An improved mass spectrometric method for multicomponent analysis of metabolites in urine, well-suited for clinical biochemistry, is described. The method involves solvent elution of the metabolites from an adsorbent and the concentration of the eluate on a microadsorption column. This is administered by a direct inlet probe into the ionizing source of field ionization mass spectrometry (FIMS), which yield a molecular weight profile of the metabolites. The procedure provides rapidly (within one hour) reproducible profiles from a small volume of urine. The optimization of the sampling technique and the reproducibility are discussed.

Electrospray ionization mass spectrometry (ESI MS) was used, along with gas chromatography-mass spectrometry (GC-MS), to monitor Paal-Knorr synthesis of 2,5-dimethyl-1-phenylpyrrole by condensation of aniline with 2,5-hexanedione. In addition to 2,5-dimethyl-1-phenylpyrrole observed as a single spot by TLC, unexpected dimer size compounds were observed by GC-MS. Dimers and trimers were observed by ESI MS. ESI tandem mass spectrometry was used to select plausible structures for the dimer. ESI MS with or without liquid chromatographic separation is useful for observing oligomeric byproducts with low volatility produced in organic reactions.
The purpose of this paper is to introduce the simplified linkage position determination method using tandem mass spectrometry combined with molecular modeling study. Using low energy tandem mass spectrometric experiments and molecular modeling, it has been suggested that significant differences in glycosidic bond cleavage may occur due not only to ionic considerations but also may have contributions from steric hindrance of the absorbance of collision energy, leading to a statistically higher bond cleavage for sterically crowded linkages. Permethylated derivatives of the linkage-isomeric trisaccharides give useful fragmentation ratios and productions, including a 3-linkage specific ion. The ratios of fragment ions are related to the ability of each linkage position in the oligosaccharide to absorb collisional energy.

Contamination of microcystins, a family of heptapeptide hepatotoxins, in eutrophic water bodies is a worldwide problem. Due to their poisoning effects on animals and humans, there is a requirement to characterize and quantify all microcystins present in a sample. As microcystins are, for most part, intracellular toxins produced by some genera of cyanobacteria, lysing cyanobacterial cells to release all microcystins is considered an important step. To date, although many cell lysis methods have been used, little work has been conducted comparing the results of those different methods. In this study, various methods for cell lysis and toxin extraction from the cell lysates were investigated, including sonication, bead beating, freeze/thaw, lyophilization and lysing with TritonX-100 surfactant. It was found that lyophilization, followed by extraction with 75% methanol, was the most effective for extracting toxins from Microcystis aeruginosa cells. Another important step prior to the analysis is removing impurities and concentrating the target analyte. For these purposes, a C18 Sep-Pak solid phase extraction cartridge was used, with the percentage of the eluent methanol also evaluated. As a result, methanol percentages higher than 75% appeared to be the best eluting solvent in terms of microcystin-leucine-arginine (MC-LR) recovery efficiency for the further chromatographic and mass spectrometric analyses.
A high-performance liquid chromatography-electrospray ionization-tandem mass spectrometric method was developed to determine simultaneously eight marker constituents of Forsythiae fructus, and subsequently applied it to classify its two botanical origins. The marker compounds of Forsythia suspensa were phillyrin, pinoresinol, phillygenin, lariciresinol and forsythiaside; those of F.viridissima were arctiin, arctigenin and matairesinol. Separation of the eight analytes was achieved on a phenyl-hexyl column (150${\times}$2.0 mm i.d., 3 ${\mu}M$) using gradient elution with the mobile phase: (A) 10% acetonitrile in 0.5% acetic acid, (B) 40% aqueous acetonitrile. A few fragment ions specific to the types of lignans, among the product ions generated by collisonally induced dissociation (CID) of molecular ion clusters, such as [M-H]$^-$ or [M+OAc]$^-$ were used not only for fingerprinting analysis but for the quantification of each epimer by using multiple-reaction monitoring mode. It was shown good linearity ($r^2{\geq}$ 0.9998) over the wide range of all analytes; intra- and inter-day precisions (RSD, %) were within 9.14% and the accuracy ranged from 84.3 to 115.1%. The analytical results of 40 drug samples, combined with multivariate statistical analyses - principal component analysis (PCA) and hierarchical cluster analysis (HCA) - clearly demonstrated the classification of the test samples according to their botanical origins. This method would provide a practical strategy for assessing the authenticity or quality of the herbal drug.
본 연구는 콩 함유 사포닌의 함량을 쉽고 빠르게 정량할 수 있는 방법을 개발하기 위하여 시도하였다. 사포닌 표준 물질인 soyasaponin I은 대두에서 직접 분리하여 동정하였고, 분석은 HPLC/MS/MS를 이용하였으며 그 결과는 다음과 같다. 1. DAD 또는 ELSD를 이용하여 분석할 때보다 전처리 과정을 획기적으로 줄일 수 있어 신품종 육성의 선발과 같은 대량의 분석에 적합하였다. 2. Soyasaponin I의 함량은 나물콩과 같은 소립종에서 대립종에 비해 함량이 유의하게 높은 것으로 나타났다.

