Sphingomonas chlorophenolica ATCC39723 was successfully labeled with the gfp (green fluorescent protein) gene inserted into the pcpB gene by homologous recombination. As the gfp recombinant was easily distinguished from other indigenous organisms, the population of gfp recombinant was monitored after being released into the soil microcosms. Their population density dropped from 10$\^$8/ to 10$\^$6/ (cfu/$m\ell$) in the non-sterilized soil microcosms during the first 6 days. Moreover, the gfp recombinant was not detected even at lower dilution rates after a certain time period. The recombinant, however, survived for at least 28 days in the sterilized soil microcosms. Although the gfp recombinant did not degrade pentachlorophenol (PCP), this experiment showed the possibility of using gfp as a monitoring reporter system for S. chlorophenolica ATCC39723 and potentially other species of Sphingomonas.
Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, and energy in selected carbohydrate sources including wheat flour (WF), ${\alpha}-potato$ starch (PS), ${\alpha}-corn$ starch (CS), Na alginate (AL), dextrin (DEX), and carboxymethyl cellulose (CMC) were determined for olive flounder. The olive flounder averaging $150{\pm}8.0g$ were held in 300-L tanks at a density of 30 fish per tank. Chromic oxide was used as the inert marker. Feces were collected from the flounder by a fecal collector attached to a fish rearing tank. Apparent dry matter and energy digestibilities of flounder fed WF, PS, CS, and DEX diets were significantly higher than those of fish fed AL and CMC diets. Apparent crude protein digestibility coefficients of flounder fed PS and CS diets were significantly higher than those of fish fed AL, DEX, and CMC diets. Apparent crude lipid and nitrogen-free extract digestibility coefficients of flounder fed PS and DEX diets were significantly higher than those of fish fed WF, CS, AL, and CMC diets. The present findings indicate that PS and DEX could be effectively used as dietary carbohydrate energy compared to WF, CS, AL, and CMC for olive flounder.

Background: Molecular prognostic markers have been under investigation for the last decade and no validated marker to date has been proven to be used in daily clinical practice for urinary bladder cancers. The aim of the present study is to evaluate the significance of HYAL-1 expression in prediction of recurrence and progression in pT1 urothelial carcinomas. Materials and Methods: Eighty-nine urothelial carcinoma cases staged as T1 according to 2004 WHO classification were studied. Representative sections from every case were stained immunohistochemically for HYAL-1 and scored between 0 and +3, according to staining density, and graded as low and high for the scores 0-1 and 2-3, respectively. Results: Of the 89 pT1 bladder cancer patients, HYAL-1 expression was high in 92.1% (82 patients; 72 patients +3 and 10 patients +2) and low in 7.9% (only 7 patients; 6 patients +1 and 1 patient 0) of the cases. Of the 89 patients, 38 (42.7%) had recurrence and 22 (24.7%) showed progression. HYAL-1 staining did not show significant characteristics for tumor grade, accompanying CIS, multiplicity, tumor size, age and sex. HYAL-1 expression did not have any prognostic value in estimating recurrence or progression. Conclusions: HYAL-1 expression was found to be high, but did not have any prognostic importance in T1 bladder urothelial carcinomas.
The purpose of the present study was to examine the relation of total antioxidant status (TAS) to metabolic risk factors in Korean adults. Anthropometric measures, blood pressure, serum lipids and fasting glucose were determined in 406 men and women. TAS was measured by using commercially available Randox kit. Serum TAS was significantly positively correlated with body weight (p=0.004), body mass index (BMI) (p=0.033), waist circumference (p=0.017), total cholesterol (p=0.038) and triglyceride (TG) (p<0.001). The mean TAS of hypertriglyceridemic subjects (TG ${\geq}$150 mg/dl) was significantly higher than that of subjects whose TG was lower than 150 mg/dl (p=0.001). When central obesity, TG, high density lipoprotein cholesterol, fasting glucose and blood pressure were considered as metabolic risk factors, TAS was shown to be elevated with increased number of metabolic risk factors (p=0.004). The positive association between TAS and a number of metabolic risk factors suggests that increased TAS may not always indicate one's healthier condition. In order to help understand TAS as a marker of total antioxidant capacity in humans with various metabolic conditions, it is needed to clarify the factors affecting TAS in relation to changes in metabolic risk factors.
Quantitative traits are mostly controlled by a large number of genes. Some of these genes tend to have a large effect on quantitative traits in cattle and are known as major genes primarily located at quantitative trait loci (QTL). The genetic merit of animals can be estimated by genomic selection, which uses genome-wide SNP panels and statistical methods that capture the effects of large numbers of SNPs simultaneously. In practice, the accuracy of genomic predictions will depend on the size and structure of reference and training population, the effective population size, the density of marker and the genetic architecture of the traits such as number of loci affecting the traits and distribution of their effects. In this review, we focus on the structure of Hanwoo reference and training population in terms of accuracy of genomic prediction and we then discuss of genetic architecture of intramuscular fat(IMF) and marbling score(MS) to estimate genomic breeding value in real small size of reference population.
