독도의 해안에 군락을 이룬 해안식물 근권에서 호염성 및 염내성을 가지는 세균의 분리를 위해 3종의 해안식물의 군락을 선정한 후 각 식물의 군집 하부에서 토양시료를 채취하였다. 시료는 marine broth 한천배지를 이용하여 형태학적인 구분을 통해 순수분리 되었다. 분리된 161개 세균들을 NaCl 9.0% 농도로 조정된 배지에서 생존하는 26개 균주를 선발하여 genomic DNA를 얻은 후, 16S rRNA gene sequence를 증폭하여 부분동정 하였다. 이들의 유연관계 확인을 위해 계통수를 작성한 결과, 이들은 각각 Firmicutes (30.8%), Gamma proteobacteria (53.8%), Bacteroidetes (7.7%), Alpha proteobacteria (7.7%), Actinobacteria (7.7%)에 속하였으며, 이는 기존의 독도 토양 및 해수 미생물상 연구와 특징적 차이를 보인다. 또한, 분리된 세균의 종 조성도 기존 독도 토양 및 해양연구와 유의적으로 상이함을 보였다. 이에 더하여 선발된 26개 균주들 중에서 4균주가 12.0% 이상의 염농도에서 생장하였으며, 이들 중에서 3개 균주가 15.0% 이상의 염농도에서 생장하여 극호염성의 특성을 나타내었으며, 광범위한 염분농도에서도 생장하는 특성을 보였다. 이들은 해안식물 근권에서 독도 특유의 고염분 및 염분변화라는 환경적 스트레스를 극복하며 해안식물과 어떠한 상호작용을 하는 것으로 생각된다.
A novel inhibitory protein against blood coagulation factor Va (FVa) was purified from muscle protein of granulated ark (Tegillarca granosa, order Arcoida, marine bivalvia) by consecutive FPLC method using anion exchange and gel permeation chromatography. In the results of ESI-QTOF tandem mass analysis and database research, it was revealed that the purified T. granosa anticoagulant protein (TGAP) has 7.7 kDa of molecular mass and its partial sequence, HTHLQRAPHPNALGYHGK, has a high identity (64%) with serine/threonine kinase derived from Rhodopirellula baltica (order Planctomycetales, marine bacteria). TGAP could potently prolong thrombin time (TT), corresponding to inhibition of thrombin (FIIa) formation. Specific factor inhibitory assay showed that TGAP inhibits FVa among the major components of prothrombinase complex. In vitro assay for direct-binding affinity using surface plasmon resonance (SPR) spectrometer indicated that TGAP could be directly bound with FVa. In addition, the binding affinity of FVa to FII was decreased by addition of TGAP in dose-dependant manner ($IC_{50}$ value = 77.9 nM). These results illustrated that TGAP might interact with a heavy chain of FVa ($FVa_H$) bound to FII in prothrombin complex. The present study elucidated that non-cytotoxic T. granosa anticoagulant protein (TGAP) bound to FVa can prolong blood coagulation time by inhibiting conversion of FII to FIIa in blood coagulation cascade. In addition, TGAP did not significantly (P < 0.05) show fibrinolytic activity and cytotoxicity on venous endothelial cell line (ECV 304).
Neoagaro-oligosaccharides are produced only by enzymatic degradation of agarose by ${\beta}-agarase.^{1)}$ Neoagaro-oligosaccharides inhibit the growth of bacteria, slow the rate of degradation of starch, are used as low-calorie additives to improve food quality, and have macrophage-stimulating activity. Furthermore, neoagarobiose is a rare reagent that has both moisturizing effect on skin and whitening effect on melanoma $cells.^{2)}$ An agar-degrading marine bacterium was isolated from the sea water at the northeast coast in Cheju island, Korea. The strain was gram negative, aerobic, and motile rod. The 16S rRNA of the strain had the closest match of 98% homology, with that from Agarivorans albus. On the basis of several phenotypic characters and a phylogenetic analysis, this strain was designated Agarivorans sp. JA-1. In solid agar plate, Agarivorans sp. JA-1 produced a diffusible agarase that caused agar softening around the colonies. Agarivorans sp. JA-1 was cultured for 36 hr in marine broth 2216 (Difco, USA) and the supernatant that containing an extracellular ${\beta}-agarase$ was prepared by centrifugation of culture media. The enzyme exhibited relatively strong activity at $40^{\circ}C$ and was stable up to $60^{\circ}C$. Using PCR primers derived from the ${\beta}-agarase$ gene of Vibrio sp., the gene encoding ${\beta}-agarase$ from Agarivorans sp. JA-1 was cloned and sequenced. The structural gene consists of 2931 bp encoding 976 amino acids with a predicted molecular weight of 107,360 Da. The deduced amino acid sequence showed 99% and 34% homology to $agaA^{2)}$ and $agaB^{2)}$ genes for ${\beta}-agarase$ from Vibrio sp., respectively. The expression plasmid for ${\beta}-agarase$ gene of Agarivorans sp. JA-1 is being constructed and the recombinant enzyme will be biochemically characterized.
