A Least Squares Regression Model to Detect Quantitative Trait Loci with Polar Overdominance in a Cross of Outbred Breeds: Simulation

  • A least squares regression interval mapping model was derived to detect quantitative trait loci (QTL) with a unique mode of genomic imprinting, polar overdominance (POD), under a breed cross design model in outbred mammals. Tests to differentiate POD QTL from Mendelian, paternal or maternal expression QTL were also developed. To evaluate the power of the POD models and to determine the ability to differentiate POD from non-POD QTL, phenotypic data, marker data and a biallelic QTL were simulated on 512 F2 offspring. When tests for Mendelian versus parent-of-origin expression were performed, most POD QTL were classified as partially imprinted QTL. The application of the series of POD tests showed that more than 90% and 80% of medium and small POD QTL were declared as POD type. However, when breed-origin alleles were segregating in the grand parental breeds, the proportion of declared POD QTL decreased, which was more pronounced in a mating design with a small number of parents ($F_0$ and $F_1$). Non-POD QTL, i.e. with Mendelian or parent-of-origin expression (complete imprinting) inheritance, were well classified (>90%) as non-POD QTL, except for QTL with small effects and paternal or maternal expression in the design with a small number of parents, for which spurious POD QTL were declared.

  • The screw compressor is first invented by a Swedish engineer, Alf Lysholm in 1934. Since then, the development of the screw compressor idea for industrial applications has been continued by the Swedish research organization Svenska Rotor Maskiner, often identified by its initials SRM. The first industrial application of the machine was marketed as an air compressor in the 1950s. The screw compressor which is a positive displacement type compressor compresses gases by the rotation of a pair of mating rotors. The operation of this compressor is entirely rotary and dynamically in balance. Also there is no need for any valve mechanism and there exists less mechanical wear between the parts compared to the conventional reciprocating compressors. Due to these prominent features, the screw compressor has been rapidly spread into the air compressor market replacing the conventional reciprocating compressors and begun to be applied as a refrigerant compressor since the 1960s. In this work, the operation principle of the screw compressor is described in brief and the major design parameters affecting the compressor performance are classified. The international research trend in screw compressor development is introduced and the current situation in our country is described.

Minimization of Tilting Moment of Co-Rotating Scroll Compressor by Design of Back Pressure Chamber (배압실의 설계를 통한 상호회전 스크롤 압축기의 전복 모멘트 최소화)

  • In a co-rotating scroll compressor, both scrolls rotate on their fixed axes contrary to the conventional orbiting type scroll machine. This implies fixed locations and directions of the gas pressure force and sealing force. Because the tilting moment is mainly caused by interplay between the resultant force of above forces and bearing reaction force, the variation during one cycle is relatively small. Under real operation, this moment is balanced by the restoring moment created by the reaction between the baseplate and thrust bearing or between the scroll tip and baseplate. If these reactions become too large, greater torque is required due to increased friction in addition to the wear of mating parts. Consequently, appropriate study and minimization of tilting moment is important in the design of scroll machines. In this study, taking into account of the small variation of tilting moment during one cycle, we minimize the moment and thrust bearing reaction force by a properly designed back pressure chamber. As a result, for both the driving and driven scrolls, the tilting moment and the reaction force of thrust bearing can be minimized. And the stability is improved for all cases.

  • Ubiquinone compounds of strains in Leucosporidium scottii, Leucosporidium fellii, Leucosporidium lari-marini, and Rhodosporidium fluviale were analyzed by the high performance liquid chromatography. The Q-9 or Q-l0 compounds, independent on mating or self-sporulating type, were determined in the analyses of Leucosporidium scottii strains. Particularly, there is heterogenous in the quinone compounds in the same strain of L. scottii. These results showed reassessment of the quinone compounds as a taxonomic criterion; L. fellii had the Q-9 compound, L. lari-marini the Q-8 compound, and R. fluviale the Q-10 compound, Taxonomic position of L. lari-marini should be elucidated on the basis of analysis of Q-compound with other aspects.

