한국산 너도바람꽃속의 종자 비교형태 (Comparative seed morphology of Korean Eranthis (Ranunculaceae))

  • 미나리아재비과(Ranunculaceae) 너도바람꽃속(Eranthis)중에서 우리나라에 자생하는 너도바람꽃(E. stellata Maxim.)과 변산바람꽃(E. byunsanensis B. Sun)을 대상으로 종자의 외부형태와 해부형태를 관찰하였다. 그 결과, 두 종간에 종자표면의 모용 발달 유무에서 뚜렷한 차이가 관찰되었다. 즉, 너도바람꽃의 종자는 표면에 단세포성 모용이 발달하지 않는 반면, 변산바람꽃은 종자표면에 표피세포 유래의 단세포성 모용이 잘 발달하는 특징이 확인되었다. 이것은 변산바람꽃을 신종으로 발표하면서 제시했던 형태적 특징 외에, 변산바람꽃을 특산종으로 강하게 지지할 수 있는 또 다른 명확한 형태적 특징이라고 판단된다. 아울러 이번 연구에서 재료의 미확보로 연구하지 못한 풍도바람꽃과 일본종 E. pinnatifida의 종자형태도 비교 연구되어야 할 것으로 생각된다.

Effect of Harvest Time on Yield Components and Quality in 'Heuksaeng', the Purple-Skin Peanut Variety

  • This study was conducted to investigate the impact of harvest time on yield and seed quality in purple seed coat peanut. Compared to the average Virginia peanut variety, 'Heuksaeng', purple peanut variety has been reported to experience yield loss and reduced seed quality under delayed harvest. We examined the yield components and seed quality at different harvest times from 70 to 110 days after flowering (DAF), with harvesting occurring at intervals of 10 days. The number of mature pods per plant reached a maximum of 51.0 at 80 DAF, and average pod and seed yield peaked at 80 DAF with values of 5,229 and 3,532 kg per ha, respectively. Seed quality drastically decreased after 100 DAF, and the concentrations of two major anthocyanin compounds were significantly lower under delayed harvest. The antioxidative activities of DPPH and ABTS decreased to only 69 and 83% of their maximum values at 110 DAF. On the basis of these results, we recommend that 'Heuksaeng' is harvested approximately 10 to 15 days earlier that average Virginia-type cultivars to obtain the highest yield while minimizing the deterioration of seed quality.

Effects of Rhizobium Inoculant, Nitrogen, Phosphorus, and Molybdenum on Nodulation, Yield, and Seed Protein in Pea

  • The effects of Rhizobium inoculant, nitrogen, phosphorus, and molybdenum on nodulation, dry matter production, yield attributes, pod and seed yields, protein and phosphorus contents in seed of pea (pisum sativum) var. IPSA Motorshuti-3 were assessed by a field experiment. Among the treatments Rhizobium inoculant in combination with 25kg P and 1.5kg Mo/ha performed best in recording number of nodules/plant, total dry matter yield, number of pods/plant, number of seeds/pod, 1000-seed weight, green pod yield, green and mature seed yields of pea. The highest green pod yield of 15.37 t/ha ($97.05\%$ increase over control) and green seed yield of 9.6t/ha ($69.31\%$ increase over control) were obtained by inoculating pea with Rhizobium inoculant in association with 25kg P and 1.5 Mo/ha. The effects of 60 or 120kg N/ha were comparable to Rhizobium inoculant in most cases. There were positive correlations among yield attributes, yield, protein and phosphorus contents in seeds of pea. From the viewpoint of yield attributes, yield, and seed quality, application of Rhizobium inoculant along with 25kg P and 1.5kg Mo/ha was considered to be the balanced combination of nutrients for achieving the maximum output from cultivation of pea in Shallow-Red Brown Terrace Soil of Bangladesh.

올콩의 등숙단계별 종실 수분함량 및 품질 변화 (Changes of Seed Moisture Content and Quality during Grain-filling in Early Maturing Soybean Genotypes)

  • 올콩의 수확시기에 따른 품질변화를 밝히고자, 대립종인 Yuhsuzumi와 소립종인 담양재내를 공시하여 수확시기별 종실의 수분함량, 입색, 발아율, 지방산 조성 등을 조사하여 다음과 같은 결과를 얻었다. 1. 협과 종실의 수분함량은 R7단계 전후까지는 협의 수분함량이 뚜렷이 높았으나, R8단계 이후에는 종실의 수분함량이 협보다 다소 높은 경향을 보였으며, 종실의 수분함량이 14%(종실 수분함량으로 본 수확적기) 내외가 되는 시점은 Yuhsuzumi에서는 생리적 성숙기(R7단계)로부터 24일, 담양재내는 8일이 소요되었다. 2. R8단계(고유 성숙협색기)에서 종실의 수분함량은 대립종인 Yuhsuzumi는 25.3%, 소립종인 담양재내는 14.5%를 나타내어 R8단계에 수확시 Yuhsuzumi는 철저한 건조과정이 필요하였다. 3. 수확기 지연에 의하여 Yuhsuzumi는 청색의 종실이 황변하고 입질이 저하되어 발아력이 낮아졌으나, 담양재내는 입질과 발아력의 차이가 없었다. 4. 수확기 지연에 의하여 Yuhsuzumi는 지방산 함량이 낮아졌고, 지방산 조성에 있어서는 Lino-lenic 산은 낮아지고 Oleic산은 증가되었다.