The effect of calcium and boron supplementation on bone strength was determined in growing and ovariectomized (OVX) Sparague-Dawley rats. Rats were divided into 9 groups and fed diet with different intake levels of calcium and boron for 4 weeks. About fifty percentages of rats in each group were OVX and the others were sham-operated. The rats were fed same diets after operation for 8 weeks. The feed intake, body weight gain, and FER were significantly higher in OVX rats than those in sham-operated ones. Serum osteocalcin, bone formation biomarker, was significantly increased with increment in calcium and boron intakes. Serum estradiol was lower in OVX rats than in sham-operated ones. Bone mineral density of femur was significantly lower in OVX rats than in other group. The breaking forces of bones were not significantly different among the groups. The urinary excretion of deoxypyridinoline, osteolytic marker was significantly increased with increment in calcium intake and ovariectomy. The urinary calcium excretion was significantly increased with increment in calcium intake, but decreased with increment in boron intake. According to theses results, the boron supplementation resulted in higher serum osteocalcin and lower urinary calcium excretion. Therefore, it could be suggested that the boron supplementation may be complementary and useful to calcium nutrition for bone health.
This paper deal with development of dredging equipment that proper water head elimination method changing transitional dredging method. Numerical simulation and model test were accomplished for development of seabed sediment inhalation equipment using water head. Sediment elimination system by the difference of water head has been analysed by numerical method. Model test was achieved for inhalation equipment performance test using water head by primary design. Also sediment elimination system has designed and developed that doesn't occur secondary pollution phenomenon. To verify the numerical results are compared with experimental results.
Age at first calving is an important trait for achieving earlier reproductive performance. To detect quantitative trait loci (QTL) for reproductive traits, a genome wide association study was conducted on the 96 Hanwoo cows that were born between 2008 and 2010 from 13 sires in a local farm (Juk-Am Hanwoo farm, Suncheon, Korea) and genotyped with the Illumina 50K bovine single nucleotide polymorphism (SNP) chips. Phenotypes were regressed on additive and dominance effects for each SNP using a simple linear regression model after the effects of birth-year-month and polygenes were considered. A forward regression procedure was applied to determine the best set of SNPs for age at first calving. A total of 15 QTL were detected at the comparison-wise 0.001 level. Two QTL with strong statistical evidence were found at 128.9 Mb and 111.1 Mb on bovine chromosomes (BTA) 2 and 7, respectively, each of which accounted for 22% of the phenotypic variance. Also, five significant SNPs were detected on BTAs 10, 16, 20, 26, and 29. Multiple QTL were found on BTAs 1, 2, 7, and 14. The significant QTLs may be applied via marker assisted selection to increase rate of genetic gain for the trait, after validation tests in other Hanwoo cow populations.
Numerical simulations are made for the three-dimensional flow around a wing in ground effect craft haying the complex geometry. A numerical tool is developed for the primary design of hull and wing shape of practical Wing-In-Ground effect(WIG) stop. The finite-difference method is utilized to descretize the governing equations and pressure field is obtained by using Marker-And-Cell(MAC) method. The air and water flows are simultaneously simulated in the time-marching solution procedure for the Navier-Stokes equation. The porosity technique and the density function are devised for the implementation of the three-dimensional body-boundary and the free-surface conditions, respectively. In this paper, a craft is modeled simply by three blocks containing a wing mounted on a main body horizontally, with the endplate. The numerical calculations of a WIG advancing in a calm water are performed and the WIG-generated wave profiles are also obtained. In the final paper, details of the numerical methods employed for the present study and calculated results are discussed.
Purpose: This study was done to identify the effects of weight-bearing exercise(WBE) on bone metabolism. Method: WBE was performed for 12 weeks by healthy college women. Bone-related parameters were measured four times during this period by evaluating the immunoradiometric assay and enzyme immunoassay. Bone mineral densities(BMDs) were measured by dual energy x-ray absorptiometry before and after the WBE program. Data was analyzed using t-test, paired t-test, $x^2$-test, and repeated measures ANOVA. Result: Osteocalcin, a bone formation marker, increased more in the experimental group than in the control group based on the interaction between time and group(F=3.29 p=.024). Little difference between the two groups was found for the other parameters: urinary deoxypyridinoline, insulin-like growth factorI, parathormone, serum calcium, and serum phosphorus without showing any time interaction between the groups. The femoral trochanter BMD rose in the experimental group while that of the control group fell, showing a significant difference for BMD(t=3.06 p=.005). However, there was no significant difference between the two groups for changes in BMD of the forearm, lumbar spine, femoral neck, and femoral ward's triangle. Conclusion: These findings supported the WBE is beneficial for increasing bone formation in college women and long-term application is needed to substantiate the effects of WBE as a intervention in promotion of bone-health.
