Survey on Plant Parasitic Nematodes in Cropping by Controlled Horticulture (시설원예에 있어서 식물기생선충 조사)

  • Distribution and population of plant parasitc nematodes were investigated using 70 soil and root samples in total collected from six locations in Gyeongbug and Gyeongnam provinces. Among nematodes identified, Meloidogyne spp. was the most prevalent with $91.4\%$ in frequency cf detection. Frequency of detection was $87.1\%$ for Tylenchus sp., $24.3\%$ for Hirschmanniella, $23.3\%$ for Aphelenchus sp., and $20\%$ for Aphelenchoides sp.. Meloidogyne spp. was most abundant from samples collected in Namji and the number of nematodes dectected was 2,104 per $300cm^3$ soil. The number of Meloidogyne spp. detected was 671 from Milyang, 401 from Samrangjin. The number of nematodes was 57 from Samrangjin for Tylenchus sp., 127 from Gupo for Pratylenchus, 26 for Aphelenchoides sp. and 34 for Criconemoides from Samrangjin. Population of Meloidogyne spp. was the highest in Namji and Milyang where pepper and cucumber or pepper and chinese cabbage was cultivated in multiple croppings, however, population of Pratylenchus spp., and Meloidogyne spp. was the highest in Samrangjin where strawberry was cultivated as a single crop. Population in Pratylenchus and Meloidogyne was the highest in Gupo where tomato and chinese cabbage were cultivated alternatively, however, population of nematodes was generally low in Seongju and Yucheon where watermelon-rice or pepper-rice were cultivated in sequence. Meloidogyne spp. and Pratylenchus spp. appear to be economically important nematode species in cropping by controlled horticulture. In this study, 15 species of plant parasitic nematodes were identified.

Molecular Identification of Meloidogyne spp. in Soils from Fruit and Vegetable Greenhouses in Korea (분자기법을 이용한 과채류 시설재배지 토양 내 분포하는 뿌리혹선충의 종 동정)

  • In this study, we analyzed the phylogenetic characterization of root-knot nematodes (Meloidogyne spp.) in soils from fruits and vegetables greenhouses in Korea. Soil samples were collected from 12 greenhouse fields in which tomato, cucumber, watermelon, and Oriental melon were being cultivated. Meloidogyne spp. were detected in all the soil samples at an average number of $72{\pm}6$ nematodes/300 g of soil to $2,898{\pm}468$ nematodes/300 g of soil. Phylogenetic analysis using polymerase chain reaction-restriction fragment length polymorphism was attempted for the second-stage juveniles (J2) of Meloidogyne spp. collected from the greenhouse soils. Twelve Meloidogyne spp. from the greenhouse soils were classified into two groups by using HinfI digestion of mitochondrial DNA, resulting in 900, 410, 290, and 170 bp fragments (group A) and 900, 700, and 170 bp fragments (group B). Phylogenetic analysis based on mitochondrial DNA sequences (1,483-1,521 bp) showed that nine group A isolates were identified as Meloidogyne incognita (99.73-99.93%) and three group B isolates showed 99.54-99.73% similarity to Meloidogyne arenaria.

Incidence of Plant-Parasitic Nematodes in Strawberry Nursery and Nematode Dispersal by Daughter Plant (딸기 육묘장 토양 내 식물기생선충의 감염현황과 자묘를 통한 선충의 분산)

