  • The purpose of this research was to study the kinetics of a packed bed aerobic biofilm process. Experiments were carried out by feeding an aerated packed bed reactor with a synthetic wastewater. The reactor packed with glass beads as media had a nominal hydraulic detention time of 5 hours. The flow pattern in the reactor was determined by a tracer test using a NaCl solution to be a completely-mixed type. The results of the research showed that the F/M ratio in the reactor was almost constant since the the biomass increased due to the growth of biofilm as the volumetric organic loading increased. It was also proved theoretically and experimentally that packed bed aerobic biofilm process could be analyzed by the kinetics of completely-mixed activated sludge process with sludge recycle.

  • Scale effects of stability, run-up, run-down and reflection of layered coastal structures are investigated through the experiments with 7 kinds of hydraulic scale models. The occurrence mechanism and the control method of scale effects are also discussed. As a result, it is found that the similarity of permeability of inner layers plays an important role in the occurrence of scale effects, which has been neglected in the most of conventional model tests. To assure the best scale effects for permeable coastal structures, control of Reynolds numbers of the porous media flow in each layer is recommended. It is also found that Reynolds numbers in revetment, filter, and core layer must be greater than $2{\times}10^4$, $3{\times}10^3$, and $1{\times}10^3$, respectively.

  • An aim of the study is to develop an efficient numerical simulation technique that can handle the two-scale analysis of fluid permeation filters fabricated by the partial sintering technique of small spherical ceramics. A solid-fluid mixture homogenization method is introduced to predict the mechanical characters such as rigidity and permeability of the porous ceramic filters from the micro-scale geometry and configuration of partially-sintered particles. An extended finite element (X-FE) discretization technique based on the enriched interpolations of respective characteristic functions at fluid-solid interfaces is proposed for the non-interface-fitted mesh solution of the micro-scale analysis that needs non-slip condition at the interface between solid and fluid phases of the unit cell. The homogenization and localization performances of the proposed method are shown in a typical two-dimensional benchmark problem whose model has a hole in center. Three-dimensional applications to the body-centered cubic (BCC) and face-centered cubic (FCC) unit cell models are also shown in the paper. The 3D application is prepared toward the computer-aided optimal design of ceramic filters. The accuracy and stability of the X-FEM based method are comparable to those of the standard interface-fitted FEM, and are superior to those of the voxel type FEM that is often used in such complex micro geometry cases.

  • Emerging issues related with fully coupled Thermo-Hydro-Mechanical (THM) behavior of unsaturated soil demand the development of a numerical tool in diverse geo-mechanical and geo-environmental areas. This paper presents general governing equations for coupled THM processes in unsaturated porous media. Coupled partial differential equations are derived from three mass balances equations (solid, water, and air), energy balance equation, and force equilibrium equation. With Galerkin formulation and time integration of these governing equations, finite element code is developed to find nonlinear solution of four main variables (displacement-u, gas pressure-$P_g$), liquid pressure-$P_1$), and temperature-T) using Newton's iterative scheme. Three cases of numerical simulations are conducted and discussed: one-dimensional drainage experiments (u-$P_g-P_1$), thermal consolidation (u-$P_1$-T), and effect of pile on surrounding soil due to surface temperature variation (u-$P_1$-T).

  • In this study, the coupled-numerical model has been newly developed to investigate numerically scouring and deposition around a coastal structure like a submerged breakwater using a numerical wave model and a lagrangian particle model for sand transport. As a numerical wave model, LES-WASS-2D (Hur and Choi, 2008) is adopted. The model is able to consider the flow through a porous midium with inertial, laminar and turbulent resistance term and determine the eddy viscosity with LES turbulence model. Distinct element method (Cundall and Strack, 1979), which is able to apply to many dynamical analysis of particulate media, as a lagrangian particle model for sand transport is newly coupled to the numerical wave model. The numerical simulation has been carried out to examine the scour problem in front of an impermeable submerged breakwater using the newly coupled-numerical model. The numerical results has been compared qualitatively with an existing experimental data and then its applicability has been discussed.

