Effects of Maternal Folic Acid Nutritional Status on the Expression of Myelin Basic Protein in the Offspring (어미 쥐의 엽산 영양상태가 자손 쥐의 수초기본단백질(Myelin Basic Protein)발현에 미치는 영향)

  • Myelin basic protein (MBP), a major structural protein of the myelin, is thought to be important for the maintenance of myelin in the central nervous system (CNS). We investigated the effect of maternal folic acid nutritional status on the folate level and the synthesis of MBP in the offspring. In order to test this hypothesis, female Sprague-Dawley rats were fed either folic acid sufficient (8 mg/kg diet) or deficient (0 mg/kg diet) diet from 2 wks prior to the mating throughout the entire pregnancy, lactation and weaning period. We examined plasma folate level by the radioimmunoassay and homocysteine level by HPLC, respectively. The MBP expression was measured by the western blot analysis. The maternal folic acid deficiency decreased plasma folate level with a concomitant increase in plasma homocysteine level in their offspring. The maternal folic acid deficiency decreased hepatic levels of SAM and SAM/SAH ratio with a concomitant increase in hepatic levels of SAH and the MBP expression of spinal cord in their offspring at 7 wks of age. These results suggest that maternal folic acid nutritional status affect plasma folate and homocysteine level in their offspring. Moreover, the maternal folic acid deficiency mi호t inhibit the MBP expression of the spinal cord and disrupt many other vital CNS reactions in their offspring.

Comparative Early Life History of Two Pipefish, Urocampus nanus and Syngnathus schlegeli (Syngnathidae) in Laboratory Culture from Korea (실험실 사육에 의한 한국산 실고기과(Syngnathidae) 2종, 풀해마(Urocampus nanus)와 실고기(Syngnathus schlegeli)의 초기생활사 비교)

  • This study provides a detailed morphological description of larvae obtained from Urocampus nanus and Syngnathus schlegeli male brood fish over 20 days of culture in the laboratory. In both species, mating takes place when several males each spread their brood pouch like a wing to attract a female's attention. When the female begins to swim upward, the males follow her and receive her eggs in their brood pouches. Newborn larvae of U. nanus and S. schlegeli had already completed formation of dorsal and caudal fin rays, but not of pectoral fin rays. Pectoral fin rays were completely formed 15 days after release in S. schlegeli and 20 days after release in U. nanus. The ratio of caudal fin length to standard length increased until 8 days and decreased thereafter in S. schlegeli, while in U. nanus this ratio declined continuously after hatching. The larvae of the two species were very similar in external morphology, but well distinguished by the number of dorsal fin rays (15-16 in U. nanus vs. 39-43 in S. schlegeli), the presence of a membrane under the tail (absent in U. nanus vs. present in S. schlegeli), and the presence of melanophores in the dorsal fin (present in U. nanus vs. absent in S. schlegeli). Based on this study, U. nanus appears to be evolutionally more similar to pipefish than to seahorse.

Estimation of Genetic Parameter and Growth Traits by Sex of Pacific Abalone, Haliotis discus hannai (북방전복 (Haliotis discus hannai) 의 성별에 따른 성장형질 및 유전모수 추정)

  • The purpose of this study is to estimate genetic parameter and growth traits by sex of Pacific abalone, Haliotis discus hannai. The abalone 10 family produced using 1:1 mating system of male and female for analyses of sex ratio and growth traits (shell length, shell width, and total weight) by sex. Overall mean in phenotypic traits at 30-month-old showed 85.01 mm of shell length, 57.49 mm of shell width and 73.34 g of total weight respectively. The sex ratio (female : male) was 1:0.93 (n = 191:177). The values of growth traits by sex showed significant difference (P < 0.05). The each value of female growth traits were shown to be higher than the values of male growth traits. The heritability of growth traits by sex were estimated that the heritability of female growth traits are higher than male that. The results suggest a possibility of improving the growth of cultured abalone using selection breeding by sex.

