To evaluate the renal toxicity of the antitumor agent, 1-(N-methyl) piperazinyl-3-phenyl-isoquinoline(CWJ-$\alpha$-5), rats were terated with CWJ-$\alpha$-5 (acute : 100mg/kg, i.p., single and subacute : 10mg/kr, i.p., daily for 7 days). The changes in the body weights, water consumption, kidney weights and urine volume after and during the treatment were observed. The concentrations of urinary creatinine, the activities of N-acetyl-$\beta$-D-glucosaminidase (NAG), alanine aminopeptidase (AAP), $\gamma$-glutamyl transpeptidase ($\gamma$-GT) and lactate dehydrogenase (LDH) in 24 hr urine were also determined. The body weight and water consumption were decreased after the acute and subacute administration. However, the excretion of urine was not changed except the 1 day after the acute treatment. The excretion of creatinine was significantly decreased from 1 day after acute administration and continuously decreased. Also the excretion of creatinine was decreased during subacute administration. However, the protein excretion did not changed in both treatment. Those indicate that CWJ-$\alpha$-5 might decrease the metabolic rate of muscle. The urinary activities of NAG, AAP, $\gamma$-GT, and LDH were significantly affected by the drug treatment. The urinary activities of NAG, AAP and $\gamma$-GT were significantly increased 1 and 3 days after the acute administration and then returned to the control value. However, the urinary activities of LDH were increased 7 days after acute treatment. During subacute treatment, the urinary activities of $\gamma$-GT were not changed. However, the urinary activities of NAG, AAP and LDH were only significantly increased after the third administration. These results indicate that either the high acute dose or the subacute administration with low dose of the compound might induce a temporal damage in the kidney cells.

The isolated plantamajoside from Plantago asiatica that is often used as a marker compound in chemotaxonomic studies has various bioactivites such as the inhibitions of cyclic AMP phosphodi-esterase and 5-lipoxygenase, microbial growth and inflammation, and currently demands the generation of toxicity data. The purpose of this study was to examine the toxicities of the single and 14 days repeated dose toxicity in Sprague-Dawley rats orally administrated with plantamajoside at dose levels of 0, 500, 1000, and 2000 mg of dried material/kg body weight/day. The results showed that there was no difference in body weight change, food intake, water consumption, or relative organ weight among different dose groups. Also we observed no death and abnormal clinical signs were observed during the experimental period. Between the groups orally administered Plantago asiatica and the control group, there was no statistical significance in hematological test or serum biochemical values. There were no gross findings at final sacrifice. There was no evidence of histopathological alteration mediated by 14 days treatment with Plantago asiatica. These results suggest that no observed adverse effect level (NOAEL) of the oral application was considered to be more than 2000 mg/kg in rats under the conditions employed in this study. Another observation was performed to investigate the safety of Plantago asiatica in respect of genotoxicity. This substance was examined that Salmonella typhimurium reversion assay (Ames test) in strain TA98, TA100, TA1535. In the reverse mutation test, Plantago asiatica did not induce mutagenicity in Samonella typhimurium with and without metabolic activation. These results indicated that Plantago asiatica had no genotoxicity.

In this study, the researcher tried to present the groundwork to prepare the oriental medical yangseng plan for the future obesity with the comparison between yangseng level and the obesity of workers. The researcher made up the questionnaire which asks the general character, health-related character and yangseng level, projected among 560 people. All collected material was analyzed by SPSS and tested by T-test and ANOVA. 1. The general yangseng level average is 3.27, morality yangseng 3.90, sleep yangseng 3.39, mind yangseng 3.31, sex life yangseng 3.30, exercise yangseng 3.15, activities and rest yangseng 3.08, diet yangseng 2.94, seasonal yangseng 2.84. The highest is morality yangseng and seasonal yangseng is the lowest. 2. In the aspect of yangseng level: Having a spouse, Non-smoking, Regular exercising, Sufficient sleeping have higher yangseng levels. 3. Obesity related index is changing into the higher level when he/she is older, more paid, more job experience, more education background but less metabolic calory. And drinking and regular exercising have relativity with body composition analysis. 4. After comparing yangseng level with body composition analysis, we can easily find that the more visceral fat, the higher WHR has a high yangseng level. The more mineral also has a high mind yangseng and a low diet yangseng. The visceral fat level has a high yangseng level when higher morality yangseng and mind yangseng are getting higher and higher. And sex life yangseng shows that the highest yangseng level is from 9-10 visceral fat and the lowest yangseng level is from below 4 visceral fat. The higher WHR, the higher morality, mind and sleep yangseng. The heavier, the lower diet yangseng. The mind yangseng was very high when body fat rate was higher. The more body fat, the higher morality yangseng. The higher yangseng, when we have more muscle. Yangseng level and obesity of laborer has a close relationship with individual character and daily habits. Also, relevance can be easily found between yangseng level and obesity. Now the researcher came into the conclusion that we need to control over laborers' health and prevention of their obesity.
