경남지역의 체세포수 문제 목장에서의 젖소 유방염 관리실태 및 발생양상 조사 (Survey of mastitis management and incidence of mastitis in high somatic cell count of bulk milk at dairy farms in the Gyeongnam)

  • Survey of mastitis management and incidence of mastitis in the Gyeongnam was started in May to September 2009 to solve mastitis problem statistically valid data for use in estimating mastitis management, isolation and antimicrobial drug susceptibility in 30 dairy farms having over 350,000/ml somatic cell count. In investigation on recognition of farmer about bovine mastitis, the ratio of understanding of differences between infectious and environmental origin, understanding of correlation between superbacteria and using indiscriminate, necessity of pathogen identification, and necessity of antimicrobial sensitivity tests were 80.0%, 73.3%, 33.3%, and 53.3%, respectively. In survey of mastitis management type, regular california mastitis test (CMT), conducting CMT test and empirical self-treatment, when detecting suspected cows, were 30.0%, 40.0%, and 46.7%, respectively. Checking and cleaning pulsators biweekly, cleaning vacuum system and replacing liners every 3~6 month, and getting milking system checked by engineers showed 80.0%, 76.7%, and 76.7% in the questionnaires, respectively. In recognition of farmer about milking hygiene for prevention of bovine mastitis, using individual towels, separated milking (milking order of cows), and teat-dipping disinfection after milking exhibited 13.3%, 86.7%, and 93.3%, respectively. In conclusion, through the questionnaires and laboratory test, we suggest that recognition of farmer about management and incidence of mastitis was very low, thus systemic educational program and public relations about mastitis management were need for dairy farmers.

대규모 젖소목장의 임상형 유방염 발생에 관한 후향적 연구 (A retrospective study of the incidence of clinical mastitis found in large-scale dairy herds in Korea)

  • Incidence of mastitis is highest in dairy cows and it is a leader of all diseases and the economic losses, in dairy farms. The objective of this study is to analyse the incidence of clinical mastitis using the lactation number, lactation month and season of calving. In this study, we used 3,779 lactation data and animal health records collected from 1990 to 2006 at the National Institute of Animal Science. Out of 3,779 cows, a total of 1,721 cows were reported with clinical mastitis, which was 46.3%. The frequency of mastitis increased from 36.9% from first lactation to 56.0% by the fifth lactation. As many as 766 cows (46.9%) showed a recurrence of mastitis after 14 days, apart for two or more and chronic mastitis that were recorded for 657 cows (20.3%). This came to a total of 3,010 cases that had clinical mastitis in the herd for sixteen years. The distribution of incidence of clinical mastitis was highest during the first month of lactation at 24.4%, and it reached to 43.4% by the third month. The incidence of mastitis was even higher for cows in first lactation, showing 28.6% in the first month, and 42.9 percent by third month of lactation. Cows calved during the summer and winter months showed higher rate of incidence with 59.9% and 57.9% within 30 days of lactation, respectively. It is urgently needed to establish a preventative management for heifer cows during their expected first lactation and the clinical specific dry-period management strategies that influence the rate of clinical mastitis during the next lactation. The aim of this study is to present information that might be useful to improve clinical mastitis prevention.

젖소 유방염 유래 Staphylococcus aureus의 PCR을 이용한 Genomic Fingerprinting (Genomic Fingerprinting of Staphylococcus aureus Isolated from Bovine Mastitis Milk by PCR)

  • A total of 137 strains of Staphylococcus aureus were isolated from dairy cow's milk with subclinical mastitis from 33 herds in 5 provinces and 36 strains of S aureus from clinical mastitis from 4 herds where the mastitis were severe problem. Arbitrary primed polymerase chain reactions with 10 bp oligonucleotide primer were performed and the PCR products were analysed with image analyzer, The S aureus strains were genotyped into 20 distinct DNA fingerprinting profiles. The size of PCR products ranged from 163 to 2,479 bp and PCR products of 506, 770, 784 and 2,479 bp were the most prevailing bands. Genotype 3 was founded in all 5 provinces. The various genotypes were identified in newly founded dairy herds, however, only one or two genotypes were identified in the closed herds. In clinical mastitis, only a limited number of different S aureus genotype was founded in each of the herds in comparision with subclinical mastitis. The results demonstrated that PCR-based DNA fingerprinting analysis of S aureus strain can be used to study epidemiology of mastitis, in addition, common genotype in geographic region can be useful for the development of an effective S aureus bacterin.

