The dietary effect of medicinal herbs extract and multiple probiotics mixture on the growth performance, innate immune response and antibacterial activity of nile tilapia Oreochromis niloticus

  • The study investigated the dietary effects of medicinal herbs extract and multiple probiotics mixture on the growth performance, innate immune response and antibacterial activity of nile tilapia Oreochromis niloticus. Tilapia were divided in four groups. The first is a fish group fed a basal diet added with 40% medicinal herbs extract (MHE). The second is a fish group fed a basal diet supplied with $2{\times}10^8CFU/g$ of 2 Bacillus sp, 2 Lactobacillus sp and 2 Yeast sp, respectively (PB). The third group was fed with a mixture of probiotics (2 Bacillus sp, 2 Lactobacillus sp and 2 Yeast sp) with the medicinal herbs extract added in basal diet (MHE+PB). The fourth group was fed only a basal diet (C). In a non-specific immune parameters analysis, respiratory burst activity, lysozyme activity, phagocytic activity (PA), alternative complement pathway activity ($ACH_{50}$) and superoxide dismutase (SOD) activity were significantly (p<0.05) increased in the group MHE+PB compared to other groups. Both PB and MHE groups showed a significant (p<0.05) increase in respiratory burst activity, lysozyme activity compared to the control C group, whereas no significant differences were observed in PA, $ACH_{50}$ and SOD activity compared to the control group. In challenging test, fish were administered with Edwardsiella tarda (E. tarda) on 30 days after feeding with each experimental diet and viable E. tarda cell reduction was checked over 21 days post injection. MHE+PB group showed a significantly (p<0.05) reduced E. tarda cells compared to other groups. No significant antibacterial difference (p>0.05) was observed between PB and MHE only treated group. Compared to the control, a significant antibacterial difference (p<0.05) appeared in PB but not in MHE (p>0.05). The results suggest that the probiotics and MHE mixture could be utilized as an alternative to antibiotics in the control of fish diseases caused by E. tarda.

한약재발효액의 항생제 다제내성균에 대한 항균활성 및 항산화활성 (Antibacterial Activities of Fermented Sayuksan Ingredient Extracts for Multidrug-resistant Strains)

  • Sayuksan has been widely applied to treat a variety of diseases such as acute hepatitis, gastritis, and colitis. Sayuksan consists of medicinal herbs such as Glycrrhizae uralensis Fischer, Paeonia lactiflora Pallas, Bupleurum falcatum Linne, and Poncirus trifoliata Rafinesqul. Methanol extracts (1 mg/mL) from the four kinds of medicinal herbs did not show antibiotic activities against general test strains and multi-drug resistant strains. The antibacterial activity of fermented medicinal herbs extracts with Lactobacillus spp. strain was confirmed as Gram-positive bacteria which are higher than Gram-negative bacteria. Extracts of Glycrrhizae uralensis Fischer fermented with Lb. casei KCTC 3109 displayed inhibitory diameters of 16 mm against Pseudomonas aeruginosa P01828. Superoxide dismutase (SOD)-like activity of the medicinal herb extracts was not determined, but the extract of Paeonia lactiflora Pallas fermented with six strains of Lactobacillus spp. had the highest antioxidant activity. SOD-like activity of Paeonia lactiflora Pallas extracts fermented by Lb. brevis KCTC 3498 was $41.4{\pm}0.8%$, which was the highest antioxidant activity among the fermented extracts with the other medicinal herbs.

한약재의 벤조피렌 함량 및 추출물에서의 이행률에 대한 연구 (Study on Benzo(a)pyran Content and its Transfer Ratio in Extracts of Medicinal Herbs)