선버들의 생장 특성 (Growth Characteristics of Salix nipponica)

  • The morphology, seed germination, and life history of Salix nipponica were investigated in the wetland of the Nam-River, Jinju, Gyeongsangnam-do, Korea from 2000 to 2001 to provide the basic data necessary for riverine ecosystem restoration through understanding the growth characteristics of Salix nipponica. Salix nipponica had stomata on only lower side and stomata type was paracytic. Salix nipponica produced many small and light seeds. The seed number per mature ament was 1599.4, seed mass of 0.04 mg, and fertilization rate of 66.1%. Seed germination was little affected by light. Germination rate was high and mean germination time was short Under flooding condition, seeds were germinated normally and were viable after as much as 14 days of flooding. But there were no differences under various water depths on germination rates. Tree age was closely correlated with more stem diameter than tree height and there was no difference of growth rate between male and female tree. Growth rate was most rapid for 2 to 3 years after germination, and length growth was almost stopped for more than 11 years even though mass growth was done. Besides even a second-year indivisual was flowered.

벼 종자배양에서 Copper가 캘러스 형성 및 식물체 재분화에 미치는 영향 (Effect of Copper on Callus Formation and Plant Regeneration in Seed Culture of Rice)

  • 벼의 종자배양에서 식물체 재분화 능력을 향상시키기 위하여 2 mg/L의 2,4-D와 여러 가지 농도의 copper (0~5.0mg/L)가 첨가된 MS배지에 종자배양 한 바 캘러스 생장정도는 copper 농도에 따라 다르게 나타났는데 특히, 2.5 mg/L의 copper가 첨가된 배지에서 가장 양호하였다. 그리고 캘러스 형성률과 copper 농도 간에는 유의성 있는 차이가 인정되지 않았다. 식물체 재분화 배지에 2.5 mg/L의 copper의 첨가는 Indica 품종들의 식물체 재분화 효율을 크게 향상시켰다. Copper가 첨가되지 않은 N$_{6}$배지에서 Indica 6품종의 평균재분화율은 2.4%이었으나, copper가 첨가된 배지에서 27.4%로 높게 나타났다.

국내 채종 및 수입 녹비작물 종자의 발아 및 활력 특성 구명 (Germination and Viability of Green Manure Crop Seeds Produced from Domestic and Foreign Countries)

  • 녹비종자의 입모증진 및 지속재배 가능성을 구명하고자 국내산 녹비종자의 종자의 성숙시기별 발아 및 휴면 특성과 수입녹비종자의 종자활력유지기간을 구명한 결과는 다음과 같다. 1. 녹비종자의 채종시기별 발아율은 5월 25일에서 6월 4일로 종자 성숙이 진전 될수록 경실율이 높아져 낮아졌다. 6월 4일 수확한 녹비종자의 무처리 발아율은 마메초 30%, 크림손클로버 16%, 살갈퀴는 2%로 낮았으나 종피 파상처리후 종자활력은 마메초 85%, 크림손 클로버 94%, 살갈퀴는 95%로 높아 녹비종자의 휴면은 종자경실에 기인한 것으로 확인되었다. 2. 녹비종자 성숙시기별 종자 활력은 크림손클로버와 살갈퀴는 5월 30일 이후에는 90%이상으로 높았으나 마메초는 77~85%로 낮아 크림손클로버와 살갈퀴는 밀양에서 5월 30이후 완전히 성숙이 되었으나 마메초는 6월 4일까지도 성숙이 되지 않았다. 3. 6월초에 수확한 녹비종자를 상온저장 후 매월 발아율 및 경실율(휴면율)을 조사한 결과, 저장기간이 경과함에 따라 경실종자의 비율이 감소하여 발아율이 증가하였으나 녹비종자 파종시기인 9월까지도 발아율이 26~34%로 낮았다. 따라서 국내 생산 녹비종자 이용시 9월 파종직전 휴면타파 처리를 해야 안전 입모수 확보가 가능할 것으로 판단되었다. 4, 수입 녹비종자의 광조건이 암조건보다 발아율이 4~5% 정도 높았으나 통계적인 유의성은 없어 녹비종자의 발아에 광의 영향은 크게 받지 않았다. 5. 수입 헤어리베치와 크림손클로버 종자를 상온보관시 1년후에는 발아율이 헤어리베치는 18∼32%, 크림손 크로버는 30~40%가 낮았고, 2년후에는 발아율이 헤어리베치는 5~13% 정도로 낮고 크림손클로버는 전혀발아가 되지 않아 수입후 2년이 경과한 종자는 사용하지 말아야 한다.

