Effects of Rubus coreanus Miquel on the Expressions of Tyrosinase, TRP-1 and TRP-2 in B16 Melanoma Cells (복분자가 B16 세포주의 Tyrosinase, TRP-1 and TRP-2 발현에 미치는 영향)

  • Melanogenesis is induced mainly by ultraviolet radiation of sunlight and ${\alpha}-melanocyte$-stimulating hormone (${\alpha}-MSH$) which binds to a specific G protein coupled receptor. The purpose of this study was to investigate the mechanism of melanogenesis inhibition in B16/F10 cells by methanol extract of Rubus coreanus Miquel (RCM). In the present study, ${\alpha}-MSH$ and forskolin led to a stimulation of melanin synthesis that appeared to result from an increased tyrosinase activity and melanin content. However, RCM inhibited the ${\alpha}-MSH$- and forskolin-induced melanin synthesis. In addition, RCM abolished the ${\alpha}-MSH$- and forskolin-induced cytoplasmic dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase and tyrosinase-related protein 1 (TRP-1) were increased after incubation with α-MSH and forskolin. The treatment of RCM decreased the ${\alpha}-MSH$- and forskolin-induced expression levels of tyrosinase and TRP-1. Based on these findings, it is likely that RCM exerts its depigmenting effects in B16/F10 cells through the suppression of tyrosinase and TRP-1 expression, which are key enzymes for melanogenesis.

Effects of Lycii Fructus Extracts(LFE) on Skin whitening and Elasticity using Melanoma cells (구기자 추출물이 피부 미백 및 주름에 미치는 영향)

  • Objective : Lycii Fructus Extracts(LFE) can do Anti-hypertension activity, Antidepressant, Anti-diabetic activity. This study was designed to investigate effects of LFE on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of LFE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : More than $250{\mu}g/ml$ of LFE treated group showed lowered proliferation rates significantly compared to non-treated group. More than $125{\mu}g/ml$ of LFE treated groups were lower levels of melanin synthesis respectively. LFE showed inhibitory effect on tyrosinase activities in vitro. And, LFE suppressed tyrosinase activities in B16F10 cells significantly. Finally, LFE suppressed elastase type I and IV activities in dose-dependent manner in vitro. And LFE also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that LFE can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin elasticity or whitening.

Skin lightening effect of fermented Panax ginseng extract (자연삼 발효 추출물의 미백 활성에 대한 연구)

  • Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.

Effect of Antioxidant and Skin Whitening of Ethanol extracts from Ultrasonic Pretreated Abeliophyllum distichum Nakai (초음파 처리 미선나무 에탄올 추출물의 항산화 및 미백효과)

  • This research evaluate antioxidant and skin-whitening effect of Abeliophyllum distichum Nakai by extraction processes. First, antioxidant effects were follows: EE (70% ethanol extract) showed higher DPPH scavenging activity of 69.66% than WE (hot water exract) 59.13% at $0.3mg/m{\ell}$, also UE's (70% ethanol extract by sonication process) higher than EE. Reducing power was that also EE showed higher than WE, and it was the highest value with UE's because of ultrasonic pretreatment. Next, the whitening effect tyrosinase inhibition activity was measured that EE was 23.88%, WE's was 16.69%, and UE was 23.34%. Ultrasonic pretreatment did not influence to tyrosinase inhibition activity. Cell viability showed low cell toxicity in all groups. UE's inhibited melanin synthesis, 55.1%, that is higher than EE and WE, 52.7% and 39.5%, respectively. As a result, we confirmed that antioxidant activities and skin-whitening effect by extraction process. Also, this results confirmed that the Abeliophyllum distichum Nakai extracts worth as cosmetic materials.

Inhibitory Effects on Melanogensis of Scrophularia koraiensis Nakai in Melanocytes

  • Scrophularia koraiensis Nakai (S. koraiensis) has used its roots as traditional herbal medicine. Some research is reported to be effective in allergic inflammation and osteoporosis. In a present study, we conducted to investigate the bioactivity of the ethanol extract of S. koraiensis (ESK) on the inhibition of melanogenesis and the apoptosis of melanocytes. We analyzed Harpagoside of ESK by using LCMS and HPLC-PDA and investigated the regulation of ESK on reactive oxygen species. Also, the expressions of melanin synthesis-related factors and apoptosis-related factors were confirmed. As a result, the quantification results of quercetin and rutin in ESK were 77.2 and 7.4 mg/g. IC50 on DPPH and ABTS radical scavenging activity is 33.1 and 9.5 ug/mL. ESK attenuated not only the expression of tyrosinase, TYRP-1, TYRP-2, and MITF in melanogenesis. It is thought that ESK may be effective in the inhibition of melanogenesis through MAPK cell signaling pathway in melanocytes. These study results suggest that ESK has the ability to inhibit melanin production and induce apoptosis.

