규조류 및 유산균 첨가 사료 공급에 따른 넙치(Paralichthys olivaceus)의 성장 및 비특이적 면역 촉진 반응에 미치는 영향 (Effect of Dietary Supplementation of Diatom Melosira nummuloides and Lactic Acid Bacteria Lactobacillus plantarum on the Growth and Immune Stimulation Responses of Olive Flounder Paralichthys olivaceus)

  • The diatom Melosira nummuloides is a microalga that is widely distributed in freshwater and seawater is used is used in the production of silicon and fucoxanthin. The objective of this experimental study was to determine the effects of diatom powder on the physiology of olive flounder Paralichthys olivaceus. In four feeding groups consuming 0%, 1%, 2% and 3% diatom powder. After 8 weeks of feeding, we investigated P. olivaceus growth rate, feed efficiency rate, survival rate, anti-oxidant enzyme rate, non-specific immune activity and immune gene expression. The rates of growth rate, feed efficiency rate and survival were significantly higher for olive flounder in all diatom groups than in the control. The results for anti-oxidant enzyme, superoxide dismutase and catalase showed no significance, but glutathione was significant, depending on the concentration of diatom addition. The galectin and lysozymes of immune genes were increased in the control group. Galectin and lysozymes were thought to have increased due to infections by from pathogens during the experiment period. These results suggest that the addition of diatoms to olive flounder diets is effective in enhancing growth rate and innate immunity.

Morphological observations and phylogenetic position of the parasitoid nanoflagellate Pseudopirsonia sp. (Cercozoa) infecting the marine diatom Coscinodiscus wailesii (Bacillariophyta)

  • During a sampling at Nokdong harbor, southern coast of Korea in January 2017, the marine diatom Coscinodiscus wailesii cells infected by a novel parasitoid nanoflagellate were observed. While the development process of the trophosomes of the parasitoid was more similar to that of Pseudopirsonia mucosa, division pattern of the auxosomes was similar to that of Pirsonia species. Phylogenetic analyses inferred from 18S rRNA gene sequences revealed that the parasitoid infecting C. wailesii fell within the cercozoan groups and branched as a sister lineage of the clade consisting of Pseudopirsonia mucosa and the undescribed Cercomonas sp. SIC7235, with the sequence dissimilarity of 7.3% with Pseudopirsonia mucosa. All of these developmental and molecular characteristics suggest that the parasitoid nanoflagellate infecting the diatom C. wailesii is a new Pseudopirsonia species.

Isolation and Physiological Characterization of a Novel Algicidal Virus Infecting the Marine Diatom Skeletonema costatum

  • Diatoms are a major component of the biological community, serving as the principal primary producers in the food web and sustaining oxygen levels in aquatic environments. Among marine planktonic diatoms, the cosmopolitan Skeletonema costatum is one of the most abundant and widespread species in the world's oceans. Here, we report the basic characteristics of a new diatom-infecting S. costatum virus (ScosV) isolated from Jaran Bay, Korea, in June 2008. ScosV is a polyhedral virus (45-50 nm in diameter) that propagates in the cytoplasm of host cells and causes lysis of S. costatum cultures. The infectivity of ScosV was determined to be strain- rather than species-specific, similar to other algal viruses. The burst size and latent period were roughly estimated at 90-250 infectious units/cell and <48 h, respectively.

  • There are a number of pieces of evidences that suggest a link between marine diatoms and microorganisms, but knowledge about related microbial communities is greatly lacking. The present study investigated the microbial community structures related to the growth of the marine diatom Cyclotella meneghiniana. We collected free-living bacteria (FLB) and particle-associated bacteria (PAB) at each growth stage (e.g., lag, exponential, stationary and death) of the diatom, and analyzed their bacterial 16S rDNA using pyrosequencing. Metagenomics analysis showed that community structures of FLB and PAB differed considerably with the progress of growth stages. FLB showed higher diversity than PAB, but variation in the different growth stages of C. meneghiniana was more evident in PAB. The proportion of the genus Hoeflea, belonging to the order Rhizobiales, was dominant in both FLB and PAB, and it gradually increased with the growth of C. meneghiniana. However, Enhydrobacter clade tended to considerably decrease in PAB. In addition, Marinobacter decreased steadily in FLB, but first increased and then decreased in PAB. These results suggest that Hoeflea, Enhydrobacter, and Marinobacter may be closely related to the growth of diatom C. meneghiniana.