Transmission and Reproduction of Transgenic Mice Expressing Human Growth Hormone Gene (사람 성장호르몬 유전자를 발현하는 형질전환 생쥐의 번식성 및 형질유전)

  • The transgenic mice were produced by microinjection of human growth hormone gene fused with mouse metallothionein Ⅰ promoter. They were mated with momal mice by backcross or brother-sister mating. The reproduction efficiencies of female and male n the FO transgenic mice were 17.6%(3/17 mice) and 31.2%(5/16 mice), respectively, and were very lower than that in normal mice(85.7% and 100%, respectively). Interestingly, a few of female transgenic mice were fertile which was different from the previous reports. Of 6 fertile transgenic mice, 2 mice were identified as mosaic type by the reduced frequency of genetic transmission to successive generation below Mendelian levle and the enhanced copy numbers of transgene in progeny mice compared with the transgenic parent. In the group of F1, F2, F3 transgenic mice the reproduction efficiencies of males were gradually improved, whereas females were absolutely infertile. It was consequently shown that the transgenic mice expressing human growth hormone gene were frequently infertile, but the genotypic and phenotypic characteristics of the fertile transgenic mice were normally passed on to the progeny through herm line. Therefore it must be considered wheter or not the products of foreign DNA introduced into animals will detrimentally affect their physiological aspects.

Selection of Superior Strains of Cordyceps militaris with Enhanced Fruiting Body Productivity

  • In vitro fruiting bodies were produced from ten different isolates of Condyceps militaris EFCC C-5736, EFCC C-5941, EFCC C-5976, EFCC C-6040, EFCC C-6849, EFCC C-7268, EFCC C-7342, EFCC C-7992, EFCC C-8027 and EFCC C-8549. Single ascospores were isolated from in vitro grown fruiting bodies and used for fruiting body production in brown rice medium by both intra-strain crossing and out-crossing. Length and dry wt. of stromata grown in vitro were measured. Strains producing highest dry wt. of stromata were selected. Both intra-strain crossings and inter-strain crossings of single ascospore strains were found to produce profuse fruiting bodies of C. militaris.

Root Rot of Carnation Caused by Phytophthora nicotianae (Phytophthora nicotianae(Breda de Haan)에 의한 카네이션 역병)

  • Root rot of carnation caused by isolates of Phytophthora was found at Kongju, 1996. Infected plants appeared symptoms of wilting, brown discoloration on collar area and consequently led to death. The causal pathogen was identified as Phytophthora nicotianae based on morphological, cultural and physiological characteristics. Mycelium was grown to rosette colony pattern on corn meal agar and the growth was 10.2 mm/day on 10% clarified V8 medium at $25^{\circ}C$. Swelling with radiating hyphae was formed in water and on agar within 7 days. Chlamydospores were abundantly produced on agar within 7 days. Sporangium was prominent papillate, 2~3 apex, 1.2 : 1 l/b ratio, lateral attacment on sporangiospore and was rarely produced on solid media but produced in water. The shape of sporangium was spherical or ovoid, and the size was 34~73$\times$32~60 (av. 33$\times$66.5) ${\mu}{\textrm}{m}$ in dimension. The isolates were heterothallic, and mating type A2. Oogonium was spherical, ovoid, 26~36 (av. 36 ${\mu}{\textrm}{m}$) in diameter, and antheridium was amphygynious to oogonia. The fungus was able to grow 10~35$^{\circ}C$, and optimal temperature was 27$^{\circ}C$.