  • 본 연구에서는 건조하기 어려운 원료한약재 5 품목(총 50 건)을 대상으로 벤조피렌 함량 및 물 추출물과 연조엑스 제조단계에서의 벤조피렌의 이행률을 HPLC/FLD로 측정하였다. 벤조피렌의 검량선은 3~40 ng/mL의 농도 범위에서 상관계수($R^2$)가 1.000으로서 우수한 상관관계를 나타내었다. 원료한약재의 벤조피렌 검출 범위는 불검출 ${\sim}37.54{\mu}g/kg$이고, 검출평균은 $6.73{\mu}g/kg$이었다. 그 중에서 식품의약품안전처의 고시(2009-302호) 기준인 $5{\mu}g/kg$ 이상 검출된 시료는 15건(30%)이었다. 특히 황련의 벤조피렌 함량은 $37.54{\mu}g/kg$으로서 가장 높았다. 물 추출물, 연조엑스 및 물 추출 후 찌꺼기에서 벤조피렌의 검출 범위는 각각 불검출${\sim}2.31{\mu}g/kg$, 불검출${\sim}2.28{\mu}g/kg$$2.18{\sim}21.91{\mu}g/kg$으로 분석되었다. 물 추출물(탕제)와 연조엑스의 제조에서 벤조피렌은 이행되지 않거나, 특히, 현삼에서 물 추출물과 연조엑스의 경우에 각각 최대 8.9%와 최대 9.8%의 이행률을 보였다. 따라서 원료한약재에 비해 물 추출물 및 연조엑스에서 벤조피렌 함량이 각각 90.0% 이상까지 감소하였다.

수종(數種) 한약재(韓藥材)의 항암활성(抗癌活性) 연구(硏究)

  • An extensive anticancer drug screening from natural resources has been carried out primarily using murine tumor model for past fourty years. Recently a new screening program from NCI, so called disease-oriented screening system. has been estabished to detect anticancer drugs that show selective growth inhibition toward variety of tissue specific human solid tumors originated from leukemia, lung, colon, CNS, melanoma, ovarian, renal, prostate amd breast. To develope the anticancer drugs from oriental medicinal herbs, we investigated the cytotoxic effects against human tumor cell panels with 23 kinds of MeOH extract of medicinal herbs. Evodiae Fructus, Meliae Toosendan Fructus, Saussureae Radix and Pharbitidis Semen showed strong activities against several tumor cell lines.

Escherichia coli에 대한 한약재의 항균활성 (Antimicrobial Activity of Extracts from Medicinal Herbs Against Escherichia coli)

  • 10종의 한약재 MeOH 추출물과 EtOH 추출물의 E. coli에 대한 항균력을 조사한 결과 소목과 오배자의 MeOH 추출물과 EtOH 추출물은 5 mg/ml 수준에서 항균활성을 나타내었으며 목단피와 황금등은 10 mg/ml 이상에서 항균성을 보였다. 최소저해농도는 MeOH 추출물에서는 $1.4{\sim}8mg/ml$, EtOH 추출물에서는 $1.2{\sim}12mg/ml$ 농도에서 항균활성을 보였으며 분획층 중 EtOAc층에서 성장억제효과가 가장 높게 나타났다. 항균력이 우수한 것으로 확인된 소목과 오배자 추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 각각의 추출물을 0, 100, 300 및 500 ppm의 농도로 첨가하여 균주의 증식을 조사 한 결과 배양 후 24시간에 소목 추출물 무첨가구의 $OD_{620}$값이 0.6인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.1$정도로 균증식이 현저히 억제되었고, 오배자 추출물 무첨가구의 $OD_{620}$값이 0.5인 반면 500ppm 이상의 추출물 첨가 시 $0.02{\sim}0.2$정도로 균증식이 현저히 억제되었다. 이상의 결과로 볼 때 질병을 일으키는 E. coli의 감염을 예방하거나 또는 치료할 수 있는 한약재 추출물의 개발 가능성을 확인할 수 있었다.

  • This study was performed to develop new anti-obesity agents for functional foods from medicinal plants and herbs. Methanol extracts from 560 kinds of medicinal plants and herbs, and their lipase inhibitory activities were investigated. Methanol extract from Desmodium oxyphyllum Dc showed the highest lipase inhibitory activity at 74.2%. This lipase inhibitory activity was maximally extracted when powder of Desmodium oxyphyllum Dc was treated with 80% methanol (1 : 20) for 48 h.