The hypoglycemic and metabolic effects of Commelina communis L. extract were investigated in alloxan induced diabetic rats. The increased blood glucose level in the diabetic rats was significantly reduced and the loss of body weight was recovered with the treatment of the plant protein fractions($30{\sim}70%$ ammonium sulfate precipitates). Administration of the plant protein fractions elicited the significant increase of glucose-6-phosphate dehydrogenase (G-6-P DH) activity and liver weight which were decreased in the diabetic rat liver. G-6-P DH was partially purified from extract- or insulin-treated diabetics, diabetic control, and normal rat liver and studied for the biochemical properties. The $K_m$ value(9.002 mM) of diabetic rat liver enzyme was greatly higher than that (0.033 mM) of normal enzyme indicating the affinity of enzyme for the substrate was significantly reduced in the diabetic rat liver. This reduced affinity of enzyme for the substrate in the diabetic rat was recovered in the extract- or insulin-treated rat liver enzyme having 0.164 or 0.208 mM of their $K_m$ values, respectively. Although there was no significant difference in the optimum pH(6.0) and optimum temperature($37^{\circ}C$) of enzyme among the experimental groups, the dependence of their activities on pH appeared to be slightly resistant in the extract- or insulin-treated group compared to the diabetic group. In order to investigate the antigenicity of rat liver enzyme among experimental groups, enzyme-linked immunosorbent assay was carried out by using anti-G-6-P DH anti-serum. Absorbance(0.102) shown in the normal rat liver was reduced even below zero in the alloxan-diabetic rat liver, but increased again in the extract- or insulin-treated rat liver(0.096 or 0.118, respectively). The result of this study suggested that G-6-P DH may be used as a marker enzyme to diagnose and to indicate the progress of the diabetics, and the hypoglycemic effect of the extracts of Commelina communis L. was certainly associated with action or mode of G-6-P DH on the rat liver.

An experiment was conducted to investigate the effect of dietary supplemental protease on performance, blood components, organ weight and protein digestibility. Two hundred and fifty, one day old cobb x cobb male broiler chicks were assigned to 4 treatments with 4 replicates. Four levels of protease were supplemented with 0, 40, 80 and 160U. Weight gain, feed intake, feed conversion were weekly measured for 5 weeks. Blood components and organ weight were examined at the end of experiment. Metabolic feeding trial was carried out to investigate the protein digestibility for one week at five weeks of age. Basal diets contained 21.5, 19.0% CP and 3,100, 3,200kca1/kg ME for starter and finisher, respectively. Weight gain of chicks fed 80U protease was significantly higher than other treatments for starting period(P〈0.05). Feed conversion of chicks fed protease addition diets was improved as dietary protease increased. It showed significant difference between 80U and control groups(P〈0.05). Sera protein tended to be lowered in protease added groups. HDL-cholesterol was decreased at the maximum level of protease. However, organ weight and protein digestibility were not influenced significantly by dietary protease level. In conclusion, the present study demonstrated that protease addition in broiler diets improved the feed conversion.