GzRUM1, Encoding an Ortholog of Human Retinoblastoma Binding Protein 2, is Required for Ascospore Development in Gibberella zeae

  • Gibberella zeae (anamorph: Fusarium graminearum), a homothallic (self-ferile) ascomycete with ubiquitous geographic distribution, causes serious diseases in several cereal crops. Ascospores (sexual spores) produced by this fungal pathogen have been suggested as the main source of primary inoculum in disease development. Here, we report the function of a gene designated GzRUM1, which is essential for ascospore formation in G. zeae. The deduced product of GzRUM1 showed significant similarities to the human retinoblastoma (tumor suppressor) binding protein 2 and a transcriptional repressor, Rum1 in the corn smut fungus (Ustilago maydis). The transcript of GzRUM1 was detected during the both vegetative and sexual stages, but was more highly accumulated during the latter stage. In addition, no GzRUM1 transcript was detected in a G. zeae strain lacking a mating-type gene (MAT1-2), a master regulator for sexual development in G. zeae. Targeted deletion of GzRUM1 caused no dramatic changes in several traits except ascospore formation. The ${\Delta}$GzRUM1 strain produced perithecia (sexual fruit bodies) but not asci nor ascospores within them. This specific defect leading to an arrest in ascospore development suggests that GzRUM1, as Rum1 in U. maydis, functions as a transcriptional regulator during sexual reproduction in G. zeae.

Characteristics of new mid-high temperature adaptable oyster mushroom variety 『Heuktari』 for bottle culture (중고온성 병재배용 느타리 신품종 『흑타리』의 특성)

  • The 'Heuktari', a new mid-high temperature adaptable variety of oyster mushroom for the bottle culture, was bred by mating with monokaryons isolated from 'P11056' and 'MT07156'. The optimum temperature for the mycelial growth was $23{\sim}26^{\circ}C$ on PDA medium and that for the primordia formation and the growth of fruiting body of 'Heuktari' was $18{\sim}19^{\circ}C$ on sawdust substrate. In case of bottle cultivation, the period of mycelial growth was required about 30 days. In addition, the period of primordia formation and growth of fruiting body was 4 days and 5 days, respectively. In the characteristics of fruiting body, shape and color of pilei were round type and dark grayish brown, stipe color was white color and stipe shape was short and thick. The yield of fruiting bodies was 180 g/900 ml bottle which was 15% higher than that of Suhan-1ho. The gumminess and brittleness of stipe tissue were 110% and 140% stronger than those of Suhan-1ho, respectively.

Regulation of S100G Expression in the Uterine Endometrium during Early Pregnancy in Pigs

  • Calcium ions play an important role in the establishment and maintenance of pregnancy, but molecular and cellular regulatory mechanisms of calcium ion action in the uterine endometrium are not fully understood in pigs. Previously, we have shown that calcium regulatory molecules, transient receptor potential vanilloid type 5 (TRPV6) and calbindin-D9k (S100G), are expressed in the uterine endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-specific manner, and that estrogen of conceptus origin increases endometrial TRPV6 expression. However, regulation of S100G expression in the uterine endometrium and conceptus expression of S100G has been not determined during early pregnancy. Thus, we investigated regulation of S100G expression by estrogen and interleukin-$1{\beta}$ (IL1B) in the uterine endometrium and conceptus expression of S100G during early pregnancy in pigs. We obtained uterine endometrial tissues from day (D) 12 of the estrous cycle and treated with combinations of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone ($P_4$), and increasing doses of IL1B. Real-time RT-PCR analysis showed that $E_2$ and IL1B increased S100G mRNA levels in the uterine endometrium, and conceptuses expressed S100G mRNA during early pregnancy, as determined by RT-PCR analysis. To determine if endometrial expression of S100G mRNA during the implantation period was affected by the somatic cell nuclear transfer (SCNT) procedure, we compared S100G mRNA levels in the uterine endometrium from gilts with SCNT-derived conceptuses with those from gilts with conceptuses derived from natural mating on D12 of pregnancy. Real-time RT-PCR analysis showed that levels of S100G mRNA in the uterine endometrium from gilts carrying SCNT-derived conceptuses was significantly lower than those from gilts carrying conceptuses derived from natural mating. These results showed that S100G expression in the uterine endometrium was regulated by estrogen and IL1B of conceptus origin, and affected by the SCNT procedure during early pregnancy. These suggest that conceptus signals regulate S100G, an intracellular calcium transport protein, for the establishment of pregnancy in pigs.
