Isolation and cloning of seven-band grouper (Epinephelus septemfasciatus) growth hormone cDNA from pituitary gland revealed an open reading frame of 612 bp coding for a pre-growth hormone of 204 amino acids with a 17 amino acid putative signal peptide. Deduced amino acid sequence showed that there was one possible N-glycosylation site at $Asn^{l84}$ and four cysteine residues $(Cys^{52},\;Cys^{160},\;Cys^{177},\;Cys^{185})$ on t e same positions as in some other species where they were involved in the stabilization of the tertiary structure. The seven-band grouper growth hormone (sbgGH) presented a $99.5\%$ amino acid sequence identity with the growth hormone of Epinephelus coioides and contained the conserved hormone domain region. Comparison of growth hormone sequences from evolutionarily diverse species revealed 25 amino acid residues conserved in jawless fishes to modern mammals. It also revealed an evolutionary trend to retain the same polypeptide sequence even in the distantly related animals while allowing alterations to occur in polypeptides of the closely related species. In order to create a recombinant system to produce high levels of the growth hormone, it was expressed in Escherichia coli (BL21) cells. The gel analysis revealed theoretically expected molecular weights for both mature and pre-sbgGHs.

Using a biochemical indicator, the effects of fecal pollution in the estuary of the Nakdong river and in the Chinhae Hay were investigated from summer to winter in 1997-1998. The biochemical indicator used in this study was coprostanol (5-cholestan-3-ol) which is a kind of fecal sterol. It is a useful tracer of fecal pollution, as it is only found in feces of mammals including humans. Coprostanol was less affected by surrounding environments than other indicators of fecal pollution such as fecal coliform or ammonium and was proved to be a good indicator of fecal pollution. Therefore, long-term observations of coprostanol and related compounds could be used as useful indicators for the effects of feces on marine environments and ecosystems.
Population ecological characteristics of growth and mortality play an important role in understanding the population dynamics of marine mammals. The instantaneous coefficients of natural and bycatch mortality were estimated for minke whales (Balaenoptera acutorostrata) in Korean waters using a population assessment model composed of bycatch and abundance data. The survivorship curve of this population was fitted to the data, and then the curve was revised using age-specific relative bycatchability coefficients ($q_t$). Instantaneous coefficients of natural and bycatch mortality of minke whales were estimated as 0.024/year and 0.076/year, respectively, and from this the survival rate was estimated as 0.905. This estimated survival rate was comparable to other cetaceans in other regions. The $q_t$ for this population ranged from 0.020 to 0.193. The revised survival rates were higher when the $q_t$ was taken into account. The mortality coefficient, survival rate, $q_t$ and survivorship curves had not previously been determined for minke whale in this area. This estimate could serve as fundamental information to assess the status of this population and for conservation and rational management.

Because of the shallow depth of 44 m and large area of wetland in Korea and China, the class I productivity of Yellow Sea is very high (>300 $gC/m^2/year$), which is supporting substantial populations of fish, invertebrates, marine mammals and seabirds. However, the Yellow Sea is a large marine ecosystem which is enclosed by mainland of China and Korean peninsula, so it is vulnerable to external stress such as environmental degradation and overfishing. Recently, since the Fisheries Agreement between Korea and China, overfishing and illegal fishing of China and environmental degradation caused by coastal development in Korea and China have given much stress to the marine ecosystem of Yellow Sea. This article suggests that the major factors of governance are ineffectively responding to the growing demand for exploitation of the Yellow Sea and the international cooperation for establishing network of Yellow Sea governance is urgent.
Flounder (Paralichthys olivaceus) Immunoglobulins (Igs) were purified from the serum of mouse IgG-immunized flounder by using affinity chromatography. Under denaturing conditions in SDS-PAGE, the flounder Igs appeared to be composed of 2 heavy (H) chains (72 and 77 kDa) and two light (L) chains (26 and 28 kDa). Monoclonal antibodies (MAbs) were produced by the fusion of myeloma cells (SP2/0) with Balb/c mouse spleen cells that were previously sensitized against affinity-purified flounder Igs. In a Western blot analysis, the produced MAbs, FIM511, FIM519, and FIM562 recognized both the 72 and 77 kDa H chains, 26 kDa, and 28 kDa L chain, respectively. Mouse antiserum against flounder Igs reacted more strongly with the L chain of 28 kDa than with 26 kDa, suggesting that the 28 kDa molecule is more immunogenic than the 26 kDa L chain molecule. In a FACS analysis, the ratios of the Ig+ cell population in the flounder head kidney and spleen cells were 49% and 24%, respectively. Unexpectedly, however, the ratios of the Ig+ B-like cell population in the flounder were not significantly augmented, even after the immunization of an immunogenic antigen. This suggests that the humoral immune response in fish could be considerably different from that in mammals. The produced MAbs in this study would be useful in characterizing flounder Ig+ B-like cells and in developing flounder Ig detecting an immunoassay system.
Moderately diverse, but very abundant trace fossils are found from the Late Pleistocene deposits of Jeju Island, Korea. Vertical I-, Y- and U-shaped domichnia of annelids or decapods are, over 2500, extremely abundant, 3D network domichnia of callianassids are, over 200, very abundant, and small sinuous trails of nematode repichnia are, over 50, abundant in number. Horizontal trails attributable to polychaete or worm-like animals are, less than 50, common, but horizontal spreiten burrows, fish traces and crab trackways are, less than 10, rare in occurrence. Of these trace fossils, Taenidium barretti, Undichna britannica and Undichna unisulca represent the first record from the Pleistocene in Asia. Psilonichnus upsilon is the second record in Asia. Crab trackways probably produced by underwater punting gait of sideway walking crabs may represent the first record in the world. In addition, diverse and very abundant footprints of more than 500 hominids, more than 200 birds and more than 1000 mammals are closely associated with these invertebrate trace fossils. Trace fossil assemblage integrated with sedimentary facies is interpreted to have been formed in the marginal marine foreshore to backshore environment corresponding to the Psilonichnus and Skolithos ichnofacies.
The genus Anisakis is among the most significant parasites to public health, as it causes anisakiasis, a parasitic infection in humans resulting from consuming raw or undercooked seafood. Although the infection status of Anisakis in second intermediate hosts, such as marine fishes and cephalopods, and humans have been severally reported in Korea, no information about the definitive host in Korean waters is available. In 2014, 2 adult gastric nematodes were collected from a common minke whale (Balaenoptera acutorostrata) found in the East Sea, Korea. These worms were identified as A. simplex sensu stricto (s.s.) by comparing the mitochondrial COX2 marker with previously deposited sequences. Phylogenetic and phylogeographic analyses of A. simplex (s.s.) worldwide revealed 2 distinct populations: the Pacific population and the European waters population. This is the first report on adult Anisakis and its definitive host species in Korea. Further studies on Anisakis infection in other cetacean species and marine mammals in Korean seas are warranted.

