A plantlet regeneration protocol was developed on pot-grown mature plants of Elaeocarpus robustus Roxb. cv. Dwarf from nodal and leaf explants. The best yield of adventitious shoots was achieved from the leaf-derived calli in a modified MS ($MMS_1$, half strength of major salts, full strength of minor salts, and vitamins) medium containing $4.0{\mu}M$ BA + $4.0{\mu}M$ Kn + $0.5{\mu}M$ NAA + 15% coconut water (CW). The shoot multiplication rate was amplified about twofold per culture after the addition of 15% CW to the medium. The rate of shoot multiplication reached maximum at the 5th subculture, and it maintained this rate throughout the 3 subsequent subcultures. The best rooting in vitro was investigated by subculturing the microcuttings in an $MMS_2$ (half strength of both major salts and minor salts and full strength of vitamins) medium containing $1.0{\mu}M$ IBA in the dark for one initial week at $30^{\circ}C$, followed by subculturing them in a plant-growth regulator (PGR)-free medium in the light. The plantlets raised in vitro were successfully established under ex vitro conditions.

Background: Breast cancer is the most common malignant disease in women. The patients with advanced breast cancer develop metastasis to bone. Bone metastasis and skeletal-related events by breast cancer are frequently associated with the invasiveness of breast cancer cells and osteoclasts-mediated bone resorption. Forsythia koreana is used in oriental traditional medicine to treat asthma, atopy, and allergic diseases. The aim of this study was to evaluate the inhibitory effects of F. koreana extracts on the invasion of breast cancer cells and bone resorption by osteoclasts. Methods: Cell viability was measured by an MTT assay and the migration and invasion of MDA-MB-231 cells were detected by a Boyden chamber assay. The formation of osteoclasts and pit was detected using tartrate-resistant acid phosphatase staining and calcium phosphate-coated plates, respectively. The activities of matrix metalloproteinases (MMPs) and cathepsin K were evaluated by gelatin zymography and a cathepsin K detection kit. Results: The fruit and leaf extracts of F. koreana significantly inhibited the invasion of MDA-MB-231 cells at noncytotoxic concentrations. The fruit extract of F. koreana reduced the transforming growth factor ${\beta}1-induced$ migration, invasion and MMPs activities of MDA-MB-231 cells. In addition, the fruit, branch, and leaf extracts of F. koreana also inhibited the receptor activator of nuclear factor kappa-B ligand-induced osteoclast formation and osteoclast-mediated bone-resorbing activity by reducing the activities of MMPs and cathepsin K. Conclusions: The extracts of F. koreana may possess the potential to inhibit the breast cancer-induced bone destruction through blocking invasion of breast cancer cells, osteoclastogenesis, and the activity of mature osteoclasts.

This study has been undertaken to increase availability of native camellia in Jeonnam as a medicinal resource and to isolate the effective components from them. Fresh leaf and flower of camellia, single camellia tea and camellia tea mixed with green tea, herbs were screened for cytotoxicity on MCF -7 (human breast adenocarcinoma pleual effusion), Calu-6 (human pulmonary carcinoma), SNU-601 (human gastric carcinoma) cells. Also their multidrug-resistance reversing activity were evaluated using drug sensitive AML-2/WT and multidrug-resistant AML-2/D100 cells. Among the camellia extracts, young leaf and camellia tea mixed with green tea had strong growth inhibitory effects in below 100 $\mu\textrm{g}$/mL against human cancer cells. In result, young leaf showed the strongest inhibitory effects on MCF -7 ($IC_{50}$/ = 100 $\mu\textrm{g}$/mL ↑), Calu-6 ($IC_{50}$/ = 79 $\mu\textrm{g}$/mL), and SNU -601 ($IC_{50}$/ = 39 $\mu\textrm{g}$/mL), and AML-2/WT ($IC_{50}$/ = 64 $\mu\textrm{g}$/mL). Chemosensitizing effect was the extracts of mature leaf ($IC_{50}$/ = 97 $\mu\textrm{g}$/mL, RF=3.0), roasted tea ($IC_{50}$/ = 76 $\mu\textrm{g}$/mL, RF = 2.6 ↑) and steam tea ($IC_{50}$/ = 70 $\mu\textrm{g}$/mL, RF=2.8 ↑) strongly potentiate vincristine cytotoxicity in AML-2/D100 cells. But their cytotoxicities to both sensitive AML-2/WT and resistant AML-2/D100 cells were in the same order of magnitude. This results indicate that crude extracts of camellia mature leaves would contain some principles which have chemosensitizing activity.