Breeding to Improve some of the Economic Characters of the Sex-limited Dark Egg Color Strains. (한성흑란계통누에의 몇가지 실용형질향상을 위한 교잡방법)

  • To investigate suitable breeding system for the improvement of economic characters of the sexlimited dark egg colour strains, the four kinds of breeding methods which are represented as sibmating, repeated backcrossing, multi-backcrossing every other generation, and repeated hybridization were detected from the P to the F6. 1. Pupation ratio was varied between the four kinds of breeding methods and decreased gradually in order of repeated hybridization, mult-backcrossing every other generation and sib-matinv, and repeated backcrossing. 2. Repeated backcrossing, multi-backcrossing every other generation, and repeated hybridization with the exception of sib-mating were consiered as available breeding methods to improve single cocoon weight, cocoon shell weight and cocoon shell ratio of the sex-limited dark egg colour strains. 3. As for the responses of the economic characters form the P to the F6, pupation ratio was fluctuant and unstable, on the other hand single cocoon weight, cocoon shell weight and cocoon shell ratio increased remarkably in the F2, but in the subsequant generatios the increasing rate of each characters were fluctuant irregularly. 4. After the original sex-limited dark egg colour strains were bred under the corresponding breeding methods, from the P to the F6 the economic characters were approached to the level of normal breeding silkworm lines'. 5. It came to the consideration that the autosomal fragment which was translocated on the W chromosome had no physiologically significant effect on the expression of the economic characters because the Expression index (E) of the sex-limited dark egg colour strains was similar to that of normal silkworm lines. 6. From the results, it comes to the conclusion that the useful breeding methods to improve the economic characters of the sex-limited dark egg colour strains will be a line separation including fixation of the characters.

The sexual dimorphism of Mauremys reevesii (남생이의 성적 이형성)

  • In this study, a total of 21 parts of the head, carapace, and plastron of 21 tortoises were examined to investigate the characteristics of male and female sexual dimorphism of the Reeve's turtle (Mauremys reevesii), a natural monument in Korea. The ratio values of a total of 19 areas were derived from Head Length (HL) and Carapace Length (CL). As a result of comparing the ratio values, it was found that male tortoises had larger head length, head width, and head height than females compared to the size of the carapace. Since females grow eggs inside the body through internal fertilization after mating, female carapaces are judged to be larger than males overall. As a result of statistical analysis of the ratio values of the measured values, Plastron Length (PL), Bridge Length (BL), Gular Length (GL), Interhumeral Length (IH), Interanal Length (IAN), Femoral-Anal seam (FA) and Head Height (HH) showed statistically significant differences. Statistically significant in the part of the plastron, it is judged that the tortoise acts directly on each deck of the plastron as it grows. In addition, the head size of male tortoises was smaller than that of females, but it was found to be larger in proportion. It is considered that the tortoise has a rather large head compared to the body due to the male's habit of shaking his head and courtship behavior before mating. This study, we were able to identify the characteristics of sexual dimorphism in Reeve's turtle. However, since most of them were rescued, it was not possible to accurately determine habitat and age. In the future, if additional studies are conducted on individuals with clear habitats and ages, it is expected that the characteristics of the physical differences that develop as they grow can be identified.

Effect of Crossbred Korean Native Ducks on the Retail Cut Yield, Meat Quality, and Sensory Evaluation of Duck Meats (토종오리의 교배 조합이 오리고기의 부분육 생산수율, 육질 및 관능검사에 미치는 영향)

