CD44 and CD133 as Cancer Stem Cell Markers for Gastric Cancer

  • Purpose: Currently, the two most influential gastric stem cell marker candidates are CD44 and CD133. The aim of this study was to make a comparison and determine the appropriate marker for use in gastric cancer stem cell research. Materials and Methods: We analyzed the expressions of CD44, CD133, and CD24 from the gastric cancer cell lines MKN45, MKN74, KATO-III, NCI-N87, SNU-1, SNU-216, SNU-601, SNU-638, and SNU-688 using flow cytometry. In addition, we measured the change in viability after applying 5 fluorouracil (5-FU) to the MKN45, MKN74, KATO-III, and NCI-N87 cell lines using a Cell Counting Kit 8. Results: CD133 expression was above moderate in the KATO-III, SNU-216, SNU-601 cell lines, whereas it was below 1% in the remaining cell lines. CD44 was expressed at levels above 5% in all gastric cancer cell lines. The effect of 5-FU on viability and CD133 or CD44 expression in the cell lines were not related. Conclusions: Expression of CD133 positive cells was insufficient in the gastric cancer cell lines. Therefore, of the cell lines tested, CD44 was the most appropriate tumor maker for research on gastric cancer stem cells.

Predicting Ability of Dynamic Balance in Construction Workers Based on Demographic Information and Anthropometric Dimensions

  • Background: Difficulties in walking and balance are risk factors for falling. This study aimed to predict dynamic balance based on demographic information and anthropometric dimensions in construction workers. Methods: This descriptive-analytical study was conducted on 114 construction workers in 2020. First, the construction workers were asked to complete the demographic questionnaire determined in order to be included in the study. Then anthropometric dimensions were measured. The dynamic balance of participants was also assessed using the Y Balance test kit. Dynamic balance prediction was performed based on demographic information and anthropometric dimensions using multiple linear regression with SPSS software version 25. Results: The highest average normalized reach distances of YBT were in the anterior direction and were 92.23 ± 12.43% and 92.28 ± 9.26% for right and left foot, respectively. Both maximal and average normalized composite reach in the YBT in each leg were negatively correlated with leg length and navicular drop and positively correlated with the ratio of sitting height to leg length. In addition, multiple linear regressions showed that age, navicular drop, leg length, and foot surface could predict 23% of the variance in YBT average normalized composite reach of the right leg, and age, navicular drop, and leg length could predict 21% of that in the left leg among construction workers. Conclusion: Approximately one-fifth of the variability in the normalized composite reach of dynamic balance reach among construction workers using method YBT can be predicted by variables age, navicular drop, leg length, and foot surface.

Sex hormones alter the response of Toll-like receptor 3 to its specific ligand in fallopian tube epithelial cells

  • Objective: The fallopian tubes play a critical role in the early events of fertilization. The rapid innate immune defense is an important part of the fallopian tubes. Toll-like receptor 3 (TLR3), as a part of the innate immune system, plays an important role in detecting viral infections. In this basic and experimental study, the effect of sex hormones on the function of TLR3 in the OE-E6/E7 cell line was investigated. Methods: The functionality of TLR3 in this cell line was evaluated by cytokine measurements (interleukin [IL]-6 and IL-1b) and the effects of sex hormones on TLR3 were tested by an enzyme-linked immunosorbent assay kit. Additionally, TLR3 small interfering RNA (siRNA) and a TLR3 function-blocking antibody were used to confirm our findings. Results: The production of IL-6 significantly increased in the presence of polyinosinic-polycytidylic acid (poly(I:C)) as the TLR3 ligand. Using a TLR3-siRNA-ransfected OE-E6/E7 cell line and function-blocking antibody confirmed that cytokine production was due to TLR3. In addition, 17-${\beta}$ estradiol and progesterone suppressed the production of IL-6 in the presence and absence of poly(I:C). Conclusion: These results imply that sex hormones exerted a suppressive effect on the function of TLR3 in the fallopian tube cell line when different concentrations of sex hormones were present. The current results also suggest that estrogen receptor beta and nuclear progesterone receptor B are likely to mediate the hormonal regulation of TLR3, as these two receptors are the main estrogen and progesterone receptors in OEE6/E7 cell line.

