Developmental Toxicity Study in the Embryos/Fetuses with a Recombinant Human Granulocyte Colony-Stimulating Factor (YHB6211) in Pregnant Rabbits (임신토끼에 있어서 새로운 Recombinant Human Granulocyte Colony-Stimulating Factor(YHB6211)의 배.태자 발생독성평가)

  • YHB6211, a newly developed recombinant human granulocyte colonystimulating factor, was administered at dose levels of 0, 3, 15, and 75 $\mu$g/kg/day intravenously to the pregnant New Zealand White rabbits (20 rabbits per group) during the organogenetic period, days 6 to 18 of gestation. All dams were subjected to Caesarian section on day 28 of gestation and their fetuses were examined for external, visceral, and skeletal abnormalities. No abnormalities in clinical signs, body weight changes, gross findings, mortality, and external appearance were found in all dams and fetuses exposed to 0, 3, and 15 $\mu$g/kg/day of YHB6211. However, in the group treated with 75 $\mu\textrm{g}$/kg/day of YHB6211, maternal body and uterine weights, fetal body weights and length, and the number of live fetuses were significantly decreased and further fetal mortality was remarkably increased. It is suggested that YHB6211 may have no side effect up to the dose level of 15 $\mu$g/kg/day, and there would be no teratogenicity for fetuses of rabbits up to 75 $\mu\textrm{g}$/kg/day even if it may have some toxic effects over 75$\mu\textrm{g}$/kg/day for dams and fetuses of rabbits.

The Effects of Invasive Procedure Education Program on the Anxiety and Supportive Behavior of Mothers with Hospitalized Children (침습적 처치에 대한 교육 프로그램이 입원환아 어머니의 불안 및 지지행위에 미치는 영향)

  • Purpose: This study was to examine how an invasive procedures education program affected the anxiety and supportive behavior of mothers with hospitalized children, and to help provide an efficient to improve their maternal role performance by boosting their supportive behavior. Method: To evaluate their anxiety, Spilberger(1972)'s Anxiety Inventory that was translated by Kim Jeong-taek, et al.(1978) was employed, and Weon Dae-yeoung(1999)'s modified version of Melnyx(1994)'s Index of Parent Support During Instructive Procedure was utilized to assess their supportive behavior. The collected data were analyzed with SPSS program. Statistical data on real number and percentage were acquired, and $x^2$-test and t-test were conducted. Result: There was a more favorable change in the anxiety(trait anxiety and state anxiety) of the experimental group that participated in an invasive procedures education program(a mean of -0.14) than in that of the control group that didn't(a mean of 0.16). The experimental group(an average of 4..65) outstripped the control group(a mean of 3.42) in that regard. Conclusions: The invasive procedures education program that catered to the mothers with hospitalized children turned out to be effective in easing their anxiety and enhancing their supportive behavior.

Effects of Maternal Massage to Newborn on Stress, Immune Function and Self-confidence of Mothers during the Postpartum Period (신생아 마사지를 통한 모아상호작용이 어머니의 스트레스, 면역기능 및 영아기르기 자신감에 미치는 효과)

  • Purpose: To investigate the effect of infant massage on stress, immune function and self-confidence in postpartum mothers. Method: From September, 2003 to February, 2004, 65 women who had delivered babies and were enrolled in the postpartum clinic were assigned to the experimental group or control group. In the experimental group, infant massage was given for 20 minute sessions twice a day from 4 days to 14 days after delivery. The instruments were Parental Stress Index, serum cortisol levels, WBCs, lymphocytes, and Self Confidence Scale. Results: Stress scores, WBCs and lymphocytes showed significant differences by the time, but there weren't significant differences according to the group and interactions between the group and the time. Cortisol and self-confidence scores showed significant differences by the time and the group. Conclusions: Applying massage to their newborn baby influenced cortisol levels of postpartum mothers. Therefore stress, cortisol, and immune functions in postpartum mothers should be generally screened and follow up studies are needed for the effective application of massage in postpartum mothers.