  • This work was carried out to investigate on the retail cut yield and the meat quality of crossbred ducks. A total of 360 pullets that were produced from 4 mating methods used in this work. Four mating method were A) native ducks (female) $\times$ native ducks (male), B) meat-type ducks (female) $\times$ native ducks (male), C) native ducks (female) $\times$ meat-type ducks (male), and D) meat-type ducks (female) $\times$ meat-type ducks (male). Ducks were bred at the flat house, and selected nine ducks with similar weights from each treatment at the certain weeks (A, B, C 8 weeks; D 6 weeks). Selected ducks were slaughtered, calculated the retail cut yield(wing, back, neck, breast, leg), analyzed the physico-chemical compositions, and tested the sensory evaluation. Wing and neck meat ratios of D treatment were lowest (P<0.05), and breast meat ratio of B treatment was high (P<0.05) compared to other treatments. pH of duck meat was no difference among treatments, fat and protein contents of B treatment was highest, and collagen contents of D treatment was highest among all treatments. Meat color and physical compositions was no difference among treatments. Juiciness of D treatment meats was highest, and there was no difference between B and D treatments. Finally, mating method of native duck and meat-type duck affected on the retail cut yield, but did not improve on the physico-chemical compositions and sensory evaluation.

Chemical Synthesis and Orientation Disruption Bioassay of Sex Pheromone of the Oriental Fruit Moth, Grapholita molests (Busck) (복숭아순나방(Grapholita molests) 성페로몬 합성과 유인력 교란생물검정)

  • Three sex pheromone components (cis-8-dodecenyl acetate (Z12Ac), trans-8-dodecenyl acetate (E12Ac), cis-8-dodecenol (Z12OH)) of the Oriental fruit moth, Grapholita molesta, were chemically synthesized. Especially to increase the composition of cis-stereoisomer, a triple bond intermediate was hydrogenated at $-20^{\circ}C$ with catalytic $Pd/BaSO_{4}$. The resulting product consisted of the acetates with a stereoisomer ratio in 92:8 (Z:E). The biological activity of the synthesized pheromone compounds was analyzed both in male responses and orientation disruption. The indoor pheromone effect was determined by male flight behavior showing wing movement in response to lure. Different mixtures of the synthetic pheromone components were prepared by mixing acetate and alcohol components in 100:0, 99:1, and 90:10 (g/g) and tested with a comparison of a standard commercial pheromone lure. The highest pheromone effect was observed in only acetate mixture (100:0) and the effect was reduced with the addition of the alcohol component. This indoor pheromone effect could be observed in field monitoring trial, in which 100:0 mixture showed the highest trap catches. Orientation disruption assay was conducted indoor by using a cage, in which the center had a commercial lure on sticky plate and the four candidates were placed at 6 cm away from the central lure on each of four directions. Test males were released to the arena during overnight (12 h) and then the caught males on the sticky plate were counted. The synthesized pheromone as well as the commercial pheromone showed 100% orientation disruption. However, the orientation disruption effect was reduced with decrease in the number of the surrounding disrupting pheromone baits. These results clearly suggest that the synthesized sex pheromone of G. molesta is biologically active and can be used for field mating disruption.

Production of ${\alpha}$1,3-Galactosyltransferase (GalT) Double Knock-out (-/-) Transgenic Pigs for Xenotransplantation (${\alpha}$1,3-Galactosyltransferase(GalT) 유전자가 완전 Knock-out(-/-)된 바이오장기용 형질 전환 돼지 생산)

  • This study was conducted to analyze the transgenic efficiency and sex ratio in ${\alpha}$-1,3-galactosyltransferase (GalT) knock-out (KO) transgenic pigs according to generation. GalT KO piglets were produced by artificial insemination or natural mating. The transgenic confirmation of GalT KO was evaluated by PCR amplification using specific primers. After electrophoresis, three types of bands were detected such as 2.3 kb single band (Wild), 2.3 and 3.6kb double bands (GalT KO -/+; heterozygote), and 3.6kb single band (GalT KO -/-; homozygote). Transgenic efficiency in F1 generation was 64.5% (23/35) of GalT KO (-/+). In F2 generation, GalT KO transgenic efficiency was 36.4% (21/57, Wild), 47.5% (28/57, GalT KO -/+), and 16.1% (8/57, GalT KO -/-), respectively. Interestingly, no homozygote piglets were born in 6 deliveries among total 11 deliveries, although they were pregnant between male (M) and female (F) $F_1$ heterozygote. In the 5 litters including at least one GalT KO -/- piglet, the transgenic efficiency was 13.3% (2/24, Wild), 51.3% (14/24, GalT KO -/+), and 35.3% (8/24, GalT KO -/-), respectively. The sex ratio of M and F was 40:60 in $F_1$ and 49:51 in $F_2$ generation, respectively. Based on these results, GalT KO transgenic pigs have had a reproductive ability with a normal range of transgenic efficiency and sex ratio.