  • Objectives : Inflammation is important event in the development of vascular diseases including hypertension, atherosclerosis, and restenosis. Bodusan (BDS) was a traditional Korean herbal medicine and widely used in treatment of gastrointestinal complaint and stomach ulcer. The aim of this study was to determine whether BDS and its components inhibit production of nitrite, PGE2 and proinflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages. Methods : Cytotoxic activity of BDS and its components on RAW 264.7 cells was using 5-(3caroboxymcrophages. eth-oxyphenyj)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were detected by western blot. Results : Our results indicated that BDS and its components significantly inhibited the LPS-induced NO and PGE2 production. Moreover. BDS and its components inhibited iNOS and COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-11${\beta}$, IL-6 and MCP-1 formation in macrophages. Conclusions: These results indicate that BDS and its components have potential as an anti-inflammatory agent.

Down-regulation of Phosphoglucose Isomerase/Autocrine Motility Factor Enhances Gensenoside Rh2 Pharmacological Action on Leukemia KG1α Cells

  • Aims and Background: Ginsenoside Rh2, which exerts the potent anticancer action both in vitro and in vivo, is one of the most well characterized ginsenosides extracted from ginseng. Although its effects on cancer are significant, the underlying mechanisms remain unknown. In this study, we sought to elucidate possible links between ginsenoside Rh2 and phosphoglucose isomerase/autocrine motility factor (PGI/AMF). Methods: $KG1{\alpha}$, a leukemia cell line highly expressing PGI/AMF was assessed by western blot analysis and reverse transcription- PCR (RT-PCR) assay after transfection of a small interfering (si)-RNA to silence PGI/AMF. The effect of PGI/AMF on proliferation was measured by typan blue assay and antibody array. A cell counting kit (CCK)-8 and flow cytometry (FCM) were adopted to investigate the effects of Rh2 on PGI/AMF. The relationships between PGI/AMF and Rh2 associated with Akt, mTOR, Raptor, Rag were detected by western blot analysis. Results: KG1${\alpha}$ cells expressed PGI/AMF and its down-regulation significantly inhibited proliferation. The antibody array indicated that the probable mechanism was reduced expression of PARP, State1, SAPK/JNK and Erk1/2, while those of PRAS40 and p38 were up-regulated. Silencing of PGI/AMF enhanced the sensibility of $KG1{\alpha}$ to Rh2 by suppressing the expression of mTOR, Raptor and Akt. Conclusion: These results suggested that ginsenoside Rh2 suppressed the proliferation of $KG1{\alpha}$, the same as down-regulation of PGI/AMF. Down-regulation of PGI/AMF enhanced the pharmacological effects of ginsenoside Rh2 on KG1${\alpha}$ by reducing Akt/mTOR signaling.

Diagnostic Values of Serum Levels of Pepsinogens and Gastrin-17 for Screening Gastritis and Gastric Cancer in a High Risk Area in Northern Iran

  • Background: Gastric cancer (GC) is the second cause of cancer related death in the world. It may develop by progression from its precancerous condition, called gastric atrophy (GA) due to gastritis. The aim of this study was to evaluate the accuracy of serum levels of pepsinogens (Pg) and gastrin-17 (G17) as non-invasive methods to discriminate GA or GC (GA/GC) patients. Materials and Methods: Subjects referred to gastrointestinal clinics of Golestan province of Iran during 2010 and 2011 were invited to participate. Serum levels of PgI, PgII and G17 were measured using a GastroPanel kit. Based on the pathological examination of endoscopic biopsy samples, subjects were classified into four groups: normal, non-atrophic gastritis, GA, and GC. Receiver operating curve (ROC) analysis was used to determine cut-off values. Indices of validity were calculated for serum markers. Results: Study groups were normal individuals (n=74), non-atrophic gastritis (n=90), GA (n=31) and GC patients (n=30). The best cut-off points for PgI, PgI/II ratio, G17 and HP were $80{\mu}g/L$, 10, 6 pmol/L, and 20 EIU, respectively. PgI could differentiate GA/GC with high accuracy (AUC=0.83; 95%CI: 0.76-0.89). The accuracy of a combination of PgI and PgI/II ratio for detecting GA/GC was also relatively high (AUC=0.78; 95%CI: 0.70-0.86). Conclusions: Our findings suggested PgI alone as well as a combination of PgI and PgI/II ratio are valid markers to differentiate GA/GC. Therefore, Pgs may be considered in conducting GC screening programs in high-risk areas.

