Preliminary Structure Determination of the theonellapeptolide Ie from the marine sponge Theonella swinhoei Using NMR Methods

  • A known theonellapeptolide Ie, previously reported in other research group, was isolated from the methanolic extract of the Philippine sponge Theonella swinhoei. The planar structure of this compound was determined on the basis of NMR methods including HMBC and selective HMBC experiments. This is fast and efficient for the dereplication of natural products compared with the MS studies of fragments obtained from complete and partial hydrolysis. The sequence of thirteen amino acids including six N-methyl amino acids in the compound was clearly determined from correlations of extensive HMBC experiments.

  • Fish is a major source of protein for the Senegalese population. Fishing plays a dominant role in the Government's policy towards generating employment. It currently generates about 100,000 direct jobs (fishermen) for nationals, of which more than 90% are in small-scale fishing. The fishing industry also contributes to Government revenue through different agreements. In addition to associated dues, fishing agreements imply a series of economic, trade and technical counterparts. Under the latest fishing agreement concluded by Senegal and the European Union (1997-2001), direct financial compensation amounts to about CFAF 32 billion. Despite its economic and social importance, the sector has to face serious disequilibria both in resource exploitation and market supply: the coastal demersal (deep lying fish) stocks with high market value (mostly exported) are fully and even over-exploited, with a serious risk of local market supply shortages looming ahead as the fishing effort shifts from locally consumed species to export-oriented ones.

  • Out of 20 primers, 6 generated 1349 highly reproducible RAPD markers, producing approximately 5.2 polymorphic bands per primer in catfish(Sizurus asotus) population from Kunsan. The electrophoretic analysis of polymerase chain reaction-random amplified polymorphic DNAs (PCR-RAPD) products showed the middle levels of similarity between different individuals in population from Kunsan. That is to say, the degree of similarity varied from 0.40 to 0.54, with the average of 0.46, as calculated by bandsharing analysis. (omitted)

  • Wetlands perform various functions of vital socio-ecological significance playing fundamental role in moderating monsoon tidal floods and coastal protection and generate goods and products such as fish and forest resources. The depletion of mangroves is a cause of serious environmental and economic concern to many developing countries. Problems of sustainability of mangrove ecosystems are not only technical but also socio-economic. Functions played by mangrove wetlands are of fundamental importance for society. The present study aims to identify the challenges of the mangrove wetlands of Southeast coast of Chittagong, their uses and socio-economic influence on local people, and the value of ecosystem services, and to suggest how to conserve this ecosystem in a more equitable way.

The GOCI-II Early Mission Marine Fog Detection Products: Optical Characteristics and Verification (천리안 해양위성 2호(GOCI-II) 임무 초기 해무 탐지 산출: 해무의 광학적 특성 및 초기 검증)

  • This study analyzes the early satellite mission marine fog detection results from Geostationary Ocean Color Imager-II (GOCI-II). We investigate optical characteristics of the GOCI-II spectral bands for marine fog between October 2020 and March 2021 during the overlapping mission period of Geostationary Ocean Color Imager (GOCI) and GOCI-II. For Rayleigh-corrected reflection (Rrc) at 412 nm band available for the input of the GOCI-II marine fog algorithm, the inter-comparison between GOCI and GOCI-II data showed a small Root Mean Square Error (RMSE) value (0.01) with a high correlation coefficient (0.988). Another input variable, Normalized Localization Standard (NLSD), also shows a reasonable correlation (0.798) between the GOCI and GOCI-II data with a small RMSE value (0.007). We also found distinctive optical characteristics between marine fog and clouds by the GOCI-II observations, showing the narrower distribution of all bands' Rrc values centered at high values for cloud compared to marine fog. The GOCI-II marine fog detection distribution for actual cases is similar to the GOCI but more detailed due to the improved spatial resolution from 500 m to 250 m. The validation with the automated synoptic observing system (ASOS) visibility data confirms the initial reliability of the GOCI-II marine fog detection. Also, it is expected to improve the performance of the GOCI-II marine fog detection algorithm by adding sufficient samples to verify stable performance, improving the post-processing process by replacing real-time available cloud input data and reducing false alarm by adding aerosol information.

  • This study aims to review the progress of biotechnological applications of sea cucumber and presents fundamental validation data on a range of methods and applications to determine the most promising fields for future exploitation and application. Sea cucumbers from different regions, subjected to numerous industrially applicable drying methods, were evaluated for their biochemical composition. The study highlights the nutritional excellence of Jeju red sea cucumber, particularly the high amino acid and fatty acid composition of its intestinal organs and the possibility of the industrial utilization of these organs. In addition, an efficient extraction method to extract collagen and chondroitin sulfate from red sea cucumber was established. The extracted collagen and chondroitin sulfate can be used as commercially valid biomaterials. Accordingly, the red sea cucumber was confirmed as a valuable source of raw food material of varied functionality and bioactivity. Upon conducting an analysis of patents thus far relating to the sea cucumber, it was found that the associated technology was limited to some parts of primary and secondary industries. Therefore, there is a clear need for the strategic development of technologies to produce specialized functional products from sea cucumber's raw materials, with a view to promote other related industries.

