Indigo crop, Persicaria tinctoria H. Gross is an annual plant containing natural colorant, the blue dye indigo, and local cultivars had been cultivated to produce natural indigo for textile dyeing in Korea since ancient times. Naju No. 2 is a new mass-selected line from the mother population, Naju Local cultivar. In this study, two breeding lines of Naju Local and Naju No. 2, have been cultivated in four different locations, the South regions of Korea, to compare plant growth and yield characteristics between two lines. Naju No. 2 was higher in plant height, and Naju Local has more 1st branches. Naju No. 2 has larger leaf area and higher width/length ratio of leaf, showing the round leaf type as morphological stable character without regional differences. Though there was considerable regional variation in fresh and dry top weight of harvested plant, the significant difference of plant weight between two lines were not shown. The ratio of leaf to total shoot of dry weight of Naju No. 2 was higher than one of Naju Local, indicating that Naju No. 2 has better yielding of colorant which is synthesized mostly in leaf. Naju No. 2 contained more Niram (crude indigo extract) and indigo, and showed much blueness at dyeing of silk using fresh leaves than Naju Local. We concluded that a new line, Naju No. 2 could be a superior cultivar due to having higher leaf yield and better quality of natural colorant than Naju local cultivar.
Because of the high water content of the household garbage, it is difficult to compost it. Therefore, this study was conducted to investigate the possibility of using coal fly ash as humidity conditioner for the household garbage composting. The summarized results are as follows : 1. The maximum temperatures were $35^{\circ}C$ in spring, $47^{\circ}C$ in summer, and $43^{\circ}C$ in winter during the composting periods. 2. The mass was reduced to 70.5% after 60 days. The average volume reduced down to 74.7% after 60 days. 3. The seasonal variation of pH values showed a similar tendency and reached 8.5 after 60 days. 4. The water content was reduced at an early stage of composting and not much changed thereafter. It had 49.7% in spring, 33.9% in summer, and 56.5% in winter after 60 days. Ash contents were not much changed during the composting periods. 5. The contents of inorganic compounds were in the range of $0.06{\sim}4.03%$$P_2O_5$, $0.01{\sim}2.87%$ CaO, $0.18{\sim}1.43%$ MgO, and $0.39{\sim}2.03%$$K_2O$. Heavy metal contents were in the range of $ND{\sim}14.08$ Hg ${\mu}g/kg$, $ND{\sim}0.80$ Cd mg/kg, $4.99{\sim}28.95$ Cu mg/kg, $ND{\sim}242.62$ Cr mg/kg, $ND{\sim}20.24$ Pb mg/kg, and $ND{\sim}59.87$ Zn mg/kg.

To understand the cross-sectional structures of temperature, salinity and current across the Korea Strait, field measurements were carried out for the period of May 2 to 20, 1994. Using the R/V Tam Yang, detailed CTD profiles and ADCP records were obtained and used to examine the mean and variability field on two time scales (15 days and 25 hours). A sharp coastal front in the middle of the Korea Strait exists across which two different water masses, i.e., warm and saline water in the eastern side and cold and less saline water in the western side are neighboring. We observed highly variable field of T and S apparently caused by the westward movement of warm and saline water mass. Short-term fluctuations of T and S in the middle layer are remarkable and their importance was analysed as the first Eigen mode accounting for more than 50% of total variances. The currents in th Korea Strait are strongly influenced by tidal currents with spring and neap variation whose maximum speed ranges 80-90 and 60-70 cm/s respectively near the central portion of the channel. Strong southward tidal current could even mask the Tsushima Current completely. Results of harmonic analysis show that the magnitudes of semidiurnal, diurnal and mean components of currents are comparable to each other at spring and neap tide conditions. The volume transport across the western channel of the Korea Strait were estimated to be 2.1 Sv at neap tide condition and 3.4 Sv at spring tide condition.
