Cytological Studies of Diapause in the Silkworm, Bombyx mori L. (가잠의 휴면성에 관한 세포학적 연구)

  • In present study, radioautography and electron-microscopy were applied by the author to the elucidation of the mechanism of diapause in Bombyx mori L. (1). Patterns of nucleic acid and protein syntheses during embryonic development of silkworms, incubated at high and at low temperatures, were demonstrated by means of radioautography with labelled precursors of nucleic acid and protein. On the third day after blastokinesis the embryo incubated at high temperature generally incorporated much of the $^3$H-glycine into the brain and the suboesophageal ganglion, and some into other regions. When incubated at low temperature, no difference in the pattern between the brain, the suboesophageal ganglion and other parts was observed. Radioautography with $^3$H-thymidine revealed no significant difference in DNA synthesis in embryos incubated at high and low temperatures. In diapausing eggs twenty days after deposition, only a few cells of the mesoderm incorporated the labelled material into their nuclei, but in the hibernated eggs all the nuclei of the mesoderm had thymidine incorporation. After blastokinesis only the anterior portion of the embryo around the brain and the suboesophageal ganglion had thymidine incorporation; this was not observed in the posterior portion.

  • Dermoid cyst is a benign neoplasm, so called by teratoma that is derived from both ectoderm and mesoderm during embryonic period. The majority of dermoid cysts are found in the ovaries and approximately only 7% of dermoid cysts are found in the head and neck. The growth of dermoid cysts in the floor of the mouth is considered a rare condition. We report one case of huge dermoid cyst in the floor of mouth suffered with double chin appearance and difficulty in pronunciation and respiration. The cyst was excised successfully only intra-oral incision. Clinical progress was uneventful and postoperative recovery excellent without recurrence.

  • The malignant germ cell tumor found in the gonad can originate in the anterior mediastinum. Endodermal sinus tumor[Yolk sac tumor] is a kind of malignant germ cell tumor and is derived from extra-embryonic mesoderm. We experienced a case of primary mediastinal endodermal sinus tumor occurred in 22 year old male patient. His chief complaint was anterior chest pain for 2 days. The tumor located in the anterior mediastinum and invaded upper lobe of the deft lung and pericardium. A left upper lobe resection including phrenic nerve and pericardium was performed and the tumor in the anterior mediastinum was excised. The patient has been treated with combination chemotherapy[Cis-platinum, Vincristine, Actinomycin-D, & Cyclophosphamide} and followed up for 4 months with partial remission.

  • The skin(皮膚) separates epidermis(皮) and dermis(膚) in Oriental medicine. epidermis and hair come from ectoderm as dermis comes from mesoderm in embryology. Epidermis and hair belong to the Metal(金) and dermis belongs to the Earth(土) in Oriental medicine. The lung is related to the skin and hair. The skin surface is nourished by the essence from the lung, and the. lung has the function of sending wei-qi(衛氣.: defensive Energy) to the skin surface to guarantee body resistance. The skin takes nourishment from blood(血) and express by qi(氣). The supply of nutrients to the lung and skin relies on the function of the spleen, pancreas and subcutis. Flaming of the heart fire may scorch the essence of the lung-metal, inducing dermatosis.

  • Dermoid cysts are benign neoplasm that is derived from both ectoderm and mesoderm. They may be found at various sites of the body, but are extremely rare to occur in the lower neck. We present the case of a 22-year old woman with a midline cystic mass anterior to the cervical trachea. The mass was excised under local anesthesia. Histopathologically the diagnosis was confirmed as a dermoid cyst.

  • The trend of organogenesis in Xenopus presumptive ectoderm was studied by combined dose of activin A and IGF-1(insulin-like growth factor-1). In reference study of Asashima and his colleagues, the inductive patterns of various organs were reported with activin, the potent mesoderm inducing factor. In present study, the inducing pattern was cleared with combined-dose of concentration 1-100 ng/ml activin A and IGF-1. In addition, the result from single treatment of activin A was compared with former study. As a result, eye was differentiated in 5-20% of explants at 10 and 50 ng/ml concentrative combination of activin A. Otic vesicle was appeared in the entire concentrative combination of IGF-1. Pronephric duct was induced 19-38% of explants at the concentration of activin A 100 ng/ml by adding IGF-1. The comparison of single treatment of activin A was showed some difference in dose-dependent inducing pattern.

