The Expression of Adhesion Molecules on Alveolar Macrophages and Lymphocytes and Soluble ICAM-1 Level in Serum and Bronchoalveolar Lavge(BAL) Fluid of Patients with Diffuse Interstitial Lung Diseases(DILD) (간질성 폐질환환자들의 기관지 폐포세척액내 폐포 대식세포와 임파구의 접착분자 발현 및 Soluble ICAM-1 농도에 관한 연구)

  • Background: The expression of the adhesion molecules on the cell surface is important in the movement of cells and the modulation of immune response. DILD starts as an alveolitis and progresses to pulmonary fibrosis. So adhesion molecules in these patients is expected to be increased. There are several reports about adhesion molecules in DILD in terms of the percentage of positive cells in immuno-stain, in which the interpretation is subjective and the data were variable. Methods: So we measured the relative median fluorescence intensity(RMFI) which is the ratio of the FI emitted by bound primary monoclonal antibody to FI emitted by isotypic control antibody of the cells in BALF of 28 patients with DILD(IPF:10, collagen disease:7, sarcoidosis:9, hypersensitivity pneumonitis:2) and 9 healthy control. Results: RMFI of the ICAM-1 on AM($3.30{\pm}1.16$) and lymphocyte($5.39{\pm}.70$) of DILD were increased significantly than normal control($0.93{\pm}0.18$, $1.06{\pm}0.21$, respectively, p=0.001, P=0.003). RMFI of the CD18 on lymphocyte was also higher($24.9{\pm}14.9$) than normal($4.59{\pm}3.77$, p=0.0023). And there was a correlation between RMFI of ICAM on AM and the % of AM(r=-0.66, p=0.0001) and lymphocyte(r=0.447, p=0.0116) in BALF. Also RMFI of ICAM on lymphocyte had a significant (r=0.593, p=0.075) correlation with the % of IL-2R(+) lymphocyte in BALF. The soluble ICAM(sICAM) in serum was also significantly elevated in DILD($499.7{\pm}222.2\;ng/ml$) compred to normal($199.0{\pm}38.9$) (p=0.00097) and sICAM in BAL fluid was also significantly higher than normal control group($41.8{\pm}23.0\;ng/ml$ vs $20.1{\pm}13.6\;ng/ml$). There was a Significant correlation between sICAM level in serum and the expression of ICAM-l on AM(r=0.554, p=0.0259).Conclusion: These data suggest that in DILD the expression of adhesion molecules is increased in the AM and BAL lymphocytes with elevated serum sICAM, and these parameter may be useful in determining disease activity.

Antimicrobial Effects against Food-Borne Pathogens and Antioxidant Activity of Rhododendron brachycarpum Extract (만병초 추출물의 식중독 유발균에 대한 항균효과 및 항산화활성)

  • This study was performed to investigate the antimicrobial effects against food-borne pathogens and antioxidant activity of Rhododendron brachycarpum ethanol-extract. The antimicrobial activity of the extract was determined using a paper disc-diffusion method, and the diameter of the clear zone was measured. The diameter of the clear zone in the presence of 10 mg of extract was maximal against Bacillus cereus among the three tested Gram-positive bacteria and against Escherichia coli O157:H7 among the five tested Gram-negative bacteria. Analysis of the minimum inhibitory concentration (MIC) showed that the extract exhibited a similar efficacy as that of sorbic acid, a well-known chemical preservative. The growth inhibitory effects of the extract at concentrations of 250, 500, 1,000, and 2,000 mg/L on food-borne pathogens were determined against Staphylococcus aureus, Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7. Growth of the microorganisms was not affected by the extract at concentrations up to 250 mg/L, but it was significantly (p<0.05) inhibited by the extract at concentrations higher than 1,000 mg/L. The antioxidant effects of the extract were examined via measurement of DPPH radical scavenging activity, inhibition of reactive oxygen species (ROS) generation using fluorescent dichlorofluorescien (DCF) assay, and prevention of peroxyl radical- and hydroxyl radical-induced supercoiled DNA breakage. The $IC_{50}$ of the extract for DPPH radical scavenging activity was about half that of ${\alpha}$-tocopherol, which was used as a positive control. DCF fluorescence intensity decreased as the concentration of the extract increased, demonstrating that ROS generation was inhibited in a concentration-dependent manner. The ROS inhibitory effect of the extract was higher than that of ascorbic acid. The extract prevented supercoiled DNA strand breakage induced by peroxyl radical and hydroxyl radical. Thus, the results of the present study demonstrate that the extract exhibits antimicrobial effects against food-borne pathogens as well as potent antioxidant capacity, suggesting that R. brachycarpum could be used as a natural antibacterial agent and effective antioxidant in food.