Tristetraprolin Inhibits the Growth of Human Glioma Cells through Downregulation of Urokinase Plasminogen Activator/Urokinase Plasminogen Activator Receptor mRNAs

  • Urokinase plasminogen activator (uPA) and urokinase plasminogen activator receptor (uPAR) play a major role in the infiltrative growth of glioblastoma. Downregulatoion of the uPA and uPAR has been reported to inhibit the growth glioblastoma. Here, we demonstrate that tristetraprolin (TTP) inhibits the growth of U87MG human glioma cells through downregulation of uPA and uPAR. Our results show that expression level of TTP is inversely correlated with those of uPA and uPAR in human glioma cells and tissues. TTP binds to the AU-rich elements within the 3' untranslated regions of uPA and uPAR and overexpression of TTP decreased the expression of uPA and uPAR through enhancing the degradation of their mRNAs. In addition, overexpression of TTP inhibited the growth and invasion of U87MG cells. Our findings implicate that TTP can be used as a promising therapeutic target to treat human glioma.

Intercellular Adhesion Molecule-1 Gene Polymorphism (K469E) in Korean Preeclamptic Women

  • 목 적: 자간전증은 임신 특이적 질환으로, 면역 반응 관련 결합 요소의 증가에 의한 광범위한 혈관내피 기능손상을 나타낸다. 자간전증을 이끄는 생태병리학적 기전에 관여하는 세포내부 결합요소-1 (intracellular adhesion molecule-1; ICAM-1)은 면역반응의 시작을 유도하고, 그것의 혈장 정도와 기능은 유전적으로 영향을 받는다. 그러므로 우리는 이번 연구에서 한국인 자간전증 임산부에서 ICAM-1의 유전자 다형성 부위인 K469E의 분포를 평가하고 자간전증과 이 유전자 다형성 사이의 연관성을 조사하였다. 대상 및 방법: 이 유전자 다형성은 SNapShot kit와 ABI Prism 3100 Genetic analyzer를 사용하여 197명의 자간전증임산부와 193명의 정상 임산부의 말초 혈액에서 분석하였다. 결 과: ICAM-1 유전자 다형성인 K469E의 유전자형과 대립유전자 빈도는 자간전증 임산부와 정상 임산부 사이에 차이가 없었다. 유전자형인 KK, KE, EE는 자간전증 임산부에서 각각 40.6%, 43.7%, 15.7%였고, 정상 임산부에서 38.9%, 45.1%, 16.1%였다. 그리고 K와 E 대립유전자 빈도는 자간전증 임산부에서 각각 0.62, 0.38이었고 정상 임산부에서 0.61, 0.39였다. 다중회귀분석결과에서 자간전증 발생 위험도는 K469E의 이종접합 유전자형 (KE)이나 돌연변이 유전자형(EE)을 수반하는 그룹에서 증가되지 않았다. 결 론: 이 연구는 ICAM-1 유전자 다형성인 K469E가 한국인 임신부의 자간전증 발생과 연관이 없음을 나타낸다.