The Influences of Parental Attachment on Social Competence of School-Aged Children : The Mediating Role of Empathy (부모에 대한 애착이 학령기 아동의 사회적 능력에 미치는 영향 : 공감 능력의 매개적 역할)

  • The effects of parental attachment and empathy on social competence were examined with a sample of 403 fifth and sixth graders (192 males, 211 females) attending elementary schools in Gyoung-gi province. All research variables were measured using self-reported questionnaires conducted by the children concerned. Data were analyzed by means of structural equation modeling. The results indicated that parental attachment had no direct effect, but there was an indirect effect on children's social competence mediated by children's empathy. Maternal attachment influenced children's social competence relatively more than paternal attachment. As compared to parental attachment, however, children's empathy was the more influential variable when it came to social competence. These results were consistent in both genders. The results of this study emphasize that children's own characteristics such as empathy tend to be important factors in developing social competence compared to the parental role in middle childhood. Accordingly, interventions aimed at improving children's empathy need to be emphasized in order to develop children's social competence regardless of gender.

Advantages of the outgrowth model for evaluating the implantation competence of blastocysts

  • The implantation process is highly complex and difficult to mimic in vitro, and a reliable experimental model of implantation has yet to be established. Many researchers have used embryo transfer (ET) to assess implantation potential; however, ET with pseudopregnant mice requires expert surgical skills and numerous sacrificial animals. To overcome those economic and ethical problems, several researchers have tried to use outgrowth models to evaluate the implantation potential of embryos. Many previous studies, as well as our experiments, have found significant correlations between blastocyst outgrowth in vitro and implantation in utero by ET. This review proposes the blastocyst outgrowth model as a possible alternative to animal experimentation involving ET in utero. In particular, the outgrowth model might be a cost- and time-effective alternative method to ET for evaluating the effectiveness of culture conditions or treatments. An advanced outgrowth model and further culture of outgrowth embryos could provide a subtle research model of peri- and postimplantation development, excluding maternal effects, and thereby could facilitate progress in assisted reproductive technologies. Recently, we found that outgrowth embryos secreted extracellular vesicles containing specific microRNAs. The function of microRNAs from outgrowth embryos should be elucidated in further researches.

Possible Epithelial Ovarian Cancer Association with HPV18 or HPV33 Infection

  • The present study was conducted to investigate the prevalence of HPV infection in epithelial ovarian cancer (EOC) in Hunan province. DNA samples were collected from paraffin embedded ovarian tissue from 322 patients with EOC, 99 with ovarian benign tumors and 199 normal persons. The polymerase chain reaction and direct sequencing were used to identify the HPV types in the samples. The relationship between the infection of human papillomavirus (HPV) and the epithelial ovarian carcinoma (EOC) was investigated combined with clinical data. The prevalence of HPV18 and HPV33 in EOC group and benign group was higher than in the normal group. HPV18 and HPV33 may play a role in the development of both EOC and ovarian benign tumor and may participate in the development of EOC with traditional risk factors, family history and abortion, possibly exerting synergistic effects.

  • Extensive studies have shown that breast milk is the best source of nutrition for infants, especially during the first six months, because it fulfills almost all of their nutritional needs. Among the many functional building blocks in breast milk, human milk oligosaccharides (HMOs) have been receiving more attention recently. Furthermore, it is the third most common group of compounds in human milk, and studies have demonstrated the health benefits it provides for infants, including improved nutritional status. HMOs were previously known as the 'bifidus factor' due to their 'bifidogenic' or prebiotic effects, which enabled the nourishment of the gastrointestinal microbiota. Healthy gastrointestinal microbiota are intestinal health substrates that increase nutrient absorption and reduce the incidence of diarrhea. In addition, HMOs, directly and indirectly, protect infants against infections and strengthen their immune system, leading to a positive energy balance and promoting normal growth. Non-modifiable factors, such as genetics, and modifiable factors (e.g., maternal health, diet, nutritional status, environment) can influence the HMO profile. This review provides an overview of the current understanding of how HMOs can contribute to the prevention and treatment of nutritional issues during exclusive breastfeeding.

