Susceptibility of marine medaka Oryzias dancena to fish pathogenic viruses (어류병원바이러스에 대한 해산 송사리 Oryzias dancena의 감수성)

  • The susceptibility of marine medaka, Oryzias dancena to fish pathogenic viruses (infectious pancreatic necrosis virus (IPNV), viral hemorrhagic septicemia virus (VHSV), hirame rhabdovirus (HIRRV), infectious hematopoietic necrosis virus (IHNV), and lymphocystis disease virus (LCDV)) was investigated. The cumulative mortalities of fish immersed with IPNV (experimental condition: $15^{\circ}C$ sea water (SW)), VHSV ($15^{\circ}C$ SW), HIRRV ($15^{\circ}C$ fresh water (FW)) were 30%, 40% and 60%, respectively. In the fish immersed with IPNV ($15^{\circ}C$ FW, $18^{\circ}C$ FW and SW), VHSV ($15^{\circ}C$ FW, $18^{\circ}C$ FW and SW), HIRRV ($15^{\circ}C$ SW), IHNV ($15^{\circ}C$ FW and SW), LCDV ($15^{\circ}C$ FW and SW, $18^{\circ}C$ FW and SW), and mock-challenged group, mortality rate was less than 10%. IPNV, VHSV and HIRRV were re-isolated from the dead fish. These results suggest that marine medaka is susceptible to IPNV, VHSV and HIRRV, although their susceptibility depends on the environmental conditions.

Antiviral Effects of Sulfated Exopolysaccharide from the Marine Microalge Gyrodinium impudicum strain KG03

  • The sulfated exopolysaccharide p-KG03, which is produced by the marine microalga Gyrodinium impudicum strain KG03, exhibited impressive antiviral activity in vitro ($EC_{50}$ = 26.9 g/ml) against the encephalomyocarditis virus (EMCV). Depending on the p-KG03 concentration, the development of cytopathic effects in EMCV-infected HeLa cells was either inhibited completely or slowed. Moreover, p-KG03 did not show any cytotoxic effects on HeLa cells, even at concentrations up to 1,000 g/ml. The polysaccharide was purified by repeated precipitation in ethanol, followed by gel filtration. The p-KG03 polysaccharide had a molecular weight of $1.87\;{\times}\;10^6$, and was characterized as a homopolysaccharide of galactose with uronic acid (2.96%, w/w) and sulfate groups (10.32% w/w). The biological activities of p-KG03 suggest that sulfated metabolites from marine organisms are a rich source of antiviral agents. This is the first reported marine source of antiviral sulfated polysaccharides against EMCV. The p-KG03 polysaccharide may be useful for the development of marine bioactive exopolysaccharides for use in biotechnological and pharmaceutical products.

Production of monoclonal antibodies against viral hemorrhagic septicemia virus (VHSV, genotype IVa) from olive flounder (넙치 유래 viral hemorrhagic septicemia virus (VHSV, genotype IVa)에 대한 단클론 항체 생산)

  • Mouse monoclonal antibodies (MAbs) were produced by using viral hemorrhagic septicemia virus (VHSV, genotype IVa) as an immunogen, isolated from diseased olive flounder (Paralichthys olivaceus). Four hybridoma clones secreting MAbs against VHSV were established. The MAbs were recognized the nucleoprotein (MAb 4), phosphoprotein (MAb 1) and matrix protein (MAbs 2 and 3) of VHSV by western blot analysis. Among them, the MAbs 1 and 4 strongly reacted with the VHSV-infected FHM cells, but not normal FHM cells. In enzyme linked immunosorbent assay, the four MAbs reacted with the VHSV, but not different six fish viruses (infectious hematopoietic necrosis virus, hirame rhabdovirus, spring viraemia of carp virus, infectious pancreatic necrosis virus, marine birnavirus and nervous necrosis virus). These results indicate that the MAbs are useful for diagnosis of VHSV infection.

A Simple Method for the Concentration of Fish Pathogenic Virus in Sea Water (한외여과막을 이용한 해수내 어류 병원바이러스 농축법)

  • A method was developed for concentrating fish pathogenic virus from sea water using membrane ultrafiltration system and centricon. The method consists of passing large volumes (Ca. 20 liter) of sea water through ultrafiltration (PAN) filter followed by cross-flow filtration method and centrifugation use the centricon (Plus-20). This procedure permitted the processing of 20 liter of sea water which resulted in a 20,000-fold reduction in the volume of water and greater than 90% recovery of the seeded MABV.

