Lipoxygenase Inhibitors Suppressed Carrageenan-Induced Fos-Expression and Inflammatory Pain Responses in the Rat

  • Lipoxygenase (LO) metabolites are generated in inflamed tissues. However, it is unclear whether the inhibition of the LO activity regulates the expression of c-Fos protein, a pain marker in the spinal cord. Here we used a carrageenan-induced inflammation model to examine the role of LO in the development of c-Fos expression. Intradermally injected carrageenan caused elevated number of cells exhibiting Fos-like immunoreactivity (Fos-LI) in the spinal dorsal horn, and decreased the thermal and mechanical threshold in Hargreaves and von Frey tests. Pretreatment with an inhibitor of phospholipase A2, that generates the LO substrate, prior to the carrageenan injection significantly reduced the number of Fos-(+) cells. A general LO inhibitor NDGA, a 5-LO inhibitor AA-861 and a 12-LO inhibitor baicalein also exhibited the similar effects. Moreover, the LO inhibitors suppressed carrageenan-induced thermal and mechanical hyperalgesic behaviors, which inidcates that the changes in Fos expression correlates with those in the nociceptive behaviors in the inflamed rats. LO products are endogenous TRPV1 activators and pretreatment with BCTC, a TRPV1 antagonist inhibited the thermal but not the mechanical hypersensitivity. Overall, our results from the Fos-LI and behavior tests suggest that LO products released from inflamed tissues contribute to nociception during carrageenan-induced inflammation, indicating that the LO pathway is a possible target for modulating inflammatory pain.

Grain size measurement based on marked watershed algorithm (유역분할 알고리즘을 이용한 결정립 크기 측정)

  • Grain size of material is important factor in evaluating mechanical properties. Methods for grain size determination are described in ASTM grain size standards. However, conventional method require pretreatment of the surface to clarify grain boundaries. In this study, the grain size from the surface image obtained from scanning electron microscope was measured using the watershed algorithm, which is a region-based method among image segmentation techniques. The shapes of the crystals are similar to each other, but the size and growth height are different. In addition, crystal grains are adjacent to each other, so it is very similar to the shape image of the topography. Therefore, grain boundaries can be efficiently detected using the Watershed algorithm.

Effects of Bupivacaine Pretreatment at the Periphery on the Expression of Mechanical Hyperalgesia in a Rat Model of Peripheral Neuropathy (백서의 말초신경 통증 모델에서 신경결찰 전 말초에 주입된 Bupivacaine이 기계적 통각과민의 발현에 미치는 영향)

  • Background: Although several mechanisms of causalgia, which results from a partial injury to the peripheral nerve trunk, have been proposed, whether or not antidromic impulses from the injured neurons contribute to the development of the mechanical hyperalgesia has not been studied. The purpose of this experiment is was to investigate the role of antidromic impulses to the peripheral sensory receptor site on the development of mechanical hyperalgesia in a rat model of peripheral neuropathy. Methods: Rats were prepared with tight ligation of by tightly ligating the left fifth and sixth lumbar spinal nerves. The effect of bupivacaine pretreatment on the development of mechanical hyperalgesia was evaluated by injecting 0.5% bupivacaine (0.3 ml) into the plantar surface of the left hind paw before the skin incision was made. For the control group, normal saline (0.3 ml) was injected instead of bupivacaine. To measure the mechanical hyperalgesia, paw withdrawal thresholds were measured using a series of von Frey hairs. Mechanical hyperalgesia was measured a the day before, and 1, 2, 3, 4, 7, and 14 days after the surgery. Results: The control group showed decreased withdrawal thresholds from the day after the surgery (the values were $14.0{\pm}0.5$, $8.9{\pm}1.3$, $8.4{\pm}1.6$, $6.9{\pm}1.2$, $8.8{\pm}1.5$, $10.5{\pm}1.3$, and $8.6{\pm}1.3$ g; at -1, 1, 2, 3, 4, 7, and 14 days after the surgery, respectively). However, withdrawal thresholds of the bupivacaine-pretreated group showed increased withdrawal thresholds for three days after the surgery ($14.5{\pm}0.3$, $12.6{\pm}1.4$, $12.7{\pm}1.1$, $10.5{\pm}1.3$ g; at 1, 1, 2, 3 days after the surgery). Conclusions: Our result suggests that antidromic impulses to the peripheral sensory receptors are at least partly responsible for the initial development of mechanical hyperalgesia in a rat model of peripheral neuropathy.

