The tube formation assay is a widely used in vitro experiment model to evaluate angiogenic properties by measuring the formation of tubular structures from vascular endothelial cells (ECs). In vitro experimental results are crucial when considered the advisability of moving forward to in vivo studies. Thus, the additional attentions to the in vitro assay is necessary to improve the quality of the pre-clinical data, leading to better decision-making for successful drug discovery. In this study, we improved the tube formation assay system in three aspects. First, we used human endothelial colony forming cells (ECFCs), which are endothelial precursors that have a robust proliferative capacity and more defined angiogenic characteristics compared to mature ECs. Second, we utilized a real-time cell recorder to track the progression of tube formation for 48 hours. Third, to minimize analysis error due to the limited observation area, we used image-stitching software to increase the microscope field of view to a $2{\times}2$ stitched area from the $4{\times}$ object lens. Our advanced tube formation assay system successfully demonstrated the time-dependent dynamic progression of tube formation in the presence and absence of VEGF and FGF-2. Vatalanib, VEGF inhibitor, was tested by our assay system. Of note, $IC_{50}$ values of vatalanib was different at each observation time point. Collectively, these results indicate that our advanced tube formation assay system replicates the dynamic progression of tube formation in response to angiogenic modulators. Therefore, this new system provides a sensitive and versatile assay model for evaluating pro- or anti-angiogenic drugs.
When a building suffers damages under moderate to severe loading condition, its physical properties such as damping and stiffness parameters will change. There are different practical methods besides various numerical procedures that have successfully detected a range of these changes. Almost all the previous proposed methods used to work with translational components of mode shapes, probably because extracting these components is more common in vibrational tests. This study set out to investigate the influence of using both rotational and translational components of mode shapes, in detecting damages in 3-D steel structures elements. Three different sets of measured components of mode shapes are examined: translational, rotational, and also rotational/translational components in all joints. In order to validate our assumptions two different steel frames with three damage scenarios are considered. An iterative model updating program is developed in the MATLAB software that uses the OpenSees as its finite element analysis engine. Extensive analysis shows that employing rotational components results in more precise prediction of damage location and its intensity. Since measuring rotational components of mode shapes still is not very convenient, modal dynamic expansion technique is applied to generate rotational components from measured translational ones. The findings indicated that the developed model updating program is really efficient in damage detection even with generated data and considering noise effects. Moreover, methods which use rotational components of mode shapes can predict damage's location and its intensity more precisely than the ones which only work with translational data.
In general, the design of structures and its construction processes are fundamentally dependent on their foundation and supporting ground. Thus, it is imperative to understand the behavior of the soil under certain stress and drainage conditions. As it is well known that certain characteristics and behaviors of soils with fines are highly dependent on water content, it is critical to accurately measure and identify the status of the soils in terms of water contents. Liquid limit is one of the important soil index properties to define such characteristics. However, liquid limit measurement can be affected by the proficiency of the operator. On the other hand, dynamic properties of soils are also necessary in many different applications and current testing methods often require special equipment in the laboratory, which is often expensive and sensitive to test conditions. In order to address these concerns and advance the state of the art, this study explores a novel method to determine the liquid limit of cohesive soil by employing video-based vibration analysis. In this research, the modal characteristics of cohesive soil columns are extracted from videos by utilizing phase-based motion estimation. By utilizing the proposed method that analyzes the optical flow in every pixel of the series of frames that effectively represents the motion of corresponding points of the soil specimen, the vibration characteristics of the entire soil specimen could be assessed in a non-contact and non-destructive manner. The experimental investigation results compared with the liquid limit determined by the standard method verify that the proposed method reliably and straightforwardly identifies the liquid limit of clay. It is envisioned that the proposed approach could be applied to measuring liquid limit of soil in practical field, entertaining its simple implementation that only requires a digital camera or even a smartphone without the need for special equipment that may be subject to the proficiency of the operator.