Leaf Shape Index, Growth, and Phytochemicals in Two Leaf Lettuce Cultivars Grown under Monochromatic Light-emitting Diodes (단색 발광다이오드에서 자란 축면상추 두 품종의 엽형, 생장 및 기능성 물질)

  • As an artificial light source, light-emitting diode (LED) with a short wavelength range can be used in closed-type plant production systems. Among various wavelength ranges in visible light, individual light spectra induce distinguishing influences on plant growth and development. In this study, we determined the effects of monochromatic LEDs on leaf shape index, growth and the accumulation of phytochemicals in a red leaf lettuce (Lactuca sativa L. 'Sunmang') and a green leaf lettuce (Lactuca sativa L. 'Grand rapid TBR'). Lettuce seedlings grown under normal growing conditions ($20^{\circ}C$, fluorescent lamp + high pressure sodium lamp, $130{\pm}5{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod) for 18 days were transferred into incubators at $20^{\circ}C$ equipped with various monochromatic LEDs (blue LED, 456 nm; green LED, 518 nm; red LED, 654 nm; white LED, 456 nm + 558 nm) under the same light intensity and photoperiod ($130{\pm}7{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod). Leaf length, leaf width, leaf area, fresh and dry weights of shoots and roots, shoot/root ratio, SPAD value, total phenolic concentration, antioxidant capacity, and the expression of a key gene involved in the biosynthesis of phenolic compounds, phenylalanine ammonia-lyase (PAL), were measured at 9 and 23 days after transplanting. The leaf shape indexes of both lettuce cultivars subjected to blue or white LEDs were similar with those of control during whole growth stage. However, red and green LEDs induced significantly higher leaf shape index than the other treatments. The green LED had a negative impact on the lettuce growth. Most of growth characteristics such as fresh and dry weights of shoots and leaf area were the highest in both cultivars subjected to red LED treatment. In case of red leaf lettuce plants, shoot fresh weight under red LED was 3.8 times higher than that under green LED at 23 days after transplanting. In contrast, the accumulation of chlorophyll, phenolics including antioxidants in lettuce plants showed an opposite trend compared with growth. SPAD value, total phenolic concentration, and antioxidant capacity of lettuce grown under blue LED were significantly higher than those under other LED treatments. In addition, PAL gene was remarkably activated by blue LED at 9 days after transplanting. Thus, this study suggested that the light quality using LEDs is a crucial factor for morphology, growth, and phytochemicals of two lettuce cultivars.

The Effect of Mean Brightness and Contrast of Digital Image on Detection of Watermark Noise (워터 마크 잡음 탐지에 미치는 디지털 영상의 밝기와 대비의 효과)

  • Watermarking is a widely employed method tn protecting copyright of a digital image, the owner's unique image is embedded into the original image. Strengthened level of watermark insertion would help enhance its resilience in the process of extraction even from various distortions of transformation on the image size or resolution. However, its level, at the same time, should be moderated enough not to reach human visibility. Finding a balance between these two is crucial in watermarking. For the algorithm for watermarking, the predefined strength of a watermark, computed from the physical difference between the original and embedded images, is applied to all images uniformal. The mean brightness or contrast of the surrounding images, other than the absolute brightness of an object, could affect human sensitivity for object detection. In the present study, we examined whether the detectability for watermark noise might be attired by image statistics: mean brightness and contrast of the image. As the first step to examine their effect, we made rune fundamental images with varied brightness and control of the original image. For each fundamental image, detectability for watermark noise was measured. The results showed that the strength ot watermark node for detection increased as tile brightness and contrast of the fundamental image were increased. We have fitted the data to a regression line which can be used to estimate the strength of watermark of a given image with a certain brightness and contrast. Although we need to take other required factors into consideration in directly applying this formula to actual watermarking algorithm, an adaptive watermarking algorithm could be built on this formula with image statistics, such as brightness and contrast.

