, Perillae Herba and Moutan Cortex Radicis slightly increased revertible colonies on any of the test strains with a metabolic activation system (S9 mix), These results indicate that most herbal medicines might be carefully used in obstetrics and gynecology, although they do not have the potent mutagenic potential under the present test conditions.
1. Objectives The purpose of this study was to investigate the correlation of Metabolic syndrome(MetS) and Intima-media thickness(IMT) of common carotid artery with Sasang constitution. 2. Methods 197 subjects who had taken health examinations and diagnosis of Sasang constitution from Jan 1, 2006 to Dec 31, 2006 at East-west health examination center of Kyung-Hee medical center were included and underwent B-mode ultrasonography for measurement of intima-media thickness of common carotid arteries. MetS was defined by the criteria of the criteria of the National Cholesterol Education Program Adult Treatment Panel III. 3. Results and Conclusions Mean intima-media thickness of common carotid artery was significantly higher in subjects with MetS compared with control subjects($0.070{\pm}0.017$ vs $0.063{\pm}0.014cm$) and a significant trend towards increased IMT was observed with increasing numbers of metabolic syndrome components(r=0.331, p=0.000). After adjustment for age and sex, IMT of common carotid arteries was significantly different among three constitutional groups (Soyangin:$0.065{\pm}0.015cm$, Taeumin:$0.068{\pm}0.016cm$, Soeumin:$0.056{\pm}0.010cm$), however, after adjustment for age, sex and existence of metabolic syndrome, there was no significant different. But, factor of Taeumin was the effective determinant of the increase of mean IMT of common carotid arteries.

Autonomous ultradian metabolic oscillation (T$\simeq$50 min) was detected in an aerobic chemostat culture of Saccharomyces cerevisiae. A pulse injection of GSH (a reduced form of glutathione) into the culture induced a perturbation in metabolic oscillation, with respiratory inhibition caused by $H_2S$ burst pro-duction. As the production of $H_2S$ in the culture was controlled by different amino acids, we attempted to characterize the effects of GSH on amino acid metabolism, particularly with regard to branched chain and sulfur-containing amino acids. During stable metabolic oscillation, concentrations of intra-cellular glutamate, aspartate, threonine, valine, leucine, isoleucine, and cysteine were observed to oscil-late with the same periods of dissolved $O_2$ oscillation, although the oscillation amplitudes and maximal phases were shown to differ. The methionine concentration was stably maintained at 0.05 mM. When GSH (100 $\mu$M) was injected into the culture, cellular levels of branched chain amino acids increased dramatically with continuous $H_2S$production, whereas the cysteine and methionine concentrations were noticeably reduced. These results indicate that GSH-dependent perturbation occurs as the result of the promotion of branched chain amino acid synthesis and an attenuation of cysteine and methionine synthesis, both of which activate the generation of $H_2S$. In a low sulfate medium containing 2.5 mM sulfate, the GSH injections did not result in perturbations of dissolved $O_2$ NAD(P)H redox oscillations without burst $H_2S$ production. This suggests that GSH-dependent perturbation is intimately linked with the metabolism of branched-chain amino acids and $H_2S$ generation, rather than with direct GSH-GSSG redox control.

Overconsumption of fructose results in dyslipidemia, hypertension, which have documented as a risk of cardiovascular diseases. This experimental study was designed to investigate the beneficial effects of Rubus coreanus Miq.(RCM) in high-fructose diet-induced metabolic syndrome. Animals were divided into three groups; Control group fed regular diet and tap water, fructose groups were fed the 65% high-fructose (HF) diet with/without RCM 100 mg/kg/day for 8 weeks, respectively. Chronic treatment with RCM significantly decreased body weight, fat weight and adipocyte size. Moreover, RCM significantly prevented the development of the metabolic disturbances such as hyperlipidemia and hypertension. RCM also led to increase in high density lipoprotein level in the HF group. In addition, RCM suppressed vascular cell adhesion molecule-1 (VCAM-1) expression and significantly recovered the levels of endothelial nitric oxide synthase (eNOS) expression in aorta. These results demonstrates that RCM may be a beneficial therapeutic for metabolic syndrome through the improvement of hyperlipidemia, obesity, and hypertension.
