Quantitative analysis of cordycepin in Cordyceps militaris under different extraction methods

  • Cordyceps militaris (CM) is one of the most important medicinal mushrooms known to possess various biological activities. Cordycepin (CP) is a bioactive compound present in the fruiting bodies of CM and is known to have anti-tumor, anti-metastatic immunomodulatory and anti-inflammatory activities. In this study, we aim to analyze CP quantitatively under various CM extraction conditions. CP was measured using high-performance liquid chromatography, quantified using a reversed phase column using a gradient elution system of water and acetonitrile, and detected with a UV absorbance wavelength of 260 nm. The CP content of CM was the highest in 100% ethanol extract of the fruiting bodies and 60% ethanol extract of the mycelium. This study provides an efficient analysis method to determine the optimal extraction conditions for CP that can be used as a basis for developing functional foods and pharmaceutical products derived from CM.

An Indole Alkaloid from the Fruiting Body of Boletus umbriniporus (암갈색그물버섯(Boletus umbriniporus)에서 분리한 indole alkaloid)

  • Mushrooms are valued as a nutritional food and also as an important source of useful medicinal components. They produce various secondary metabolites which have interesting biological activities and unique chemical structures. As part of our ongoing investigation on chemical constituents and bioactive components of Korean native mushrooms, compound 1, an indole alkaloid, was isolated from the fruiting body of Boletus umbriniporus. B. umbriniporus is characterized by its yellow flesh, which changes to pallid blue when exposed to air, and its chemical constituent has not been reported. Chemical structure of compound 1 was determined to be flazin on the basis of ESI-mass, $^1H$ NMR, $^{13}C$ NMR, $^1H-^1H$ COSY, HMQC, and HMBC analysis. This compound was isolated from the fruiting body of B. umbriniporus for the first time.

Growth of Chaga Mushroom (Inonotus obliquus) on Betula platyphylla var. japonica (자작나무시루뻔버섯(차가버섯)을 접종한 자작나무로부터 버섯의 생장)

  • Chaga mushroom (Inonotus obliquus), which has invaluable medicinal uses, grows only on living trees. To date, it is still harvested from its natural habitat and is not cultivated artificially. We artificially cultivated chaga mushrooms by inoculating its sawdust spawns on Betula platyphylla var. japonica in 2007, and monitored mushroom growth on the inoculated trees for 9 years. The mushrooms grew less than 1 cm per year, with the largest mushroom growing up to 9 cm in the 9 years of study. There was no difference in the growth (diameter at breast height) of trees with viable and non-viable I. obliquus. In conclusion, artificial cultivation of chaga mushroom was successful. Our findings suggest that selection of large B. platyphylla var. japonica as host tree could lead to better I. obliquus productivity. Further improvements of the method are needed to increase the success rate of I. obliquus inoculation.

A Rapid and Universal Direct PCR Method for Macrofungi

  • Macrofungi are valuable resources as novel drug candidates, new biomaterials, and edible materials. Recently, genetic approaches pertaining to macrofungi have been continuously growing for their identification, molecular breeding, and genetic engineering. However, purification and amplification of fungal DNA is challenging because of the rigid cell wall and presence of PCR inhibitory metabolites. Here, we established a direct PCR method to provide a rapid and efficient method for PCR-grade macrofungal DNA preparation applicable to both conventional PCR and real-time PCR. We first optimized the procedure of lysis and PCR using the mycelia of Lentinula edodes, one of the most widely consumed macrofungal species. Lysates prepared by neutralizing with (NH4)2SO4 after heating the mycelia in a mixture of TE buffer and KOH at 65℃ for 10 min showed successful amplification in both conventional and real-time PCR. Moreover, the addition of bovine serum albumin to the PCR mixture enhanced the amplification in conventional PCR. Using this method, we successfully amplified not only internal transcribed spacer fragments but also low-copy genes ranging in length from 500 to 3,000 bp. Next, we applied this method to 62 different species (54 genera) of macrofungi, including edible mushrooms, such as Pleurotus ostreatus, and medicinal mushrooms such as Cordyceps militaris. It was found that our method is widely applicable to both ascomycetes and basidiomycetes. We expect that our method will contribute to accelerating PCR-based approaches, such as molecular identification, DNA marker typing, gene cloning, and transformant screening, in macrofungal studies.