The Involvement of Protein kinase C in Glutamate-Mediated Nociceptive Response at the Spinal Cord of Rats (흰쥐의 척수에서 Glutamate가 매개하는 Nociceptive Response에 있어서 Protein kinase C의 관련성)

  • When glutamate was infected intrathecally, the result is similar to those produced by TPA injected. The involvement of protein kinase C (PKC) in the nociceptive responses in rat dorsal horn neurons of lumbar spinal cord was studied. In test with formalin, a PKC inhibitor (chelerythrine) inhibited dose-dependently the formalin-induced behavior response. Neomycin also inhibited it significantly. But, a PKC activator (12-O-tetradecanoylphorbol-13-ester, TPA) showed reverse effect. When gluatamate was injected intrathecally, we observed the result is smilar to those produced by TPA injection. On the other hand, intrathecal injection of glutamate induced thermal and mechanical hyperalgesia. In Tail-flick test, we examined the involvement of PKC on the glutamate-indeced thermal hyperalgesia. Chelerythrine showed an inhibitory effect and TPA enhanced thermal response. Glutamate decreased the mechanical threshold significantly. A pretreatment of chelerythrine and neomycin inhibited glutamate-induced mechanical hyperalgesia, but the effect of neomycin was not significant. TPA had little effect on the mechanical nociceptive response. These results suggest that the PKC activation through metabotropic receptor at postsynaptic region of spinal cord dorsal horn neurons may influence on the persistent nociception produced by chemical stimulation with formalin, thermal and mechanical hyperalgesia induced by glutamate.

Effect of Mechanical Polishing Pretreatment on Tribological Properties of Manganese Phosphate Coating of Carbon Steel (기계적 연마 전처리가 인산망간 피막의 윤활 특성에 미치는 영향)

  • In this study, the effect of mechanical polishing of carbon steel on the tribological properties of manganese phosphate coating on carbon steel has investigated. The microstructure, surface morphology and chemical composition were analyzed by SEM, EDS, and XRD. The surface roughness test was carried out in order to calculate Rvk value by 3D laser microscopy. Also, the tribology property of manganese phosphate coating was tested by ball-on disk. In the results of EDS analysis, coating layer consists of elements such in Mn, P, Fe, and O. XRD showed that (Mn,Fe)5H2(PO4)4·4H2O in manganese phosphate coating layer was formed by the chemical reaction between manganese phosphate and elements in carbon steel. As the mechanical polishing degree increased, the friction coefficient was reduced. The rougher the mechanical polishing degree, the better corrosion resistance was obtained.

The Effect of Solubilization Pretreatment Process on Anaerobic Digestion of Waste Activated Sludge (전처리 가용화 공정이 잉여슬러지 혐기성 소화효율에 미치는 영향)

  • COD properties of waste activated sludge (WAS) were investigated for various solubilization rate of mechanical pretreatment method in anaerobic digestion process. Inert COD was 37.0% of total COD in untreated WAS. Particulate biodegradable COD was converted to soluble biodegradables and particulate unbiodegradables as solubilization was processed. Particulate unbiodegradable portion of COD in WAS can be increased as particulate biodegradable portion is decreased in case of relatively long SRT of biological treatment. Thus, COD properties of WAS should be investigated in case of relatively low particulate biodegradable COD, because of possible low effect of solubilization. COD removal rate in anaerobic digester was enhanced as much as 2.1% and 15.1% for solubilization rate 5% and 35% due to pretreatment, respectively. COD removal rate was increased from 25% to 40%, and methane gas generation was increased from $607m^3/d$ to $907m^3/d$ as particulate COD of WAS was solubilized to 35% in pretreatment facilities.