In general, the proliferation of orchids via somatic embryos has been used for mass production of somatic clones because of high propagation efficiency. In spite of high propagation rate, this method often brings somaclonal variation, especially polyploid frequency. Therefor we here concentrated to investigate the relationship between endopolyploidization patterns of explants and the occurrence of tetraploid variant in clonally proliferated Doritaenopsis via somatic embryo regeneration system. In the fully developed somatic embryo, upper part contained 2C to 16C while middle and lower parts showed 2C to 32C DNA content. Two-week-old embryo contained 2C to 16C, whereas those regenerated after 4 to 10-week-old contained 2C to 64C nuclei. Results showed that endoreduplication was variable depending upon tissue types, ages, and parts in one species. lower part of somatic embryo having high endoreduplication degree increased the regeneration of tetraploid variants by about 3-fold comparing to upper part of somatic embryo culture. polyploid frequency occurrence might be closely related to the high levels of endoreduplication of somatic embryos used as explant. It suggested that the upper part of somatic embryo having comparatively low endoreduplication degree is suitable for the stable in vitro propagation system.
Ergosterol is an indicator chemical of fungi and involves in fungal cell growth as a major component in fungal cell membranes. Thus, this chemical can be used to estimate live fungal biomass within various solid substrates. Ergosterol content in liquid culture of Lentinula edodes had a linear relationship (r=0.98) with the hyphal mass of the fungus. This chemical content differed depending on the fungal strains, age of culture and water content levels of sawdust substrate. Ergosterol content was 0.13% in the 10 weeks old liquid culture while it was 0.10% in 20 weeks old one. The chemical content in the sawdust cultures of the fungus varied 0.015% to 0.042% depending upon strains and water content levels within sawdust substrate. Ergosterol content in the culture of a L. edodes strain, Sanrim 4, was higher by $20{\mu}g$ to $140{\mu}g$/g dry substrate than those of strains, Mok-H and Sanrim 6. And the chemical contents in the sawdust cultures with 125% or 175% water, 297 to 425 ${\mu}g$/g dry substrate, were higher than those with 75% or 225% water, 148 to $286{\mu}g$/g dry substrate. We conclude that ergosterol analysis can estimate the fungal biomass within solid substrate such as logs and sawdust.
Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.
The spreading fire of very small floating particles after they are ignited is fast and t therefore dangerous. The research on this area has been limited to experiments and global simulations which treat them as dusts or gaseous fuel with certain concentration well m mixed with air. This research attempted micro-scale analysis of ignition of those particles modeling them as liquid droplets. For the beginning, the in-line array of fuel droplets is modeled by two-dimensional, unsteady conservation equations for mass, momentum, energy and species transport in the gas phase and an unsteady energy equation in the liquid phase. They are solved numerically in a generalized non-orthogonal coordinate. The single step chemical reaction with reaction rate controlled by Arrhenius’ law is assumed to a assess chemical reaction numerically. The calculated results show the variation of temperature and the concentration profile with time during evaporation and ignition process. Surrounding oxygen starts to mix with evaporating fuel vapor from the droplet. When the ignition condition is met, the exothermic reactions of the premixed gas initiate a and burn intensely. The maximum temperature position gradually approaches the droplet surface and maximum temperature increases rapidly following the ignition. The fuel and oxygen concentration distributions have minimum points near the peak temperature position. Therefore the moment of ignition seems to have a premixed-flame aspect. After this very short transient period minimum points are observed in the oxygen and fuel d distributions and the diffusion flame is established. The distance between droplets is an important parameter. Starting from far-away apart, when the distance between droplets decreases, the ignition-delay time decreases meaning faster ignition. When they are close and after the ignition, the maximum temperature moves away from the center line of the in-line array. It means that the oxygen at the center line is consumed rapidly and further supply is blocked by the flame. The study helped the understanding of the ignition of d droplet array and opened the possibility of further research.