BACKGROUND/OBJECTIVES: Several previous studies have investigated whether regular walnut consumption positively changes heart-health-related parameters. The aim of this study was to investigate the effects of daily walnut intake on metabolic syndrome (MetS) status and other metabolic parameters among subjects with MetS. SUBJECTS/METHODS: This study was a two-arm, randomized, controlled crossover study with 16 weeks of each intervention (45 g of walnuts or iso-caloric white bread) with a 6 week washout period between interventions. Korean adults with MetS (n = 119) were randomly assigned to one of two sequences; 84 subjects completed the trial. At each clinic visit (at 0, 16, 22, and 38 weeks), MetS components, metabolic parameters including lipid profile, hemoglobin A1c (HbA1c), adiponectin, leptin, and apolipoprotein B, as well as anthropometric and bioimpedance data were obtained. RESULTS: Daily walnut consumption for 16 weeks improved MetS status, resulting in 28.6%-52.8% reversion rates for individual MetS components and 51.2% of participants with MetS at baseline reverted to a normal status after the walnut intervention. Significant improvements after walnut intake, compared to control intervention, in high-density lipoprotein cholesterol (HDL-C) (P = 0.028), fasting glucose (P = 0.013), HbA1c (P = 0.021), and adiponectin (P = 0.019) were observed after adjustment for gender, age, body mass index, and sequence using a linear mixed model. CONCLUSION: A dietary supplement of 45 g of walnuts for 16 weeks favorably changed MetS status by increasing the concentration of HDL-C and decreasing fasting glucose level. Furthermore, consuming walnuts on a daily basis changed HbA1c and circulating adiponectin levels among the subjects with MetS. This trial is registered at ClinicalTrials.gov as NCT03267901.
This study assessed the amount of energy consumed and fat deposition after endurance training in order to review the effect of 4-week endurance exercise on resting metabolic rate of a mouse during and after exercise and the effect of exercise. A total of 19 seven-week-old ICR male mice were used as the study subject. Those mice were divided into sedentary group (Sed) and trained group (Tr) after a week of environment adaption. The Tr group was trained with endurance exercise five times a week for four weeks. Weight and the amount of food intake were daily weighed and resting metabolic rate and metabolic rate after exercise were assessed before starting exercise and on the fourth week after training. Metabolic rate during exercise were measured four weeks after training. At the end of breeding period, statistically significant difference was shown in weights of trained and sedentary groups (p < 0.05). During a resting period, no significant difference was shown in oxygen intake, respiratory exchange ratio, and the amount of carbohydrate and fat oxidized. Moreover, no significant difference was shown in excess post-exercise oxygen consumption (EPOC) of an hour period after training. In contrast, the maximal oxygen uptake (VO2 max) was approximately 11.1% higher in trained group after training compare to before. However, there was no significant difference in respiratory exchange ratio and carbohydrate and fat oxidization. During exercise, oxygen uptake, carbon dioxide production, and respiratory exchange ratio in energy metabolism during exercise showed no significant difference. However, significant difference was exhibited in the amount of fat oxidized in both groups. Summing up those results, endurance exercise could be concluded to be effective in weight control. However, weight loss is thought to be resulted from increase in fat oxidization during exercise unlike the conclusion made from previous studies where weight loss is prominently influenced by energy metabolism during a resting period and increased fat oxidation during post-exercise recovery. All experimental procedures were carried out at the Animal Experiment Research Center of Konkuk University. This study was conducted in accordance with the ethical guidelines of the Konkuk University Institutional Animal Care and Use Committee.