Time Course Variation of Liver 25-Bydroxyvitamin $D_3$ Content in Broiler Chicks Exposed to UVB Light with Different Dobe (상이한 선양의 자외선을 조사한 브로일러 병아리에 있어서 간장 25-Hydroxyvitamin $D_3$회량의 계치적 변화)

  • This research was carried out to determine the 25-Hydroxyvitamin $D_3$[25(OH)$D_3$] content in liver of broiler Hubbard chicks fed vitamin VD-deficient diet for 31 days in a subdued light room and exposed to UVB light (maximum intensity at 297nm) with dose of 0.204 or 0.408 mJ/$\textrm{cm}^2$(30 or 60 min irradiation) . The lipid in liver collected at 0~138 hr after irradiation was extracted by chloroform-methanol(2:1, v /v) and 25(OH)$D_3$ fraction was separated by Sep-Pak silica cartridge. The 25(OH)$D_3$ concentration was measured by normal phase HPLC. The negative control chicks Presented 25(OH)D$_3$17.5 ng/g liver. When 0.204mJ/$\textrm{cm}^2$ was treated to whole body of chicks, the 25(OH)$D_3$ level was increased to 37.8 ng/g at 12 hr after irradiation, the peak concentration, 40.5 ng /g was appeared at the time of 86 hr, and decreasing trend was shown thereafter until 138 hr, the final time in this study. When 0.408 mJ/$\textrm{cm}^2$ was applied, the 25(OH)$D_3$ content was 36.7 ng /g liver at 12 hr, 61.4 ng/g(maximum value ) was appeared at 42 hr, and 39.5 ng /g at 138 hr. The increased absolute amounts in liver 25(OH)$D_3$ were 23 and 43.9 ng/g as chicks were exposed to UVB light with dose of 0.204 and 0.408mJ/$\textrm{cm}^2$, respectively. Consequently, it was found that when double dose of UVB light was irradiated to the chicks, their liver samples produced nearly double 25(OH)$D_3$ at 42 hr after exposure, and the peak value was presented earlier by 24 hr than that in the low dose treatment.

The Effect of Salicylic Acid on $Cd^{2+}$-induced Physiological Toxicity in Commelina communis L. ($Cd^{2+}$에 의한 닭의장풀의 생리적 독성에 salicyclic acid가 미치는 영향)

  • The effect of salicylic acid (SA) on C $d^{2+}$ - induced physiological toxicity in Commelina communis was investigated. 3- weeks old Commelina communis was transferred to and grown in Hoagland solution in the presence or absence of 100 $\mu$M C $d^{2+}$ and SA for 3 weeks. In the treatment of C $d^{2+}$ + SA, the length of stem was increased to 0.7 cm for 3 weeks (C $d^{2+}$, 2.1cm; control, 7.2 cm). C $d^{2+}$ + SA reduced total chlorophyll content up to 86%, and changed chlorophyll a/b ratio below 1.6. C $d^{2+}$ + SA also reduced about 40-78% of water potential, but C $d^{2+}$ increased 16-39% from 1 week to 3 weeks. C $d^{2+}$ + SA also inhibited 27% of Fv/Fm, but in case of C $d^{2+}$, Fv/Fm was not changed. The treatment of C $d^{2+}$ + SA showed about 37-58% inhibition of photosynthetic activity when measured at various light intensity (500-1000 $\mu$mol $m^{-2}$ $s^{-1}$ ). In the case of C $d^{2+}$ treatment, photosynthetic activity was inhibited to 12-15%. Similar effect was found in terms of stomatal conductance. Therefore, it could be concluded that the treatment of C $d^{2+}$ + SA into plant decrease or block various physiological activities and lend to die by double effects of both chemicals.cts of both chemicals..