Infection of marine diatom Coscinodiscus wailesii(Bacillariophyceae) by the parasitic nanoflagellate Pirsonia diadema(Stramenopiles) from Yongho Bay in Korea

  • The infection of marine diatom Coscinodiscus wailesii by a parasitic protist from the Yongho Bay of Busan, Korea was observed during the diatom bloom events in 2017 through 2018. The morphological and molecular features suggested that the parasitic nanoflagellate Pirsonia diadema was responsible for the infection. During the study period, the parasite prevalence ranged from 0.3% to 3.3%, and infected C. wailesii cells were observed only at surface seawater temperatures ranging between 10.9 and 19.9℃, although the host population appeared at temperatures above 25℃. The parasite and host system was successfully established as cultures. Using the cultures, we determined the morphological features over the infection cycle, parasite generation time, parasite prevalence as a function of inoculum size, and zoospore infectivity and survival time. The diatom C. wailesii was readily infected by the parasite P. diadema, with a parasite prevalence reaching up to 100% and a zoospore to host inoculum ratio above 20:1. The survival and infectivity of the parasite zoospores decreased with age. While the zoospores could survive up to 88 hours, they quickly lost their ability to infect after 48 hours. These results could lead to a better understanding of the biology and ecology of the parasitoid infecting the giant-sized diatoms in coastal waters.

Next-generation sequencing reveals the diversity of benthic diatoms in tidal flats

  • Benthic diatoms are ubiquitous in tidal flats and play major roles in maintaining coastal ecosystems. Spatio-temporal variations in diatom diversity have not been well-studied, mainly because of difficulties in morphological identification and the lack of appropriate genetic tools. To overcome these problems, we used the gene encoding the ribulose bisphosphate carboxylase large-subunit (rbcL) as a molecular marker, and sequenced these genes with the aid of the MiSeq platform. In this manner, we explored the genetic diversity of benthic diatoms in tidal flats of Guenso Bay on the west coast of Korea; differences in the spatial distributions of benthic diatoms were evident. The diatom communities were dominated by Nitzschia, Navicula, and Amphora; their relative distributions were affected by the sand proportion, grain size, and air exposure time. Our results suggest that meta-barcoding of the rbcL gene and next-generation sequencing can be used to explore the diversity of benthic diatoms.

  • Allenic and epoxy carotenoid, fucoxanthin (1) was isolated from the marine bacillariophycean microalga Hantzschia marina and the structure was assigned on the basis of comprehensive spectroscopic analyses. Fucoxanthin was detected only from diatom among three families (green algae, diatom and blue-green algae) of the marine microalgae tested. Fucoxanthin showed free radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and peroxynitrite $(ONOO^-)$ with $IC_{50}$ values of $32\;{\mu}M\;and\;60\;{\mu}M$, respectively.

오메가-3 지방산 및 푸코잔틴 고함량 토착 규조류 아크난티디움 균주의 분리 및 특성 (Isolation and Characterization of Indigenous Marine Diatom Achnanthidium sp. BS-001 Producing a high Content of Omega-3 Fatty Acid and Fucoxanthin Production)