우리나라 젖소 유방염(乳房炎) 원인균(原因菌)의 역학적조사(疫學的調査) 및 치료(治療)에 관(關)한 연구(硏究) (Studies on Epidemiological Investigations and Therapy of Mastitis)

  • A total of 835 quarter milk samples of 212 dairy cows from 14 herds were examined for mastitis and the results obtained were as follows; 1. Three hundred and fifty-eight quarters(42.9%) from 149 cows(70.3%) were found to be infected with mastitis. It was found that 11(1.3%) of the infected quarters were clinical mastitis and all of the rest were subclinical mastitis. 2. Streptococcus agalactiae(62 quarters) and Staphylococcus aureus(42 quarters) were the main two causative organisms of the mastitis. Streptococcus dysgalactiae, Streptococcus uberis, other streptococci, Corynebacteria, and Yeast were also found to cause the infection. 3. The majority of Staphylococcus aureus strains were sensitive to penicillin, orbenin, terramycin, and leucomycin, however, the most of Streptococcus strains were sensitive to penicillin and orbenin only. 4. Penicillin and orbenin were highly effective in the treatment of mastitis, especially orbenin for Staph ylococcus aureus infection and penicillin for Streptococcal infection. 5. A mastitis control program for dairy farms in Korea was discussed and recommended.

유우(乳牛) 유방염(乳房炎) 진단(診斷)에 있어서 N-acetyl-β-D-glucosaminidase 치(値)와 분리(分離) 원인균(原因菌)과의 관계(關係) (Relationship between the N-acetyl-β-D-glucosaminidase levels and the presence of mastitis pathogens in bovine mastitis milk samples)

  • A study was carried out to define the relationship between the N-acetyl-$\beta$-D-glucosaminidase(NAGase) levels and isolated pathogenic bacteria in 379 quarter fore milk of mastitis suspected samples collected in this clinics. All samples were tested the NAGase, California mastitis test(CMT), Somatic cell count(SCC) and bacterial culture. Except 111 from 379 samples, 268 bacteria-positive quarter fore milk samples were classified into the latent and mastitis infection group by SCC($500,000cells\;per\;m{\ell}$), and the mean NAGase levels($nmol/min/m{\ell}$) of each isolated pathogen in mastitis infection group were Staphylococcus aureus 3.067, Coagulase-negative staphylococci 4.083, Staphylococcus aureus 3.594, Str. uberis 3.513, Str. dysgalactiae 1.640, E coli 4.441 and gram negative rods 4.560, respectively. Most of the relationship between mean SCC and NAGase in each pathogen group were highly significant using a student t test(p<0.05). When the mastitis pathogens were classified into minor(Coagulase-negative staphylococci, Corynebacterium sp.) and major pathogen group(Staphylococcus aureus, Streptococcus agalactiae, Str. uberis, Str. dysgalactiae, gram negative rods), the NAGase levels were higher at major than minor pathogen group. On the other hand, when the mastitis milk samples were classified by SCC($500,000cells\;per\;m{\ell}$) and by the presence of pathogen(IDF scheme), the NAGase levels were also higher at the mastitis than latent infection. The possibility of combining SCC and NAGase data in order to give the more difinitive diagnosis is discussed.

Molecular characteristics and antimicrobial susceptibility profiles of bovine mastitis agents in western Türkiye

  • Importance: Identifying bovine mastitis agents using molecular methods to reveal their phylogenetic relationships and antimicrobial resistance profiles is essential for developing up-to-date databases in mastitis cases that cause severe economic losses. Objective: This study examined bacterial mastitis agents in cows with clinical and subclinical mastitis observed in various dairy cattle farms to reveal their phylogenetic relationships and antibiotic resistance properties. Methods: Sixty-two clinical and subclinical bovine mastitis milk samples were collected from 15 dairy farms. The polymerase chain reaction (PCR) was used to amplify the 16S rRNA gene regions of the bacteria. The 16S rRNA gene sequences obtained from sequencing include the V4-V6 regions. The strains were compared using a similarity analysis method that produced phylogenetic trees using the Molecular Evolutionary Genetics Analysis 11 program. Antibiotic susceptibilities were determined using the Kirby-Bauer disk diffusion method. Results: Sixty-three bacteria were isolated and identified in this study. The most isolated bacteria from all mastitis cases were Staphylococcus spp. (30.2%), Escherichia coli (25.4%), Streptococcus spp. (14.3%), and Aerococcus spp. (7.9%), respectively. The phylogenetic trees were drawn from the 16S rRNA sequences. Some of these bacteria showed resistance to different types of antibiotics at varying rates. Conclusions and Relevance: The bacteria isolated in this study originated from environmental sources. Regular cleaning of barns and proper hygiene practices are essential. Regular screenings for mastitis should be conducted in herds instead of the random or empirical use of antibiotics.