Dietary Reference Intake of n-3 polyunsaturated fatty acids for Koreans

  • This paper examines the process and evidence used to create the Dietary Reference Intake (DRI) of alpha-linolenic acid (ALA) and eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) for Koreans. ALA (18:3n3) is an essential fatty acid, and EPA and DHA are known to have beneficial effects on cardiovascular disease risk and reduction of triglyceride levels. Various international organizations have suggested dietary recommendations for n-3 polyunsaturated fatty acids (PUFAs), including ALA, EPA, and DHA. A DRI for Koreans was established for the first time in 2020, specifically for the adequate intake (AI) of ALA and EPA + DHA. This recommendation was based on the average intake of ALA and EPA + DHA from the Korea National Health and Nutrition Examination Survey 2013-2017. For Korean infants, the AI of ALA and DHA was based on the fatty acid composition of maternal milk. Estimated average requirement and a tolerable upper intake level have not been set for n-3 PUFA due to insufficient evidence. In addition, the intake level of n-3 PUFA for prevention of chronic disease has also not been determined. Future studies and randomized controlled trials are required to establish the UL and to define the level for disease prevention.

EFFECTS OF CHILDREN'S PROBLEMATIC BEHAVIORS AND INTELLIGENCE ON MATERNAL DEPRESSION (아동의 문제행동과 지능이 어머니의 우울증에 미치는 영향)

  • Objectives : The purpose of the study was to evaluate the related factors of depression in mothers bringing their offspring for evaluation or treatment of problematic behaviors. Methods : Fifty mothers were recruited through their offspring's evaluation or treatment for offspring's problematic behaviors. Psychological tests battery including Korean Wechsler Adult Intelligence Scale and the Korean Educational Development Institute-Wechsler Intelligence Scale for Children was administered to all mothers and their offspring. Mothers completed Korean version of Child Behavior Checklist (K-CBCL), and teachers completed Teacher's Report Form (TRF). On the basis of the maternal depression, the mothers were divided into two groups. Results : 1) Among the fifty mothers, $42\%$ of the mothers had depressive disorder, $16\%$ had anxiety disorder, and $42\%$ had no psychiatric disorder. 2) Depressed mothers rated significantly higher in aggressive behaviors, externalizing problems of K-CBCL in their children. 3) Children of depressed mothers had lower score in picture arrangement and coding subtests of KEDI-WISC than those of no psychiatric disorder mothers. 4) After controlling for the total problematic behavior scores of TRF, mother's depression is positively correlated with aggressive behaviors, externalizing problems, total problematic behaviors of K-CBCL, and negatively correlated with coding of KEDI-WISC. Conclusion : Children's aggressive behavior, externalizing behavior, total problematic behaviors and low coding score may be associated with mother's depression.

Age- and Area-Dependent Distinct Effects of Ethanol on Bax and Bcl-2 Expression in Prenatal Rat Brain

  • Cell proliferation and differentiation are critical processes in a developing fetal rat brain, during which programmed cell death (PCD) also plays an important role. One of the decisive factors for PCD is Bcl-2 family proteins, where Bax induces cell death, whereas Bcl-2 acts as an inhibitor of PCD. As maternal drinking is known to cause fetal alcohol syndrome (FAS) or malformation of the fetal brain during pregnancy, the objective of the present study was to investigate whether maternal ethanol exposure alters the PCD-related Bax and Bcl-2 protein expression during fetal brain development. Pregnant female rats were orally treated with 10% ethanol and the subsequent expressions of the Bax and Bcl-2 proteins examined in the fetal brain, including the forebrain, midbrain, and hindbrain, from gestational day (GD) 15.5 to GD 19.5, using Western blots, in situ hybridization, and immunohistochemistry. With regard to the ratio of Bcl-2 to Bax proteins (Bcl-2/Bax), the Bax protein was dominant in the forebrain and midbrain of the control GD 15.5 fetuses, except for the hindbrain, when compared with the respective ethanol-treated groups. Moreover, Bcl-2 became dominant in the midbrain of the control GD 17.5 fetuses when compared with the ethanol-treated group, representing an alternation of the natural PCD process by ethanol. Furthermore, a differential expression of the Bcl-2 and Bax proteins was found in the differentiating and migrating zones of the cortex, hippocampus, thalamus, and cerebellum. Thus, when taken together, the present results suggest that ethanol affects PCD in the cell differentiation and migration zones of the prenatal rat brain by modulating Bax and Bcl-2 expression in an age- and area-dependent manner. Therefore, this is the first evidence that ethanol may alter FAS-associated embryonic brain development through the alteration of Bax and Bc1-2 expression.
