Catalytic Ability Improvement of Phenylalanine Hydroxylase from Chromobacterium violaceum by N-Terminal Truncation and Proline Introduction

  • Phenylalanine hydroxylase from Chromobacterium violaceum (CvPAH) is a monomeric enzyme that converts phenylalanine to tyrosine. It shares high amino acid identity and similar structure with a subunit of human phenylalanine hydroxylase that is a tetramer, resulting in the latent application in medications. In this study, semirational design was applied to CvPAH to improve the catalytic ability based on molecular dynamics simulation analyses. Four N-terminal truncated variants and one single point variant were constructed and characterized. The D267P variant showed a 2.1-fold increased thermal stability compared to the wild type, but lower specific activity was noted compared with the wild type. The specific activity of all truncated variants was a greater than 25% increase compared to the wild type, and these variants showed similar or slightly decreased thermostability with the exception of the $N-{\Delta}9$ variant. Notably, the $N-{\Delta}9$ variant exhibited a 1.2-fold increased specific activity, a 1.3-fold increased thermostability and considerably increased catalytic activity under the neutral environment compared with the wild type. These properties of the $N-{\Delta}9$ variant could advance medical and pharmaceutical applications of CvPAH. Our findings indicate that the N-terminus might modulate substrate binding, and are directives for further modification and functional research of PAH and other enzymes.

Feasibility of clay-shielding material for low-energy photons (Gamma/X)

  • While considering the photon attenuation coefficient (${\mu}$) and its related parameters for photons shielding, it is necessary to account for its transmitted and reflected photons energy spectra and dose contribution. Monte Carlo simulation was used to study the efficiency of clay ($1.99g\;cm^{-3}$) as a shielding material below 150 keV photon. Am-241 gamma source and an X-ray of 150 kVp were calculated. The calculated value of ${\mu}$ for Am-241 is higher within 5.61% compared to theoretical value for a single-energy photon. The calculated half-value layer (HVL) is 0.9335 cm, which is lower than that of ordinary concrete for X-ray of 150 kVp. A thickness of 2 cm clay was adequate to attenuate 90% and 85% of the incident photons from Am-241 and X-ray of 150 kVp, respectively. The same thickness of 2 cm could shield the gamma source dose rate of Am-241 (1 MBq) down to $0.0528{\mu}Sv/hr$. For X-ray of 150 kVp, photons below 60 keV were significantly decreased with 2 cm clay and a dose rate reduction by ~80%. The contribution of reflected photons and dose from the clay is negligible for both sources.

Signal-to-noise Ratio in Time- and Frequency-domain Photoacoustic Measurements by Different Frequency Filtering (주파수 필터링 함수에 따른 시간 및 주파수 영역 광음향 측정에 대한 신호 대 잡음비 분석)

  • We investigate the signal-to-noise ratios (SNRs) of time-domain (i.e. pulsed illumination) and frequency-domain (i.e. chirped illumination) photoacoustic signals measured by a spherically focused ultrasound transducer for spherical absorbers. The simulation results show that the time-domain photoacoustic SNR is higher than that of frequency-domain photoacoustic signals, as reported in the previous literature. We understand the reason for this SNR gap between the two measurement modes by analyzing photoacoustic-signal spectra, considering the incident beam energy controlled by the maximum permissible exposure. As the result of this approach, we find that filtering off the DC term in the chirped signal's spectrum improves frequency-domain photoacoustic SNRs by up to approximately 5 dB. In particular, it is observed that photoacoustic SNRs are highly sensitive to an upper-frequency value of frequency filtering functions, and the optimal upper-frequency values maximizing the SNR are different in time- and frequency-domain photoacoustic measurements.

  • Cardiac arrest is a series of conditions occur when the heart is stopped, regardless of the cause. one of the only ways to save a patient's life in the event of cardiac arrest is cardiopulmonary resuscitation, which is very important beacause it can maintain circulation through this technique, and high-quality CPR affects the survival rate and neurological prognosis of the patient. For infant cardiopulmonary resuscitation, use two finger to compress the chest. Hower, this method can be diffcult to reach the chest commpressions recommended by the American Heart Association because of the anatomically increased fatigue of the fingers and diffculty of vertical pressure. The study aims to verify the effects of new chest compressions in the implementation of chest compressions during infant cardiopulmonary resuscitation. The study also showed singnificant differences in chest depth and average rate of pressure(p<0.001). Based on the results of this study, we can see that the accuracy of the new chest compressions during infant cardiopulmonary resuscitation is increased, and the depth of chest compressions is improved, improving the quality index of chest compressions.

Development and Effect of Simulation Education Program including Interdisciplinary Communication for Nursing Students (간호대학생을 위한 학제간 의사소통을 포함한 시뮬레이션 교육 프로그램 개발 및 효과)

  • Communication is an important factor for nurses to lead successful nursing interventions by establishing therapeutic relationships with the subjects and cooperative relationships between the various occupations. The purpose of this study was to develop a communication education program to nurse college students to improve communication skills among medical professionals and to analyze the effects of the program. The subjects of this study were 54 students in the 4th grade of P nursing. The results of this study showed that there was a significant difference between the subjects' communication clarity(t=-7.75, p<.001) and self-leadership (t=-4.43, p<.001). Satisfaction with communication increased after the training but there was no significant difference (t=-1.22, p=.226). In conclusion, communication program using SBAR has a positive effect on clarity of communication and self-leadership of nursing students, and it is necessary to develop continuous education that can develop various scenarios and repeat training.

