Optimization of Medium and Fermentation Conditions for Mass Production of Bacillus licheniformis SCD121067 by Statistical Experimental Design (Bacillus licheniformis SCD121067 균체 생산성 증가를 위한 통계적 생산배지 및 발효조건 최적화)

  • In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.

  • The subframe type rear suspension consisting of a side member and a front/rear cross member is widely used in a medium car and full car. In the small car case, the beam of tubular type without independent suspension system is used to reduce manufacturing cost. In this study, a subframe type rear suspension by hydroforming has been developed. In designing suspension, a driving stability and durability should be considered as an important factor for the performance improvement, respectively. Thus, we focus on increasing the stiffness of suspension and decreasing the maximum stress affecting a durability cycle life. Several optimization design techniques such as shape, size, and topology optimization are implemented to meet these requirements. The shapes of rear suspension obtained from optimization are formed by using hydroforming process. Through commercial software based on the finite element, the superiority of this design method is demonstrated.

  • Pitch drive system in wind turbine is composed by the planetary gear system to satisfied its required performance such as long life and light weight for gear train. When the planetary gear system can reduce its volume and weight, the power consumption of the wind turbine can be reduced. In this study, the planetary gear system of the pitch drive system in medium-sized wind turbine is obtained for weight reduction by shape optimization method. And the planetary gear system is verified for their strength by the structural analysis.

  • This study deals with an Excel-based scheduling system for a small and medium sized manufacturing factory without sufficient capability for managing full-scale information systems. The factory has the bottleneck with identical machines and unique batching characteristics. The scheduling problem is formulated as a variation of the parallel-machine scheduling system. It can be solved by a two-phase method: the first phase with an ant colony optimization (ACO) heuristic for order grouping and the second phase with a mixed integer programming (MIP) algorithm for scheduling groups on machines.

  • Polynomial is used to optimize crown bowl shape of a marine medium speed diesel engine piston. The primary goal of this paper is that it's for an original design through a thermal stress and highest temperature minimum. Piston is modeled using solid element with 6 design variables defined the positional coordinate value. Global optimum of design variables are found and evaluated as developed and integrated with the optimum algorithm combining genetic algorithm(GA) and tabu search(TS). Iteration for optimization is performed based on the result of finite element analysis. After optimization, thermal stress and highest temperature reduced 0.68% and 1.42% more than initial geometry.

  • Media optimization for the production of thermostable protease specifically hydrolyzing defatted soybean meal (DSM) from Bacillus licheniformis NS70 was performed by two methods, one-at-a-time method and response surface methodology (RSM). The best carbon source and nitrogen source for the protease production were lactose and DSM, respectively. The maximum protease production estimated by RSM was 606 U/L at 1.11% lactose and 0.43% DSM, the value of which was nearly consistent to the experimental value of 599 U/L. Yeast extract suppressed the protease production. The medium pH was slightly increased at the beginning stage of fermentation, and it tended to decrease after 8 hours. The optimal pH for the protease production was 7.2 in the batch fermentation.

  • Optimization of the protocorm-like body (PLB) formation of Phalaenopsis and Dendrobium hybrids was performed by determining the effects of plant growth regulators (PGRs) and different parts and division sizes of the PLB. For both genera, the base part was the best for the proliferation of PLBs, yielding the highest number of PLBs on a PGR-free medium for Phalaenopsis and medium containing $0.1{\mu}M$ NAA and $10.0{\mu}M$ BAP for Dendrobium. As regards the division size, four-division sections resulted in a higher PLB formation efficiency for Phalaenopsis, while two-division sections produced a higher PLB formation efficiency for Dendrobium. It is expected that these findings will be applicable to efficient PLB formation of other Phalaenopsis and Dendrobium orchids.

Improved Ectoine Production from Methane by Optimization of the Bio-milking Process in Engineered Methylomicrobium alcaliphilum 20Z

  • Methane is one of the major greenhouse gases, recently, the biotechnological conversion from methane to high-value added chemicals have emerged as an effort to reduce methane gas emission. In this study, we optimized ectoine bio-milking conditions in which cells were repeatedly used to improve intracellular and extracellular ectoine yield from methane by using Methylomicrobium alcaliphilum 20ZDP2. First, the cultivation and intracellular ectoine accumulation conditions were optimized with respect to the growth phase and medium salinity to achieve the highest yield of synthesis. Second, ectoine excretion was optimized by determining the ectoine secretion time (15 min) in appropriate medium salinity under hypoosmotic conditions (1% NaCl). Finally, bio-milking of ectoine was successfully repeated more than 10 times using M. alcaliphilum 20ZDP2, and the ectoine yield was improved up to 129.29 mg/ DCW g.

  • The Pneumococcus, Streptococcus pneumoniae, has an ample polysaccharide (PS) capsule that is highly antigenic and is the main virulence factor of the organism. The capsular PS is the source of PS vaccine. This investigation was undertaken to optimize the culture conditions for the production of capsular PS by type 4 pneumococcus. Among several culture media, brain heart infusion (BHI) and Casitone based medium were found to support luxuriant growth of pneumococcus type 4 at the same level. Therefore in this study, the Casitone based medium was used to study optimization of the culture condition because of BHI broth's high cost and complex nature. The phase of growth which accomodated maximum PS production was exponential phase. Concentrations of glucose greater than 0.8% did not enhance growth or PS production. Substitution of nitrogen sources with other resources or supplementation of various concentrations of metal ion (with the exception of calcium, copper, and magnesium ions) had adverse effects on growth and PS production. On the other hand, low level aeration and supplementation of 3 mg/l concentration of asparagine, phenylalanine, or threonine were beneficial for increased PS production. The synergistic effect of all the favorable conditions observed in pneumococcal growth assays provided a two-fold cumulative increase in capsular PS production.

Enhanced Antibiotic Production by Streptomyces sindenensis Using Artificial Neural Networks Coupled with Genetic Algorithm and Nelder-Mead Downhill Simplex

  • Antibiotic production with Streptomyces sindenensis MTCC 8122 was optimized under submerged fermentation conditions by artificial neural network (ANN) coupled with genetic algorithm (GA) and Nelder-Mead downhill simplex (NMDS). Feed forward back-propagation ANN was trained to establish the mathematical relationship among the medium components and length of incubation period for achieving maximum antibiotic yield. The optimization strategy involved growing the culture with varying concentrations of various medium components for different incubation periods. Under non-optimized condition, antibiotic production was found to be $95{\mu}g/ml$, which nearly doubled ($176{\mu}g/ml$) with the ANN-GA optimization. ANN-NMDS optimization was found to be more efficacious, and maximum antibiotic production ($197{\mu}g/ml$) was obtained by cultivating the cells with (g/l) fructose 2.7602, $MgSO_4$ 1.2369, $(NH_4)_2PO_4$ 0.2742, DL-threonine 3.069%, and soyabean meal 1.952%, for 9.8531 days of incubation, which was roughly 12% higher than the yield obtained by ANN coupled with GA under the same conditions.