This study was carried out to investigate the distribution of ${\alpha}-glucosidase$ inhibitor (AGI) activity and to evaluate horticultural characteristics of pepper (Capsicum spp.). AGI activities of pepper fruits and leaves were different from 1.0 to 20.5 times and 1.0 to 5.9 times, respectively. Weight, length and width of evaluated pepper fruit were distributed from 0.5 to 56.0 g, 0.8 to 15.4 cm and 0.5 to 6.3 cm per fruit respectively. Stem colors before transplanting varied from green to violet. Length and width of leaf were distributed from 3.1 to 5.0 cm and 2.1 to 3.0 cm. Immature fruit color was almost green and mature fruit color was almost red. In horticultural characteristics of selected pepper lines with high AGI activity, the fruit position was downward position. The immature fruit color was green in all lines except one and the mature fruit color was red in all lines. Fruit weight and fruit length of selected pepper lines with high AGI activity were distributed from 5.9 to 41.1 g and 5.9 to 17.0 cm and leaf width and leaf length were distributed from 5.8 to 29.7 cm and 3.9 to 8.7 cm, respectively. The AGI activities of pepper is widely variable between leaf and fruit. According to this result, it suggested the possibility of developing a new pepper line with high AGI activity.

Incidence of leaf blast on nursery plants and pitting disease on maturing banana bunches has been recorded in banana plantations during rainy season in Eastern India during 2014 to 2015. Taxonomical identification as well as DNA sequence analysis of the internal transcribed spacer region of fungus isolated from affected tissue culture derived plantlets and fruits confirmed the pathogen to be Pyricularia angulata Hashioka "in both the cases". Koch's postulates were proved on young plantlets as well as on maturing fruits of cv. Grand Naine under simulated conditions. Evolutionary history was inferred and presented for our P. angulata strain PG9001 with GenBank accession no. KU984740. The analysis indicated that the P. angulata is phylogenitically distinct from other related species related to both Pyricularia and Magnaporthe. Detailed symptoms of blast lesions on young leaves, transition leaves, mid rib, petioles, peduncle, maturing bunches, bunch stalks and cushions were documented. Notably, the distinct small pitting spots on maturing bunches reduced the visual appeal of mature fruits. Appearance of pitting symptoms on fruits in relation with age of fruits and their distribution pattern on bunch and fingers was also documented in detail. Further, the roles of transitory leaves, weed hosts, seasonality on disease occurrence have also been documented.
We investigated the variation of morphological features, density, biomass and characteristics of reproductive shoot on Zostera marina. This species was monthly monitored and collected during a year in Tongyeong (from January 2016 to December 2016). Morphological features, biomass and density showed significantly seasonal variation (p<0.001). Dimensions of Z. marina were highest in spring and early summer but lowest in winter. Biomass and leaf density of Z. marina showed highest value in May (4,700.5 g w·wt m-2) and June (858.0 leaves m-2) respectively and lowest value in November (515.9 g w·wt m-2 and 312.0 leaves m-2). Reproductive shoot was monitored from April (13℃) to June (21℃) and grew up to ca. 200 cm. Spathe length, spadix weight, numbers and weight of fruit showed significantly different on monthly. None mature flowers were monitored in April but matured flowers, fruits were founded in May and seed releasing were occurred in June.