High-Performance Compton SPECT Using Both Photoelectric and Compton Scattering Events

  • In conventional single-photon emission computed tomography (SPECT), only the photoelectric events in the detectors are used for image reconstruction. However, if the $^{131}I$ isotope, which emits high-energy radiations (364, 637, and 723 keV), is used in nuclear medicine, both photoelectric and Compton scattering events can be used for image reconstruction. The purpose of our work is to perform simulations for Compton SPECT by using the Geant4 application for tomographic emission (GATE). The performance of Compton SPECT is evaluated and compared with that of conventional SPECT. The Compton SPECT unit has an area of $12cm{\times}12cm$ with four gantry heads. Each head is composed of a 2-cm tungsten collimator and a $40{\times}40$ array of CdZnTe (CZT) crystals with a $3{\times}3mm^2$ area and a 6-mm thickness. Compton SPECT can use not only the photoelectric effect but also the Compton scattering effect for image reconstruction. The correct sequential order of the interactions used for image reconstruction is determined using the angular resolution measurement (ARM) method and the energies deposited in each detector. In all the results of simulations using spherical volume sources of various diameters, the reconstructed images of Compton SPECT show higher signal-to-noise ratios (SNRs) without degradation of the image resolution when compared to those of conventional SPECT because the effective count for image reconstruction is higher. For a Derenzo-like phantom, the reconstructed images for different modalities are compared by visual inspection and by using their projected histograms in the X-direction of the reconstructed images.

  • This paper presents a topology based on packet tracer and a five-step scenario model to make it easier for students to understand the network on which VLANs are applied. Virtual LAN (VLAN), developed as an alternative solution to the Routers that distribute broadcast traffic, is a virtual local area network that logically configured regardless of the physical network. The VLAN prevents the network performance degradation resulting from the broadcast traffic by dividing the broadcast domain so that the bandwidth can be used more efficiently. In addition, it enhances the security because on communication between the devices belonging to different VLANs is impossible. The five-step scenarios in this paper presented an efficient implementation case for students to understand and validate the various functions of VLANs through ping/telnet/tracert test and simulation, after setting up each step of programming switches and routers in the virtual network.

Accuracy evaluation of threshold rainfall impacting pedestrian using ROC (ROC를 이용한 보행에 영향을 미치는 한계강우량의 정확도 평가)

  • Recently, as local heavy rains occur frequently in a short period of time, economic and social impacts are increasing beyond the simple primary damage. In advanced meteorologically advanced countries, realistic and reliable impact forecasts are conducted by analyzing socio-economic impacts, not information transmission as simple weather forecasts. In this paper, the degree of flooding was derived using the Spatial Runoff Assessment Tool (S-RAT) and FLO-2D models to calculate the threshold rainfall that can affect human walking, and the threshold rainfall of the concept of Grid to Grid (G2G) was calculated. In addition, although it was used a lot in the medical field in the past, a quantitative accuracy analysis was performed through the ROC analysis technique, which is widely used in natural phenomena such as drought or flood and machine learning. As a result of the analysis, the results of the time period similar to that of the actual and simulated immersion were obtained, and as a result of the ROC (Receiver Operating Characteristic) curve, the adequacy of the fair stage was secured with more than 0.7.

Reference dosimetry for inter-laboratory comparison on retrospective dosimetry techniques in realistic field irradiation experiment using 192Ir

  • The Korea Retrospective Dosimetry network (KREDOS) performed an inter-laboratory comparison to confirm the harmonization and reliability of the results of retrospective dosimetry using mobile phone. The mobile phones were exposed to 192Ir while attached to the human phantoms in the field experiment, and the exposure doses read by each laboratory were compared. This paper describes the reference dosimetry performed to present the reference values for inter-comparison and to obtain additional information about the dose distribution. Reference dosimetry included both measurement using LiF:Mg,Cu,Si and calculation via MCNP simulation to allow a comparison of doses obtained with the two different methodologies. When irradiating the phones, LiF elements were attached to the phones and phantoms and irradiated at the same time. The comparison results for the front of the phantoms were in good agreement, with an average relative difference of about 10%, while an average of about 16% relative difference occurred for the back and side of the phantom. The differences were attributed to the different characteristics of the physical and simulated phantoms, such as anatomical structure and constituent materials. Nevertheless, there was about 4% of under-estimation compared to measurements in the overall linear fitting, indicating the calculations were well matched to the measurements.

Suppression of side lobe and grating lobe in ultrasound medical imaging system (의료용 초음파 영상 시스템에서 부엽과 격자엽의 억제)

  • We propose an effective method for suppressing both side and grating lobes by applying 2-dimensional Fourier Transform to the received channel data during the receive focusing process of an ultrasound imaging system. When the signal from the image point is focused, the channel signals have the same DC value across the channels. However, even after echoes from outside an imaging point are focused, they are manifested as having different spatial frequencies depending on their incident angles. Therefore, after the receive focusing delay time is applied, 2-D Fourier Transform is performed on the time-channel data to separate the DC component and other frequency components in the spectral domain, and the weighting value is defined using the ratio of the two values. The side lobe and grating lobe were suppressed by multiplying the ultrasound image by a weighting value. Ultrasound images with a frequency of 5 MHz were simulated in a 64-channel linear array. The grating lobe appearing in the ultrasound image was completely removed by applying the proposed method. In addition, the side lobe was reduced and the lateral resolution was greatly increased. Results of computer simulation on a human organ mimicking image show that the proposed method can aid in better lesion diagnosis by increasing the image contrast.