  • A total of 210 dairy cows of Seongwhan area were epidemiologically investigated for mastitis by Milk Quality Test(MQT) method. Milk samples from cows infected with mastitis were also bacteriologically studied. Seventy four (42.5%) of 174 dairy cows were positive for mastitis and fifty nine (79.7%) among them were infected with subclinical mastitis and fifteen (20.3%) were clinical mastitis. High infective rate were shown on August (48.7%) and July (45.7%) and it was lowered on December (35.8%). The incidence rate of subclinical mastitis for herd shown tendency of increase at the barn in which located on lower place (30.7~43.9%), whereas, the barn in which was on higher place and with bucket type milking system shown tendency of decrease causative organisms. A total of 230 strains of etiological agents were isolated from 578 infected duarters which shown MQT score (+) or more. The relevant causative organisms isolated in this study were Staphylococcus aureus (18.3%), Staphylococcus epidermidis (16.9%), fungi (16.5%), Pseudomonas aeruginosa (15.2%), Escherichia coli (7.8%) and Serratia marcescens (2.27), respectively.

EFFECT OF IMMUNOPOTENTIATING AGENTS ON SUBCLINICAL MASTITIS IN CATTLE AND BUFFALOES

  • Different parameters of subclinical mastitis were compared in 327 cattle and 493 buffaloes and the effect of immunopotentiating agents on subclinical mastitis in these animals was studied. Subclinical mastitis was detected in 8.2 percent buffalo and 24.0 percent cow quarter by modified whiteside test (MWT). In both the species there was decrease in lactose contents with increase in the degree of MWT reactivity i.e. $4.8{\pm}1.14$ to $2.31{\pm}0.82$ in cattle and $5.01{\pm}1.47$ to $2.36{\pm}1.02$ in buffaloes. While the chloride contents of the milk increased with increase in the MWT reactivity i.e. $0.19{\pm}0.4$ to $0.30{\pm}0.06$ in cattle and $0.20{\pm}0.04$ to $0.31{\pm}0.13$ in buffaloes. Micro-organisms belonging to Staphylococcus, Micrococcus, Bacillus, Streptococcus, Enterobacteria, Corynebacterium groups and yeasts were isolated from subclinical mastitis cases. Vitamin E and Levamisole cured 64.5 and 60.0 percent cases of subclinical mastitis in buffaloes but only 32.0 and 24.0 percent cases in cattle. Cure was not affected by the degree of MWT and the type of organisms involved.

乳牛의 乳房炎에 관한 疫學的 연구 -전남지역을 중심으로- (Epidemiological Study on the Bovine Mastitis in Chonnam Area)

  • Epidemiological study on the bovine mastitis was carried out on 1,493 raw milk samples from 7,406 dairy cows of dairyfarms in Chonnam province. And, it were classified by R. B. V. test during the period from July, 1984 to June, 1986. Of these positive samples, the mastitis causative agents were isolated, and examined for susceptibility to 14 antibiotics by disc method. The results are summarized as follows 1. Incidence rate of bovine mastitis from raw milk examined was observed in 1,393 dairy cows (18.8%) among 7,406 dairy cows of dairy farms. These outbreaks according to years were increased to 17.1% in 1984, 19.7% in 1985, and 20.5% in 1986. And, outbreaks of bovine mastitis according to seasons were high observed during the period from August to October. 2. Isolated rate of mastitis causative agents from raw milk was observed in the order of Staphylococcus sp. (50.2-51.7%), Streptococcus sp. (35.6-48.3%), Bacillus sp. (2.2-6.7%), Pseudomonas sp. (0.3-5.7%), and others (0.6%). 3. Generally, antibiotics susceptibility of isolated mastitis agents from raw milk was observed high susceptible in CP, CL, NB, PC, SM, NM, EM, GM etc.