  • A mushroom as a functional food material is being increasingly used in these days. It has been that about 1,500 species of mushrooms grow naturally in Asia. Among them about 700 species are edible, but about 30-50 species are poisonous. Even a poisonous mushroom is a very interesting subject for the dietary life research from biochemical viewpoint due to the fact that any mushroom can be the physiological active material. Not a few identified poisonous materials are being used as research reagents thus far. It is suggested that more useful materials could be obtained by progresses in future researches. The identification of the poisonous ingredient can open up a route for mushroom intoxication cure. It is known that cell destruction and troubles on skin, liver, and kidney troubles can be occurred from mushroom intoxication. Recently the author reported as a collection of treatises regarding anti-tumor substances of mushroom (2005), antibiotic substances of mushroom (2006), living-body functional and control substance (2007), and medicinal mushroom commonly used in Korean herbal medicines (2008). In this sequel, published research data on protoplasm, nerve, stomach and intestines, malodor, and acute pain poison of mushroom are collected and reported.

Comparative analysis of nitrite scavenging activity and anti-inflammation effects in the fruiting bodies of medicinal mushrooms (약용버섯류 자실체의 아질산염 소거능 및 항염증 효능 분석)

  • This study was carried out to compare the anti-inflammation effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. We concentrated Ganoderma species and other medicinal mushrooms by extracting with ethanol. And We made it $100{\mu}g/ml$ concentration. As a result of nitrite scavenging activity, in the contrast to the positive control; Ascorbic acid was 25%, ASI 7080 of Ganoderma species was disappeared up to around 40%. And in the contrast to Ascorbic acid was 55%, ASI 7002 was 78.5% that was the highest anti-inflammation effect in the result of "No assay test". The Cordyceps militaris showed 75% and Hericium erinaceus showed 59.7% of anti-inflammation effect. As a result of the fungus yield control test of $TNF-{\alpha}$ through ELISA method to ASI 7002 of Ganoderma species that showed the highest anti-inflammation, it was reduced as same as LPS non-treatment. We extracted RNA from ASI 7002 Ganoderma species 10, 50, $100{\mu}g/ml$ concentration and LPS $10{\mu}g/ml$ of Raw 264.7 cell. And we tested the expression of iNOS, COX-2 and TNF-a that are kinds of inflammation gene after synthesizing RNA with cDNA. Finally we could find that iNOS, COX-2 and TNF-a were all controlled expression in the result of above experiment.

Protective effect of ethanolic extract of antler-shaped Ganoderma lingzhi against oxidative stress in PC12 neuronal cell line (PC12 신경세포주에서 녹각영지버섯 주정 추출물의 산화 스트레스 개선 효과)

  • This study was carried out to identify medicinal mushrooms with protective effects against oxidative stress in PC12 neuronal cell line, followed by evaluation of their antioxidant property. Extracts of medicinal mushrooms, including Ganoderma lucidum extract (GLE), antler-shaped Ganoderma lingzhi extract (AGLE), Hericium erinaceus extract (HEE), and Sanghuangporus baumii extract (SBE), were screened for cytotoxicity using MTT assay. None of the extracts up to $10{\mu}g/ml$ concentration affected cell viability. These extracts were further checked for their protective effect against oxidative stress-induced reactive oxygen species (ROS) production. Exposure to $50{\mu}M$ $H_2O_2$ induced ROS generation in PC12 cells, which was inhibited only by treatment with AGLE. In addition, inhibition of $H_2O_2-induced$ ROS generation by AGLE was found to be in a dose-dependent manner (2.5, 5, and $10{\mu}g/ml$). Microscopic examination of DCF fluorescence for detection of ROS showed a similar pattern. Further, antioxidant activity of AGLE was determined by ABTS radical cation assay, and its $IC_{50}$ was found to be $46.90{\pm}0.31{\mu}g/ml$. Taken together, these results suggest that AGLE may help to alleviate oxidative stress in PC12 neuronal cells.