The purpose of this study was to investigate the possibility of Meles meles oil as an functional resource. To assess the effects of Meles meles oil in 25 non-insulin dependent diabetes mellitus (DM) persons, we examined changes of fat intake level, hematological and chemical variables, serum DM indices and lipid contents during the Meles meles oil supplementation. Polyunsaturated fatty acid and $\omega$3 fatty acid intake were significantly increased by Meles meles oil intakes. The levels of LDL-cholesterol and triglyceride were significantly decreased while HDL-cholesterol was significantly increased. Iron status improved during Meles meles oil intakes. These results show that modest dose of Meles meles oil supplementation can decrease serum triglyceride, cholesterol level without any changes in blood glucose level in NIDDM patients. These results indicated that Meles meles oil diet is effective therapeutic regimen for the control of metabolic derangements in diabetes mellitus. Also, these results imply that Meles meles oil can be used as possible food resources and functional food materials. However, large amounts of Meles meles oil should be used cautiously in NIDDM patients.

The in vivo digestibility of mulberry leaves (Morus alba) and the effects of the partial replacement of cotton seed with fresh mulberry leaf in rations on the performance of growing Vietnamese cattle was investigated. For the in vivo digestibility trial, twenty castrated rams of Phanrang breed (a local prolific breed) with an initial weight of 23-25 kg, were first assigned to four groups according to weight and then randomly assigned to one of four dietary treatments to determine digestibility of nutrients in mulberry leaves (M. alba), natural Bermuda grass (Cynodon dactylon), elephant grass (Pennisetum purpureum) and buffalo grass (Panicum maximum cv. TD 58). All forages were cut and chopped daily before being offered (at 120% maintenance) to the sheep. In the feeding trial, 20 Laisind (Vietnam yellow cows${\times}$Red Sindhy bulls) crossbred bulls averaged 18 month old and 184 kg were used to investigate the effect of partial replacement of cottonseed in the diet by mulberry leaves on live weight gain and feed conversion rate. The experiment was a randomized complete block design with four levels of fresh mulberry leaves which varied from 0 to 15% of total dietary dry mater and five animals per treatment over an 84 day period. The in vivo digestion trial showed the superior quality of mulberry leaves compared with the grasses. Chemical analysis indicated that mulberry leaves had the highest CP and the lowest NDF contents (22.3 and 31.1% DM, respectively) among the four forages tested. Digestibility of DM and OM of the mulberry leaf (66.4 and 71.8%, respectively) was also the highest but that of CP (58.2%) and NDF (58.4%) was the lowest of the four forages evaluated (p<0.05). Consequently, the ME value and therefore net energy (NE) and unit feed for lactation (UFL) values of the mulberry leaves, which was estimated from chemical composition and digestibility values, were the highest among the forages investigated in the present study. Results of the feeding trial showed no treatment effect on average daily gain (ADG) of the cattle. The values were 554, 583, 565 and 568 g/d for animals in the diets of 0, 5, 10, and 15% mulberry leaves inclusion, respectively. Total DM intake of the animal was not affected by the treatment when expressed as kg/animal/d. However, when adjusted for metabolic weight of the animal the DM intake was reduced (p<0.05) as whole cottonseed was replaced by mulberry leaves in the ration. When the level of mulberry leaves in the ration increased from 5 to 15% of dietary DM at the expense of whole cottonseed, CP and ME intakes of the cattle were significantly decreased (p<0.05) and the feed to gain ratio reduced by 8 to 14% as compared with the control diet (p<0.05). Mulberry leaf is a good feed ingredient for ruminants because of its high level of crude protein and high digestibility of nutrients and energy. Mulberry leaves can be efficiently used as a source of protein supplement to replace cottonseed, a more expensive animal feeds ingredient, in the diet for Vietnamese cattle.
This study was to determine genotoxicity from the extracts of fermented Acanthopanax koreanum. The bacterial reverse mutation assay, the extracts of fermented A. koreanum did not induce mutagenicity in Salmonella typhimurium TA98, TA100, TA1535, TA1537 and Escherichia coli WP2uvrA with or without metabolic activation of S-9 mixture. In addition, the micronucleus formation in ICR mice, the extracts of fermented A. koreanum treated with dose of 500, 1,000 and 2,000 mg/kg did not affected micronucleated polychromatic erythrocytes (MNPCE/2,000 PCE) and polychromatic erythrocytes (PCE)/200 polychromatic erythrocyte+normochromatic erythrocyte (RBC). The cytotoxicity effects using CHO-K1 cells observed no significant changes compared with negative control group (p < 0.05). Moreover, the extracts of fermented A. koreanum did not cause a significant chromosome aberration on CHO-K1 cells in the chromosome aberration assay. Therefore, these results suggest that the extracts of fermented A. koreanum did not induce any harmful genotoxic effects.