  • Fibroblast growth factors (FGFs) are known to induce multiple functions in early development, including mesoderm formation, gastrulation movement and antero-posterior patterning. The induction of mesoderm from Xenopus presumptive ectoderm and the combination effect on inducing organs of bFGF(basic FGF) and HGF (Hepatocyte Growth Factor) were studied. Explants were cultured in the combined solution for 3 days to normal embryo arrive at St. 43. These effects on combined dose were examined by histological experiment and by immunohistochemical method. The concentrations of growth factors were tested in 0, 0.5, 1, 10 and also tested in 50 ng/ml of bFGF, and 0, 1, 10, 50 and 100ng/ml of HGF respectively. The synergistic effects were seen in the combined-dose of bFGF and HGF rather than in single dose. Various organs were differentiated and highest inducing effects were seen at the combined concentration of 1 ng/ml of bFGF and 10ng/ml of HGF, and at the concentration 10ng/ml of bFGF and 1 ng/ml of HGF. The bFGF induces various organs from cultured animal cap explants and the effects are time and dose-dependent. HGF is also a potent mitogen for renal tubular cells and for mature hepatocytes in primary culture. Eyes were developed in high percentage at the combined concentration of 1 and 10ng/ml of bFGF, and 1 and 10 ng/ml of HGF. From the induced eye and normal embryonic eye, RPE65 was commonly detected by monoclonal antibodies 40All and 25F5 and the localization of RPE65 was seen by AP reaction.

Spatio-temparal Pattern Formation of Abdominal Muscle in Xenopus Iaevis

  • The final pattern of the skeletal muscle of a vertebrate depends on the position-specific behavior of the muscle precursor cells during early developmental process and the abdominal muscle is made of cells which migrate a relatively long distance from their original tissue, myotome of dorsal mesoderm. We report the spatia-temporal migration pattern of abdominal muscle in Xenopus laevis by in situ hybridization and immunohistological studies. Shortly after hatching tadpole stage (stage 31/32), a group of myotomal cells detaches from the lower tip of the second somite and migrates ventrally to the lower position of abdomen. At stage 34/35, a second cell group migrates away from the third somite. Total 7 myotomal cell groups migrate ventrally one by one from the second to eighth myotome along their own pathways through the cell free space located between epidermis and subepidermal layer of the abdomen. During migration, the sizes of the cell groups (abdominal muscle anlagens) are increased to several tens fold. Around stage 40 all the abdominal muscle anlagens reaches their final positions and are interconnected side by side rostrocaudally. They are also connected to other types of muscles, forming a large multisegmented abdominal muscle. Heat shock study suggests that the disruption of segmentation of somites does not block the detachment of abdominal muscle anlagen, though the treatment gave stage- and dosagedependent effects on the migration speed.

  • Incontinentia pigmenti is characterized by irregular linear blisters with erythematous lesions which are developed from birth or later, verrucous papules on the extremities which are noted after a few weeks or months, and the appearance of streaks, spots and swirls of hyperpigmentation. This disorder is known as a systemic disorder caused by a defect at the developmental stage of organs originated from ectoderm or mesoderm. We experienced an incontinentia pigmenti in a male newborn infant who did not have any family history and had cutaneous lesions, which were in bullous and verucous stages, ophthalmic problems, and neurologic abnormalities.

The potential of mesenchymal stem cells derived from amniotic membrane and amniotic fluid for neuronal regenerative therapy

  • The mesenchymal stem cells (MSCs), which are derived from the mesoderm, are considered as a readily available source for tissue engineering. They have multipotent differentiation capacity and can be differentiated into various cell types. Many studies have demonstrated that the MSCs identified from amniotic membrane (AM-MSCs) and amniotic fluid (AF-MSCs) are shows advantages for many reasons, including the possibility of noninvasive isolation, multipotency, self-renewal, low immunogenicity, anti-inflammatory and nontumorigenicity properties, and minimal ethical problem. The AF-MSCs and AM-MSCs may be appropriate sources of mesenchymal stem cells for regenerative medicine, as an alternative to embryonic stem cells (ESCs). Recently, regenerative treatments such as tissue engineering and cell transplantation have shown potential in clinical applications for degenerative diseases. Therefore, amnion and MSCs derived from amnion can be applied to cell therapy in neuro-degeneration diseases. In this review, we will describe the potential of AM-MSCs and AF-MSCs, with particular focus on cures for neuronal degenerative diseases.