13 weeks repeated oral dose toxicity studies with LMK02-Jangwonhwan in SD rats (LMK02의 Sprague-Dawley 랫드를 이용한 13 주간 반복 경구투여 독성시험)

  • Objectives : The oriental medicine Jangwonhwan, a boiled extract of 12 medicinal herbs/mushrooms, has been prescribed to patients with cognitive dysfunction, as originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan (LMK02-Jangwonhwan) consisting of seven medicinal plants/mushrooms, was shown to reduce the ${\beta}$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer's disease. The toxicity of LMK02-Jangwonhwan was investigated in SD rats, by a daily oral administration for 13 weeks and NOAEL(No observed adverse effect dose), a definite toxic dose and target organ, as well. Methods : Quality control of the tablet form of LMK02-Jangwonhwan was established by estimating the indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02-Jangwonhwan was investigated in 6 week old, specific pathogen free (SPF), Sprageu-Dawley rats by oral administration. Each test group consisted of 10 male and 10 female rats. The groups received doses of 500, 1,000 or 2,000 mg/kg/day of test substance for 13 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of the control group. Results : The 13-week repeated oral treatment doses didn't result in any specific symptoms or death. There were no significant changes in the rat's weight and food consumption. Further, ophthalmic examination, urinalysis, hematological, serum biochemistry test and organ weight revealed no significant differences. Conclusions : The no-observed-adverse-effect level(NOAEL) of LMK02 for male and female Sprague-Dawley rats was determined as 2,000mg/kg/day and the target organ wasn't confirmed. Because no significant adverse effects were observed, the target organ could not be determined.

Trends of commercial strain development and spawn industry in mushrooms (버섯의 품종 육성과 종균 산업의 동향)

  • According to UPOV (International Union for Protection of New Varieties of Plants), mushroom spawn market have to be opened by the year 2009. Number of commercial strains distributed by the year 2005 were 179 of 24 species of edible and medicinal mushrooms. Only nine strains of oyster mushrooms were registered as protected variety, which is not compatible with those recorded in other advanced countries. Meaning of spawn in broad sense contains commercial strains. Development of commercial strains faces two main problems in Korea : One is the complicated genetic patterns and sexuality of mushroom species, and the other is expensive experimental equipments and fruiting body growing houses. Resolution of these problems leads to development of mushroom strains. This could be achieved as follows; genetic resources collection and assessment, molecular characterization of useful genetic characters, development of new commercial strains by hybridization using typical genetic resources, strengthening of breeding research using "Mushroom Breeding Group", management of spawn research company by consortium, foundation of mushroom general industry, promotion of consumption, and upgrade of competition ability for other countries. These points are under discussion.

Effects of Dried Medicinal Mushroom (Sparassis crispa) Powder on Wheat Flour Dough and Bread Properties (약용버섯(꽃송이버섯) 분말의 첨가가 밀가루 반죽과 빵의 품질에 미치는 영향)

  • The effects of medicinal mushroom (Sparassis crispa) powder supplementation on the rheological property of dough and the quality of bread were investigated. Naturally dried S. crispa powders (NDSCP) and freeze-dried ones (FDSCP) were prepared. Farinograph profiles of dough showed that mushroom powder addition at levels higher than 1% led to reduced dough stability suggested by decreased development time and increased weakness. The incorporation of S. crispa powders was significantly (p<0.05) lower compared to other mushrooms which might be attributed to its high content of beta-glucan. FDSCP was chosen over NDSCP because of its fragrant flavor which could be ascribed to the preservation of volatile polyphenol components. The quality of FDSCP bread was evaluated with respect to specific volume, texture, color, and organoleptic qualities. The specific volume of bread with S. crispa powder decreased compared to the control, while textural properties, such as hardness, chewiness, and gumminess, were enhanced. Sensory evaluation showed that 0.3% FDSCP incorporation presented the highest bread quality. With its daily consumption, S. crispa supplemented bread can provide consumers with multiple health benefits.