Purpose: Matrix metalloproteinases (MMPs) have been implicated in atherosclerosis, and therefore, are considered risk factors for metabolic dysfunction in adults. However, there is little data on circulating levels of MMPs and tissue inhibitors of MMPs (TIMPs) with regard to obesity-related biomarkers in the general adolescent population. In the present study, we determined the associations of MMP-8, MMP-9, and TIMP-1 levels and MMP-8/TIMP-1 and MMP-9/TIMP-1 ratios with obesity-related biomarkers in apparently healthy adolescent boys. Methods: We measured MMP and TIMP concentrations in plasma samples using the enzyme-linked immunosorbent assay and analyzed their associations with obesity-related biomarkers, such as liver enzymes and lipid profiles, in a sample of 91 Korean boys aged 13-14 years who participated in a general health check-up. Results: The mean age of the boys was $13.8{\pm}0.3years$; 72 boys were normal weight and 19 were overweight/obese. The Pearson correlation coefficients revealed a significant correlation between MMP-8 and aspartate aminotransferase (r=0.217, P=0.039) and alanine aminotransferase (r=0.250, P=0.017) and between TIMP-1 and aspartate aminotransferase (r=0.267, P=0.011). In a multivariate linear regression analysis, serum alanine aminotransferase was positively associated with the MMP-8 level. There were no significant differences in the MMP-8, MMP-9, and TIMP-1 levels or MMP-8/TIMP-1 and MMP-9/TIMP-1 ratios between control and overweight/obese subjects. Conclusion: We found a significant association between the MMP-8 level and alanine aminotransferase in the apparently healthy adolescent boys. These findings indicate that there may be a pathophysiological mechanism underlying the relationship between MMP-8 and liver enzymes in young adolescents.
Volatile organic compounds (VOCs) are increasingly been recognized as the chemical mediators of mold interactions, shaping their community dynamics, growth, and metabolism. Herein, we selectively examined the time-correlated (0 D-11 D, where D = incubation days) effects of intraspecies VOC-mediated interactions (VMI) on Aspergillus oryzae KCCM 60345 (S1), following co-cultivation with partner strain A. oryzae KACC 44967 (S2), in a specially designed twin plate assembly. The comparative evaluation of $S1_{VMI}$ (S1 subjected to VMI with S2) and its control ($S1_{Con}$) showed a notable disparity in their radial growth ($S1_{VMI}$ < $S1_{Con}$) at 5 D, protease activity ($S1_{VMI}$ > $S1_{Con}$) at 3-5 D, amylase activity ($S1_{VMI}$ < $S1_{Con}$) at 3-5 D, and antioxidant levels ($S1_{VMI}$ > $S1_{Con}$) at 3 D. Furthermore, we observed a distinct clustering pattern for gas chromatography-time of flight-mass spectrometry datasets from 5 D extracts of $S1_{VMI}$ and $S1_{Con}$ in principle component analysis (PC1: 30.85%; PC2: 10.31%) and partial least squares discriminant analysis (PLS-DA) (PLS1: 30.77; PLS2: 10.15%). Overall, 43 significantly discriminant metabolites were determined for engendering the metabolic variance based on the PLS-DA model (VIP > 0.7, p < 0.05). In general, a marked disparity in the relative abundance of amino acids ($S1_{VMI}$ > $S1_{Con}$) at 5 D, organic acids ($S1_{VMI}$ > $S1_{Con}$) at 5 D, and kojic acid ($S1_{VMI}$ < $S1_{Con}$) at 5-7 D were observed. Examining the headspace VOCs shared between S1 and S2 in the twin plate for 5 D incubated samples, we observed the relatively higher abundance of C-8 VOCs (1-octen-3-ol, (5Z)-octa-1,5-dien-3-ol, 3-octanone, 1-octen-3-ol acetate) having known semiochemical functions. The present study potentially illuminates the effects of VMI on commercially important A. oryzae's growth and biochemical phenotypes with subtle details of altered metabolomes.
