Optimization of Medium Components for Cell Mass Production of Saccharomyces cerevisiae JUL3 using Response Surface Methodology (반응표면분석법을 이용하여 Sacharomyces cerevisaeJUL3의 균체량 생산을 위한 배지조성 최적화)

  • [ ${\beta}-Glucan$ ], one of the cell wall components, is most plentiful polysaccharides in cell wall and has several advantages in immune system. In yeast ${\beta}-glucan$ is mainly contained in the yeast cell wall, and thus it is important to produce high levels of cell mass for the mass production of yeast ${\beta}-glucan$. The best carbon and nitrogen sources on cell mass production were high fructose syrup and yeast extract. Response surface methodology (RSM) was very potential tool for the optimization of process factor and medium component. It was applied to estimate the effects of medium components on the production of cell mass. Optimal concentrations of high fructose syrup and yeast extract by response surface methodology were 8.0% (v/v) and 5.2% (w/v), respectively and the cell mass predicted was $17.0\;g/{\ell}$ at 20 h of cultivation.

  • Mass production system of most manufacturing companies has been supplying us with various useful needs in life for many years. It leads our life to live in comfortable and prosperous circumstances. And we can consider that the contributions of the mass production system to the reduction of different stratification in community remarkable . However, The needs of many clients in these days are various, different, and segmented. It is undesirable to consider every Product on the same basis anymore. So these new requirements of the customers let manufacturing companies be ready for the new production systems which are able to satisfy all the customers, but differently. In this aspects, the customization system could meet new needs of the customers. As we know, cost increase and flexibility matter for the production are main difficulties for the introduction to mass customization. However, to make arrangements for the mass customization systems of the firms is an indispensible factor for maintaining competitiveness and coping with new needs in the future market. We should remember that customers' needs in market will be changing, segmenting, and diversifying, continuously. For this purpose this thesis is tried for studying strategical approach methods of the mass customization.

  • Quickness and precision are the two most important requirements for an industrial scale used in production lines. In this paper, a new approach, "Dynamic-Mass measurement control System of Acceleration and Displacement(DMS-AD) sensing", is presented to improve some of drowbacks in conventional scales. The system, consisted of acceleration and displace- ment sensors, spring scale and microcomputer, is based on full utilization of dynamic mass measurement of acceleration and displacement via microcomputer-assisted real time monitoring. The rsulting system, when combined with appropriate dynamic mass estimation algorithm software, has shown its effectiveness in terms of two desirable characteristics required. required.

Dynamic response of a beam on multiple supports with a moving mass

  • The dynamic behavior of an Euler beam with multiple point constraints traversed by a moving concentrated mass, a "moving-force moving-mass" problem, is analyzed and compared with the corresponding simplified "moving-force" problem. The equation of motion in matrix form is formulated using Lagrangian approach and the assumed mode method. The effects of the presence of intermediate point constraints in reducing the fluctuation of the contact force between the mass and the beam and the possible separation of the mass from the beam are investigated. The equation of motion and the numerical results are expressed in dimensionless form. The numerical results presented are therefore applicable for a large combination of system parameters.

Litter Production and Decomposition in the Querces acutissima and Pinus rigida Forests (상수리나무림과 리기다소나무림의 낙엽 생산과 분해)

  • Litter production and decomposition were investigated for 2 years in the oak, Quercus acutissima, and the pitch pine, Pinus rigida, stands in the vicinity of Kongju, Chungnam Province. Litter production was measured with litter trap at monthly basis. Litterbag method was used for the measurement of litter decomposition. Litter producion continued throughout the year, but showed a peak in autumn. Second peak in May or June was caused by falling of bud scales and reproductive organs. Average litter production in the oak and the pitch pine stands were $567.1g{\cdot}m^{-2}{\cdot}yr^{-1}\;and\;653.2g{\cdot}m^{-2}{\cdot}yr^{-1}$, respectively. Litter production in this study area were higher than those in other reports. Nutrient concentrations in litter were the highest in summer when the least litter production occurred, and the lowest in autumn when the greatest litter production occurred, except for calcium in the oak stand. Nutrient concentrations of the oak litter were higher than those in the pitch pine litter. After 1 year, % remaining mass of oak and pitch pine litter was 43.6% and 58%, respectively. After 21 months elapsed, % remaining mass of oak and pitch pine litter was 22.2% and 33.2%, respectively.

Influence of Additives on the Yield and Pathogenicity of Conidia Produced by Solid State Cultivation of an Isaria javanica Isolate

  • Recently, the Q biotype of tobacco whitefly has been recognized as the most hazardous strain of Bemisia tabaci worldwide, because of its increased resistance to some insecticide groups. As an alternative control agent, we selected an Isaria javanica isolate as a candidate for the development of a mycopesticide against the Q biotype of sweet potato whitefly. To select optimal mass production media for solid-state fermentation, we compared the production yield and virulence of conidia between 2 substrates (barley and brown rice), and we also compared the effects of various additives on conidia production and virulence. Barley was a better substrate for conidia production, producing $3.43{\times}10^{10}$ conidia/g, compared with $3.05{\times}10^{10}$ conidia/g for brown rice. The addition of 2% $CaCO_3+2%$ $CaSO_4$ to barley significantly increased conidia production. Addition of yeast extract, casein, or gluten also improved conidia production on barley. Gluten addition (3% and 1.32%) to brown rice improved conidia production by 14 and 6 times, respectively, relative to brown rice without additives. Conidia cultivated on barley produced a mortality rate of 62% in the sweet potato whitefly after 4-day treatment, compared with 53% for conidia cultivated on brown rice. The amendment of solid substrate cultivation with additives changed the virulence of the conidia produced; the median lethal time ($LT_{50}$) was shorter for conidia produced on barley and brown rice with added yeast extract (1.32% and 3%, respectively), $KNO_3$ (0.6% and 1%), or gluten (1.32% and 3%) compared with conidia produced on substrates without additives.

Development of Solid Culture Media for the Mass Production of Beauveria bassiana Conidia (흰굳음병균(Beauveria bassiana) 포자의 대량배양을 위한 고체배지의 개발)

  • To develope a solid culture media for the mass culture of Beauveria bassiana conida, wheat bran was selected as C source and N source. The pellet media were prepared without(P-I) and with(P-II) solidified ingredient. The moisture for the growth of B. bassiana was required over 4 : 2 of Media : D.W (W/V). The growth of Beauveria bassiana on the media type was more effective in media of pellet type than that of powder type. In addition, production of Beauveria bassiana conidia on the media size was more effective in media of S type($\Phi$3mm) than that of L type($\phi$7mm). And the yield of Beauveria bassiana conidia at 2$0^{\circ}C$ cultivation on 3 weeks post inoculation was similar to that of $25^{\circ}C$. Above-mentioned results showed that pellet medium (P-I) was effective to growth of Beauveria, suggesting that the pellet media may be useful in the mass production of Beauveria bassiana conidia.