  • Objectives: This study evaluated the effect of aqueous extract from roots of Siberian ginseng on mTORC1 pathway. Methods: mTORC1 activity was measured by the phosphorylation status of p70 S6 kinase (S6K) in HeLa cells as well as the brain, liver and muscle tissues in diabetic db/db mice. Autophagy induction after the treatment of Siberian ginseng extract was evaluated by monitoring the conversion of cytoplasmic LC3I into lipidated LC3II in cultured human HeLa GFP-LC3 cells. Cell cycle analysis was performed in HeLa cells treated with Siberian ginseng using flow cytometry. Results: Among >2,800 plant products used for oriental medicine, Siberian ginseng was found to inhibit mTORC1 to phosphorylate S6 kinsase (S6K) in HeLa cells as well as the brain, liver and muscle tissues in diabetic db/db mice. Siberian ginseng-mediated mTORC1 activity was reversible unlike the prolonged suppression of mTORC1 by rapamycin when HeLa cells were grown in fresh media after the removal of the inhibitors. Siberian ginseng extract at concentrations to inhibit mTORC1 was not overly cytotoxic in cultured HeLa cells whereas rapamycin was obviously cytotoxic. The conversion of cytoplasmic LCI into lipidated LCII was increased by fivefold in HeLa GFP-LC3 cells treated with Siberian ginseng extract. Progression of cell cycle was attenuated at G2/M phase by the treatment of Siberian ginseng extract. Conclusions: These results suggest that the aqueous extract of Siberian ginseng possibly plays a good therapeutic role in various diseases involving mTORC1 signaling.

  • Water quality in the public water course has been polluted more seriously than ever before due to the increase of the number and aremount of pollution sources such as domestic and industrial wastewater. For water quality conservation, the Korean government has been trying to construct sewage treatment facilities continually, of which treatment capacity reached to 11,452,400m$^{3}$/day in 1996. Night soil treatment facilites of m nationwide have the treatment capacity of 24,038m$^{3}$/day. But water quality has not been improved because the sewer systems were insufficient and the treatment efficiencies of sewage were not high, enough. For renovation of water quality, miscellaneous domestic wastewater must be treated because 27g BOD/day out of total 40g BOD/person-day come from miscellaneous wastewater, comparing to 13g BOD/day from night soil. However, sole treatment purifier treat only night soil from the flushing toilet. Therefore, it may be desirable to treat the miscellaneous domestic wastewater and the night soil from flushing toilet together by joint treatment purifier system as on-site domestic wastewater treatment technology. In Korea, the joint treatment purifier system, introduced in 1997, have the benefit as follows; i) good water poiluion control effect, ii ) good effect on river water flow, iii) water pollution control with sewage treatment facility, and iv) rapid pollution control effect, etc. In order to achieve a good effect as stated before, i ) strengthening effluent guideline including BOD, nitrogen and phosphorus, ii ) specializing operation to maintain high performance, and iii) supporting its construction and maintenance costs by the governmental level may be necessary: In addition, automation system of joint treatment purifier, technology for its package and compactness, and a new bio-media bio-filter with higher capacity should be further developed in agreement with a more stringent effluent guideline.

  • Particle morphology change and different experimental condition analysis during composite fabrication process by traditional ball milling with discrete element method (DEM) simulation were investigated. A simulation of the three dimensional motion of balls in a traditional ball mill for research on the grinding mechanism was carried out by DEM simulation. We studied the motion of the balls, the ball behavior energy and velocity; the forces acting on the balls were calculated using traditional ball milling as simulated by DEM. The effect of the operational variables such as the rotational speed, ball material and size on the flow velocity, collision force and total impact energy were analyzed. The results showed that increased rotation speed with interaction impact energy between balls and balls, balls and pots and walls and balls. The rotation speed increases with an increase of the impact energy. Experiments were conducted to quantify the grinding performance under the same conditions. Furthermore, the results showed that ball motion affects the particle morphology, which changed from irregular type to plate type with increasing rotation speed. The evolution was also found to depend on the impact energy increase of the grinding media. These findings are useful to understand and optimize the particle motion and grinding behavior of traditional ball mills.