Abalone Haliotis discus hannai Intestine Digests with Different Molecule Weights Inhibit MMP-2 and MMP-9 Expression in Human Fibrosarcoma Cells

  • The abalone Haliotis discus hannai, is one of the economically important species in the fisheries industry. Abalone intestines are one of the by-products of its processing. To investigate its bioactive potential, abalone intestine was digested using an in vitro gastrointestinal (GI) digestion system containing pepsin, trypsin, and ${\alpha}$-chymotrypsin. The abalone intestine G1 digests (AIGIDs) produced by the GI digestion system were fractionated into AIGID I (> 100 kDa), AIGID II (10-100 kDa), and AIGID III (1-10 kDa) using an ultrafiltration membrane system. Of the three digests, AIGID II and AIGID III exhibited inhibitory effects against matrix metalloproteinase-2 and -9 (MMP-2, MMP-9) in HT1080 human fibrosarcoma cells. Both fractions potently inhibited gelatine digestion by MMP-2 and MMP-9 treated with phorbol 12-myristate 13-acetate (PMA) and migration of HT1080 cells in dose dependently. Furthermore, AIGID II and III attenuated expression of p65, a component of nuclear transcription factor kappa B. These results indicate that of the abalone intestine digests inhibit MMP-2 and MMP-9. Thus, the AIGIDs or their active components may have preventive and therapeutic potential for diseases associated with MMP-2 and MMP-9 activation in fibrosarcoma cells.

Sanitary Characterization of Commercial Boiled-dried Pacific Herring Clupea pallasi and Boiled-dried Anchovy Engraulis japonicus and Proposal of Quality Standards (시판 마른청어(Clupea pallasi) 및 마른멸치(Engraulis japonicus)의 위생 특성 비교 및 품질기준 제시)

  • The objective of this study was to estimate the food quality of commercial boiled-dried Pacific herring Clupea pallasi as a substitute for boiled-dried anchovy. Standards for controlling quality of boiled-dried Pacific herring were suggested based on international (US FDA and CODEX) and domestic (Korean FDA, Standards on Quality of Seafood and Seafood Products, KS) standards for boiled-dried anchovy. The standards included requirements for moisture [less than 30% (less than 35 in very tiny sizes)], water activity (less than 0.85), salinity (less than 10%), water-phase salt (less than 20%), acid-insoluble ash (less than 1.5%), yeast and mold (fewer than 1.0×103 CFU/g), and different size and breakage (less than 5%). Based on the standards suggested, commercial boiled-dried Pacific herring passed nine levels (all levels) in water activity, acid-insoluble ash, mold and yeast concentrations; seven levels (L-1, 2, 3, 4, M-1, 2, S) in water-phase salt, and three levels (L-1, 3, 4) in the ratio of different size and breakage categories. These results suggest that the quality of commercial boiled-dried Pacific herring is similar to that of commercial boiled-dried anchovy.

Utilization of a Soluble Protein Recovered from Surimi Wastewater by Calcium Powder of Cuttle, Sepia esculents Bone (갑오징어(Sepia esculenta)갑 칼슘으로 회수한 surimi 가공폐수 단백질의 어묵소재로서 이용)

  • Utilization of soluble protein recovered from surimi wastewater using calcium powder of cuttle bone were examined. The crude ash content of the heat-induced surimi gel was increased linearly by increasing substitution ratio of recovered protein-ATC toward commercial surimi. Moisture (approximately $76\%$) and lipid $(0.2\%)$ contents were not change, but their protein contents were decreased 15.7 to $14.3\%$ depend on increasing of substitution ratio. The white index of the heat-induced surimi gel by color meter was increased up to $10\%$ of substitution ratio. There were no difference between $0\%\;and\;5\%$ substituted surimi gel in the gel strength. The sensory score on white index and texture of the heat-induced surimi gel did not change in 0 to $10\%$ as a substitution ratio of recovered protein-ATC toward commercial surimi, while decreased in more $15\%.$ The optimal substitution ratio of recovered protein-ATC as a bulking agent was $10\%.$ The heat-induced surimi gel prepared with $10\%$ substitution of recovered protein-ATC was similar to the content and composition of total amino. acids, and superior to calcium content and the ratio of calcium and phosphorus toward those of commercial surimi.

Processing and Characteristics of Canned Seasoned Boiled Oyster Crassostrea gigas and Canned Seasoned Roasted Oyster Crassostrea gigas (조미 자숙굴(Crassostrea gigas) 통조림 및 조미 구운굴(Crassostrea gigas) 통조림의 제조 및 품질특성)

  • The purpose of this study was to collect basic data that could be used in the manufacture of two kinds of canned oyster Crassostrea gigas. The steamed oyster was prepared by shucking after boiling for 6 min at $105^{\circ}C$ and then washing and dewatering. The roasted oyster was prepared by baking steamed oyster at $140^{\circ}C$ for 20 min. The manufacturing methods of canned seasoned boiled oyster and canned seasoned roasted oyster were as follows. The boiled or roasted oyster (50 g) was added to a can (RR-90) along with a mixture of seasoning sauce 40 and then seamed using a vacuum seamer under 20 cm Hg after pre-exhausting at $90^{\circ}C$ for 20 min. The two kinds of canned oyster products produced under sterilization of Fo 12 min were tested for cultured bacteria, external appearance, proximate composition, pH, VBN (Volatile basic nitrogen), TBA (Thiobarbiuric aicd) value, amino-N, salinity, color value sensory evaluation, etc. Results showed that the canned seasoned roasted oyster had higher overall acceptability than the canned seasoned boiled oyster. The reason for this was judged to be that the process of roasting at $140^{\circ}C$ for 20 min influenced the sensory evaluation.