  • To survey the incidence of plant-parasitic nematode in strawberry nursery, 117 soil samples were collected from main producing area of strawberry nursery plants in 2016. The incidences of two major problematic nematodes such as root-lesion nematodes (Pratylenchus spp.) and root-knot nematodes (Meloidogyne spp.) were 11% and 3%, respectively. Strawberry nurseries using upland soil either in fields or in plastic-houses showed were higher infection rates of Pratylenchus spp. or Meloidogyne spp. than others. On the other hand, a strawberry nursery using pots filled with bed soil in plastic-house was the lowest infection rate with 0%. Phylogenetic relationships and distance analysis were performed for identification of Pratylenchus spp. and Meloidogyne spp. detected from strawberry nurseries. As the results, they were identified as Pratylenchus penetrans, Pratylenchus vulnus and Meloidogyne hapla. Meanwhile, when nursery plants of strawberry infested with P. vulnus transplanted to pots filled with sterilized soil, P. vulnus was detected from soil in pots as 33% frequency. Thus, P. vulnus can be spread through nursery plants of strawberry infested with that. These studies suggested that Pratylenchus spp. and Meloidogyne spp. were ploblematic nematodes in major areas of strawberry nursery plants and P. vulnus could be spread through nursery plants of strawberry.

Incidence of the Rice Root-Knot Nematode, Meloidogyne graminicola, in Guangxi, China

  • Root-knot nematodes (Meloidogyne spp.) are the most destructive group of plant-parasitic nematodes. Plants infected by Meloidogyne spp. develop above-ground symptoms, stunting, yellowing, nutrient deficiencies, and gall formations with typical hook-shaped root tips. Infected plants experience yield losses. During 2018-2019 survey, leaf chlorosis rice plants were found in 206 fields of 67 counties in Guangxi, China, around 30 days after transplanting. Galls and hooked tips on the roots and pear-shaped females were observed. About 32.04% of fields were infested with the nematode. The nematodes were identified as Meloidogyne graminicola base on morphological and molecular analysis. To the best of our knowledge, this is the first report of M. graminicola on rice plants in Guangxi, China. The results of this study urge the discovery of resistant cultivars and the development of management strategies.

A Study on the Root-knot Nematodes (Meloidogyne spp.) Affecting Economic Crops in Korea (경제작물에 영향을 미치는 뿌리혹 선충에 관한 연구)

  • A study on the root-knot nematodes (Meloidogyne spp.) affecting economic crops in Korea was undertaken to know the distribution of the nemic fauna. Total 66 samples were taken from soil and root of 24 host plants at different localities in 4 provinces. Four Meloidogyne species such as Meloidogyne hapla, Meloidogyne arenaria, Meloidogyne incognita and Meloidogyne javanica were identified and some morphological characteristics were described. One of these, Meloidogyne javanica was reported newly in Korea from Horticultural Experimental Station, Suweon, Gyeong Gi on potato, Geomsadong, Daegu on chinese cabbage, Sangeogdong, Daegu, Gyeong Bug on violet and Choeumri, Namhae, Gyeong Nam on pumpkin. In Jae Ju province, Meloidogyne incognita was only found except the other three species. The most common and widely distributed Meloidogyne species in Korea is Meloidogyne hapla by $50\%$ in total, next Meloidogyne incognita $33.3\%$ Meloidogyne arenaria $10.6\%$ Meloidogyne javanica $6.0\%$ in turn, The root-knot nematodes infected the most severely in Jae Ju province and Gyeong Nam, Gyeong Bug and Gyeong Gi province in turn. Twenty four plants were attacked by root-knot nematodes among them important economic crops are soybean, peanut, potato, tomato, cucumber, carrot, pumpkin, wateremelon, edible burdock, pepper, eggplant, cabbage, lettuce and tobacco in Korea.

The major plant-parasitic nematodes in plastic vinyl house field (하우스시설 재배지에서 발생하는 주요 선충)

  • To know how much damages occurred by nematodes in plastic vinyl house field, soil samples were collected from strawberry cultivation areas at Buyeo and Nonsan in Chungnam and Jinju in Kyeongnam, melon cultivation area at Gocksung in Junnam and cucumber field at Gongju in Chungnam. And then, nematode samples were separated from each soil sample, and identified the kind of plant-parasitic nematodes. Plant-parasitic nematodes were separated from 52 soil samples. Among samples, Meloidogyne spp., Pratylenchus spp. and Helicotylenchus spp. were isolated from 45 (86.5%%) and 33 (63.5%) and 47 collected soil samples (90.4%), respectively. As a result of identification of plant-parasitic nematodes from regional collected soil samples, distribution of Helicotylenchus spp. was higher than any other plant-parasitic nematode. And the population of Meloidogyne spp. and Pratylenchu spp. were also higher, and a occurrence ratio of Meloidogyne spp. is higher than Pratylenchu spp. except the cucumber growing area at Gongju.