당뇨병 예방을 위하여 한약재 추출액으로 제조한 현미밥의 품질특성 (Quality characteristics of brown rice boiled with medicinal herbs extract for diabetes prevention)

  • 한국인의 식생활에서 매일 섭취 할 수 있는 방안을 제시하기 위해 당뇨병에 효능이 있는 한약재를 이용하여 열수추출액을 제조하고 현미밥을 제조하여 그 품질특성을 연구한 결과는 아래와 같다. 수분함량은 현미의 첨가량이 증가할수록 수분함량은 낮아졌으며, $H_2$에서도 같은 수분함량을 보였다. 물성특성은 $H_1$$H_2$ 모두가 유사한 경향을 보였는데 단단한 정도(hardness)는 대조구인 백미 100%의 경우가 가장 낮았고, 현미 첨가량이 높아질수록 증가하였다. 탄력성(springiness), 응집성(cohesiveness), 점착성(gumminess)은 현미 첨가량이 증가할수록 감소하는 경향을 보였으며, 씹힘성(chewiness)은 현미 첨가량이 증가할수록 증가하는 경향을 보였다. 색도는 $H_1$$H_2$ 모두가 유사한 경향을 보였으며 L값은 현미 첨가량이 증가할수록 감소하였으며 a값은 현미 첨가량이 증가할수록 감소하는 경향을 보였고 b값은 반대로 현미 첨가량이 증가할수록 증가하는 경향을 보였다. $H_2$를 이용하여 제조한 현미밥의 관능평가 결과는 외관과 맛, texture, 전반적 기호도는 현미 첨가량이 증가할수록 감소하는 경향을 보였다. 전체적인 기호도는 $H_1$과 유사한 경향을 보여 현미 30% 첨가구에서 가장 높게 나타났다.

Escherichia coli에 대한 한약재추출물의 항균활성 (Antimicrobial activity of extracts from medicinal herbs against Escherichia coli)

  • The extracts from 40 different traditional medicinal herbs were used to investigate the antimicrobial activities against Escherichia coli. Among them, the extracts from Paeonia suffruticosa (PS), Siegesbeckia orientalis (SO), Schizandra chinensis (SC), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited high antimicrobial activities against E. coli, Minimum inhibitory concentrations (MIC) of the RJ extract against E. coli were 0.8 mg/ml. After heating treatment of these extracts, the antimicrobial activities against E. coli were significantly reduced in case of the CS extract. After alkaline or acid treatment of these extracts, the antimicrobial activities against E. coli were significantly increased in the PS extract but reduced in both SO and RJ extract. Since extracts from RJ and CS exhibited the highest antimicrobial activities, bacterial growth-inhibiting activities against E. coli by these two extracts were further examined. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of RJ extract ranged from 0.1 to 0.2 compared to 0.35~0.65 in the absence of RJ extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 100 ppm of RJ extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 or 500 ppm of CS extract ranged from 0.01 to 0.25 compared to 0.5~0.55 in the absence of CS extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 300 ppm of CS extract. In conclusion, these findings suggest that extracts from RJ and CS may play important roles for antimicrobial activities against E. coli causing various animal diseases.

황금, 산조인, 백출 추출물의 생리활성 (Biological Activities of Extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala. Methods : Three kinds of medicinal herbs were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidant activity and growth inhibition activity against cancer cells. Results : Electron donating abilities (EDA) of the water and ethanol extract from Zizyphus jujuba and Atractylodes macrocephala were over than 87% at 1,000 ${\mu}g/mL$, respectively. The highest nitrite scavenging abilities (NSA) of the water and ethanol extracts from Scutellaria baicalensis were 68.9 and 79.2% at 1,000 ${\mu}g/mL$, respectively. Atractylodes macrocephala extracts had shown the highest growth inhibition activity against both of MDA and A549 cells and the water extract had higher activity than ethanol extract. Conclusions : These results suggest that the extracts of medicinal herbs used for this experiment had specific biological activity and they can be used as natural antioxidant to prevent oxidative damage in normal cells.

약용식물 추출물이 저온식품의 미생물에 대한 항균효과 (Antimicrobial Activity of Medicinal Herbs Extracts on Cooling Preservation Foods.)

  • In order to study the natural preservation of food, sixteen kinds of medicinal plants were extracted by 80% methanol and then antimicrobial activity of the extracts was investigated against various kinds of microorganisms. The extracts of medicinal plants showed good effects of antimicrobial activity against bacteria except Bacillus cereus, Escherichia coli O-157. Especially, the extracts of Artemisia annua, Aster scaber, Solidago virganrea shower good inhibitory effects on the growth of bacteria to the cooling preservation foods, and Artemisia annua was decreased of the TMA(Trimethylamine) test. the result thus indicate that Artemisia annua, Aster scaber, Solidago fischeri extract inhibitory effects on cooling preservation foods, so that it may be potential as a natural preservatives.