The total burnup in the spent U$_3$Si/Al fuel samples from Hanaro reactor was determined by destructive methods using $^{148}$ Nd, the sum of $^{143}$ Nd and $^{144}$ Nd, the sum of $^{145}$ Nd and $^{146}$ Nd, and the sum of total Nd isotopes($^{143}$ Nd, $^{144}$ Nd, $^{145}$ Nd, $^{146}$ Nd, $^{148}$ Nd and $^{150}$ Nd) monitors. The fractional($^{235}$ U) turnup in the spent fuel samples was also determined by U and Pu mass spectrometric method. The samples were dissolved in a mixture of 4 M HCI and 10 M HNO$_3$ without any catalyst. The separation of U, Pu and Nd from the spiked and unspiked sample solutions was achieved by two sequential anion exchange separation methods. The isotope compositions of these elements, after their separation from the fuel samples were measured by mass spectrometry. The contents of the elements in the spent fuel samples were determined by isotope dilution mass spectrometric method(IDMS) using $^{233}$ U, $^{242}$ Pu and $^{150}$ Nd as spikes. The effective fission yield was calculated from the weighted fission yields averaged over the irradiation period. The difference between total turnup values determined by various Nd monitors were in the range of 1.8%.

Formation and characterization of self-assembled monolayers (SAMs) on various surfaces are the essential basis for many other applications, including molecular switches, biosensors, microfluidics, and fundamental studies in surfaces and interfaces. To improve the performance at these applications, it is a key to control the quantity of each molecule in various mixed SAMs on the surface. In this study, using mixed SAM of carbamate-based hydroquinone (HQ)-PhBr and11-mercaptoundecanol, the quantitative mass spectrometric method of mixed SAM was developed based on comparison study with XPS and FT-IR methods. In addition, our method was applied to another mixed SAM of biotinylated PEG alkane thiol and 11-mercaptoundecanol for verification purpose. Time-of-flight secondary mass spectrometry (ToF-SIMS) analysis was performed to identify and quantify each molecule of mixed SAM along with principal component analysis (PCA). Since there is no matrix effect in the X-ray photoelectron spectroscopy (XPS) and Fourier transform-infrared (FT-IR) techniques, we compared ToF-SIMS results with XPS and FT-IR results. Because PCA results from ToF-SIMS analysis are well matched with XPS and FT-IR results from both mixed SAMs, we are expecting that our method will be useful to identify and quantify each molecule in various mixed SAMs.
Quadrupole mass spectrometer를 이용한 발효 배기가스의 분석을 통해 세포의 증식을 on-line monitoring하고자 model 균주로 Candida utilis에 대해 연구하였다. Quadrupole mass spectrometer와 interface된 16-bit 개인용 컴퓨터 (IBM PC-AT)에서 산소 소비속도(OUR)와 이산화탄소 발생속도(CER)를 on-line 계산할 수 있었고 계속해서 이들 계산치로부터 세포농도와 증식속도 및 비증식속도를 계산하였다. 계산된 값들은 실험적으로 측정한 세포농도 및 비증식속도와 잘 일치함을 알 수 있었다.
네일락카와 헤어스프레이 같은 화장품에 존재하는 미량의 프탈레이트를 정량분석하기 위하여, 가스크로마토그래피와 질량분석기를 사용한 효과적이면서 환경친화적인 분석방법을 개발하였다. 이들 화장품들은 다량의 유기용매를 함유되어 프탈레이트를 분석하기 위하여 널리 사용되는 시료의 클린업 방법이 적합하지 않았다. 더군다나 미량의 프탈레이트 분석시에는 실험과정 중에서의 오염으로 인해 실제보다 높은 분석값을 산출하게 되는 경우가 매우 많다. 이에 정확한 함량분석 및 이차오염을 방지하기 위해 유기용매를 사용하여 시료를 직접 희석하는 시료 전처리를 적용하였다. 이 분석방법은 높은 정확성, 분석감도, 그리고 시료전처리를 간략히 할 수 있는 이점을 가진다. 화장품에서의 검출되는 빈도가 높고, 사람과 동물에 영향을 미치는 환경호르몬으로 보고되는 dibutyl phthalate (DBP)와 di (2-ethylhexyl) phthalate (DEHP) 두 종의 프탈레이트를 분석대상으로 선정하였다. 정량시 그 정확도 향상을 위해서 내부표준물질로 두 물질의 중수소치환체인 DBP-$d_4$,와 DEHP-$d_4$를 사용하였다. 시험법의 유효화를 시행한 결과 본 시험법이 ppm 농도의 프탈레이트 정량분석에 적합함을 확인하였으며, 네일락카와 헤어스프레이 제품에 약 $25{\mu}g/g$의 농도로 표준물질을 첨가하여 분석한 회수율은 95 - 106.1 % 범위였고, % 상대표준편차 값은 3.9 % 이하였다.
