Natural killer T cell and pathophysiology of asthma

  • Natural killer T (NKT) cell is a special type of T lymphocytes that has both receptor of natural killer (NK) cell (NK1.1, CD161c) and T cell (TCR) and express a conserved or invariant T cell receptor called $V{\alpha}14J{\alpha}18$ in mice or Va24 in humans. Invariant NKT (iNKT) cell recognizes lipid antigen presented by CD1d molecules. Marine-sponge-derived glycolipid, ${\alpha}-galactosylceremide$ (${\alpha}-GalCer$), binds CD1d at the cell surface of antigen-presenting cells and is presented to iNKT cells. Within hours, iNKT cells become activated and start to secrete Interleukin-4 and $interferon-{\gamma}$. NKT cell prevents autoimmune diseases, such as type 1 diabetes, experimental allergic encephalomyelitis, systemic lupus erythematous, inflammatory colitis, and Graves' thyroiditis, by activation with ${\alpha}-GalCer$. In addition, NKT cell is associated with infectious diseases by mycobacteria, leshmania, and virus. Moreover NKT cell is associated with asthma, especially CD4+ iNKT cells. In this review, I will discuss the characteristics of NKT cell and the association with inflammatory diseases, especially asthma.

Epi-Leptosphaerin: A New L-Isoascorbic Acid Derivative from Marine Sponges

  • A new L-isoascorbic acid derivative epi-leptosphaerin (1) and two known compounds leptosphaerin (2), and verongamine (3) were isolated from sponges of the orders Verongida and Thorectidae. Compounds 1 and 2 are most likely of sponge-associated fungal origin. In the present study, isolated compounds were investigated for their inhibition of soluble epoxide hydrolase (sEH), which is considered a promising target for the management of pain, inflammation, and comorbidities associated with diabetes. Compound 3, verongamine, displayed weak inhibitory activity against sEH with an $IC_{50}$ value $51.5{\pm}1.0{\mu}M$.

A New Species of the Genus Penares (Demospongiae: Astrophorida: Ancorinidae) from Korea

  • A new marine sponge in the family Ancorinidae, Penares hongdoensis n. sp. was collected from Hongdo Island, Korea by SCUBA diving in 2004. P. hongdoensis n. sp. is closely related to P. cortius de Laubenfels, 1930 in their spicules. However, they differ in kind and size of spicules and growth form. The dichotriaene rhabds of new species are larger than that of P. cortius. And the new species has oxyaster, but P. cortius has oxyspheraster. Although the growth forms of both species are irregular massive, this new species shows conspicuous long projection toward upper part.

  • Sponges are host organisms for various symbiotic microorganisms such as archaea, bacteria, cyano-bacteria and microalgae. Sponges are also sources of a wide variety of useful natural products like cyto-toxins. antifouling agents, antibiotics, and anti-inflammatory and antiviral compounds, Symbiotic microorganisms is sponges can be sources of various natural products, because metabolites previously ascribed to sponges have recently been demonstrated to be biosynthesized by symbionts. If a symbiotic microorganisms from which some natural products are derived can be cultured, the microorganism could be used in a mass production of the bioactive comopounds. We summarize recent research on iso-lation and cultivation of sponge-symbiotic microorganisms and the symbiotic relationship.

Coupling of ent-Cyclic Peroxide and Ircinol A, Two Biologically Active Natural Marine Products

  • An acidic ent-cyclic peroxide was isolated from a sponge, Plakotis sp., and showed activity against leishmaniasis and pathogenic fungi. To improve the activity of this compound, we coupled the acidic ent-cyclic at the C1 position of ircinol A. Compound 3 exhibited significant activity against Leishmania mexican a and fungi with $IC_{50}$ values of 0.7 and $0.3-34{\mu}g/mL$, respectively. The yield of compound 3 was 98%.