비배양에 근거한 16S rDNA-DGGE fingerprinting 방법을 적용하여 공생세균 군집구조를 조사하였다. Zobell 배지와 천일염 배지를 사용하여 총 164균주를 선별하였다. 이들 균주로 부터 증폭한 16S rDNA를 제한효소 HaeIII와 MspI을 사용하여 절단한 후 각각의 다른 RFLP 패턴으로 구분하였다. RFLP패턴으로부터 유래한 16S rDNA의 염기서열 분석 결과, 알려진 염기서열들과 95% 이상의 유사도를 나타내었으며 Proteobacteria (Alphaproteobacteria, Gammaproteobacteria), Actinobacteria, Firmicutes, Bacteroidetes의 4개의 문이 나타났으며 우점 군집은 Alphaproteobacteria였다. 해면에서 분리한 total gDNA로 부터 증폭한 16S rDNA의 DGGE 분석 결과 5개의 DGGE band가 확인 되었고, 각각의 band의 염기서열 분석 결과 알려진 염기서열들과 96% 이상의 유사도를 나타내었으며 band로부터 밝혀진 모든 서열들은 배양되지 않은 세균 클론들과 높은 상동성을 나타내었다. DGGE에 의한 공생세균 군집은 Proteobacteria (Alphaproteobacteria, Gammaproteobacteria), Actinobacteria, Spirochetes, Chloroflexi로 4개의 문(phylum)으로 나타났다. Spirastrella abata의 공생세균 군집에 대한 RFLP와 DGGE 적용 결과, Alphaproteobacteria, Gammaproteobacteria, Actinobacteria의 공통 세균 그룹이 발견되었으나 전체적인 공생세균 군집구조는 분석 방법에 따른 차이를 나타내었다.
We have studied bioremediation of effective microorganisms on crude oil spill in Taean, west-coast of Korea. Oil contaminated soil samples were collected on December 14, 2007, seven days after the Hebei Spirit oil-spilled accident. Total petroleum hydrocarbon (TPH) was measured to evaluate the effectiveness of effective microorganisms (EM) which were composed with yeast, photosynthetic bacteria and lactic acid bacteria on oil degradation. TPH concentration before EM treatment was 323.8 mg/kg, whereas TPH concentrations on 2 days after EM treatment and that of control (without EM) was 102.1 mg/kg and 170.6 mg/kg, respectively. On six days after EM treatment TPH was 91.3 mg/kg and that of control was 127.7 mg/kg. Percentages of degraded crude oil were 47.3% without EM and 68.5% with EM, 60.6% without EM and 71.8% with EM on 2 and 6 days after EM treatment, respectively. These results clearly showed that the application of effective microorganisms toward oil-contaminated soil was quite useful to degrade crude oil spill. These results were derived from the effects of biostimulation of microbial media nutrients and bioaugmentation of effective microorganisms. If we carefully apply these effective microorganisms, it can be a useful bioremediation method to recover oil-contaminated marine ecosystems.