  • Omega-3 fatty acids and carotenoids, which are known as representative high-value substances derived from microalgae, are being studied from various diatoms. Most of the diatoms contain fucoxanthin and omega-3 fatty acid. Fucoxanthin produced by diatom has been reported as bioactive compounds exhibiting strong antioxidant, anticancer and anti-inflammatory activities. However, the low growth rate and fucoxanthin content of diatoms are one of the big obstacles to the industrial application. In this study, indigenous marine diatom Achnanthidium sp. BS-001 was isolated for a candidate of fucoxanthin producer. Light intensity and temperature for the culture of Achnanthidium sp. BS-001 were optimized on PhotoBiobox. Optimization of silicate concentration for increasing BS-001 biomass productivity was confirmed in F/2 medium with various concentration of sodium silicate. As a result, condition of light intensity, temperature, and silicate concentration for optimal cultivation were $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, $18^{\circ}C$ and 0.106 mM, respectively. Maximum biomass productivity reaches to $154.3mg{\cdot}L^{-1}{\cdot}day^{-1}$, and then the content of omega-3 fatty acids and fucoxanthin were $19.4mg{\cdot}g^{-1}$, $9.05mg{\cdot}g^{-1}$, respectively. These results indicate that Achnanthidium sp. BS-001 has the potential to be used as a source of omega-3 fatty acids and fucoxanthin.

북방전복 종묘생산을 위한 농축 규조류 Caloneis schroederi와 Rhaphoneis sp.의 효과 (Effect of the concentrated-diatom of Caloneis schroederi and Rhaphoneis sp. for seedling production of Haliotis discus hannai)

  • Even though seedling production of Haliotis discus hannai has fully developed, the culture of benthic diatom as a live food for larvae is still a barrier to solve in commercial hatchery. The farmer depends on mixed microalgae which are non-selectively attached on the plate by flowing of natural seawater. The adequate diatom on the plate for the larvae in terms of quality and quantity is always significant bottleneck in the hatchery. In this study, two benthic diatom species, Caloneis schroederi and Rhaphoneis sp. were separately cultured in mass and inoculated to four tons' settlement tank of the larvae. And the larvae and the spats were cultured for nineteen and nine weeks, respectively. The result on seedling production of H. discus hannai with this method was compared to that of the farmer's traditional method as a control. With regard to variation of species composition of benthic diatom on the plate, C. schroederi and Rhaphoneis sp. were dominant for first three weeks after inoculation. But the diverse diatoms mainly, Navicula, Amphora, Cylindrotheca, Licmophora, Pleurosigma began to attache on the plate from the 4th week. The larvae attached 2.5 times more in C. schroederi tank than in the control tank. The final total biomass of the seeds in Rhaphoneis sp. tank was 3.2 times more than that of the control tank. The retared-spats in the seedling production also showed significantly higher growth and survival in the spat fed Rhaphoneis sp. or C. schroederi than those in the control group. We suggest that C. schroederi is proper for settlement of the larvae and Rhaphoneis sp. is appropriate for the growth of the larvae and spats in the commercial hatchery of H. discus hannai.

  • 돌말류는 미세조류의 일종으로서 담수 및 해양의 여러 다양한 환경의 수생태계에서 가장 중요한 1차 생산자 중 하나이다. 환경을 대표할 수 있는 생물 지표로서 널리 이용되고 있으며, 여러 지역에서 서로 다른 과학자들에 의한 연구 결과들이 과학적 통일성과 객관성을 갖추도록 하기 위해서는 돌말류 분석 자료의 정도 관리가 매우 중요하다. 현재 돌말류 분석은 형태적인 특징으로 분류하는 형태 기반의 분석과 DNA 서열로 종을 식별하는 DNA 바코드 분석이 사용되고 있다. 형태학 기반의 돌말류 분류는 분석 자료를 해석하는 분류학자들 간의 일관된 종 식별이 요구되는 한편 분자 분류 기반의 돌말류 분류는 신뢰성 있는 참조데이터가 필수적이다. 본 논문에서는 돌말류 분석 자료 정도관리의 국내외 현황을 우선 살펴보았으며, 그에 기반하여 국내 돌말류 분석 자료의 정도 관리에 대한 의견을 또한 제안하였다.