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral disease of farmed rainbow trout and some marine fishes in Europe and North America. It has been reported in various marine fish species of Asian countries and induced cause mass mortality in Japanese flounder (Paralichthys olivaceus) culturing in Korea. The aims of this study were to monitor VHSV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR results showed that VHSV were detected in 17 (10.6%) out of 160 fish. G gene sequences of viral strains isolated in this study were closely related to that of a reference strain, KVHS01-1, belonging to VHSV genotype Ⅰ. The results suggest that some of wild marine fishes are VHSV carriers and may spread the pathogen directly to fish farmed in coastal area.

  • We developed and subsequently characterized mouse monoclonal antibodies (MAbs) against nervous necrosis virus (NNV, RGNNV genotype). We established six hybridoma clones secreting MAbs against NNV antigen: 2B1, 2B11, 2C12, 13C1-1, 13C1-2 and 14D11. All six MAbs belonged to the IgG2a isotype with a kappa light chain and their reactivity recognized against the 41 kDa coat protein of NNV by Western blot analysis. The affinity constants of the six MAbs were measured by enzyme-linked immunosorbent assay (ELISA). All six MAbs reacted with two NNV isolates (SgNag05 and Gemunodo06), while no reactivity was observed with five know fish viruses, namely marine birnavirus, infectious pancreatic necrosis virus, viral hemorrhagic septicemia virus, hirame rhabdovirus, and infectious hematopoietic necrosis virus. Moreover, high ELISA optical density (OD) values (0.87-1.42) were observed in the brain tissues of NNV-infected sevenband grouper, while low OD values (less than 0.12) were recorded in the brain tissues of uninfected fish. These results suggest that these six MAbs are highly competent and useful for the detection of NNV with the RGNNV genotype.

Resistance against white spot syndrome virus (WSSV) infection in wild marine crab Gaetice depressus by injection of recombinant VP28 protein

  • The resistance against white spot syndrome virus (WSSV) infection in wild marine crab Gaetice depressus by the immunization of a recombinant glutathione-S-transferase (GST) fused VP28 protein (GST-VP28) was evaluated. The cumulative mortalities of GST-VP28 injected groups were lower than those of the control groups at 10 days of post-challenge, and the time to death of 50% crab ($TD_{50}$) was delayed by the immunization using GST-VP28. The group boosted with GST-VP28 after 2 weeks of primary immunization clearly showed longer $TD_{50}$ than non-boosted group against challenge with WSSV. This result suggests that boosting with the antigen protein elicit stronger immune responses similar to adaptive immune responses of vertebrates. However, the short $TD_{50}$ was observed in the group challenged at 3 weeks post boosting comparing to the group challenged at 1 week post boosting. This suggests that the protective strength of immunization decreased by the time.

The Factor of Influencing Cervical Cancer and Human Papilloma Virus(HPV) Infection Preventive Behavioral Intention of Nursing Students (간호대학생의 자궁경부암 및 인유두종 바이러스감염 예방행위의도 영향요인)

  • The purpose of this study was to identify factors of influencing cervical cancer and Human papilloma virus(HPV) infection preventive behavioral intention of nursing students. The research design for this study was a descriptive survey design. Data collection was done using self-report questionnaires with 218 nursing students in B city from November 30 to 15 December, 2015. Data were analyzed using percentage, mean, standard deviation, t-test, ANOVA, Pearson's correlation analysis and multiple regression with SPSS Win 21.0 program. The significant factors of cervical cancer and HPV infection preventive behavioral intention was cervical cancer knowledge(${\beta}=.246$, p<.001), health belief about HPV vaccination(${\beta}=-.223$, p<.001), HPV vaccination(${\beta}=.173$, p=.008), and Gynecological Disease History(${\beta}=.145$, p=.026). And these factors explained 15.4% of the variance in the cervical cancer and HPV Infection preventive behavioral intention. In conclusion, the results of this study indicated that it's a need to develop the effective education program in addition to knowledge improvement on HPV and cervical cancer for nursing students.

Comparison of lymphocystis disease virus proteins between marine and freshwater fish

  • Lymphocystis disease virus (LCDV) was detected from olive flounder Paralichthys olivaceus, painted glass fish Chanda baculis, gourami Trichogaster leeri and rockfish Sebastes schlegeli, and proteins of the viruses were compared. The major capsid protein (MCP) gene-specific primer sets successfully amplified approximately 1300 bp nucleotides from the olive flounder and 600 bp nucleotides from painted glass fish, gourami and rockfish isolates, respectively. In western blotting analysis using anti-LCDV mouse polyclonal serum, major antigenic proteins had 21, 26, 45, 50, 80, 110 and 120 kDa in olive flounder, 26, 47 and 80 kDa in painted glass fish, 26, 46, 80 and 92 kDa in gourami, 26, 44, 49, 80 and 105 in rockfish, respectively. All the marine and freshwater isolates showed only common antigens of approximately 26 kDa and 80 kDa. These results suggest that antigenic protein profiles of LCDVs may vary depending upon fish species.