This study was undertaken to evaluate the effect of bacteria-derived $\beta$-glucan fiber on serum lipids, adiposity and uncoupling protein (UCP) expression in rats. In order to induce obesity, Sprague-Dawley weanling male rats were allowed free access to AIN-76A diet until 4 weeks of age, and fed high-fat diet (beef tallow, $40\%$ of calories as fat) for 6 weeks until 10 weeks of age. Rats were then fed with $0\%$ thigh- fat control group), $1\%$, or $5\%$ bacterial ~-glucan supplemented high-fat diets (w/w) for another 6 weeks. For comparison, normal control group was fed with AIN-76 diet $11.7\%$ fat). Supplementation with bacterial $\beta$-glucan resulted in a significant reduction of high-fat-induced white fat (i.e., visceral and peritoneal fat) development, adipocyte hypertrophy, and development of hyperinsulinemia and hyperleptinemia. Serum triglyceride, total cholesterol, and free fatty acid levels were greatly reduced, but, HDL-cholesterol concentrations were increased by bacterial $\beta$-glucan supplementation. Serum leptin level was lower in the $\beta$-glucan groups than in the high-fat group. The expression of UCPs (UCP1, UCP2, and UCP3) in brown adipose tissue (BAT) were significantly increased by $5\%$ bacterial $\beta$-glucan-containing diet. This study suggests that the anti-obesity effect of $5\%$ bacterial $\beta$-glucan is attributed to upregulation of UCPs and inefficient energy utilization.
2012년 2월 남태평양 미크로네시아 축(Chuuk)에서 채집한 두 종의 해양해면 Cinachyrella sp.와 Plakortis sp.의 공생세균의 군집구조를 PCR-DGGE 방법을 사용하여 조사하였다. Cinachyrella sp.와 Plakortis sp. 해면의 total genomic DNA에서 16S rRNA gene-V3 부분을 증폭하여 DGGE를 수행하였으며, 두 종의 해면에서 서로 다른 밴드 패턴이 나타났다. DGGE밴드로부터 16S rRNA gene의 부분 염기서열을 분석한 결과, 알려진 균주의 염기서열들과 87-100%의 유사도를 나타내었다. Cinachyrella sp.의 공생세균 군집구조는 Actinobacteria, Bacteroidetes, Chloroflexi, 그리고 Proteobacteria (Alpha-, Gamma-, Delta-), 6강으로 구성되었다. Plakortis sp.의 공생세균 군집구조는 Actinobacteria, Chloroflexi, Firmicutes, Spirochaetes 그리고 Proteobacteria (Alpha-, Gamma-, Delta-), 7강으로 구성되었다. 두 종의 해면에서 Actinobacteria, Chloroflexi와 Proteobacteria가 공통적으로 존재하였으나 주요 세균군집은 서로 다른 것으로 나타났다. 즉 Cinachyrella sp.의 경우 Proteobacteria가, Plakortis sp.의 경우, Chloroflexi가 주요 세균 군집이었다. 동일지역에서 채집한 서로 다른 두 종의 해면은 각각 다른 공생세균 군집구조를 나타내어 해면 종에 따른 숙주 특이적 분포를 보이는 것으로 나타났다.
고형 양식사료에 포함된 질소와 인의 미생물학적 제거 공정을 알아보기 위하여 벤치규모의 실험을 수행하였다. CK-10과 CK-13 균주가 새우양식장의 물시료로부터 분리되었다. CK-10과 CK-13의 혼합배양에서 N/P의 동시제거 실험을 실시하였다. 그 결과, $400\;{\mu}M\;NH^{+}_4$ 와 $NO^{-}_2$는 12시간 이내에 제거되었고, $NO^{-}_3$는 36시간 이내에 각각 제거되었으며, $500\;{\mu}M\;PO^{3-}_4$ 36시간 이내에 제거되었다. CK-10과 CK-13 배양을 새우양식사료에서 용출된 N와 P의 제거에 적용하였다. HPAEC-PAD 시스템을 이용하여 양식사료의 당을 분석하였으며, glucose, galactose, galatosamine, mammonse, fucose 등의 여러 가지 당이 분석되었다. 세균에 의한 질소와 인 제거를 수행하기 위하여 인공 해수에서 $0.2\%$(w/v)의 양식사료를 용출시켰으며, 72 시간동안 용출된 질소의 양은 대략 $33.3\;{\mu}M\;NH^{+}_4,\;12.9\;{\mu}M\;NO^{-}_2.\;81.5\;{\mu}M\;NO^{-}_3,\;248\;{\mu}M\;PO^{-3}_4$였다. 혼합배양은 $0.2\%$ 사료에 포함된 질소와 인을 84시간 이내에 완전히 제거하였으나, 단일배양은 주어진 배양기간동안 질소와 인을 완전제거 하지 못하였다. 이 연구에서 CK-10과 CK-13 배양은 새우양식사료에서 유래하는 질소와 인을 효과적으로 제거하는 것이 입증되었다.