Quantitative Measurement of Carbon Dioxide Consumption of a Whole Paprika Plant (Capsicum annumm L.) Using a Large Sealed Chamber (대형 밀폐 챔버를 이용한 파프리카(Capsicum annumm L.) 개체의 이산화탄소 소비량 측정 및 정량화)

  • This study was carried out to clarify precise $CO_2$ demands of paprika plants (Capsicum annumm L.) by measuring photosynthesis rates of the leaves in high, low positions, and the $CO_2$ consumption of a whole plant in a large sealed chamber. A photosynthesis measuring system (LI-6400) was used to measure the photosynthetic rates of the leaves located in different positions. A large sealed chamber that can control inside environmental factors was developed for measuring $CO_2$ consumption by a whole paprika plant. With increase of radiation, photosynthetic rates of the leaves in higher position became larger than those in lower position. The $CO_2$ consumption by the plant was estimated by using decrement of $CO_2$ concentration from initial level of 1500 ${\mu}mol{\cdot}mol^{-1}$ in the chamber with increase of integrated radiation. A regression model for estimating $CO_2$ consumption by the plant (leaf area = 7,533.4 $cm^2$) was expressed with integrated radiation (x) and was suggested as $y=-0.06234+3.671^*x/(2.589+x)$ ($R^2=0.9966^{***}$). The photosynthetic rate of the whole plant measured in the chamber was 3.4 ${\mu}mol\;CO_2{\cdot}m^{-2}{\cdot}s^{-1}$ under 300 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ light intensity, which is in-between photosynthetic rates of the leaves in high and low positions. For this reason, some differences between required and supplied $CO_2$ amounts in greenhouses might occur when depending too much on photosynthetic rates of leaves. Therefore, we can estimate more accurately $CO_2$ amount required in commercial greenhouses by using $CO_2$ consumption model of a whole plant obtained in this study in addition to leaf photosynthetic rate.

The Effect of Qigong Exercise on SF-36 and Psychological Factors of Middle-aged Obese (기공 운동이 비만인의 건강관련 삶의 질(SF-36)과 심리적 요인에 미치는 영향)

  • Obese people experience dissatisfaction of body, depression, anxiety, and emotional disturbance by social prejudice and discrimination, further they represent decline of health-related quality of life. Exercise therapy is recommended as one of the positive treatments to improve the emotional pain of obese people. This study is to provide guideline of exercise prescription for obese people by investigating the effects of qigong exercise on psychological factors such as health-related quality of life (SF-36), self-efficiency, depression, anxiety, and fatigue in obese people. The subjects of this study were 50~60s adults participated in Golden-Wellbeing program at S university in G gu of S city and were devided into exercise (N=17) and control (N=16) group. Qigong exercise was performed twice per week for 12 weeks and SF-36, self-efficiency, depression, anxiety, and fatigue were measured before and after 12 weeks of exercise. In the 8 items of SF-36 before and after 12 weeks of qi-gong exercise, there was no significance in role limitation-emotional (F=0.187, p=0.668), mental health (F=2.043, p=0.163) between groups, but there was significance in physical functioning (F=15.151, p<0.001), role limitation-physical (F=18.278, p<0.001), social functioning (F=4.957, p=0.033), vitality (F=11.485, p=0.002), bodily pain (F=6.623, p=0.015), and general health (F=4.498, p=0.042) between groups. Also, anxiety (F=0.631, p=0.433) was not significant, whereas self-efficiency (F=6.124, p=0.019), depression (F=5.109, p=0.031), fatigue (F=7.998, p=0.008) was significant between groups. Even though qigong is slow motion and low intensity of exercise, it was found that qi-gong has exercise effect which induces mental and psychological improvement through this study.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The strip loader and cassette were made to obtain more accurate reflectance parameters. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6~13 times higher than that of the microspheres synthesized by conventional seeded batch copolymerization. As a previous step for biomedical application, adsorption experiments of bovine albumin on microspheres were carried out under various conditions.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