Introgression of Sex-Limited Larval Markings to a Productive Multivoltine Strain of Silkworm Bombyx mori L.

  • A breeding programme was initiated during 2001 to introduce sex-limited larval markings to a productive multivoltine breed - BL67 from an inbred sex-limited line, MY1 (SL) maintained at Central Sericultural Research and Training Institute, Mysore. Introgressive hybridization, recurrent backcrossing for six generations followed by sib-mating resulted in synthesis of a new multivoltine silkworm breed BL67 (SL) with sex-limited larval markings. The new breed was studied for combining ability by crossing with eight bivoltine breeds viz., $NB_4D_2,\;CSR_2,\;CSR_2 (SL),\;CSR_2,\;CSR_4,\;CSR_8,\;CSR_{18}\;and\;CSR_{19}$. General combining ability effects of the new breed showed its superiority over the popular Pure Mysore by expressing significant GCA effects for six out of twelve characters whereas the results are on par with the original multivoltine breed. The hybrid $BL67(SL){\times}CSR2(SL)$ excelled in several quantitative characters such as pupation rate (90.2%), cocoon weight (1.97 g), cocoon shell weight (40 cg), cocoon shell ratio (20.3%), filament length (918 m), denier (2.96), raw silk percentage (14.96%) and neatness (90 p). Studies on cocoon size variability revealed that the cocoons of $BL67(SL){\times}CSR2(SL)$ were found comparatively uniform showing less standard deviation of 6.55 and co-efficient of variation of 3.91 %. The suitability of newly developed breed for easy grain age operation and commercial exploitation with promising hybrid have been discussed.

Genetic Variation in Growth and Body Dimensions of Jersey and Limousin Cross Cattle. 1. Pre- Weaning Performance

  • During a 5-yr period, 1994-1998, pre-weaning and weaning data were collected on 591 calves produced by mating either straightbred Jersey, straightbred Limousin or $F_1$ (Limousin${\times}$Jersey) bulls to mature purebred Jersey or Limousin cows. Traits recorded included birth and weaning weight, height, length, girth, fat depth and a measure of muscle (ratio of stifle to hip width expressed as a percentage). All traits were analyzed assuming a model with sire and dam random effects that included effects of year and date of birth, sex, breed and year${\times}$sex interaction. Main effects were generally significant with few exceptions. Direct genetic effects were large for weight, height, girth and muscle with a breed trend from purebred Jersey (small) to purebred Limousin (large). At weaning, the maternal effect of the Jersey dam was positive for weight (10.9${\pm}$4.9 kg), girth (3.7${\pm}$1.0 cm) and muscle (6.0${\pm}$0.9%). Heterosis was highly significant and positive only for fat depth (1.5${\pm}$0.2 mm) with the $F_1$ progeny being the fattest, followed by the backcrosses, then purebred Jersey and purebred Limousin. Also, significant (p<0.001) but negative heterosis was observed for weight, girth and muscle. The change in ranking for fat depth relative to other traits is a reflection of the large heterotic effects relative to direct effects on fat depth. Epistatic effects were not significant on any trait at birth or weaning. This study has indicated the possibility of exploiting the positive heterotic and maternal effects for fat depth and muscularity to meet specific meat quality and quantity demand by consumers.