  • The galactolipid M-5, which showed anti-inflammatory activity is glycoglycerolipid isolated from the Okinawa marine sponge Phyllospongia foliascens. Glycolipids have been synthesized by various methods, especially it were generally known that synthetic method of M-5 analogue and synthetic method of various glycolipids by glycosidation after synthesis of glycerolipid part. The others, it was not suggested that synthetic method via glycosylglycerol obtained by degradation from diglycoside. This study was carried out to investigate the synthesis of galactosylglycerol from melibiose as M-5 intermediate. Synthesis of galactosylglycerol was accomplished by selective protection of hydroxy group of sugar and diol cleavage by Pb(OAc)$_4$. As a result, galactosylglycerol was synthesized by 8 steps pathway and their structures were elucidated by analysis instrument.

A New Sponge of the Genus Haliclona (Gellius) (Haplosclerida: Chalinidae) from Gageodo Island (So-Huksando), Korea

  • A taxonomic study on marine sponges was conducted by SCUBA diving at Gageodo Island, Korea from August 1999 to July 2001. Among them, Haliclona (Gellius) coreana n. sp. turned out to be new to science. This new species seems to be close to H. (G.) arcoferus Vosmaer, 1885, based on the type of spicules. However, it is different in size of toxa and growth form. Toxa of the new species is half in length compared to H. (G.) arcoferus. The growth form is jar shaped in this new species, but flat in H. (G.) arcoferus.

  • Three new genera and species of siphonostomatoid copepods are described as associates of sponges from shallow water of Samar Island in the Philippines: Samarus filipes n. gen., n. sp., Paurocheres dentatus n. gen., n. sp., and Platymyzon umbonatum n. gen., n. sp. A new family Samarusidae is proposed to accommodate Samarus n. gen. which has rudimentary legs 1-5 represented only by filiform setae. Paurocheres n. gen. is characterized by 2-segmented endopod of leg 4 and reduced setation of legs, and Platymyzon n. gen. by missing of mandibular gnathobase.

A New Record of Hydrodendron stechowi (Hydrozoa: Leptothecata: Phylactothecidae) from Korea

  • In 2020, during a survey of cnidarian fauna around Chuja-do locating in the northern part of Jeju-do, Korea, several hydroids were collected from the subtidal zone by SCUBA diving. Through taxonomic examination, one unrecorded species, Hydrodendron stechowi Hirohito, 1995, was found in Korea. It was newly added to the list of Korean hydrozoan fauna. This species was discovered for the first time since its first report as a new species in Japan. Their vivid orange-colored colonies were attached to the sponge on rocky substrate between depths of 15 and 20 m. Thus, a total of five species belonging to genus Hydrodendron, including the species reported in this study, have been reported in Korean waters so far.

생리활성물질을 생성하는 해양미생물의 동정 -I. 항미생물 물질을 생산하는 해양방선균 분리균주 No. 101의 분리 및 배양조건- (Identification of the Marine Microorganisms Producing Bioactives -I. Isolation and Cultural Conditions of the Marine Actinomycetes No. 101 Producing Antimicrobial compounds-)

  • 본 연구에서는 생리활성물질을 생성하는 해양미생물을 분리하기 위하여 연근해역의 해면을 조사하였다. 해면 중의 미생물은 $1.2{\times}10{\sim}2.1{\times}10^3/g$ 범위에 있었고, 이들 중 방선균은 $0.01{\sim}0.5\%$(data 생략)로 나타났다. 분리된 미생물은 각각 배양하여 항균물질을 검토하였으며, 검토하던 중에 비교적 강한 항균물질을 생산하는 균주 No. 101을 분리, 선발하였다. 선발된 균의 특징은 sclerotia로 형태가 다양하였고, 다양한 배지에서 생육이 가능하였고, 기균사의 색깔은 회백색, 배면의 색깔은 주로 회백색에서 갈색으로 나타났으며, 젤라틴 액화능과 전분 분해능을 가지나 myo-inositol은 이용하지 못하였다. 이 균의 배양학적 특성은 최적 pH 및 온도가 $6.5{\sim}7.0$$30^{\circ}C$였으며, 탄소원으로는 starch가 30g/l, 질소원으로는 casamino acid 10g/l가 적합한 것으로 나타났다. 방선균 No. 101의 균체 및 균배양액의 추출물에 대한 항균 및 항진균성에서는 Staphylococcus aureus와 Bacillus subtilis에서 강한 항균력을, 효모와 곰팡이에 대하여는 낮은 항진균성을 나타내었다.