Studies on the Properties of Mechanical Pulp from Italian Poplar Wood(Populus euramericana I-476) by the Age of Tree (수령(樹齡)에 의한 포플러펄프의 성질(性質)에 관(關)한 연구(硏究))

  • The first step to utilize the growing resources of Italian poplar (Populus euramericana I-476) for pulp-Woods, its characteristics and adaptabilities to the pulp industry must be investigated completely. The plantation methods are important for its fast growing in stock, and no less important is the cutting age for its utilization as pulpwood. In this paper, the stone groundwood pulping, refiner groundwood pulping and chemi-groundwood pulping characteristics by the age of tree, along with their physical and chemical characterstics were tested, and relationships between the age groups were analyzed to find out the optimum felling age. The results obtained are as follows: 1. The coefficient of pliability was a little higher in the case of younger trees. 2. The water retention value of each pulp was directly proportional to its physical strength, but this tendency was not detected between the age groups of sample woods. 3. Generally, the physical strength of younger wood pulp was lower regardless of the pulping process. But in the case of pretreatment with NaOH, Asphund and CGP pulp from 5 year old sample wood were stronger in physical strengths than those of GP and Asplund pulp with no pretreatment from 10 years old sample wood. 4. The tear factor of Asplund pulp with alkali pretreatment was higher than that of CGP pulp but the breaking length and the burst factor was similar in all processes. Considering the pulp yield and its brightness, CGP process seems to be advantageous. 5. The dissimilarity of physical strength between 7 and 10 years old wood pulp was not very large in all pulping processes but the physical strength of 5 year old wood pulp was very weak. In the of groundwood pulping from Italian poplar woods, 5 year old wood pulp should be mixed with other long fiber pulp for making a good paper.

Effect of Radix Trichosanthis on the Melanogenesis (天花粉이 멜라닌형성에 미치는 影響)

  • Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Radix Trichosanthis on the basal melanogenic activities of B16/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Radix Trichosanthis alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Radix Trichosanthis also suppressed the increase of ${\alpha}$-MSH (10 nM) or forskolin (20${\mu}M$)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Radix Trichosanthis also inhibited the increase of forskolin($20{\mu}M$) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Radix Trichosanthis showed less intensity than B16 melanoma cells stimulated with ${\alpha}$- MSH or forskolin. These results suggest that Radix Trichosanthis inhibits melanogenesis and abrogates ${\alpha}-MSH and cAMP-induced melanogenesis in B16 melanoma cells.

Available Technology and Integrated Management Plan for Energy-positive in the Sewage Treatment Plant (에너지 생산형 하수처리장을 위한 가용 기술과 통합관리 방안)

  • Because of the intensified environmental problems such as climate change and resource depletion, sewage treatment technology focused on energy management has recently attracted attention. The conversion of primary sludge from the primary sedimentation tank and excessive sludge from the secondary sedimentation tank into biogas is the key to energy-positive sewage treatment. In particular, the primary sedimentation tanks recover enriched biodegradable organic matter and anaerobic digestion process produces methane from the organic wastes for energy production. Such technologies for minimizing oxygen demand are leading the innovation regarding sewage treatment plants. However, sewage treatment facilities in Korea lack core technology and operational know-how. Actually, the energy potential of sewage is higher than sewage treatment energy consumption in the sewage treatment, but current processes are not adequately efficient in energy recovery. To improve this, it is possible to apply chemically enhanced primary treatment (CEPT), high-rate activated sludge (HRAS), and anaerobic membrane bioreactor (AnMBR) to the primary sedimentation tank. To maximize the methane production of sewage treatment plants, organic wastes such as food waste and livestock manure can be digested. Additionally, mechanical pretreatment, thermal hydrolysis, and chemical pretreatment would enhance the methane conversion of organic waste. Power generation systems based on internal combustion engines are susceptible to heat source losses, requiring breakthrough energy conversion systems such as fuel cells. To realize the energy positive sewage treatment plant, primary organic matter recovery from sewage, biogas pretreatment, and co-digestion should be optimized in the energy management system based on the knowledge-based operation.