A tomato zinc-finger protein gene, LeZFP1, encoding the Cys2/His2-type zinc-finger transcription factor was searched from cDNA microarray analysis of gene expression following induction of the overexpressed tomato transgenic plants showing resistance for pathogen and abiotic stresses. The full-length cDNA of LeZFP1 encoded a protein of 261 amino acid residues. Analysis of the deduced amino acid sequence of LeZFP1 revealed that it shares high sequence identity with pepper CAZFP1 (81% identity). We found that single copy of LeZFP1 gene is present in the tomato genome through southern blot analysis. The LeZFP1 transcripts were constitutively expressed in the tomato mature and young leaves, but were detectable weakly in the flower, stem and root. The LeZFP1 transcripts were significantly reduced in treated leaf tissues with NaCl and mannitol. The LeZFP1 gene was induced by oxidative stress especially. Our results indicated that LeZFP1 may play a role function involved in oxidative stress signaling pathways.
The role of Ca2+ on benzyladenine (BA)-induced senescence retardation in mature wheat (Triticum aestivum L.) primary leaves was investigated. When an extracellular calcium chelator, ethylene glycol-bis-($\beta$-aminoethylether)-N, N'-tetraacetic acid (EGTA) together with BA, was applied to senescing leaves for 4 days of dark incubation, the content of chlorophyll and soluble protein decreased rapidly. And, the content of malondialdehyde (MDA), known to be a degradation product of membrane lipids, increased compared with the BA alone control. The BA-EGTA combination also caused the stimulation of protease and RNase activity and a rapid loss of catalase activity owing to the decling of BA effects. In the case of treatment with only intracellular calcium antagonist 3, 4, 5-trimethoxybenzoic acid 8-(diethylamino) octyl ester (TMB-8) without the BA addition, the chlorophyll content at day 4 after dark incubation decreased in paralled with the increasing concentration of the antagonist. In addition, the chlorophyll content at 10-5 M calcium ionophore A23187 treatment in the absence of BA was similar to that of the BA alone treatment. These results suggest that calcium may mediate the retardation effect of BA on leaf senescence by acting as a second messenger and that the calcium input from cell organelles, as well as the calcium inflow from intercellular spaces and cell walls, may be involved in modulating cytosolic calcium levels related to BA action.
To explain the horizontal expansion of a rhizomatous perennial, Convallaria keiskei(lily-of-the-valley), in a study site of Chunchon, Kangwon Province, Korea, ramet growth and clonal structure were studied. Remarkable growth stategies were clarified. First, the timing for the successive phenological events such as sprouting. flowering and rhizome growth for lily-of-the-valley was fitted to exploit early spring when the canopy of overstory was opened. Second, these events were supported by effective matter allocation pattern: for example, two-year investment for new rhizomes enabled the first year ramets to mature in six weeks after sprouting and to grow up to 85% of the leaf area of perennial ramets. Finally, the ramet population was increased by local disturbances such as freezing, herbivory and collection by human. The rule that a clone was supposed to produce one new thizome per year was broken by occasional disturbances. Then, up to 5rhizomes from latent bur could be redeveloped. Based on clonal structure, 80% or total clones have from 1 to 4 ramets. this means there have occurred minor disturbances. Therefore, in conclusion, the successful flourishing of lily-of-the-valley came from its effective frowth strategy to take advantage of site disturbance.
Acidovorax citrulli causes bacterial fruit blotch in Cucurbitaceae, including watermelon. Although A. citrulli is a seed-borne pathogen, it can cause diverse symptoms in other plant organs like leaves, stems and fruits. To determine the infection routes of A. citrulli, we examined the virulence of six isolates (Ac0, Ac1, Ac2, Ac4, Ac8, and Ac11) on watermelon using several inoculation methods. Among six isolates, DNA polymorphism reveals that three isolates Ac0, Ac1, and Ac4 belong to Clonal Complex (CC) group II and the others do CC group I. Ac0, Ac4, and Ac8 isolates efficiently infected seeds during germination in soil, and Ac0 and Ac4 also infected the roots of watermelon seedlings wounded prior to inoculation. Infection through leaves was successful only by three isolates belonging to CC group II, and two of these also infected the mature watermelon fruits. Ac2 did not cause the disease in all assays. Interestingly, three putative type III effectors (Aave_2166, Aave_2708, and Aave_3062) with intact forms were only found in CC group II. Overall, our results indicate that A. citrulli can infect watermelons through diverse routes, and the CC grouping of A. citrulli was only correlated with virulence in leaf infection assays.