Synthesis of Novel Kojic Acid Derivative and Its Anti-pigmentation Effect

  • A kojic acid derivative, kojic acid 7-O-$\beta$-D-tetraacetylglucopyranoside(KTG) was synthesized. Regio-and stereo-selective glycosylation at 7-postion in kojic acid with $\beta$-D-pentaacetylglucose was achieved with high yield(80%) by the use of Lewis acid and organic base in nonpolar solvent. KTG was hydrolyzed in methanol by the aid of sodium methoxide to give kojic acid 7-O-$\beta$-D-glucopyranoside(KGP). KGP is freely soluble in water and soluble in methanol and ethanol. Its structure was comfirmed by $^1$H-NMR and $^{13}$ C-NMR. Tyrosinase activity inhibition of KGP was measured with mushroom tyrosinase compared with ascorbic acid, kojic acid and arbutin. KGP showed higher tyrosinase inhibition activity($IC_{50}$/=33.3 $\mu\textrm{g}$/ml) than ascorbic acid(63.2 $\mu\textrm{g}$/ml) and arbutin(91.8 $\mu\textrm{g}$/ml) but lower inhibition activity than kojic acid(8.3 $\mu\textrm{g}$/ml). To test free-radical scavenging activity, we used 1, 1-diphenyl-2-picrylhydrazyl(DPPH) as a free-radical source. Free-radical scavenging activity of KGP was very low($SC_{50}$/>1000 $\mu\textrm{g}$/ml) compared with ascorbic acid($SC_{50}$/=2.68 $\mu\textrm{g}$/ml) and arbutin($SC_{50}$/=180$\mu\textrm{g}$/ml). Melanin formation inhibition of KGP was measured in B16 melanoma, compared with kojic acid, arbutin and Vitamin C. Inhibition activity of KGP for melanin formation was not found within test concentrations.

  • Vitamin C's function and mechanism are comparatively well known among the several kinds of vitamins. Inhibition of free radical. promotion of collagen synthesis, restraint of melanin formation and resolution of melanin are its main functions. But Vitamin C is very easy to oxidize by heat, moisture or air so it causes stability problem to make formulation. To solve these problems unique formulating method or derivatives using method could be effective. Object: First, stabilize 10% of Ascorbic acid as polyol/silicone emulsion and try to make polyol/silicone/polyol anhydrous multiple emulsion secondly. And then, encapsulate with porous power to enhance the stability of Ascorbic Acid from formulating method.

Anti-Pigmentation Effects of Eight Phellinus linteus-Fermented Traditional Crude Herbal Extracts on Brown Guinea Pigs of Ultraviolet B-Induced Hyperpigmentation

  • We have previously found that mycelia culture broth of eight kinds of traditional herbal extracts fermented with Phellinus linteus (previously named as 8-HsPLCB) not only inhibited melanin and tyrosinase activity, but also reduced the contents of melanogenesis-related proteins, including tyrosinase and microphthalmia-associated transcription factor, in 3-isobutyl-1-methylxanthine-stimulated B16F0 melanoma cells. For a further study, the effect of 8-HsPLCB against skin pigmentation in brown guinea pigs with ultraviolet B (UVB)-induced hyperpigmentation was investigated. 8-HsPLCB (3%) and arbutin (2%) as positive controls were applied topically twice daily for 4 weeks to the hyperpigmented areas. 8-HsPLCB showed skin-lightening effect as effective as arbutin, one of the most widely used in whitening cosmetics. Melanin index values as the degree of pigmentation showed a significant reduction week by week post 8-HsPLCB treatment and then substantially reduced by 4 weeks. The degree of depigmentation after 4 weeks of topical application with 8-HsPLCB was 32.2% as compared with before treatment (0 week). Moreover, using Fontana-Masson staining and hematoxylin-eosin staining, 8-HsPLCB reduced melanin pigmentation in the basal layer of the epidermis and epidermal thickness changes exposed to the UV-B irradiation as compared with non-treatment and vehicle treatment. The intensity of the skin-lightening effect of 8-HsPLCB was similar to arbutin. These results suggest that the skin-lightening effect of 8-HsPLCB might be resulted from inhibition of melanin synthesis by tyrosinase in melanocytes. To conclude, 8-HsPLCB treatment showed reduction of the melanin pigment and histological changes induced by UV irradiation in brown guinea pigs.

The Inhibitory Effects of Water Chestnut Extracts on Melanogenesis through Regulation of Tyrosinase Expression (Tyrosinase 발현 조절을 통한 마름열매 추출물의 Melanin 생성 억제 효과)

  • Water chestnut is fruits of Trapa natans var. bispinosa grown wild in Korea. In this study, water chestnut was investigated for anti-pigmentation. Treatment with ethanolic extracts of water chestnut significantly reduced production of melanin in α-MSH simulated B16F10 cells. At 200 ㎍/mL ethanolic extracts of water chestnut, melanin contents were repressed by 43.26% compared to the control group. Additionally, ethanolic extracts of water chestnut reduced expression and activity of tyrosinase, key enzyme in melanogenesis, in α-MSH simulated B16F10 cells. Ethanolic extracts of water chestnut downregulated tyrosinase activity and expression to 23.65% and 62.35%, respectively. These results suggest that ethanolic extracts of water chestnut might be used as a promising whitening ingredients for inhibition of α-MSH-induced melanin synthesis and pigmentation.

  • Tyrosinase is one of the important enzymes in the mammalian melanin synthesis. In the process of melanin synthesis, tyrosine is oxidized to DOPA (3,4-dihydroxyphenylalanine), and DOPA is further oxidized to dopaquinone. Tyrosinase is an enzyme catalyzing this oxidation of tyrosine, so chemicals that inhibit the activity of tyrosinase can be used as skin whitening agents. In this study, we isolated five constituents from the 80% MeOH extract of Morus bombycis cortex by bioactivity-guided fractionation. We performed mushroom tyrosinase inhibition assay. As a result, 7,2',4'-trihydroxyflavanone (1), 2',4',2,4,-tetrahydroxychalcone (2), and oxyresveratrol (3) showed the more potent inhibitory effect compared to kojic acid, a well-known skin whitening agent with antityrosinase effect. Moracinoside M (4) and moracin M-3'-O-${\beta}$-D-glucopyranoside (5) also showed the moderate tyrosinase inhibitory activities.