After the United Nations Convention on the Law of the Sea (UNCLOS) and the United Nations Fish Stocks Agreement (UNFSA) came into effect, international cooperation through Regional Fisheries Management Organizations (RFMOs) was required, and each RFMO established and adopted the Conservation and Management Measure (CMM) for the regional Observer Programs to collect data on fishing activities and biological information and to monitor compliance with its CMMs. The observer coverage required by RFMO is set differently for each organization, ranging from 5% to 100%. In addition, tuna-RFMOs recommend increasing observer coverage in longline fisheries by at least 20% for reliable quantitative analysis of not only target species but also bycatch species and ecologically related species such as sharks, seabirds, sea turtles, and marine mammals. Therefore, in this study, we discussed ways to improve the national observer programs of Korean distant water fisheries that should be addressed in the future to respond to the RFMO trends.
The gossypol existed in cottonseed meal is a well known antispermatogenic agent which can impair reproductive performances of male fish as well as mammals. Two feeding experiments were conducted to examine a toxic effect of dietary supplementation of cottonseed meal on reproduction in juvenile olive flounder (the first experiment) for 19 weeks and growing olive flounder (the second experiment) for 26 weeks. After each feeding study, females and males were sampled for histological examination in gonads and liver to verify any negative effects by the dietary supplementation of cottonseed and soybean meal on reproduction. After two feeding trial, the gonad somatic index (GSI) of male and female (from the first feeding trial) were not significantly different among all the dietary treatments. The GSI values of female (from the second feeding trial) were not significantly different among all the dietary treatments. However, males fed cottonseed and soybean meal containing diets exhibited significantly lower GSI than that fed the control diet after the second feeding trial. Histological examination of gonads and liver of fish fed cottonseed and soybean meal did not show any negative effects compared to those of fish ffd the control diet. Hepatosomatic index of fish in the first and second feeding trials were not significantly different among all the dietary treatments. The findings in this study suggest that dietary supplementation of cottonseed and soybean meal up to 40% fish meal replacement might not deteriorate the gametogenesis of juvenile and growing olive flounder. However, the supplementation in diets over 30% fish meal replacement might reduce GSI of male in growing olive flounder.

Anisakis spp. (Nematoda: Anisakidae) parasitize a wide range of marine animals, mammals serving as the definitive host and different fish species as intermediate or paratenic hosts. In this study, 18 fish species were investigated for Anisakis infection. Katsuwonus pelamis, Euthynnus affinis, Caranx sp., and Auxis thazard were infected with high prevalence of Anisakis type I, while Cephalopholis cyanostigma and Rastrelliger kanagurta revealed low prevalence. The mean intensity of Anisakis larvae in K. pelamis and A. thazard was 49.7 and 5.6, respectively. A total of 73 Anisakis type I larvae collected from K. pelamis and A. thazard were all identified as Anisakis typica by PCR-RFLP analysis. Five specimens of Anisakis from K. pelamis and 15 specimens from A. thazard were sequenced using ITS1-5.8S-ITS2 region and 6 specimens from A. thazard and 4 specimens from K. pelamis were sequenced in mtDNA cox2 region. Alignments of the samples in the ITS region showed 2 patterns of nucleotides. The first pattern (genotype) of Anisakis from A. thazard had 100% similarity with adult A. typica from dolphins from USA, whereas the second genotype from A. thazard and K. pelamis had 4 base pairs different in ITS1 region with adult A. typica from USA. In the mtDNA cox2 regions, Anisakis type I specimens from A. thazard and K. pelamis showed similarity range from 94% to 99% with A. typica AB517571/DQ116427. The difference of 4 bp nucleotides in ITS1 regions and divergence into 2 subgroups in mtDNA cox2 indicating the existence of A. typica sibling species in the Makassar Strait.