With the recent rapid increase in obesity worldwide, metabolic syndrome (MetS) has gained significant importance. MetS is a cluster of obesity-related cardiovascular risk factors including abdominal obesity, atherogenic dyslipidemia, high blood pressure and impaired glucose tolerance. MetS is highly prevalent and strongly associated with an increased risk of developing diabetes and cardiovascular disease, putting a great burden on human society. Therefore, it is very important to reduce MetS risk, which can improve patients' cardiovascular prognosis. The primary and most effective strategy to control each component of MetS is lifestyle change such as losing body weight, keeping regular exercise, adopting a healthy diet, quitting smoking and alcohol drinking in moderation. Many studies have shown that lifestyle modification has improved all components of MetS, and reduces the incidence of diabetes and cardiovascular disease. Here, the Korean Society of CardioMetabolic Syndrome has summarized specific and practical methods of lifestyle modification in the management of MetS in the healthcare field.

Objectives: This study was conducted to evaluate the effects of a nutrition education program on metabolic syndrome in middle-aged Korean adults. Methods: A total of 411 Korean adults 30-59 years of age were allocated randomly into three groups: the nutrition education group for promoting Han-sik consumption (HG), the nutrition education group for eating balanced diet (EG), and the control group (CG). The HG and EG received four face-to-face nutrition education sessions over 16 weeks to improve nutritional problems based on the individual's usual diet. Effectiveness of the program was evaluated with the differences of self-reported dietary behaviors, dietary intakes, anthropometric measurements and biochemical indices between the baseline and the end of the nutrition education program. The changes within groups were analyzed using paired t-test and McNemar test and effectiveness among three groups was analyzed by repeated analysis of variance. Results: After the nutrition education, the percentages of participants who achieved the recommended food group consumption in the Korean Food Guidance Systems significantly increased in HG (P = 0.022). Body weight (P = 0.007), body mass index (P = 0.002), and triglycerides (P = 0.002) significantly decreased in HG. Waist circumference and diastolic blood pressure decreased in all three groups (P < 0.05). Conclusions: This study found that tailored nutrition education program for middle aged Korean adults showed beneficial effects on improving dietary behaviors and metabolic syndrome risk factors. Further studies are needed to assess the long-term effects of the nutrition education programs on metabolic syndrome risks.
Objectives: We wanted to determine the distribution of the clustering of the metabolic risk factors and we wanted to evaluate the related factors in young schoolchildren. Methods: A cross-sectional study of metabolic syndrome was conducted in an elementary school in Seoul, Korea. We evaluated fasting glucose, triglyceride, HDL cholesterol, blood pressures and the body mass index, and we used parent-reported questionnaires to assess the potential risk factors in 261 children (136 boys, 125 girls). We defined the metabolic risk factors as obesity or at risk for obesity ($\geqq$ 85th percentile for age and gender), a systolic or diastolic blood pressure at $\geqq90th$ percentile for age and gender, fasting glucose at $\geqq110mg/dl$, triglyceride at $\geqq110mg/dl$ and HDL cholesterol at $\leqq40mg/dl$. Results: There were 15.7% of the subjects who showed clustering of two or more metabolic risk factors, 2.3% of the subjects who showed clustering for three or more risk factors, and 0.8% of the subjects who showed clustering for four or more risk factors. A multivariate analysis revealed that a father smoking more than 20 cigarettes per day, a mother with a body mass index of = $25kg/m^2$, and the child eating precooked or frozen food more than once per day were associated with clustering of two or more components, with the odds ratios of 3.61 (95% CI=1.24-10.48), 5.50 (95% CI=1.39-21.73) and 8.04 (95% CI=1.67-38.81), respectively. Conclusions: This study shows that clustering of the metabolic risk factors is present in young schoolchildren in Korea, with the clustering being associated with parental smoking and obesity as well as the child's eating behavior. These results suggest that evaluation of metabolic risk factors and intervention for lifestyle factors may be needed in both young Korean children and their parents.
