열대 해면동물 Lipastrotethya sp.에서 분리된 사포닌 화합물 (A New Triterpenoid Saponin from the Tropical Marine Sponge Lipastrotethya sp.)

  • Marine sponges have been a remarkably rich source of pharmacologically active and structurally diverse natural products. As a part of our continuing search for novel secondary metabolites of biomedical importance from marine invertebrate, we encountered the sponge Lipastrotethya sp. from Chuuk, Micronesia. The crude organic extract of this animal exhibited considerable cytotoxicity against the K562 cell line. Guided by the $^1H$ NMR analysis, flash chromatography of the crude extract followed by HPLC yielded a new triterpene glycoside, along with ten known saponins of the sarasinoside class. The structure of this new compound was determined by combined spectroscopic methods such as COSY, HSQC and HMBC experiment. Among these metabolites, six compounds exhibited moderate cytotoxicity against ACHN, MDA-MB-231, NCI-H23 and NUGC-3 cell lines.

시판 건 해산물 중 아황산염류 함량 조사 (Sulfites Contents Survey on Dried Marine Products in Market)

  • 서울시내 건 해산물 도매시장인 중부시장으로 반입되는 국내산, 수입산 건 해산물에 대한 아황산염류 함량을 조사 한 결과 30ppm이상을 함유하는 검체가 0.29%(8/2770)이었고, 명태알포 1 건에서 195.3ppm, 중국산 쥐치포 1 건에서 114.2ppm 그리고 베트남산 쥐치포 6건에서 130.2∼573.4ppm으로 모니어-월리암스 변법에 의한 검사 결과 나타났다. 아연분말 환원법에 의한 간이 검사 결과에서는 이황산염류 함량 1-30ppm인 경우가 전체 2,710건 가운데 259건으로 9.35%를 차지하였고, 품목별로는 건 새우 93 건, 쥐치포 53 건, 건 오징어 34건, 명태채 및 건어채 33건 그리고 홍합 살 17건 등에서 양성반응을 보여 이들 품목들의 가공에 아황산염류가 갈변방지와 방부 목적으로 상용되고 있는 것으로 나타났다. 앞으로도 건 해산물에 대한 아황산염류 등 첨가물 검사가 강화되어야 하며 특히, 수입산 건 해산물에 대한 검사가 더욱 더 강화되어야만 안전한 건 해산물이 시민에게 공급될 수 있으리라고 보여진다.

Pre-cooking and liquid smoke affect the physicochemical and microbiological quality and polyhydroxy aromatic hydrocarbon (PAH) content in smoked skipjack tuna (Katsuwonus pelamis L.)

  • This study aimed to compare the quality of smoked fish products processed with liquid smoke with and without pre-cooking when stored at room temperature (25℃) for 0, 2, 4, and 6 days. The observations included moisture content, water holding capacity, color, and total plate and mold counts. The treated samples were also analyzed for polyhydroxyaromatic hydrocarbons (PAHs) at day 0. The study found that the pre-cooked products had a lower moisture content (55.1%) than the control (59.9%) during storage. Additionally, the pre-cooked product had a higher water holding capacity than the control. The TPC value indicates that the pre-cooked product meets the Indonesian standard for smoked fish up to the 2nd day of storage, whereas the control product only meets the standard on day 0. However, the total mold of the control products meets the Indonesian standard until the 2nd day of storage, while the pre-cooked products only meet the standard on day 0. The pre-cooked product had a higher lightness value (L*) than the control. The analysis of polyhydroxy aromatic hydrocarbon (PAH) showed that the pre-cooking process did not affect the concentration of PAH.

The Synergistic Antibacterial Activity of 1-Acetyl-$\beta$-Carboline and $\beta$-Lactams Against Methicillin-Resistant Staphylococcus aureus (MRSA)

  • 1-Acetyl-$\beta$-carboline was isolated as an anti-MRSA agent from the fermentation broth of a marine actinomycete isolated from marine sediment. The producing strain was identified to be Streptomyces sp. by phylogenetic analysis of the 16S rRNA gene sequence. The anti-MRSA agent was isolated by bioactivity-guided fractionation of the culture extract by solvent partitioning, ODS open flash chromatography, and purification with a reversed-phase HPLC. Its structure was elucidated by extensive 2D NMR and mass spectral analyses. Combination of 1-acetyl-$\beta$-carboline with ampicillin exhibited synergistic antibacterial activity against MRSA.

  • A high light lantern system using LEDs is composed of power supply, flash controller, and daylight controller and proper case considering work condition. The performance of developed LED lantern is better than that of the existing marine lantern using an incandescent electric lamp and that of foreign products(Vega LED lantern made in New Zealand, Tideland system made in America). The electric consumption of our products is 30% compared with that of existing marine lantern using an incandescent electric lamp, and the luminous Intensity of them is 200% to 400% compared with that of existing marine lantern and foreign LED lantern. In addition, our LED lantern system is easy to repairing something wrong and changing the LED module and other controllers on the sea when they are in accident.