다양한 기능의 새로운 물질을 합성하고 정적 및 동적 특성을 조사하기 위해서는 매우 안정된 고온 및 고압 조건을 만드는 것이 중요하다. 새로운 물질을 제조하는 데 필요한 다이아몬드 엔빌 셀 (DAC)을 사용하여 Mbar 범위의 고압 조건은 이미 확보되었다. 이 연구에서는 DAC와 결합된 레이저 가열 시스템을 설계하였으며, DAC의 양쪽 방향에서 시료를 가열하기 위해 2 대의 1064 nm, 100 W 파이버 레이저를 사용했을 뿐만 아니라 3 대의 분광기를 사용하여 온도, 압력 및 라만 스펙트럼을 측정하였다. 고압 실험에서 시료를 가두는 구멍으로 일반적으로 사용되는 스테인리스 강 개스킷을 가열하여 열 방사선원을 만들고, 이 방사선원으로부터 방출되는 복사 스펙트럼을 획득하여 가열된 개스킷의 온도를 측정하였다. 이 기술을 적용하여 다양한 물질들을 만들고 극한 조건에서 물리적 특성을 조사 할 수 있다.
Objective: The purpose of this study was to investigate the viscoelastic properties related to handling characteristics of composite resins, Methods: A custom designed vertical oscillation rheometer (VOR) was used for rheological measurements of composites. The VOR consists of three parts: (1) a measuring unit, (2) a deformation induction unit and (3) a force detecting unit, Two medium viscous composites, Z100 and Z250 and two packable composites, P60 and SureFil were tested. The viscoelastic material function, including complex modulus $E^{*}$ and phase angle ${\delta}$, were measured. A dynamic oscillatory test was used to evaluate the storage modulus (E'), loss modulus (E") and loss tangent ($tan{\delta}$) of the composites as a function of frequency ($\omega$) from 0.1 to 20 Hz at $23^{\circ}C$. Results: The E' and E" increased with increasing frequency and showed differences in magnitude between brands. The $E^{*}s$ of composites at ${\omega}{\;}={\;}2{\;}Hz$, normalized to that of Z100, were 2.16 (Z250), 4,80 (P60) and 25.21 (SureFil). The magnitudes and patterns of the change of $tan{\delta}$ of composites with increasing frequency were significantly different between brands. The relationships between the complex modulus $E^{*}$, the phase angle ${\delta}$ and the frequency \omega were represented by frequency domain phasor form, $E^{*}{\;}(\omega){\;}={\;}E^{*}e^{i{\delta}}{\;}={\;}E^{*}{\angle}{\delta}$. Conclusions: The viscoelasticity of composites that influences handling characteristics is significant different between brands, The VOR is a relatively simple device for dynamic, mechanical analysis of high viscous dental composites. The locus of frequency domain phasor plots in a complex plane is a valuable method of representing the viscoelastic properties of composites.
본 연구의 대상 공법인 저유동성 그라우팅 주입공법은 모르타르를 주입재로 하여 비배출 치환원리에 의해 지중에 압입하는 공법으로서 지반의 밀도 증대효과를 기대할 수 있고 저소음, 저공해, 내구성 확보와 함께 경우에 따라서는 높은 주입 고결체의 강도를 이용한 기초파일로 사용할 수도 있다. 저유동성 주입공법으로 지반의 동적 특성이 향상됨을 확인하기 위하여 현장에서 시험시공을 실시하고 물리시험을 수행하였으며 코어에 대한 실내시험을 통해 지중에 형성된 개량체 품질을 확인하고 코어회수율 분석을 수행하였다. 밀도검층은 감마선의 컴프턴 산란을 이용하여 지층의 체적 밀도를 산정하였고, 음파검층은 시추공 내에서 음파발생 및 수신장치로 구성된 검층기를 이용하여 시추공 주변의 지반에 대해서 시험을 실시하였다. 그라우팅 전,후 물성변화(밀도검층, 음파검층)를 측정한 결과, 기본적으로 두 물성 모두 그라우트제 주입 후 물성이 증가하여 측정되었다. 다만, 그라우팅 후 속도 증가보다 밀도 증가의 변화 폭이 컸으며, 지반 밀도 측정 방법이 주입재의 충진 효과를 측정하는데 효과적이다. 재령 28일 후 채취한 시료로부터 강도 및 코어회수율을 측정하였으며, 시험결과 개량체의 확산 및 강도시험 결과가 설계기준을 만족시켜 내진성능보강을 만족한 것으로 확인하였다.