Control of the Root-Knot Nematode (Meloidogyne spp.) on Cucumber by a Liquid Bio-Formulation Containing Chitinolytic Bacteria, Chitin and Their Products (키틴분해세균, 키틴 및 그들의 산물이 함유된 미생물제에 의한 오이의 뿌리혹선충(Meloidogyne spp.) 방제)

  • A liquid bio-formulation containing chitinolytic bacteria, chitin and their products was assessed for its potential biological control against root-knot nematodes on cucumber. The bio-formulation was prepared by cultures of three chitinolytic bacteria, Chromobacterium sp. strain C-61, Lysobacter engymogenes and Serratia plymuthica in minimal medium supplemented with chitin. Under pot conditions, the bio-formulation showed better growth of cucumber plants, and less root galls and population density of Meloidogyne spp. than control media without the bio-formulation. In a greenhouse, 75-fold diluted bio-formulations were treated instead of water around cucumber plants through hoses for drip irrigation six times at 5-day intervals from the transplanting date. After 30 and 60 days, the treatment provided about 7% and 10% enhancement in the plant height and about 78% and 69% reduction in population density of Meloidogyne spp. in the rhizosphere, respectively. In addition, the experiments showed that the control effects occurred only in the soils contacted with the bio-formulation. Undiluted bio-formulations were drenched three times at 10-day intervals around cucumber plants severely infested with Meloidogyne spp. The treatment showed about 37% plant enhancement without dead plants compared with 37% death in the untreated control, and about 82% nematode reduction. These results suggest that the bio-formulation can be practically used to control the root-knot nematode on cucumber.

Effect of temperature and pH on the attachment of Pasteuria penetrans to Meloidogyne arenaria and the mass production (Pasteuria penetrans의 땅콩뿌리혹선층(Meloidogyne arenaria)에 대한 온도와 pH별 부착 및 증식효과)

  • The cultivating agroproducts are damaged by the Meloidogyne spp. which are gradually increasing in farm land soil. No effective control method for Meloidogyne spp., however, is available. Pasteuria penetrans which is one of the microorganisms in soil is used for biological control of Meloidogyne spp. although the method of mass production is limited. This study was conducted to investigate attachment and mass production effect of P. penetrans to M. arenaria under different temperatures (10, 30, 50 and $70^{\circ}C$) and pH values (4, 7 and 10). Attachment rates under these temperature and pH were more than 96% and 80%, respectively. In mass production rates, the number of P. penetrans attached on M. arenaria under different temperatures and pH were highly increased in root of tomato but not significantly different. Therefore, we concluded that P. penetrans can survive and attach on M. arenaria under various conditions. This method for mass production of P. penetrans can be provided to develop environmentally-friendly nematicide.

Distribution of Root-knot Nematodes, Meloidogyne spp. and Their Races in Economic Crops in Korea (주요 경제작물에 기생하는 뿌리혹 선충의 종과 Race 분포)

  • In order to investigate the distribution and density of root-knot nematodes in economic crops, samples were collected from 3,226 fields of 18 economic crops. Nematodes were extracted using a modified Baermann's funnel technique. Identification of races was based on the differential host-test for Meloidogyne spp. by Tayler and Sasser. M. hapla was dominant in fields in the middle parts of Korean peninsula; M. incognita, in the southern parts, and distribution of M. javanica was limited to Jeju island and southern seaside areas. Two races of M. arenaria identified in this study were races 1 and 2. The three races identified in M. incognita were races, 1, 2 and 3. Of these races, race 1 of M. arenaria and race 3 of M. icognita were identified for the first time in Korea.