The Y153H Variant of the STOX1 Gene in Korean Patients with Preeclampsia

  • 목 적: 자간전증은 유전적 환경적 요인에 의해 발생하는 다요인성 질환이다. 최근 네덜란드 산모에서의 자간전증 후보 유전자로 확인된 STOX1 유전자는 태반에서 발현되며 모계 대립유전자의 선택적 유전에 의해 각인된다. 본 연구에서는 STOX1 유전자의 단일염기변이형 부위인 Y153H와 한국인 자간전증의 연관성에 대하여 알아보고자 하였다. 대상 및 방법: 자간전증 산모 202과 임신기간 동안 자간전증이 발생하지 않은 정상 산모 204명의 혈액으로부터STOX1 유전자의 Y153H 변이 양상을 SNapShot assay kit와 ABI Prism 3730 DNA analyzer를 사용하여 분석하였다. 결과: STOX1 Y153H 변이의 유전자형 빈도는 자간전증환자군과 정상 대조군 간에 유의한 차이가 없었다(P>0.05). 또한 자간전증 환자군(87.1%)에서 대립유전자형인 H의 빈도도 정상 대조군(86.5%)과 유사하였다. 다중회귀분석결과 YH, HH, YH/HH 유전자형들은 자간전증 발생 위험에 있어서 연관성이 없었다. 결 론: 본 연구진은 처음으로 한국인 자간전증 환자에서 STOX1 유전자의 Y153H 변이형의 특징을 파악 하였다. 연구결과 자간전증 산모와 정상 산모 간의 유전자형과 대립유전자형의 빈도는 차이가 없었다. 비록 실험 대상군의 수적인 제한이 있지만, 이상의 결과를 통해서 한국인에서 STOX1 유전자의 Y153H 변이형은 자간전증과 연관성이 없는 것으로 사료된다.

통계적 분석을 통한 Insulin의 정상 참고치 설정 (Establishment of Reference Value of Insulin Using the Statistical Analysis)

  • Insulin은 공복시에 저혈당인 환자의평가, 당뇨병의 분류 및 예측, ${\beta}$-세포의 활성도 평가, 인슐린 내성 조사에 임상적 유용성이 있다. 이에 통계적 분석으로 insulin의 정상 참고치를 설정함으로써 insulin 검사의 유용성을 증가시키고자 한다. 2008년 5월부터 8월까지 본원 건진 센터에 내원한 환자 6648명 중 당뇨병의 가족력이 있는 사람, 당뇨 medication 중인 사람, 혈당 상승의 과거력이 있는 사람, 정상 공복혈당 100 mg/dL 이상인 사람, BMI 18.5~22.9 $kg/m^2$의 범위를 벗어나는 사람, HbA1c 6.5% 이상인 사람을 exclusion criteria로 설정하였다. Subgroup은(하위집단의 평균치 차/95% 참고범위 간격 차)${\times}$100의 산출값이 기준치 25%보다 큰 경우에 한하여 하위집단을 구분 설정하였다. 성별과 연령으로 subgroup의 참고치 설정여부를 확인하였고, Bayesian's method와 Hoffman's method로 통계적 분석을 실시하여 정상 참고치를 설정하였다. 남녀의 insulin 평균치 차이가 0.317로 산출값이 3.34%로 성별간 subgroup의 설정은 필요하지 않다. 데이터의 평균나이인 45세를 기준으로 한 산출값은 5.83%로 연령간 subgroup의 설정 또한 필요하지 않다. Bayesian 법으로 insulin의 평균은 5.403 uIU/mL로 표준편차 2.720으로 ${\pm}3SD$를 벗어난 94명을 제외한 6554명을 대상으로 분석한 결과 정상 참고치는 1.5~11.0 uIU/mL이다. Hoffmann 법을 통한 통계적 분석은 insulin값을 log insulin값으로 변환하여 정규분포를 만든 후${\pm}3SD$를 벗어난 9명을 제외한 6639명을 대상으로 분석한 결과, 정상 참고치가 1.8~12.8 uIU/mL이다. Kit 내 보고되어 있는 정상 참고치 1.7~11.8 uIU/mL와 비슷한 결과 값인 Hoffmann 법으로 통계적 분석한 결과인 1.8~12.8 uIU/mL로 insulin 정상 참고치를 설정하였다. 이는 당뇨병 평가에 중요한 역할을 하는 insulin의 정상 참고치를 설정함으로써 검사의 유용성을 높일 것으로 여겨진다.