본 연구에서는 배향도가 높은 스트랜드와 무배향성의 파티클을 혼합해서 다양한 층구조의 복합체를 제조하여 각종 탄성률을 측정하여 비교하였으며, 초음파법을 적용하여 이방성 적층재료의 층구조에 기인하는 이방성의 정도를 예측하기 위하여 초음파 전파속도와 복합체 면내의 이방성의 관계에 대하여 검토했다. 그 결과 각층구조 복합체의 정적 휨 탄생률 $E_S$는 동적 탄성률 $E_D$와 거의 일치하였으며 $E_U$는 높게 산출되는 경향을 나타내었다. 초음파법을 이용한 복합체의 최외층 스트랜드의 섬유방향(${\parallel}$)과 직각방향(${\bot}$)의 전파속도의 차는 3층 구조, 7층 구조, PB 순으로 크게 나타나 층구조에 의한 이방성의 정도를 나타내고 있었다. 또한 복합체 면내의 양방향간의 전파속도의 차는 스트랜드 표층구조가 파티클 표층구조보다 큰 값을 나타내었으며 스트랜드의 혼합률이 높을수록 각 방향의 전파속도는 빨라지는 경향이었다. 이와는 반대로, 복합체의 두께방향의 전파속도는 파티클 표층구조와 스트랜드의 혼합률이 작을수록 빨라지는 경향을 나타내었다.
$CO_2$ Welding which uses $CO_2$ instead of inert gas is most widely used in industrial sites. Welding rod for $CO_2$ Welding is roughly divided into solid wire and flux cored wire. $CO_2$ Welding has higher efficiency than any other welding methods, and also economic and speedy to handle, that's why is used frequently for welding general structures. As most of studies about $CO_2$ Welding are focused on metallurgical changes of successful joints, they developed theories about the change of configuration on weld parts. This study is especially focused on not only the change of configuration on weld parts, but also the change of the penetrating depth through changing the position of welding torch. For inspection, applied AWS A5.20 E70-1 among welding wires and fixed moving angles of torch, but controled the values of voltage and the position of welding. Also Automatic Feed Mechanism is used for exact movement of material, specimen is a piece of steel for general structures. By measuring and analyzing the configuration of sliced section and the values of welding leg length and welding throat after welding, the outcome about the changes turned out.
현재까지 서비스지향 아키텍처(SOA) 보안은 개별 표준들과 솔루션에 의한 대응에 집중해있을 뿐 전사적인 위험관리 차원의 통합적인 방법론은 부족한 실정이다. 따라서 SOA 보안 취약점 식별을 통한 다양한 보안 위협들에 대한 사전 분석과 대책 수립이 필요하다. 이를 위해 본 논문에서는 SOA 취약성을 정량적으로 분석하기 위한 SOA 동적 특성을 이용한 메트릭 기반의 취약성 평가 방법을 제안한다. SOA를 구성하는 서비스들 사이의 실행시간 종속성을 측정하여 서비스와 아키텍처 수준의 취약성을 평가한다. 서비스 사이의 실행시간 종속성은 한 서비스가 취약할 때 다른 서비스들이 얼마나 영향을 받게 되는지를 분석하기 위해 사용되는 중요한 특징이다. 한 서비스가 공격에 노출될 때 그 서비스에 종속된 서비스들도 역시 공격가능성이 높아진다. 따라서 실행시간 종속성은 SOA 아키텍처 수준의 취약성에 대한 지표로 활용할 수 있다.
불포화 폴리에스테르(unsaturated polyester : UPE) 수지를 다가알콜 (propylene glycol : PG, neopentyl glycol : NPG)의 각 몰비를 조정하여 포화 이염기산(isophthalic acid : IPA)과 불포화 이영기산 (maleic anhydride : MA)의 혼합물로 축합반응으로 제조하였다. 고체의 표면특성을 결정짓는 임계표면장력(${\gamma}_{c}$)을 Zisman plot으로 평가하였고, 또한 수지의 구조와 물성과의 관게를 레올로지 측정으로 조사하였다. 고체 glass에 의한 (${\gamma}_{c}$)은 UPE 수지 액체들에 대해서 30.5mN${\cdot}m^{-1}$를 얻었고, PG/NPG 몰비중 NPG 함량이 증가할수록 UPE 수지 용액의 접촉각과 표면장력이 감소됨을 확인할 수 있었다. 그리고 글리콜 몰비가 다른 UPE 수지의 동적 점탄성을 측정하면 정상류 점도의 전단속도 의존성, 동적 탄성율의 각 주파수 의존성에 의해 순수한 PG보다는 NPG의 함량이 증가할수록 낮은 값을 얻을 수 있었다.