Proteomic Approach of the Protein Profiles during Seed Maturation in Common Buckwheat (Fagopyrum esculentum Moench.)

  • Single seeds of common buckwheat cultivar Suwon No. 1 when subjected to SDS-PAGE revealed very high polymorphism. High variation existed for protein or protein subunits with molecular weight 54-47kDa, 45-25kDa and 16-11kDa. The electrophoregram showed variation for globulin as well as other protein fractions. About 300 proteins were separated by two-dimensional electrophoresis in common buckwheat (Fagopyrum esculentum Moench.) seed. Seed maturation is a dynamic and temporally regulated phase of seed development that determines the composition of storage proteins reserves in mature seeds. Buckwheat seeds from 5, 10, 15, 20, and 25 days after pollination and matured stage were used for the analysis. This led to the establishment of high-resolution proteome reference maps, expression profiles of 48 spots. It was identified 48 proteins from MALDI-TOF/MS analysis of wild buckwheat seed storage proteins. The 48 proteins were found identical or similar to those of proteins reported in buckwheat and other plants; it is belonging to 9 major functional categories including seed storage proteins, stress/defense response, protein synthesis, photosynthesis, allergy proteins, amino acid, enzyme, metabolism, and miscellaneous. It appears that the major allergenic storage protein separated played the important role in buckwheat breeding and biochemical characterization.

Assessment of different pretreatments to breakage dormancy and improve the seed germination in Elaeocarpus serratus L. - an underutilized multipurpose fruit tree from South India

  • The seeds of Elaeocarpus serratus, a tropical underutilized fruit tree are characterized by hard seed coat and consequent poor water uptake and low germination. To improve the regeneration through seeds, various parameters such as viability of seeds, water uptake, and effect of seed mass on germination and pretreatments were performed using a completely randomized design (CRD). Tetrazolium (TZ) test was conducted using fresh, mature seeds revealed $50{\pm}2.56%$ mean viability. Seeds of different weight classes showed similar pattern of water uptake and the saturation level was achieved at 60 hrs of soaking. Seeds belong to weight class 2.6-3.5g were germinated ($12.5{\pm}1.26%$) with $175{\pm}1.75days$ (d) of mean time taken for germination (MTG). Germination capacity of seeds varied significantly among different populations and Varkala population gave $12.5{\pm}1.1%$ germination with $174.6{\pm}2.5d$ MTG. Among various seed treatments, mechanical scarification was superior in germination and significant reduction in MTG ($p{\leq}0.05$). The mechanical scarification by complete removal of seed coat resulted in $49.2{\pm}1.52%$ germination within a short period of time ($9.52{\pm}0.89d$ MTG). However, the complete removal of seed coat without damaging to embryo is a difficult task. An alternate treatment (Mechanical scarification II) by making cracks on nut faces vertically followed by soaking in distilled water for 24 hrs gave $48.4{\pm}1.73%$ germination with significantly reduced MTG ($12.14{\pm}0.56d$) over unsoaked, untreated control ($6.5{\pm}1.84%$ germination and $197.18{\pm}1.79d$ MTG; $p{\leq}0.05$). This treatment (Mechanical scarification II) is therefore recommended for E. serratus seeds as it can adopt easily and can achieve 7 fold increases in germination over control. The recorded germination through mechanical scarification is in tune with realized viability percentage of the seeds.

배초향 종자처리가 발아성 향상에 미치는 영향 (Effect of Seed Treatment on Improved Germinability of Agastache rugosa O. Kuntze)

  • This study was carried out to establish optimal conditions for breaking dormancy of Agastache rugosa O. Kuntze seeds. A series of experiments according to seed maturity and treatment with plant growth regulators were performed to improve germination percentage and synchronize germination of the seeds. In addition, it was conducted to test whether the useful effect of seed treatment before sowing leads to healthy seedling and early vigorous growth. The average seed size was 1.85 mm (length) × 0.82 mm(width). The seed size was much smaller than other vegetable seeds. Seeds colorappeared dark brown, the shape of the seeds was oval and the weight of 1,000seeds was 352.8 mg. The optimum germination temperature was 22℃. Light exposure during germination did not affect germination promotion, suggesting that A. rugosa seeds are a kind of dark germinating seeds. Seed dormancy lasted for 40 days after harvesting, and GA3 treatment of dormant seeds could break dormancy. There were significant differences in germination percentage and rate according to the maturity of seeds. The germination percentage of mature seeds was 10 - 18% higher than that of immature seeds, and germination rate was 2 days faster. GA3 treatment during growth regulator treatment improved germinability, but BAP or ethephone treatment did not. The optimal growth regulator concentration of for germination was the combination treatment of 100 mM GA3 + 100 mM BAP.