최근에 해양자원에 있는 동물, 해조류 곰팡이 세균에서 신규 잠재적인 후보약효제가 조사되어 왔다. 본 연구에서는 치료제를 탐색하기 위하여 암전이, 관절염, 만성염증 및 주름형성에 주요한 역할을 하는 기질금속단백질분해효소(s) (MMPs)를 목적효소로 이용하였다. 5종의 녹조류, 18종의 홍조류, 4종의 갈조류를 포함한 다양한 해조류가 사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 미치는 영향을 gelatin zymography를 이용하여 조사하였다. 사람피부섬유아세포에서는 홍조류중에서 Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate 및 Sinkoraena lancifolia에서 MMP-2 억제효과가 관찰되었다. 반면에 녹조류의 Enteromorpha compressa와 Enteromorpha linza, 갈조류의 Peltaronia bighamiae and Sargassum thunbergii에서는 MMP-2 활성화가 관찰되었다. 사람섬유아육종세포에서는 MMP-9 활성화가 갈조류인 Sargassum thunbergii, 홍조류의 Polysiphonia japonica, 녹조류의 Enteromorpha compressa와 Enteromorpha linza의 존재 하에서는 감소되었다. 본 연구에서 흥미로운 발견은 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 정상세포에서는 MMP-2에 대하여 활성화 효과를 나타내었으나, 암세포에서는 MMP-9응 억제하는 효과를 나타낸 것이다. 이러한 결과는 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 항암 효능을 발휘할 수 있는 성분을 함유하고 있다는 것을 암시하고 있다.
항생제 남용의 문제점 및 사용제한에 따른 천연물 유래 항생제 대체제 개발을 위하여 우리나라와 중국, 일본에서만 서식하는 고유종인 참담치의 족부 근육 추출물로부터 항균 펩타이드를 분리 및 정제하였다. $C_{18}$ 역상 컬럼을 사용하여 항균 펩타이드를 정제하였으며, MALDI-TOF/MS로 분석 결과 분자량은 6,701 Da에 해당하였다. edman degradation 방법으로 N-말단 아미노산 서열 분석을 통하여 20개의 아미노산 서열을 밝혔으며 이는 캘리포니아 홍합(Mytilus californianus)의 sperm-specific protein 또는 protamine-like PL-II/PL-IV precursor와 100% 상동성을 지님을 밝혀냈다. 이러한 정보를 바탕으로 ORF를 분석한 결과 60개의 아미노산을 코딩하는 183 bp로 캘리포니아 홍합의 protamine-like PL-II/PL-IV precursor와 아미노산 서열이 100% 일치하였고 유전자 염기서열은 97.2%의 상동성을 나타냈다. 합성된 항균 펩타이드는 그람 양성 균주 Bacillus cereus (MEC, $20.8{\mu}g/ml$), Bacillus subtilis (MEC, $0.2{\mu}g/ml$), Streptococcus mutans (MEC, $0.2{\mu}g/ml$), 그람 음성 균주 Pseudomonas aeruginosa (MEC, $5.7{\mu}g/ml$), Escherichia coli (MEC, $2.6{\mu}g/ml$) 그리고 진균류 Candida albicans (MEC, $56.3{\mu}g/ml$)의 항균활성을 나타냈다. 또한 높은 열안정성을 지녔으며, C. albicans를 제외하고 염안정성을 지닌 항균 펩타이드로 확인되었다. 이를 통해 참담치 유래의 항균 펩타이드는 항생제 대체제의 후보소재로 높은 가능성을 지녔다고 할 수 있다. 따라서 본 연구에서 분리한 참담치 족부 근육 유래 항균 펩타이드는 향후 추가적인 연구를 통하여 항생제를 대체할 수 있는 물질의 개발이 가능할 것으로 사료된다. 그리고 해양무척추동물의 선천성 면역 기작에 대한 정보를 제공할 것으로 기대한다.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
