Detection of Marine Birnavirus (MBV) from Rockfish Sebastes schlegeli Using Reverse Transcription and Nested PCR

  • Reverse transcription (RT)-PCR and nested PCR methods (2-step PCR) were tested for their ability to detect marine birnavirus (MBV) in cultured rockfish, Sebastes schlegeli. One set of primers for RT-PCR was designed, based on a gene of infectious pancreatic necrosis virus (IPNV), and another set of primers for nested PCR was designed based on the VP2/NS junction region of MBV. This 2-step PCR method was specific for MBV and sensitivity was heightened when nested PCR was combined to RT-PCR. This 2-step PCR method was useful for detecting MBV not only in diseased fish, but also in asymptomatic fish. These results indicate that this 2-step PCR method is useful for detecting MBV in rockfish.

  • During the last two decades, the rapid increase in aquaculture world wide, particularly in finfish and shellfish culture, is the result of both area expansion and production intensification. Under these conditions, the prevalence and spread of infectious diseases have increased as a result of higher infection pressure and decreased resistance of the aquatic animals. Accordingly, the effective control of infectious diseases has become more and more important to development of aquaculture. Recently the Pathology Team of National Fisheries Research and Development(NFRDI), has played pivotal roles in establishing the national management and control system of aquatic animal health and disease by new legislative works and technical guidelines. Under the national aquatic animal disease control framework, the Pathology Team of NFRDI and MOMAF(Ministry of Maritime Affairs and Fisheries) should also establish effective management and regulations, with the support of enforcement and monitoring policies. In conjunction with effective aquatic disease national management, financial assistance or alternative compensation options for aquaculture should also be available in the event of production losses or eradication system.

저수온기 참돔(Pagrus major) EP사료 내 동·식물성단백질 혼합물의 어분 대체 (Fish Meal Replacement with a Mixture of Plant and Animal Protein Sources in Extruded Pellet (EP) Diet for Red Seabream Pagrus major at Low Water Temperature)

  • This study aimed to evaluate how fish meal (FM) replacement in diets with a mixture of animal and plant protein sources affect growth performance, feed utilization, hematological parameters and innate immunity of red seabream Pagrus major. A control FM diet was formulated to contain 65% FM (Con). Two other diets were prepared replacing FM in the control diet with a mixture of protein sources (wheat gluten, soy-protein concentrate, tankage meal, and poultry by-product meal) by 30 and 40% (FM30 and FM40, respectively). Total 300 red seabream (body weight, 77.6±0.3g) were distributed to 12 tanks (300 L) in 4 replicates per diet. The fish were fed the diets to apparent satiation for 19 weeks. After the feeding trial, no significant differences could be observed in growth performance, feed utilization, hematological parameters, innate immunity, and survivals among all the dietary treatments. This long-term feeding trial at low water temperature (13.8-17.5℃) indicates that a proper mixture ratio of wheat gluten, soy protein concentrate, tankage meal, and poultry by-product meal can replace FM up to 40% in red seabream diets.

DNA Barcoding of the Marine Protected Species Parasesarma bidens (Decapoda: Sesarmidea) from the Korean Waters

  • Parasesarma bidens(De Haan, 1835) has been designated as a marine protected species by the Act on conservation and management of marine ecosystems. This crab has been recorded only from Jeju-do and Geomun-do, Republic of Korea. In this study, we describe for the first time the mitochondrial cytochrome c oxidase subunit I(COI) sequences of P. bidens. The intra-specific genetic distance among the Korean populations and between the Korean and Chinese populations ranged from 0% to 0.9% and 1.9% to 2.7%, respectively. The inter-specific genetic distances among the four Parasesarma species ranged from 10.9% to 12.8%. The finding of this study will be helpful to better describe P. bidens using COI DNA barcodes and can be used as basic data for their restoration and conservation research.

  • Pseudohelice subquadrata (Dana, 1851) is endangered due to its restricted habitat; hence, it has been designated as a marine protected species and endangered species by law in Korea. It has been recorded only Jeju-do and Geomun-do, Republic of Korea. The present study, is the first report on a cytochrome c oxidase subunit I DNA barcode for P. subquadrata. The maximum intra-specific genetic distance among all P. subquadrata individuals was found to be 0.5%, whereas inter-genetic distance within the same genus was 17.2-21.5% compared with Helice tientsinensis (Rathbun, 1931), H. tridens (De Haan, 1835), H. epicure (Ng et al., 2018), and Helicana wuana (Rathbun, 1931). Our barcoding data can thus be used as reference for restoration and conservation studies on P. subquadrata, which are designated as marine protected species.

Effects of Dietary Supplementation of Barodon, an Anionic Alkali Mineral Complex, on Growth Performance, Feed Utilization, Innate Immunity, Goblet Cell and Digestibility in Olive Flounder (Paralichthys olivaceus)

  • A 15-wk feeding trial was conducted to examine the supplemental effects of Barodon on growth performance, gastrointestinal histology, feed digestibility and innate immunity in olive founder. A basal commercial diet was used as a control and two other diets were prepared by spraying 0.1% or 0.2% of Barodon. Triplicate groups of fish (BW, 145 g) were fed one of the test diets to apparent satiation twice daily. At the end of the feeding trial, fish growth performance was not significantly affected by dietary treatments; however, feed utilization was significantly improved (linear and quadratic, p<0.05) by Barodon supplementation. Significantly higher (p<0.05) survival rates were obtained in fish fed Barodon containing diets. Hepatosomatic index increased significantly in Barodon treated groups. Also, the use of Barodon resulted in significant increase (linear and quadratic, p<0.05) of intestine length and number of goblet cells. Significantly higher (Quadratic, p<0.05) apparent digestibility coefficient of DM was obtained by supplementation of Barodon. Lysozyme and myeloperoxidase activities increased quadratically and linearly, respectively, in Barodon treated fish. Also, significantly higher (linear and quadratic, p<0.05) superoxide dismutase activity was found in Barodon fed fish. The findings in this study show that inclusion of Barodon in diets for olive flounder improves feed utilization and digestibility, and positively affects digestive tract histology and innate immunity.

  • A feeding trial was conducted to investigate the effects of supplementation with four phosphorus (P) additives [mono-calcium phosphate (MCP), dicalcium phosphate (DCP), tricalcium phosphate (TCP) and magnesium hydrogen phosphate (MHP)] on the growth, feed utilization and whole body mineral composition of juvenile olive flounders Paralichthys olivaceus. A basal diet without P supplementation was prepared as a negative control and four supplemental P sources were added at a level of 2%. Triplicate groups of fish (initial mean body weight 11 g) were fed one of the experimental diets to apparent satiation twice a day, at 08:30 and 18:00 for 10 weeks. The final body weights of fish fed the experimental diets ranged from 29.4 g (DCP) to 34.2 g (MCP) and did not differ significantly (P>0.05) among treatments. Similar tendencies were found for all parameters including weight gain (%), specific growth rate (SGR), feed conversion ratio (FCR), protein efficiency ratio (PER), feed intake (FI) and survival rate (SR). The hematocrit (%), hemoglobin (g/dL), serum inorganic P and whole body mineral composition did not differ significantly different (P>0.05) among the treatments. Therefore, dietary P addition is not necessary for juvenile olive flounder fed a fish meal-based diet.