BACKGROUND: Pesticides are broadly used to control weeds and pests, and the residues remaining in crops are managed in accordance with the MRLs (maximum residue limits). Therefore, an analytical method is required to quantify the residues, and we conducted a series of analyses to select and validate the quick and simple analytical method for tolpyralate in five agricultural products using QuEChERS (quick, easy, cheap, effective, rugged and safe) method and LC-MS/MS (liquid chromatography-tandem mass spectrometry). METHODS AND RESULTS: The agricultural samples were extracted with acetonitrile followed by addition of anhydrous magnesium sulfate, sodium chloride, disodium hydrogencitrate sesquihydrate and trisodium citrate dihydrate. After shaking and centrifugation, purification was performed with d-SPE (dispersive-solid phase extraction) sorbents. To validate the optimized method, its selectivity, linearity, LOD (limit of detection), LOQ (limit of quantitation), accuracy, repeatability, and reproducibility from the inter-laboratory analyses were considered. LOQ of the analytical method was 0.01 mg/kg at five agricultural products and the linearity of matrix-matched calibration were good at seven concentration levels, from 0.0025 to 0.25 mg/L (R2≥0.9980). Mean recoveries at three spiking levels (n=5) were in the range of 85.2~112.4% with associated relative standard deviation values less than 6.2%, and the coefficient of variation between the two laboratories was also below 13%. All optimized results were validated according to the criteria ranges requested in the Codex Alimentarius Commission (CAC) and Ministry of Food and Drug Safety (MFDS) guidelines. CONCLUSION: In conclusion, we suggest that the selected and validated method could serve as a basic data for detecting tolpyralate residue in imported and domestic agricultural products.
Staurolite-biotite-andalusite-garnet (SBAG) assemblage and its sub-assemblages (SBA and SBG) commonly occur in the Dueumri Formation near the Chunyang granite, belonging to andalusite and sillimanite zones. The occurrence of the SBAG mineral assemblage is unusual because it is univariant in the $K_2O-FeO-MgO-Al_2O_3-SiO_2-H_2O$ (KFMASH) model system. We used projection and singular value decomposition (SVD) methods to investigate the equilibrium relationship between SBAG and its sub-assemblage. The SVD modelling of single specimen containing the SBAG assemblage suggests no reaction relationship with respect to mass-balance. Thus, the SBAG assemblages are stabilized by non-KFMASH component. On the other hand, the AFM-Mn projection suggests a reaction relationship between SBAG and its sub-assemblage because they intersect each other in this composition space. The SVD modelling, however, suggests no reaction relationship between these assemblages. Thus, the SBAG assemblages are likely to be stabilized by the variation in bulk-rock composition and/or 1.1~2,. The stable occurrence of staurolite in the sillimanite zone is compatible with pressure estimates from the garnet-plagioclase-biotite-muscovite geobarometer.

Diabetes is a disease that contains a high concentration of glucose in blood and due to defects in either insulin secretion or insulin action. Although the distinctive causes and factors of diabetes have not been clarified, the genetic factors are suggested as a main susceptibility until now. SNP (Single Nucleotide Polymorphism), as the most common genetic variation, has an influence on personal susceptibility for diseases. A nonsynonymous SNP, which changes the amino acid of the protein and its function, is especially important. Therefore, this study hypothesized that there are associations between specific SNPs of the targeted genes. Transcription factor 7-like 2 (TCF7L2) and fat mass and obesity associated (FTO) genes were selected as target genes from the results of genome-wide association and other related research studies. Second, four nonsynonymous SNPs (three in TCF7L2 and one in FTO gene) were selected as target SNPs by using public database of NCBI (National Center for Biotechnology Information). The recruited personnel was classified into three subgroups of diabetes, impaired fasting glucose (IFG) and normal groups. The individual genotypes of each group were analyzed by resequencing. None of genetic variations at four targeted SNP sites was revealed in all samples of this study. However, this study found two new SNPs that were not reported in TCF7L2 gene. One is synonymous SNP, which is heterozygous of C/T and no amino acid change of asparagine/asparagines, was located at c1641 and found in one normal person. Another is nonsynonymous SNP, which is heterozygous of G/A, was located at c1501 and found in two samples. This new discovered nonsynonymous SNP induce the amino acid change from alanine to threonine. Moreover, this new nonsynonymous SNP was found among two persons, one of whom was a diabetes patient and the other one was a person at boundary between IFG and normal, suggesting that this variant might be associated with IFG or diabetes. Even if there is a limitation of sample number for statistical power, this study has an importance due to the discovery of new SNPs. In the future study, a large sample number of diabetes cohort will be needed to investigate the frequency and association with new discovered SNP.