  • Nature is one of the most important sources of pharmacologically active compounds in the search for drugs against life threatening diseases. Even though plants and terrestrial microorganisms have played as an important source for the new drug candidates from nature, marine organisms such as tunicates, sponges, soft corals, sea horses, sea snakes, marine mollusks, seaweeds, nudibranches, sea slugs and marine microorganisms are increasingly attracting attention in recent years. Marine organisms also have the potential to develop into future drugs against important diseases, such as cancer, a range of bacterial and viral diseases, malaria, and inflammations. Even though the mechanism of action in the molecular level of most metabolites is still unclear, the mechanisms by which they interfere with the pathogenesis of a wide range of diseases have been reported. The knowledge of this is one of the key factors necessary to develop bioactive compounds into medicines. This is due to their structurally unique and pharmacologically active compounds. The potential pharmaceutical, medicinal and research applications of some of these compounds are discussed in hundreds of scientific papers, and are reviewed here.

Soft corals collected from Jeju Island inhibits the α-MSH-induced melanogenesis in B16F10 cells through activation of ERK

  • In the present study, we first evaluated the melanin inhibitory effect of four crude 70% ethanol extracts separated from soft corals abundantly growing along the seawaters of Jeju Island, South Korea, including Dendronephthya castanea (DC), Dendronephthya gigantea (DG), Dendronephthya puetteri (DP), and Dendronephthya spinulosa (DS). Among the four ethanol extracts, the ethanol extract of DP (DPE) did not possess any cytotoxic effect on B16F10 cells. However, all other three extracts showed a cytotoxic effect. Also, DPE reduced the melanin content and the cellular tyrosinase activity without cytotoxicity, compared to the ${\alpha}-MSH$-stimulated B16F10 cells. Specifically, DPE downregulated the expression levels of tyrosinase and microphthalmia-associated transcription factor by activating the ERK signaling cascade in ${\alpha}-MSH$-stimulated B16F10 cells. Interestingly, the melanin inhibitory effect of DPE was abolished by the co-treatment of PD98059, an ERK inhibitor. According to these results, we suggest that DPE has whitening capacity with the melanin inhibitory effects by activating ERK signaling and could be used as a potential natural melanin inhibitor for cosmeceutical products.

Recent Prospect of Compounds Derived from Marine Macroalgae for Medicinal Application of Anti-Inflammation for Chemoprevention of Cancer

  • Although marine living organism contains a numerious number of compounds, it is difficult to collect these compounds in a large scale for medicinal application. However, in recent years, several bioactive compounds isolated from marine macroalgae have been proved to be able to provide potential sources for development of medicinal products because they can be obtained in large amount from marine. A number of studies have reported a variety of effects of marine macroalgae but a few anti-inflammatory activity of marine macroalgae have recently been published. Herein, we reviewed novel anti-inflammatory compounds recently isolated from marine brown algae, green algae and red algae. From this survey, in particular, some compounds contained in edible macroalgae exert anti-inflammatory effects with inhibition on cyclooxygenase-2 (COX-2), inducible nitric oxide synthase(iNOS) and matrix metalloproteinases (MMPs) activity regulated by nuclear factor-kappa B transcription factor that play a key role in cancer as well as inflammation, demonstrating to be able to potentially apply to development of anti-inflammatory agent for chemoprevention of cancer. Furthermore, some macroalgae and their compounds with both excellent anti-inflammatory activity and very low toxicity can select a potential candidates capable of preventing or treating several chronic inflammation such as colitis, hepatitis and gastritis, leading to cancer.

Ecklonia cava (Laminariales) and Sargassum horneri (Fucales) synergistically inhibit the lipopolysaccharide-induced inflammation via blocking NF-κB and MAPK pathways

  • Ecklonia cava (EC) has been widely utilized as an ingredient in commercial products such as functional foods and cosmeceuticals. Recently it has been found that Sargassum horneri (SH) has been invading on Jeju Island coast area by its huge blooming. Moreover, both seaweeds are considering as important ingredients in traditional medicine specifically in East-Asian countries (China, Japan, and Korea). In the present study, we attempted to compare anti-inflammatory properties of 70% ethanolic extracts of EC (ECE), SH (SHE), and their different combinations on lipopolysaccharide (LPS)-activated RAW 264.7 cells. Results indicated that 8 : 2 combinations of ECE : SHE significantly inhibited LPS-activated inflammatory responses (cytokines, protein, and gene expression) in RAW 264.7 macrophage cells compared to the respective extracts and other combinations. The synergistic effect of ECE and SHE was found to be prominent than the effects of ECE or SHE alone. These observations provide useful information for the industrial formulation of functional materials (functional foods and cosmeceuticals) using these two particular seaweeds in Jeju Island of South Korea.
